Highly substituted, novel, 8- and 9-nitro-2,3,4,5-tetrahydro-1,5-benzodiazepin-2(1H)-ones were obtained by direct nitration of the 7-bromo-5-trifluoroacetyl (or formyl)-substituted tetrahydrobenzodiazepinones. Alkaline and acidic hydrolysis of the novel mononitro derivatives was examined. Semiempirical AM1 calculations of aromatic substituents orientation in the nitration products are presented.  相似文献

A method for stereospecific hydroxylation of 1,2-dihydro-3H-1,4-benzodiazepin-2-ones using free and immobilized cells of actinomycetes as biocatalysts was developed. The hydroxylation under the action of yeast results in the formation of racemates. Actinomyces do not hydroxylate quinazolinones, quinoxalinones, and tetrahydro-1,5-benzodiazepin-2-ones; derivatives of 1,2,3,4-tetrahydro-1,5-benzodiazocin-2-ones are transformed into 2-[N-(3-acetylaminopropionyl)amino]benzophenones. Zusammenfassung Beim Erhitzen von (2-Aminophenyl)acetamiden mit Formaldehyd in Äthanol oder Diäthylenglykoldimethyläther erhält man 2,3,4,5-Tetrahydro-1H-1,3-benzodiazepin-4-one; 2,3,4,5-Tetrahydro-1H-1,3-benzodiazepin-2-one entstehen beim Einwirken von Phosgen auf -(2-Aminophenyl)äthylamine. Die als Zwischenprodukte benötigten (2-Nitrophenyl)acetamide wurden aus den entsprechenden Nitrobenzoesäuren durchArndt—Eistert-Reaktion hergestellt.
Syntheses of New 2.3.4.5-tetrahydro-1H-1-3-benzodiazepinones
Heating (2-aminophenyl)acetamides with formaldehyde in ethanol or diethylene glycol dimethylether yields 2.3.4.5-tetrahydro-1H-1,3-benzodiazepine-4-ones; 2.3.4.5-tetrahydro-1H-1.3-benzodiazepine-2-ones are formed by reaction of phosgene with -(2-aminophenyl)ethylamines. The (2-nitrophenyl)acetamides needed as intermediate compounds were synthesized from the corresponding nitrobenzoic acids byArndt—Eistert reaction.

A new synthesis of 1H-2,3,4,5-tetrahydro-1,5-benzodiazepin-2-ones (I) by the reductive cyclization of o-nitro derivatives of N-phenyl--alanine is described. Novel mono- or dibromo-substituted tetrahydro-1,5-benzodiazepinones were obtained by direct bromination of the corresponding 2,3,4,5-tetrahydro-(1H)-1,5-benzodiazepin-2-ones and 5-N-alkyl (or formyl) derivatives with bromine. Some [1,2,4]triazolo[1,5-c]quinazolin-5(6H)-ones 7 , the corresponding isomers [1,2,4]triazolo[4,3-c]quinazo-lin-5(6H)-ones and the 5-amino derivatives 8, 9 and 11 have been synthesized starting from the acylamidrazones 5 . The novel 4H‐thiazolo[3,2‐d][1,5]benzodiazepinium salts have been synthesized in a single step by the reaction of the variously substituted 2,3,4,5‐tetrahydro‐1,5‐benzodiazepine‐2(1H)‐thiones and bromoacetaldehyde diethyl acetal. Cyclization is obviously influenced by the nature of the substituents in the benzodiazepine system. A new procedure has been proposed for the synthesis of 8-aryl[1,2,4]triazolo[1,5-d][1,2,4]-triazin-5(6H)-ones by reaction of 6-aryl-1,2,4-triazin-3(2H)-ones with hydrazides derived from aliphatic, aromatic, and heterocyclic carboxylic acids. The reactions of 5,6,7,8-tetrafluoro-4-hydroxycoumarin derivatives with o-phenylenediamine occur with pyrone heterocycle cleavage and formation of substituted benzodiazepin-2-ones. 5,6,7,8-Tetrafluoro-4-hydroxycoumarin affords 4-(3,4,5,6-tetrafluoro-2-hydroxyphenyl)-2,3-dihydro-1H-1,5-benzodiazepin-2-one, 3-acetimidoyl-5,6,7,8-tetrafluoro-4-hydroxycoumarin produces 3-(3,4,5,6-tetrafluoro-2-hydroxybenzoyl)-4-methyl-1,2-dihydro-1H-1,5-benzodiazepin-2-one, and 3-acetyl-5,6,7,8-tetrafluoro-4-hydroxycoumarin yields both these heterocycles.  相似文献

A series of novel N-benzylcarboxamide derivatives of bicyclic compounds, 3,4-dihydropyrido[3,2-f][1,4]oxazepin-5(2H)-one and 2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-one, were synthesized by cyclization of N-benzyl-2-chloro-N-(2-hydroxyethyl)- [and -(3-hydroxypropyl)-] nicotinamides, respectively. Atropisomerism was observed in 5-[3,5-bis(trifluoromethyl)benzyl]-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-ones due to steric hindrance of the carboxamide moiety and restriction of its rotation. Cyclization of N-[3,5-bis(trifluoromethyl)benzyl]-2-chloro-N-[(2S)-3-hydroxy-2-methylpropyl]-5-methyl-4-phenylnicotinamide gave (3S)-5-[3,5-bis(trifluoromethyl)benzyl]-3,8-dimethyl-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3b][1,5]oxazocin-6-one, which exists predominantly in the thermodynamically stable aR-conformer in CDCl3. This compound showed excellent NK1-antagonistic activity with IC50 value (in vitro inhibition of [125I]-Bolton-Hunter-substance P binding in human IM-9 cells) of 0.47 nM, which is ca. 200-fold more potent than that of its enantiomer, indicating that the atropisomer chirality affects NK1-receptor recognition.  相似文献

The p-toluenesulfonate of 7,8-dihydro-5(6H)quinoloneoxime( 3 ) was subjected of a Beckmann rearràngement. The resulting 2,3,4,5-tetrahydro-1H-pyrido[3,2-b]azepin-2-one ( 4 ) was reduced with lithium aluminum hydride affording 2,3,4,5-tetrahydro-1H-pyrido[3,2-b] azepine ( 5 ). 5,6-l)ihydro-8(7H)quinolone ( 7 ), obtained by oxidation of 5,6,7,8-tetrahydro-8-quinolinol ( 6 ), was converted into the p-toluenesulfonate of 5,6-dihydro-8(7H)quinolone oxitne ( 9 ). Similarly the latter compound could be rearranged into 2,3,4,5-letrahydro-1H-pyrido [2,3-b] azepin-2-one ( 10 ) which on reduction produced 2,3,4,5-tetrahydro-1H-pyrido [2,3-b] azepine ( 11 ).  相似文献

A tandem reduction‐reductive amination reaction has been applied to the synthesis of (±)‐4‐alkyl‐2,3,4,5‐tetrahydro‐1,5‐benzoxazepines and (±)‐4‐alkyl‐1‐benzoyl‐2,3,4,5‐tetrahydro‐1H‐1,5‐benzodiazepines. The nitro aldehydes and ketones required for 1,5‐benzoxazepine ring closures were prepared by nucleophilic aromatic substitution of the alkoxides from several 3‐buten‐1‐ol derivatives with 2‐fluoro‐1‐nitrobenzene followed by ozonolysis. Precursors for the 1,5‐benzodiazepines were prepared by similar addition of N‐(3‐butenyl)benzamide anions to 2‐fluoro‐1‐nitrobenzene followed by ozonolysis. Catalytic hydrogenation of the nitro carbonyl compounds using 5% palladium‐on‐carbon in methanol then gave the target heterocycles by a tandem reduction‐reductive amination sequence. The 1,5‐benzoxazepines were isolated in high yield following chromatographic purification; the 1,5‐benzodiazepines were isolated as solids directly from the hydrogenation mixture and possessed differentiated functionality on the two nitrogen atoms.  相似文献

The 1,4-cycloaddition of dichloroketene to N,N-disubstituted 6-aminomethylene-b,7,8,9-tetra-hydro-5H-benzocyclohepten-5-ones afforded N,N-disubstituted 4-amino-3,3-dichloro-3,4,6,7-tetrahydro-5H-benzo[3,4]cyclohepta[2,l-b]pyran-2-ones only in the case of aromatic or strong hindering aliphatic N-substitution. The adducts gave N,N-disubstituted 4-amino-3-chloro-b,7-dihydro-5H-benzo[3,4]cyclohepta[2,l-b]pyran-2-ones by dehydrochlorination with collidine. Upon chromatography on neutral alumina, two products were instead isolated in the case of usual aliphatic N-substitution (diethylamine, piperidine), namely 6-(2,2-dichloroethylidene)-6,7,8,9-tetrahydro-5H-benzocyclohepten-5-one and the dehydrochlorinated 2-pyrone; this latter was the sole product in the case of pyrrolidine substitution. The 1,4-cycloaddition of sulfene occurred readily to give N,N-disubstituted 4-amino-3,4,6,7-tetrahydro-5H-benzo[3,4]cyclohepta-[1,2-e]-1,2-oxathiin 2,2-dioxidesin the case of both aliphatic and partially aromatic N-substitution.  相似文献

The conformational analysis of biologically active lofendazam (7-chloro-5-phenyl-1,3,4,5-tetrahydro-2H-1,5-benzodiazepin-2-one) is carried out by means of lanthanide shift reagent assisted 1H NMR spectroscopy: the lanthanide induced shift computer simulation suggests that in deuteriochloroform the heterocyclic ring of lofendazam assumes a cycloheptene-like chair conformation, where 1-N moves away from trigonal stereochemistry to a very flattened pyramidal structure. At room temperature the conformational equilibrium is markedly shifted (85%) towards the conformer showing pseudoaxial H-1 and 5-Ph. The remarkable influence of steric requirements in controlling conformation, and the importance of 3- and/or 4-methyl groups in hindering the ring inversion at room temperature, have been verified by conformational analysis of suitable analogous 1,3,4,5-tetraydro-2H-1,5-benzodiazepin-2-ones.  相似文献
