Reaction of the amino acid D-phenylalanine ethyl ester (4) with cinnamaldehyde gave chiral Schiff base 5, which underwent an asymmetric Staudinger [2+2] cycloaddition reaction with phthalimidoacetyl chloride to give the monocyclic beta-lactam 6 as a single stereoisomer. Ozonolysis of 6 followed by reduction with lithium aluminum tri(tert-butoxy) hydride afforded the hydroxymethyl beta-lactam 8. Treatment of 8 with methansulfonyl chloride gave the mesylated monocyclic beta-lactam 9, which was converted to the bicyclic beta-lactam 10 upon treatment with 1,8-diazabicyclo[5,4.0] undec-7-ene (DBU). Deprotection of the phthalimido group in beta-lactams 6 and 10 by methylhydrazine and subsequent acylation of the free amino beta-lactams with different acyl chlorides in the presence of pyridine afforded mono and bicyclic beta-lactams 14a-d and 15a-d respectively. The compounds prepared were tested against Escherichia coli, Staphilococcus citrus, Klebsiella pneumanie and Bacillus subtillis. Some of these compounds showed potential antimicrobial activities.  相似文献

Anhydrous lanthanide(III) chlorides (Ln = Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb) react with 3 equiv of lithium 2,2,5,5-tetramethyl-2,5-disila-1-azacyclopentanide, Li[N[Si(CH3)2CH2Ch2Si(CH3)2]], in THF or Et(2)O to afford the monomeric four-coordinate heteroleptic ate complexes Ln[N[Si(CH3)2CH2CH2Si(CH3)2]]3(mu-Cl)Li(THF/Et2O)3 (Ln = Sm (1), Eu (2), Gd (3), Tb (4), Dy (5), Ho (6), Er (7), Tm (8), Yb (9)), whose solid-state structures were determined by the single-crystal X-ray diffraction technique. All complexes additionally were characterized by melting point determination, elemental analyses, and mass spectrometry.  相似文献

The syntheses of some new sugar-based monocyclic beta-lactams possessing several other functionalities in addition to the carbohydrate moiety are described. The key step was the Staudinger [2+2] cycloaddition of chiral carbohydrate Schiff base 5 with phthalimidoacetyl chloride to yield the sugar-based monocyclic beta-lactam 6 as a single isomer. Treatment of protected beta-lactams 6 and 8 with methylhydrazine afforded the free amino beta-lactams 9 and 10. Acylation of these free amino beta-lactams with benzoyl, phenoxyacetyl, cinnamoyl and phenylacetyl chloride in the presence of pyridine afforded beta-lactams 11a-d and 12a-d. Some of these novel beta-lactams were found to be active against Staphilococcus citrus, Klebsiella pneumoniae, Escherichia coli and Bacillus subtilis.  相似文献

Liquid-phase studies concerning the solid-phase synthesis of monocyclic beta-lactams via the ester-enolate imine condensation route have been conducted utilizing triazene esters 1 and 2 as model compounds. Esters were attached to benzylamine resin 6 by a triazene linker employing the respective diazonium salts. Immobilized ester-enolates 8 and 10 were reacted with various imines and imine precursors to give polymer-bound beta-lactams 14 and 17 in different substitution patterns. Traceless cleavage from the triazene linker yields the desired beta-lactams 16 and 19.  相似文献

New monocyclic β-lactams 4–6 were synthesized by a ketene-imine [2+2] cycloaddition reaction. The prepared monocyclic β-lactams 4–6 were cleaved by ceric ammonium nitrate (CAN) to give NH-β-lactams 7–9. The NH-β-lactams were converted to N-sulfonyl β-lactams 1021 by treatment with four different sulfonyl chlorides in the presence of Et3N and 4-N,N-dimethylaminopyridine (DMAP). Some of these monocyclic β-lactams were active against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa.

The reactions of 2-methylimidazoline and 2-methyl-1,4,5,6-tetrahydropyrimidine with 1,3-diacid chlorides, in the presence of Et3N in refluxing MeCN give highly functionalized potentially bioactive 1,8-naphthyridinetetraones. 2-Methylimidazoline and 2-methyl-1,4,5,6-tetrahydropyrimidine can be viewed as tridentate nucleophiles which give four consecutive tandem nucleophilic attacks on electrophiles.  相似文献

为了寻找新的含噻唑杂环的先导化合物, 利用2-取代-1,3-噻唑烷1与氯甲酸取代苯基酯2及取代苯基甲酰氯4在三乙胺存在下发生缩合反应, 合成了6个2-取代-1,3-噻唑烷-3-甲酸酯类化合物3和5个N-苯甲酰基-2-取代-1,3-噻唑烷类化合物5, 并利用1H NMR, IR, MS, 元素分析对其结构进行了表征. 用X-ray单晶衍射测定了2-硫代-1,3-噻唑烷-3-甲酸苯酯(3d)和N-苯甲酰基-2-氧代-1,3-噻唑烷(5a)的晶体结构, 证实了氯甲酸酯与2-硫代-1,3-噻唑烷的反应产物为硫酮式结构, 而非硫酯式结构. 初步生物活性试验结果表明, 在试验浓度下部分目标化合物35具有一定的杀菌和杀虫活性.  相似文献

To learn more about the bleaching action of pulps by (hydroxymethyl)phosphines, lignin chromophores, such as the alpha,beta-unsaturated aromatic aldehydes, sinapaldehyde, coniferylaldehyde, and coumaraldehyde, were reacted with the tertiary phosphines R2R'P [R = R' = Me, Et, (CH2)3OH, iPr, cyclo-C6H11, (CH2)2CN; R = Me or Et, R' = Ph; R = Ph, R' = Me, m-NaSO3-C6H4] in water at room temperature under argon. In all cases, initial nucleophilic attack of the phosphine occurs at the activated C=C bond to form a zwitterionic monophosphonium species. With the phosphines PR3 [R = Me, Et, (CH2)3OH] and with R2R'P (R = Me or Et, R' = Ph), the zwitterion undergoes self-condensation to give a bisphosphonium zwitterion that can react with aqueous HCl to form the corresponding dichloride salts (as a mixture of R,R- and S,S-enantiomers); X-ray structures are presented for the bisphosphonium chlorides synthesized from the Et3P and Me3P reactions with sinapaldehyde. With the more bulky phosphines, iPr3P, MePPh2, (cyclo-C6H11)3P, and Na[Ph2P(m-SO3-C6H4)], only an equilibrium of the monophosphonium zwitterion with the reactant aldehyde is observed. The weakly nucleophilic [NC(CH2)2]3P does not react with sinapaldehyde. An analysis of some exceptional 1H NMR data within the prochiral phosphorus centers of the bisphosphonium chlorides is also presented.  相似文献

The mixture of samarium diiodide, amine, and water (SmI2/H2O/Et3N) is known to be a particularly powerful reductant, but until now the limiting reducing power has not been determined. A series of unsaturated hydrocarbons with varying half-wave reduction potentials (E(1/2) = -1.6 to -3.4 V, vs SCE) have been treated with SmI2/H2O/Et3N and YbI2/H2O/Et3N, respectively. All hydrocarbons with potentials of -2.8 V or more positive were readily reduced with SmI2/H2O/Et3N, whereas all hydrocarbons with potentials of -2.3 V or more positive were readily reduced using YbI2/H2O/Et3N. This defines limiting values of the chemical reducing power of SmI2/H2O/Et3N to -2.8 V and of YbI2/H2O/Et3N to -2.3 V vs SCE.  相似文献

Treatment of [Et4N][(Me2Tp)W(CO)3] (Me2Tp = HB(3,5-dimethylpyrazol-1-yl)3) with S8 in DMF at room temperature afforded a tris(sulfido) complex [Et4N][(Me2Tp)WS3] (1a), while that of [Et4N][TpW(CO)3] (Tp = HB(pyrazol-1-yl)3) in MeCN resulted in the formation of [Et4N][TpWS3] (1b) along with [Et4N]2[[WO(S2)2]2(mu-S)] (6) as a byproduct. Under similar conditions, [Et4N][(Me2Tp)Mo(CO)3] gave a mixture of a sulfido-tetrasulfido complex [Et4N][(Me2Tp)MoS(S4)] (2a) and its monooxo analogue [Et4N][(Me2Tp)MoO(S4)], although a sulfido-tetrasulfido complex [Et4N][TpMoS(S4)] (2b) was exclusively obtained from [Et4N][TpMo(CO)3]. The reaction of 1a with [PtCl2(cod)] (cod = 1,5-cyclooctadiene) in MeCN at room temperature led to the formation of a sulfido-bridged mixed-metal complex [Et4N][(Me2Tp)WS(mu-S)2PtCl2] (10). The structures of new complexes have been determined in detail by the X-ray analyses for 1a.MeCN, 1b, 2a, 2b, 6, and 10.  相似文献

The monocyclic beta-lactam antibiotic nocardicin A is related structurally and biologically to the bicyclic beta-lactams comprised of penicillins/cephalosporins, clavams, and carbapenems. Biosynthetic gene clusters are known for each of the latter, but not for monocyclic beta-lactams. A previously cloned gene encoding an enzyme specific to the biosynthetic pathway was used to isolate the nocardicin A cluster from Nocardia uniformis. Sequence analysis revealed the presence of 14 open reading frames involved in antibiotic production, resistance, and export. Among these are a two-protein nonribosomal peptide synthetase system, p-hydroxyphenylglycine biosynthetic genes, an S-adenosylmethionine-dependent 3-amino-3-carboxypropyl transferase (Nat), and a cytochrome P450. Gene disruption mutants of Nat, as well as an activation domain of the NRPS system, led to loss of nocardicin A formation. Several enzymes involved in antibiotic biosynthesis were heterologously overproduced, and biochemical characterization confirmed their proposed activities.  相似文献

A novel approach to racemic and enantiopure nonconventional fused bi- and tricyclic beta-lactams has been developed by using regio- and stereocontrolled intramolecular free radical reactions in monocyclic 2-azetidinone-tethered allenynes and haloallenes. The access to allene cyclization precursors was achieved by metal-mediated carbonyl allenylation of appropriately substituted 4-oxoazetidine-2-carbaldehydes in an aqueous environment. The tin-promoted radical cyclizations of allene-beta-lactams are totally regioselective for the central allenic carbon, providing bi- and tricyclic beta-lactams containing a seven-membered ring.  相似文献

The zinc hydrazide complexes [EtZn(N(SiMe(3))NMe(2))](2), [EtZn(N(Me)NMe(2))](4), and Zn(3)Et(4)(N(Et)NMe(2))(2) were synthesized by allowing excess hydrazine, HN(R)NMe(2), to react with diethyl zinc. The product of the reaction between ZnEt(2) and HN(i-Pr)NMe(2)ortho-metalated 4-(dimethylamino)pyridine (DMAP) at room temperature, producing the complex Zn[(NC(5)H(3)-p-NMe(2))ZnEt(N(i-Pr)NMe(2))](2). At elevated temperatures, Zn(3)Et(4)(N(Et)NMe(2))(2) also ortho-metalated DMAP, but [EtZn(N(Me)NMe(2))](4) did not. Single-crystal X-ray diffraction studies revealed that the hydrazide ligands in [EtZn(N(SiMe(3))NMe(2))](2) act as bridging mono-hapto amide ligands, and in Zn(3)Et(4)(N(Et)NMe(2))(2) and Zn[(NC(5)H(3)-p-NMe(2))ZnEt(N(i-Pr)NMe(2))](2) the hydrazide ligands are di-hapto.  相似文献

Kinetic data for solvolyses of 28 acid chlorides in 97% w/w trifluoroethanol (TFE)-water spanning over 10 (9) in rate constant at 25 degrees C are obtained directly or by short extrapolation from published values. G3 calculations of the energy required for cation formation in the gas phase are validated from proton affinities and from other experimental data. G3 calculations of heterolytic bond dissociation enthalpies (HBDEs) for formation of cations from acid chlorides in the gas phase show the following trends when compared with the solvolysis rate constants: (i) electron-rich sulfonyl chlorides and most carboxylic acid chlorides, including thione derivatives, give a satisfactory linear correlation with a significant negative slope; (ii) most sulfonyl chlorides and some chloroformates and thio derivatives have higher HBDEs and fit another correlation with a small, negative slope. A significant deviation is observed for the acyl series (RCOCl), for which both solvolysis rates and HBDEs increase in the order R = Bu ( t ) < Pr ( i ) < Et < Me. The deviation may be explained either by a prior hydration mechanism or preferably by electrostatic effects on the formation of small cations. The above results of structural effects support independent evidence from solvent effects that cationic ionization reaction pathways (with nucleophilic solvent assistance or S N2 character) are involved in the solvolyses of acid chlorides.  相似文献

The bulky hydrazine t-BuN(H)NMe2 was synthesized via hydrazone and t-BuN(H)N(H)Me intermediates as the major component in a 90:5:5 mixture consisting of t-BuN(H)NMe2, t-BuN(Me)N(H)Me, and t-BuN(Me)NMe2. Reacting the mixture with n-BuLi followed by distillation and fractional crystallization led to the isolation of the ligand precursor LiN(t-Bu)NMe2. Lithium hydrazides, LiN(R)NMe2, were reacted with metal chlorides to afford the hydrazide complexes M(N(Et)NMe2)4 (M = Zr or Hf), MCl(N(R)NMe2)3 (M = Zr, R = i-Pr or t-Bu; M = Hf, R = t-Bu), and TaCl3(N(i-Pr)NMe2)2. The X-ray crystal structures of [LiN(i-Pr)NMe2]4, [LiN(t-Bu)NMe2.THF]2, ZrCl(N(R)NMe2)3 (R = i-Pr or t-Bu), and TaCl3(N(i-Pr)NMe2)2 were determined. The structural analyses revealed that the hydrazide ligands in ZrCl(N(R)NMe2)3 (R = i-Pr or t-Bu) and TaCl3(N(i-Pr)NMe2)2 are eta2 coordinated.  相似文献
