A new constituent, aloeresin H (1), was isolated from Cape aloe, a bittering and laxative agent. Its structure was elucidated by degradation experiments combined with 1D and 2D NMR data. Aloeresin H represents the first C,C-diglucoside discovered in commercial samples of the drug and its polyketide origin can be interpreted in terms of two-chain condensation. Possible conformations of the virtual aglycone were obtained as energy minima by quantum mechanical calculations and were found to be consistent with particular NOE associations observed in the original glucoside.  相似文献

From the stem bark of Bowdichia virgilioides, lupeol, lupeol acetate, sitosterol, stigmasterol and methyl-3-[2-(1-hydroxymethylvinyl)-2,3-dihydrobenzo[b]furan-5-yl]-(E)-2-propenoate have been isolated. The latter is a new compound and it has been named as bowdenol. Their structures were elucidated with the aid of spectroscopic techniques, mostly 1 and 2D NMR. Cyclization reactions of lapachol (1) isolated from Heterophragma adenophyllum have been studied under microwave irradiation under different conditions using alumina (acidic, basic and neutral)/silica gel/montmorillonite (KSF and K-10) as solid support along with neat reaction using 2-3 drops of DMF giving naturally occurring dehydro-alpha-lapachone (2), alpha-lapachone (3), beta-lapachone (4) depending upon the nature of support and irradiation time. A novel naphthoquinone derivative adenophyllone (5) can be synthesized from lapachol using DMF under microwaves.  相似文献

Four porphyrin-Re(I) conjugates, in which a pyridylporphyrin chromophore is directly coordinated to the electron-acceptor fragment [ fac-Re(CO) 3(bipy)] (+), were prepared: the dimeric and pentameric compounds [ fac-Re(CO) 3(bipy)(4'MPyP)](CF 3SO 3) ( 1) (4'MPyP = 4'-monopyridylporphyrin) and [ fac-{Re(CO) 3(bipy)} 4(mu-4'TPyP)](CF 3SO 3) 4 ( 2) (4'TPyP = 4'-tetrapyridylporphyrin), and the corresponding compounds with 3' rather than 4' porphyrins, [ fac-Re(CO) 3(bipy)(3'MPyP)](CF 3SO 3) ( 3) and [ fac-{Re(CO) 3(bipy)} 4(mu-3'TPyP)](CF 3SO 3) 4 ( 4). These adducts proved to be very stable in solution and were also structurally characterized in the solid state by X-ray crystallography. A detailed photophysical study was performed on the zincated adducts of the conjugates 1- 3, labeled 5, 6, and 7, respectively. In all adducts the typical fluorescence of the zinc-porphyrin unit was reduced in intensity and lifetime by the presence of the peripheral rhenium-bipy fragment(s) (heavy-atom effect). For the dyads 5 and 7 the photoinduced charge transfer process from the zinc-porphyrin to the Re(I)-bipy unit is only slightly exoergonic. Ultrafast spectroscopy experiments showed no evidence for electron transfer quenching in the dyads as such, whereas the addition of pyridine (that binds axially to zinc and thus affects the porphyrin redox potential) led to a moderately efficient photoinduced electron transfer process. In perspective, an appropriate functionalization of the bipy ligand and/or of the porphyrin chromophore might improve the thermodynamics and, thus the efficiency, of the photoinduced electron transfer process.  相似文献

BACKGROUND: The non-proteinogenic amino acid p-hydroxyphenylglycine is a crucial component of certain peptidic natural products synthesized by a non-ribosomal peptide synthetase mechanism. In particular, for the vancomycin group of antibiotics p-hydroxyphenylglycine plays a structural role in formation of the rigid conformation of the central heptapeptide aglycone in addition to being the site of glycosylation. Initial labeling studies suggested tyrosine was a precursor of p-hydroxyphenylglycine but the specific steps in p-hydroxyphenylglycine biosynthesis remained unknown. Recently, the sequencing of the chloroeremomycin gene cluster from Amycolatopsis orientalis gave new insights into the biosynthetic pathway and allowed for the prediction of a four enzyme pathway leading to L-p-hydroxyphenylglycine from the common metabolite prephenate. RESULTS: We have characterized three of the four proposed enzymes of the L-p-hydroxyphenylglycine biosynthetic pathway. The three enzymes are encoded by open reading frames (ORFs) 21, 22 and 17 (ORF21: [PCZA361.1, O52791, CAA11761]; ORF22: [PCZA361. 2, O52792, CAA11762]; ORF17: [PCZA361.25, O52815, CAA11790]), of the chloroeremomycin biosynthetic gene cluster and we show they have p-hydroxymandelate synthase, p-hydroxymandelate oxidase and L-p-hydroxyphenylglycine transaminase activities, respectively. CONCLUSIONS: The L-p-hydroxyphenylglycine biosynthetic pathway shown here is proposed to be the paradigm for how this non-proteinogenic amino acid is synthesized by microorganisms incorporating it into peptidic natural products. This conclusion is supported by the finding of homologs for the four L-p-hydroxyphenylpyruvate biosynthetic enzymes in four organisms known to synthesize peptidic natural products that contain p-hydroxyphenylglycine. Three of the enzymes are proposed to function in a cyclic manner in vivo with L-tyrosine being both the amino donor for L-p-hydroxyphenylglycine and a source of p-hydroxyphenylpyruvate, an intermediate in the biosynthetic pathway.  相似文献

Cytokines produced by immune cells infiltrating pancreatic islets have been implicated as one of the important mediators of beta-cell destruction in insulin-dependent diabetes mellitus. In this study, the protective effects of epigallocatechin gallate (EGCG) on cytokine-induced beta-cell destruction were investigated. EGCG effectively protected IL-1beta and IFN-gamma-mediated cytotoxicity in insulinoma cell line (RINm5F). EGCG induced a significant reduction in IL-1beta and IFN-gamma-induced nitric oxide (NO) production and reduced levels of the inducible form of NO synthase (iNOS) mRNA and protein levels on RINm5F cells. The molecular mechanism by which EGCG inhibited iNOS gene expression appeared to involve the inhibition of NF-kappaB activation. These findings revealed EGCG as a possible therapeutic agent for the prevention of diabetes mellitus progression.  相似文献

Density functional theory calculations were carried out to investigate the structural and opto-electronic properties of cyclopenta[2,1-b:3,4-b′]dithiophene (CPDT) derivatives. The ground state, cationic and anionic geometries of cyclopentadithiophene derivatives were optimized at B3LYP/6-311G(d,p) level of theory. Based on these geometries, ionic state properties such as ionization potentials, electron affinities, hole extraction potential, and electron extraction potential of cyclopentadithiophene derivatives have been calculated. The charge transfer integral, spatial overlap integral, and site energy were calculated from the matrix elements of Kohn–Sham Hamiltonian. Computed results show that the mobility of charge carriers in CPDT derivatives is strongly affected by the substitution of electron withdrawing group at bridge-head and α-positions. The excited state geometry of CPDT derivatives were optimized using configuration interaction singles method. On the basis of ground and excited states geometry, absorption and emission spectra of cyclopentadithiophene derivatives were calculated using the time-dependent density functional theory method. It has been observed that the substitution of EWG in cyclopentadithiophene core alters the absorption and emission spectra. The nonlinear optical property of CPDT derivatives have been studied through computed static polarizability and first hyperpolarizability.  相似文献

Density-functional theory studies were applied to investigate the structural, electronic, and optical properties of 9-heterofluorenes achieved by substituting the carbon at 9 position of fluorene with silicon, germanium, nitrogen, phosphor, oxygen, sulfur, selenium, or boron. These heterofluorenes and their oligomers up to pentamers are highly aromatic and electrooptically active. The alkyl and aryl substituents of the heteroatom have limited influence, but the oxidation of the atom has significant influence on their molecular structures and properties. The highest occupied molecular orbital (HOMO)-lowest occupied molecular orbital (LUMO) interaction theory was successfully applied to analyze the energy levels and the frontier wave functions of these heterofluorenes. Most heterofluorenes belong to type B of interaction with low-lying LUMO and have the second kind of wave function. Carbazole and selenafluorene have type C of interaction with high-lying HOMO and the third kind of wave function. Types C and D of heterofluorenes, such as carbazole, oxygafluorene, sulfurafluorene, and selenafluorene also have high triplet state energies. The extrapolated HOMO and LUMO for polyheterofluorenes indicate that polyselenonafluorene has the lowest LUMO; polycarbazole has the highest HOMO; polyselenafluorene has the highest bandgap (E(g)); and polyborafluorene has the lowest E(g). Heterofluorenes and their oligomers and polymers are of great experimental interests, especially those having extraordinary properties revealed in this study.  相似文献
