Eight cembranoid epimers at C-8 (1-8) were isolated from the organic extract of the Colombian Caribbean octocoral Pseudoplexaura flagellosa. Compounds 2, 4, and 6 are reported for the first time. Although compounds 1, 3, 5, 7, and 8 have been reported previously, their structures and NMR assignments are revised, completed or corrected. The structures of these compounds were established on the basis of detailed analysis of their spectroscopic data. Furthermore, the relative configurations of compounds 1-3 and 7 were confirmed by single-crystal X-ray diffraction. The absolute configurations of compounds 1-8 were determined using a combination of the modified Mosher method and unambiguous chemical interconversions. Evaluation of their antifouling properties using quorum sensing inhibition (QSI) and biofilm inhibition bioassays showed that compounds 3, 6, and 7 have excellent QSI activity against Chromobacterium violaceum, as measured by inhibition of the production of violacein pigment, without interfering with its growth. Furthermore, compounds 3, 5, 6, and 8 exhibited inhibition of biofilm maturation without interfering in the growth of Pseudomonas aeruginosa, Vibrio harveyi, and Staphylococcus aureus. This is the first report of cembranoids as inhibitors of bacterial biofilm and as compounds that interfere with QS in C. violaceum.  相似文献

Six organotin compounds with 4,4′-thiodibenzenethiol (LH2) of the type RnSnL4−nSnRn (n = 3: R = Me 1, Ph 2, PhCH23, n = 2: R = Me 4, Ph 5, PhCH26) have been synthesized. All compounds were characterized by elemental analysis, IR and NMR (1H, 13C, and 119Sn) spectra. The structures of compounds 1, 2, 4, 5 and 6 were also determined by X-ray diffraction analysis, which revealed that compounds 1 and 2 were monomeric structures, compounds 4, 5 and 6 were centrosymmetric dinuclear macrocyclic structures, and all the tin(IV) atoms are four-coordinated. Furthermore, supramolecular structures were also found in compounds 1, 2, 4, 5 and 6, which exhibit one-dimensional chains, two-dimensional networks or three-dimensional structures through intermolecular C–H?S weak hydrogen bonds (WHBs), non-bonded Sn?S interactions or C–H?π interactions.  相似文献

tert-Butyldimethylsililoxy-2-aza-1,3-butadienes react with 2H-azirine 3 leading to Diels-Alder cycloadducts in moderate yields. The reactions are endo- and regioselective with the azirine being added by its less hindered face. There is only one product in the case of 1b, 4b. There are two isomers (4 and 5) from 1a, 1c and 1d. A different result was obtained with the diene 1e. Diene 1e formed products 4e and 8. Some of compounds 4 and 5 have been hydrolysed leading to functionalised aziridines 7. Five novel compounds, psoracorylifols A-E (1-5) with important activity against Helicobacter pylori have been isolated from a well-known traditional Chinese medicine (TCM), the seeds of Psoralea corylifolia. The structures of compounds 1-5, including their absolute configurations, were established on the basis of spectral methods and biogenetic reason. The structure of 1 was confirmed by a single-crystal X-ray diffraction. Psoracorylifols D and E (4 and 5) represent an unprecedented carbon skeleton. The biogenetic origin of psoracorylifols A-E (1-5) was also postulated.  相似文献

A detailed chemical investigation of the minor metabolites produced by the endophytic fungus Pestalotiopsis sp. isolated from the Chinese mangrove Rhizophora mucronata afforded sixteen new compounds of polyketide origin, including pestalotiopyrones A-H (1-8), pestalotiopisorin A (10), pestalotiollides A-B (11-12), pestalotiopin A (13), and four amides pestalotiopamides A-D (14-17), along with three known compounds, nigrosporapyrone D (9), 2-anhydromevalonic acid (18), and p-hydroxy benzaldehyde (19). The structures of all compounds were unambiguously established from their spectroscopic data that included HR-ESIMS and 1- and 2-dimensional NMR spectroscopy, and by comparison with the literature.  相似文献

Highly selective, practical, and efficient protocols for the preparation of 4(5)-aryl-1H-imidazoles 2, 2,4(5)-diaryl-1H-imidazoles 3, and 4,5-diaryl-1H-imidazoles 1 are described. A key step of these protocols is the regioselective synthesis of 5-aryl-1-benzyl-1H-imidazoles 9 by Pd-catalyzed direct C-5 arylation of commercially available 1-benzyl-1H-imidazole (8) with aryl halides. The three-step synthesis of compounds 3 from 8 also involves the Pd-catalyzed and Cu-mediated direct C-2 arylation of imidazoles 9 with aryl halides under base-free and ligandless conditions. On the other hand, the four-step synthesis of imidazoles 1 from 8 also involves the regioselective bromination of compounds 9 and a Suzuki reaction of the resulting 5-aryl-1-benzyl-4-bromo-1H-imidazoles 11 with arylboronic acids 5 under phase-transfer conditions, followed by N-debenzylation.  相似文献

Cyclocondensation of 1-aryl-3-ferrocenyl-2-propen-1-ones (1) with hetaryl hydrazines resulted in N-hetaryl-3-aryl-5-ferrocenyl pyrazolines (3, 4). The analogous 3-aryl-1-ferrocenyl-2-propen-1-ones (5) gave the isomeric N-hetaryl-5-aryl-3-ferrocenyl-pyrazolines (6, 10), but in lower yield. The reaction of aryl-chalcones (7) with 4-hydrazino-phthalazinone led to 3,5-bis-aryl-N-hetaryl-pyrazolines (8) or to the corresponding ene-hydrazones (9). The structure of the new compounds was established by IR, 1H and 13C NMR spectroscopy, including DNOE, HMQC, HMBC and DEPT methods. For compounds 1b, 3b and 8b the stereo structure was elucidated also by X-ray diffraction.  相似文献

New methyl induced linear and angular thieno-2H-chromenes 4, 5 and 6 were prepared by reaction of new methylated 6-hydroxybenzo[b]thiophenes 2 (a, b and c) and propargylic alcohols 3a and 3b, using acidic Alumina Brockmann I as catalyst and drying agent. Compounds 2 were prepared in good to excellent yields in a ‘one pot’ three step reaction from the corresponding bromo compounds 1. The photochromic behaviour of compounds 4, 5 and 6b was evaluated with the aid of a classical set of spectrokinetic parameters, and compared to reference compounds that are benzoannellated in the 5,6 and 6,7 positions of the chromene (naphthopyrans) and also to thieno-2H-chromenes 7 and 8, previously prepared, which are analogues of 5a. The resistance to fatigue (photodegradation) under continuous irradiation was also evaluated.  相似文献

Four new compounds having an unusual 1,7-dioxadispiro[5.1.5.2]-12-ene-11-one tricyclic ring system (1-4), their potential precursor, 5R-hydroxy-1-(4-hydroxyphenyl)-eicosan-3-one (5), and two known compounds, aculeatins A (6) and B (7), have been isolated from Amomum aculeatum. All compounds were characterized by spectroscopic methods and the configurations were established by 2D NOE correlations. Compounds 1-4, 6, and 7 showed cytotoxic activity against several human cancer cell lines.  相似文献

The paper reports the first chemical study of the porostome nudibranch Doriopsilla pelseneeri collected off the Portuguese coast (Atlantic Ocean). Two new furanosesquiterpene alcohols, pelseneeriol-1 (1) and pelseneeriol-2 (2), have been isolated together with known compounds, 15-acetoxy-ent-pallescensin-A (5), and dendocarbin-A (6), from the mantle of the nudibranch, whereas euryfuran (3) and drimane ester mixture 4 were identified in the extract of the internal glands. The structures of 1 and 2 have been determined by extensive spectroscopic studies as well as by comparison with literature model compounds. In order to assess the relative stereochemistry of 1 and 2, full NMR assignment of related sponge metabolite microcionin-2 (8) and of co-occurring sesquiterpenes 9-11, that have been re-isolated from the Mediterranean sponge Fasciospongia cavernosa, has been also conducted. In particular, the relative stereochemistry of tricyclic sesquiterpene microcionin-1 (9) has now been rigorously assigned by detailed analysis of NOE difference experiments.  相似文献

Starting with 2-acetylquinoxaline a novel class of heterocyclic compounds, the 1-(thiazol-2-yl)-1H-pyrazolo[3,4-b]quinoxalines 4, were prepared by following two different synthetic procedures: 2-acetylquinoxaline reacted with thiosemicarbazide to the thiosemicarbazones 1a which was (i) cyclized with α-halogeno ketones to the thiazoles 3. These compounds were dehydrogenated in acidic medium to the title compounds 4. (ii) The thiosemicarbazone 1a could be also dehydrogenated using NH2OH·HCl to the thioamide 5a and these, finally, were cyclized with α-halogeno ketones to the title compounds 4. Only thiazole 3a was isolated, the other thiazoles 3 were dehydrogenated in a one-pot procedure. From the thioamide 5a also both the compounds 9, by reacting with dibromodiacetyl, and 10, by treatment with dimethyl acetylenedicarboxylate, were obtained. The analysis of both the 1H and 13C NMR spectra was not straightforward but could be attained finally by employing the whole arsenal of 1D and 2D NMR spectroscopy.  相似文献

Reaction of 3-acyl-4-methoxy-1-methylquinolinones 2 and 5 with hydrazines has been investigated under different experimental conditions. Compound 2 always gave rise selectively and exclusively to the regioisomeric 1,3-disubstituted- or 2,3-disubstituted-pyrazolo[4,3-c]quinolin-4(5H)one (compounds 3a,b or 4a,b, respectively), while reaction of 5 with N-methylhydrazine led to a mixture of pyrazoles 7a and 8a. With N-phenylhydrazine, compounds 7b or 8b were regioselectively obtained. Compound 8a could be selectively synthesized working in solventless conditions. Structural elucidation of all products was independently achieved by NMR spectroscopy.  相似文献

A bioassay-guided purification of the methanolic extract of Withania somnifera fruits yielded novel withanamides A-I (1-9) and withanolides (10-13). Among the withanolides, compound 10 is novel. The structures of these compounds were determined by using FABMS, HRFABMS, 1D and 2D NMR spectral and chemical methods. The withanamides possess novel chemical structures and consisted of serotonin, glucose and long-chain hydroxyl fatty acid moieties. The stereochemistry of the hydroxyl group in the long-chain fatty acid moiety in compound 1 was determined by the modified Mosher's ester method. Compounds 1-13 were tested for their ability to inhibit lipid peroxidation in a model system using large unilamellar vesicles. Withanamides 1-5 and 9 inhibited lipid peroxidation by 98, 93, 79, 94, 81 and 86%, respectively, at 1 μg/mL. However, compounds 6-8 inhibited the lipid peroxidation by 85, 82 and 90%, respectively, at 0.5 μg/mL. Withanolides 10-13 were also tested and only compound 12 inhibited the lipid peroxidation by 82% at 10 μg/mL. To evaluate the structure activity relationships of withanamides A-I, compounds 14-16 were purchased and their lipid peroxidation activity determined as in the case of compounds 1-9. Commercial antioxidants, butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and tert-butylhydroquinone (TBHQ), were also tested in this assay at 1 μg/mL and showed 80, 81 and 85% of inhibition, respectively. Our results suggest that the potent antioxidant activity exhibited by novel withanamides is probably due to the hydroxylated long-chain acyl group. This is the first report of withanamides, unique serotonin conjugates, from W. somnifera fruits.  相似文献

A total of 18 compounds 1-6 derived from triphenylamine as core group were prepared and characterized, and their mesomorphic properties were also investigated. Compounds 1-4 and 5,6 were prepared from p,p′,p″-triformyltriphenylamine and p,p′-diformyltriphenylamine with appropriate alkoxyphenylamines. The phase behavior of these mesogenic compounds was characterized and studied by differential scanning calorimetry, polarized optical microscopy, and powder XRD diffraction. Compounds 1-3 exhibited columnar mesophase, however, compounds 4-6 were nonmesogenic. The mesophases observed in compounds 1-3 were found to be side dependent. Compounds 1a, 2a, and 3a appended with one, two, or three side chains exhibited lamellar columnar (ColL) phases, and compounds 2b and 3b with four or six side chains formed hexagonal columnar (Colh) phases. The formation of the mesophases, lamellar or columnar mesophases, was probably induced by H-bonding formed between -CH2NH groups. The oxidation process determined by cyclic voltammetry showed two redox waves, one appeared at 220-255 mV and the other one at 503-677 mV, which gave energy to HOMOs range of 5.02-5.36 eV. The fluorescent properties of the compounds were examined. All λmax peaks of the absorption and photoluminescence spectra of compounds occurred at ca. 307-392 nm and 368-456 nm, respectively. Compound 4a has a larger red shift due to a better conjugation linked by CC double bonds instead of -CH2NH in other compounds.  相似文献

Two new pyridoacridine alkaloids, 13-didemethylaminocycloshermilamine D (1) and demethyldeoxyamphimedine (2), were isolated from the purple chromotype of the Western Mediterranean ascidian Cystodytes dellechiajei. This morph also contained the known shermilamine B (3), kuanoniamine D (4), N-deacetylshermilamine B (5), N-deacetylkuanoniamine D (6), styelsamines C (7), and D (8). The structures of new compounds were elucidated on the basis of spectroscopic data. A hypothetic biosynthetic pathway from the tetracyclic styelsamine D (8) was proposed for both compounds 1 and 2 and their antimicrobial potential was evaluated.  相似文献

In search of compounds that ameliorate the toxicity of amyloid-β (Aβ) peptides, new derivatives of tricyclic pyrones (1-7) were synthesized and their biological activities evaluated. The carboxylic ester and amide derivatives 1-4 were synthesized from a selective carboxylation of C3 methyl of (5aS,7S)-{7-Isopropenyl-3-methyl-1H,7H-5a,6,8,9-tetrahydro-1-oxopyrano[4,3-b][1]benzopyran (8) with LDA followed by benzyl chloroformate or carbon dioxide to provide ester 1 and carboxylic acid 9, respectively. Three isomeric tricyclic pyrone, 5-7, containing adenine moiety at C7 side chain were synthesized from the alkylation of mesylate 13 with adenine, and displacement of chloropurine 15 with amine 14. Although C3-benzyloxycarbonylmethyl analogs 1-3 have marginal ACAT and CETP activities, their modified aspartate analog 4 and C3-methyl-C7-(N3-adeninyl)-2-propyl analog 6 show a significant effect in protecting against neuron-cell death from the toxicity of intracellular accumulation of Aβ or Aβ-containing C-terminal fragments (CTF) of amyloid β precursor protein (APP). N9-Adenine analog 5 is 20-fold less effective than N3-adenine derivative 6 in the protection of neuron-cell death induced by Aβ, while N10-adenine analog 7 was inactive. As a result of this study, compounds 4 and 6 will well serve as lead compounds for further studies of the mechanism of action of Aβ-and CTF-induced neuron-cell death, studies which should enhance the future development of new drugs for the prevention and treatment of AD.  相似文献

New 1,3-propanediaminocyclotriphosphazene derivatives (7-17) were synthesized from the reactions of spiro-1,3-propanediaminocyclotriphosphazene, N3P3Cl4[NH(CH2)3NH] (1) with the cyclopropanemethylamine (2), cyclohexylamine (3), pyrrolidine (4) cyclohexanol (5), cyclopropylmethanol (6). The structures of the novel compounds (7-17) were characterized by elemental analysis, mass spectrometry, 1H and 31P NMR spectroscopy. The molecular structures of 8, 12 and 13 were determined by X-ray crystallography. The structures of all these three compounds are in the monoclinic crystal system; compounds 8 and 12 have the P21/c space group while compound 13 has the P21/n space group. The ring conformation of the cyclotriphosphazene and other external rings were investigated based on the X-ray crystal structures.  相似文献

The novel optically active derivatives of 2,2′-disubstituted-1-aminocyclopropane-1-carboxylic acid (−)-2 and (+)-3 were synthesised from the spiro-azlactone (+)-1. Oxidation of the diol moiety of (+)-3 gave by ring enlargement the racemic mixture of 2,3-dihydrofuran derivative (±)-6. This conversion is explained by stepwise rearrangement of the initially formed tetrasubstituted cyclopropanecarbaldehyde 4 through zwitterionic's reactive intermediate 5. The formation of (±)-6 is preferred energetically as established by ab initio calculations of the ground states and possible intermediates for that rearrangement. The crystal structure and absolute configuration of the compounds (+)-1, (−)-2, (+)-3 and (−)-7 were determined by single-crystal X-ray diffraction method. All four compounds possess Z-configuration of the cyclopropane ring. The dioxolane ring in the structures (+)-1 and (−)-2 adopts half-chair conformation, while the cyclopropane ring and geminally substituted groups in the structures (−)-2, (+)-3 and (−)-7 possess the anticlinal conformation. The molecules of the compound (+)-1 are connected by very weak intermolecular hydrogen bond of C-H?O type. In the compounds (−)-2, (+)-3 and (−)-7inter- and intramolecular hydrogen bonds of N-H?O type were observed. The spiro-compound (+)-1 exhibited a more pronounced inhibitory activity against the proliferation of murine leukemia and human T-lymphocytes cells than other type of tumor cell lines and normal human fibroblast cells.  相似文献

Five new meroterpenoids, chevalones A-D (1-4), aszonapyrone B (8), and a new sequiterpene alkaloid, eurochevalierine (5), together with four known compounds, sequiterpene (6), terpenoid pyrrolobenzoxazine named CJ-12662 (7), meroterpenoid, aszonapyrone A (9), and ergosterol were isolated from the fungus Eurotium chevalieri. The structures were established on the basis of spectroscopic evidence. The configurations of 1 and 5 were determined by X-ray analysis. The biosynthetic pathway of 1-3, 8, and 9 were proposed. Chemical transformation of aszonapyrone A (9) was also studied. Compounds 4, 5, and 7 exhibited antimalarial activity against Plasmodium falciparum, while 3, 5, and 7 showed antimycobacterial activity against Mycobacterium tuberculosis. In addition, compounds 2-7 showed cytotoxicity against cancer cell lines.  相似文献

Cochinchinones I-L (1-3 and 13) along with 11 known xanthones (4-12, 14, and 15) were isolated from the resin and green fruits of Cratoxylum cochinchinense. In addition, four new acetylated compounds (16-19) were derivatized from 7-geranyloxy-1,3-dihydroxyxanthone (14) and 3-geranyloxy-1,7-dihydroxyxanthone (15). All compounds were characterized on the basis of spectroscopic analyses. The structures of cochinchinone I (1), a monoacetate (18) and a dibrosylate (20), were also confirmed by X-ray diffraction analysis. The antibacterial and antifungal activities of selected compounds were evaluated as well.  相似文献
