A novel synthesis of 2,3-disubstituted indoles starting from N-alkyl-N-propvnyl anilines is described. The reactions employed are analogous to the thermal rearrangement of aryl propynyl sulfoxides.  相似文献

A straightforward and atom-economical method is described for the synthesis of 2,3-disubstituted indoles. Anilines and 1,2-diols are condensed under neat conditions with catalytic amounts of either [Cp*IrCl(2)](2)/MsOH or RuCl(3)·xH(2)O/phosphine (phosphine = PPh(3) or xantphos). The reaction does not require any stoichiometric additives and only produces water and dihydrogen as byproducts. Anilines containing methyl, methoxy, chloro and fluoro substituents can participate in the cyclocondensation. Meta-substituted anilines give good regioselectivity for 6-substituted indoles, while unsymmetrical diols afford excellent regioselectivity for the indole isomer with an aryl or large alkyl group in the 2-position. The mechanism for the cyclocondensation presumably involves initial formation of the α-hydroxyketone from the diol. The ketone subsequently reacts with aniline to generate the α-hydroxyimine which rearranges to the corresponding α-aminoketone. Acid- or metal-catalysed electrophilic ring-closure with the release of water then furnishes the indole product.  相似文献

[reaction: see text] The Stille coupling of N-acyl-2-iodoanilines with the 1-(tributylstannyl)-1-substituted allenes affected the successive one-step formation of the 2-methyl-3-substituted indoles. Alternatively, the other type of 2-alkyl-3-substituted indoles could be synthesized in a one-pot operation, which consists of the Stille coupling reaction with the 1-(tributylstannyl)-1,3-disubstituted allenes, followed by TBAF treatment. The tandem hydroformylation-Fischer indolisation protocol is used in the synthesis of 2,3-disubstituted indoles. After hydroformylation of selected olefins to form alpha-branched aldehydes in a one-pot procedure these are condensed with phenylhydrazine to give hydrazones. Upon acid-promoted [3,3]-sigmatropic rearrangement indolenine intermediates with quaternary centres in the 3-position are formed, which, after selective Wagner-Meerwein-type rearrangement of one of the substituents from the 3- to the 2-position, lead to 2,3-disubstituted indoles. Several olefins, bearing substituents with various functional groups, as well as cyclic olefinic systems are investigated.  相似文献

A series of new ethynylated benzoindoles were synthesized starting with the nucleophilic attack of naphthoquinone by lithium acetylides. The resulting 1,4-diethynylnaphthalenes were applied in palladium catalyzed CN coupling - hydroamination domino reactions. A new synthetic method for triazatruxenes from indoles is developed using N-bromosuccinimide (NBS) as a user-friendly reagent. Major reaction parameters including the amount of NBS, substrate concentration, temperature, addition rate and addition method are investigated. Additional experiments are also conducted in order to gain access toward the reaction mechanism. Compared to the use of Br2 in the conventional method, this reaction requires less reaction time, provides better yields, and displays excellent reproducibility. The reaction can be conveniently performed at 10 g scale and it is also applicable to several substituted indoles, benzoindole, and N-alkyl indoles.  相似文献

A new and efficient method has been developed for the one-pot synthesis of novel 2,9-dihydro-2-oxo-4-aryl-1H-pyrido[2,3-b]indole-3-carbonitrile derivatives by condensation of substituted (triethoxymethyl)arene, 1-methyl-1H-indol-2-ol and cyanoacetamide in the presence of a catalytic amount of crosslinked poly(2-acrylamido-2-methyl propane sulfonic acid) (AMPS) as an efficient and heterogeneous catalyst. This polymeric solid acid catalyst is stable and can be easily recovered and reused without significant change in its activity.  相似文献
