1,2,5-Thiadiazole-3-carboxaldehydes 1a-c were prepared by the acid-catalyzed decomposition of 3-azido-methyl-1,2,5-thiadiazoles 2a-c in 68-83% yields, respectively. Pyrolysis of 2a and 2b afforded the imidazoles 4a and 4b in low yields. NBS -bromination of 1a and 1b gave the corresponding carboxylic acids 10a and 10b via acid bromides 9. Azides 2a and 2b gave the s-triazines 8a and 8b on treatment with NBS.  相似文献

3,6;12,15-Di-1,4-benzo[6.6](3,4)-1,2,5-thiadiazolocyclophanes 1a-c were prepared starting from 3,4-di-p-tolyl-1,2,5-thiadiazole 3 and converted into [23]cyclophane-1,2-diones 2a-c by the reaction with Grignard reagents.  相似文献

The reaction of tetrasulfur tetranitride with alkoxybenzenes such as anisole ( 1a ), o- ( 1b ), m-( 1c ), p-dimethoxybenzenes ( 1d ), and benzyl ether ( 1e ) was investigated. Apart from the previous report, the reaction of 3-(4-nitrobenzoylformamido)-4-(4-nitrophenyl)-1,2,5-thiadiazole ( 2a ) with m-chloroperbenzoic acid in chloroform at room temperature did not proceed, whereas at reflux temperature the same reaction gave 4-nitrobenzoic acid ( 5 ) (86%) and a minute amount of a mixture of 4-nitrobenzoylformamide ( 6 ) and 3-amino-4-(4-nitrophenyl)-1,2,5-thiadiazole ( 7a ). On the other hand the same reaction in a mixture of ethanol and chloroform (1:4) at room temperature gave 3-ethoxycarbamoyl-4-(4-nitrophenyl)-1,2,5-thiadiazole ( 8a ) (24%) as an isolable product. When 3-aroylformamido-4-aryl-1,2,5-thiadiazoles 2 in tetrahydrofuran were treated with various alkoxides in the corresponding alcohols at room temperature, 3-amino-4-aryl- 7 , 3-alkoxycarbamoyl-4-aryl- 8 , and 3-aryl-4-(aryl)(hydroxy)acetamido-1,2,5-thiadiazoles 9 were isolated. The ratios of which were dependent on the kind of bases and the solvent employed. Selected compounds 2 were allowed to react with phosphorus pentasulfide in the presence of pyridine at reflux to give 3-aryl-4-arylacetarnido-1,2,5-thiadiazoles 17 (55–64%), which were also produced by the reaction of 2 with either Lawesson's reagent or hydrogen sulfide gas in the presence of pyridine at reflux.  相似文献

3-Isopropenyltropolones 1a-c were treated with bromine in carbon tetrachloride to give 3-methyl-8H-cyclohepta[b]furan-8-ones 2a-c and their corresponding 7-bromo-substituted compounds 3a-c , while reactions in acetic acid gave the bromo-substituted compounds 3a-c . On the other hand, bromination of 1a-c with N-bromosuccinimide afforded 7-bromo-3-(2-bromo-1-methylethenyl)tropolones 5a-c . The compound 2a was treated with bromine to give 2-bromo-3-methyl-8H-cyclohepta[b]furan-8-one ( 4 ). The tropolones 5a-c were heated in the presence of potassium carbonate to give the cyclized compounds 3a-c .  相似文献

Triethylamine catalyzes the reaction of 2-hydroxybenzonitrile ( 1 ) with aryl isocyanates to form the corresponding carbamates 2a-c , as well as the cyclization of the latter compounds to either 4[N-(N-arylcarbamoyl)-imino]-3-aryl-2H-1,3-benzoxazin-2-ones 4a-c , or 4-arylamino-2H-1,3-benzoxazin-2-ones 7a-c , depending on the reaction temperature. Under analogous conditions, the carbamates obtained from 1 and 2-chloroethyl isocyanate, 3-chloropropyl isocyanate and ethyl isocyanatoacetate undergo a double cyclization yielding imidazo- and pyrimido[1,2-c|1,3]benzoxazinones 13a,b,17 . Upon heating in phenyl ether, compounds 7a-c , rearrange to 2-(2-hydroxyphenyl)-4(3H)-quinazolinones 10a-c .  相似文献

Folic acid models, 1,3-dimethyl-6-(N-acylarylamino)methyllumazines 9 , were synthesized from 6-bromomethyl-1,3-dimethyllumazine ( 6 ), which was derived from 5,6-diamino-1,3-dimethyluracil ( 1 ) by the condensation with 1,3-dihydroxyacetone, followed by bromination. The bromide 6 was also prepared by the cycloaddition between 3,6,8-trioxo-5,7-dimethyl-5,6,7,8-tetrahydro-3H-pyrimido[5,4-c][1,2,5]oxadiazine ( 4 ) and 1-propenyl trimethylsilyl ether followed by bromination. The folic acid models 9 were also directly synthesized from the oxadiazine 4 and 3-(N-acylaryl)amino-1-propenyl trimethylsilyl ether 8 by cycloaddition.  相似文献

Reduction of 4,7-diphenyl-1,2,5-thia- ( 1a-i ) and 1,2,5-oxadiazolo[3,4-c]pyridines ( 3a and c-e ) gave 3,4-diamino-2,5-diphenylpyridines ( 2a-g ), which were converted into the fluorescent triazolo[4,5-c]-( 5 ), 1,2,5-selenadiazolo[3,4-c]- ( 6 ), imidazolo[4,5-c]pyridines ( 8 ), and pyrido[5,6-c]pyridines ( 11 ). In the reduction of 3a, c and e , 4,5-dihydro[1,2,5]oxadiazolo[3,4-c]pyridines ( 4a-c ) were obtained.  相似文献

Ring Transformations of 3-Substituted 5-Trifluoromethyl-1,3,4-thiadiazol-2(3H)-one with Nucleophiles The 3-chlormethyl-5-trifluoromethyl-1,3,4-thiadiazolone 3 undergoes a ring transformation to 3-acylated 2,3-dihydro-1,3,4-thiadiazoles 4 with many nucleophiles. Upon formal replacement of the chloromethyl group in the 3-position of 3 by an extended bromoalkyl chain (→9a-c) , the reaction with nucleophiles yields 4-acylated 5,6-dihydro-4H-1,3,4-thiadiazines 10 (from 9a ), 4,5,6,7-tetrahydrothiadiazepines 13 (from 9b ) and 5,6,7,8-tetrahydro-4H-1,3,4-thiadiazocines 14 (from 9c ) by ring enlargement. The 3-propargyl-thiadiazolone 17 rearranges with nucleophiles to 4-acylated 6-methylidene-5,6-dihydro-4H-1,3,4-thiadiazines 18 . The structures of the new compounds were elucidated by 1H- and 13C-NMR spectroscopy.  相似文献

6-Aminouracil and 6-aminothiouracil ( 1a, b ) were reacted with benzylidenemalononitrile 2a to afford the pyrido[2,3-d]pyrimidines 4a, b . On the other hand, the reaction of 1a, b with benzylidene ethyl cyanoacetate 2b result in a mixture of 5a,b and/or 6a,b respectively. Pyrido[2,3-d]thiazolo[1,2-b]pyrimidines 8a-c were synthesized from the reaction of 4b with α-halo compounds to give the intermediate derivatives 7a-c followed by cyclization using 70% H2SO4.  相似文献

Reactions of 4-aminofurazan-3-carboxylic acid iminoester with o-aminophenol and ethylenediamine give rise respectively to 4-(1,3-benzoxazol-2-yl)- and 1-(4,5-dihydro-1H-imidazol-2-yl)-1,2,5-oxadiazol-3-amines, with aminoethanol arises 2-[(Z)-1-amino-1-(4-amino-1,2,5-oxadiazol-3-yl)methylideneamino]-1-ethanol. Treating of 3-amino-4-(1H-benzo[d]imidazol-2-yl)-1,2,5-oxadiazole with triethyl orthoformate in acetic anhydride yielded benzo[4,5]imidazo[1,2-c][1,2,5]oxadiazolo[3,4-e]pyrimidine, and alkylation with haloalkanes furnished 3-amino-4-(1-R-benzo[d]imidazol-2-yl)-1,2,5-oxadiazoles.  相似文献

The reactions of 3-(α-chlorophenylhydrazono)hydrazinocarbonylmethyl-2-oxo-1,2-dihydroquinoxalines 4a,c with triethyl orthoesters resulted in the intramolecular cyclization to give the 3-(α-chlorophenylhydrazono-1,3,4-oxadiazol-2-ylmethyl)-2-oxo-1,2-dihydroquinoxalines 5a-d , but not the 1,2,4,5-tetrazepinylquinoxalines 6a-d . The cyclization mode into the 1,3,4-oxadiazole ring was confirmed by the alternate syntheses of 5a,c from the reactions of the 3-(1,3,4-oxadiazol-2-ylmethylene)-2-oxo-1,2,3,4-tetrahydroquinoxalines 7a,b with o-chlorobenzenediazonium chloride. Moreover, the reactions of 3-(benzimidazol-2-ylmethylene)-2-oxo-1,2,3,4-tetrahydroquinoxaline hydrochloride 8 with o-, m- and p-chlorobenzenediazonium chlorides afforded the 3-(α-chlorophenylhydrazonobenzimidazol-2-ylmethy])-2-oxo-1,2-dihydroquinoxa]ine hydrochlorides 9a-c , respectively. Compounds 5a-d and 9a-c were found to exhibit antimicrobial activities.  相似文献

The synthesis of imidazo[5,1-d]pyrrolo[1,2-b][1,2,5]benzothiadiazepine 9,9-dioxide ( 5 ), a novel sulfur-containing tetracyclic benzodiazepine, is reported starting from pyrrolo[1,2-b][1,2,5]benzothiadiazepine 5,5-diox-ide ( 6 ) by cycloaddition of tosylmethyl isocyanide to the azomethine double bond. Pyrrolobenzothiadiazepine 6 was obtained by iron powder/acetic acid reduction of 1-(2-nitrobenzenefulfonyl)pyrrole-2-carboxaldehyde ( 7 ) and subsequent ring closure of intermediate aminoaldehyde or by cyclization of 1-(2-formamidobenzene-sulfonyl)pyrrole ( 8 ) with phosphorus oxychloride via a Bischler-Napieralski reaction. Formylation of 1-(2-ami-nobenzenesulfonyl)pyrrole with acetic-formic anhydride gave 8. The structure of 5 was confirmed by oxidation with activated manganese dioxide of dihydro derivative 9 , obtained through cyclization of 11-amino-methyl-10,11-dihydropyrrolo[1,2-b][1,2,5]benzothiadiazepine 5,5-dioxide ( 10 ) with triethyl orthoformate. The last compound was prepared alternatively by catalytic reduction of nitro derivative 11 , obtained by addition of nitromethane to pyrrolobenzothiadiazepine 6 , or by lithium aluminum hydride/sulfuric acid reduction of amide 13 , synthesized starting from ethyl 10,11-dihydropyrrolo[1,2-b][1,2,5]benzothiadiazepine-11-carboxyl-ate 5,5-dioxide.  相似文献

Reaction of 2-(3-,4-)pyridinecarboxaldehydes 5 with carbomethoxymethylene triphenylphosphorane afforded predominantly E-methyl-3-(pyridinyl)-2-propenoates 7. Oxidation of 7a-c with m-chloroperbenzoic acid gave methyl E-3-(1-oxidopyridinyl)-2-propenoates 8a-c in high yield. The Darzen's reaction of 5a-c with methyl bromoacetate yielded a mixture of stereoisomers cis- 9a-c and methyl trans-3-(pyridinyl)-2,3-epoxy-propanoates 10a-c in a ratio of 2:1. Oxidation of cis- 9a-c and trans- 10a-c afforded the corresponding cis- 11a-c and methyl trans-3-(1-oxidopyridinyl)-2,3-epoxypropanoates 12a-c in good yield. The reaction of 11a and 12a with cyclic amines as piperidine gave the respective threo- 13 and methyl erythro-2-(1-piperidino)-3-hydroxy-3-(1-oxido-2-pyridino)propanoate 14. The sodium borohydride reduction of the N-alkoxylcarbonyl pyridinium salts of 9c and 10c afforded the corresponding N-alkoxycarbonyl-1,2-dihydropyridyl derivatives 15 and 16. A number of selected compounds ( 7a-c , 9a-c , 10a , 10c , 11a-c and 12a , 12c ) were found to be inactive in the P388 Lymphocytic screen. Compounds 9-12 did not react with the model nucleophile ethanethiol in phosphate buffer at 37°.  相似文献

3-Aryl-1,2,4-triazin-5(2H)-ones 1a-c react with indoles 2a-c in trifluoroacetic acid/chloroform or in boiling butanol or acetic acid to give 3-aryl-6-(indolyl-3)-1,6-dihydro-1,2,4-triazin-5(2H)-ones 3a-g . Oxidation of the dihydro-1,2,4-triazin-5(2H)-ones 3a-e afforded 6-(indolyl-3)-1,2,4-triazin-5(2H)-ones 4a-e , products of nucleophilic substitution of hydrogen in 1a-c . Refluxing 1b with N-methylpyrrote 5b in butanol for an extended time resulted in the formation of 3-(4-chlorophenyl)-6-(1-meuiylpyrrolyl-2)-1,2,4-triazin-5(2H)-one 4h. The reaction of 1a-c with indoles 2a-c , pyrroles 5a,b , 1,3-dimethyl-2-phenylpyrazol-4-one (8) and aminothiazoles 9a,b in acetic anhydride affords the 1-acetyl-3-aryl-6-hetaryl-1,6-dihydro-1,2,4-triazin-5(2H)-ones 6a-s . Reaction of 1a-c with N-methyl-pyrrole 5b in acetic anhydride gives beside the 1:1 addition products 6h-k also the 2:1 addition products 7a-c .  相似文献

Three new thiosemicarbazides (1 a-c ) were prepared from N-[4-phenyl-5-(2-thienyl)-1,2,4-triazol-3-yl]mercaptoacetohydrazide. Reaction of (1 a-c) with the appropriate phenacyl bromide afforded thiazoline derivatives (2 a-i) whereas their reaction with chloroacetic acid or maleic anhydride gave the corresponding thiazolidine derivatives (3 a,b) and (4 a,b) . Cyclodesulfurization of (1 a-c) with DCCD in toluene yielded 5-substituted-amino-1,3,4-oxadiazoles (5 a,b) , while their dehydration with PPA gave the corresponding 5-substituted-amino-1,3,4-thiadiazoles (6 a-c) . Six representative compounds were tested for their in-vitro antimicrobial activity against some pathogenic microorganisms, some of them were proved to be active.  相似文献

The reaction of diazomethane with some (E) and (Z)-2-substituted-4-methylene-5(4)-oxazolones ( 1a-c ) under two different conditions, has been studied. (E) and (Z)-1,2-disubstituted-7-oxo-6-oxa-4-azaspiro[2.4]-hept-4-enes ( 3a-c, 4a-c ) were mainly obtained, together with multiple addition compounds. The reaction showed to be stereoselective only when the substituents were aromatic. Acid hydrolysis of compounds 3a and 4a produced a mixture of (E) and (Z)-3,5-disubstituted-tetrahydrofuran-2-ones ( 8a, 9a ). Smooth methanolysis of the ring led to (E) and (Z)-1-benzamido-cyclopropanecarboxylic esters ( 10a-c, 11a-c ), which, on acid hydrolysis, gave (E) and (Z)-1-amino-2-phenylcyclopropanecarboxylic acids 12a and 13a . The pmr spectra have been analyzed by an iterative computer method, and the computed best values obtained have been used to deduce the stereochemistry of the spiroderivatives.  相似文献

Reactions of tetrasulfur tetranitride (S4N4) with aryl and alkyl bromomethyl ketones 1 in chlorobenzene at reflux temperature gave 3,5-diaroyl- and 3,5-diacyl-1,2,4-thiadiazoles 2 in 17-60% yields. No 1,2,5-thiadiazoles were detected. By heating of the two reactants at 115° without the solvent were also obtained 2 in 5-13% yields. Hydrolysis of 2 with sodium hydroxide in a mixture of ethanol, ethyl acetate, and water (v:v, 4:2:1) at 75° to 85° afforded the heretofore inaccessible 3-aroyl- and 3-acyl-1,2,4-thiadiazoles 3 in 17-79% yields.  相似文献

N-Arylisoindolines 1a-c reacted with (2,4, 7-trinitro-9H-fluoren-9-ylidene)propanedinitrile ( A ) in pyridine with admission of air via a net α-H-atom abstraction and formation of [3-(2-aryl-3-arylimino-2,3-dihydro- 1H-isoindol-1-ylidene)-2-aryl-2,3-dihydro-1H-isoindol-1-ylidene]propanedinitriles 2a-c , N-[2-aryl-3-(2-aryl-3-arylimino-2,3-dihydro-1H-isoindolyl-1-idene)-2,3-dihydro-1H-isoindol-1-ylidene]arenamines 3a, b , N, N'-[2-aryl-1H-isoindole-1,3(2H)-diylidene]bisarenamines 4a, b and N-arylphthalimides 5a-c in moderate yields. 2,4,7-Trinitro-9-fluorenone as well as one reduction product each of the latter and of A, namely compounds 6 and 7 , respectively, are also found. The structure of 2b has been unambiguously confirmed by an X-ray crystal structure analysis. A rationale for the conversions observed is presented. These involve dehydrogenation and oxidative couplings of 1a-c as well as transfer of N-aryl fragment from 1a-c to intermediate products.  相似文献

1, 1-Difluorocyclopropa[a]naphthalene ( 1b ) is prepared in three steps from 4-bromo-1,2-dihydronaphthalene ( 7 ) via carbene addition, benzylic bromination and bidehydrohalogenation. Structural evidence for formation of 1b is based on 1H- and 19F-NMR spectroscopy. Compound 1b is stable in solution at ?30°. Upon reaction with MeOH/H+ it is converted to a 1:2 mixture of 1- and 2-methylnaphthoate ( 10 and 11 , respectively).  相似文献
