The reaction of some 2-aminobenzothiazoles and 2-aminothiazoles with ethyl 3-bromo-4-oxopentanoate was investigated with the aim to obtain the corresponding imidazo[2,1-b]benzothiazole and imidazo[2,1-b]-thiazole derivatives, respectively. In some cases, two unexpected side products were obtained together with the required compound and their structures were elucidated: e.g., 2-aminobenzothiazole reacted with ethyl 3-bromo-4-oxopentanoate to give ethyl 2-methylimidazo[2,1-b]benzothiazole-3-acetate together with ethyl imi-dazo[2,1-b]benzothiazole-2-propionate and ethyl 3-(benzothiazol-2-yl)amino-4-oxopentanoate.  相似文献

The synthesis by two alternative routes of 1-(3-benzo[b]thienyloxy)-3-isopropylamino-2-propanol hydrochloride ( 1a HCl), a thiophenic isoster of Propranolol, and related compounds, is reported. The protecting and enolizing properties of the 2-methoxycarbonyl group on benzo[b]thiophene-3-one, along with its facile removal, are utilized in the first route. In the second one, conversion of 3-bromobenzo[b]thiophene in 1-(3-benzo[b]thienyloxy)-2,3-o-isopropylidenepropane is the key step. On the other hand, hydrolysis of 1-(3-benzo[b]furanyloxy)-2,3-o-isopropylidenepropane to the corresponding diol, in order to obtain a furanic isoster of Propranolol (17a ), was unsuccessful.  相似文献

Thirteen 1-(9,10-dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophen-4-yl)-3-alkylaminoazetidines 11 have been synthesized in three steps from 4-amino-9,10-dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophene ( 6 ), which was obtained from the reduction of either 4-azido 4 or 4-hydroxyimino 5 derivatives. All the compounds have been evaluated as potential antidepressive agents.  相似文献

The current interest in structures that contain a tricyclic framework with a nitrogen bridged central ring has prompted us to examine the synthesis of imino-bridged benzocycloheptapyridines. Treatment of 5,6-trans-dibromobenzo[5,6]cyclohepta[1,2-b]pyridin-11-one with anhydrous methyl amine in THF produced 6-β-bromo-5,6-dihydro-11α-hydroxy-12-methyl-11H-benzo[5,6]cyclohepta[1,2-b]pyridin-5,11-imine, the first example of the desired bridged derivatives.  相似文献

Starting from the readily available aryl 3-methyl-2-benzo[b] furyl ketones a series of 3-sub-stituted thieno[3,4-b] benzofurans and 3-substituted selenolo[3,4-b] benzofurans were prepared in high yield. The parent compound, thieno[3,4-b] benzofuran was prepared through the reaction of thioacetamide with 2-chloromethyl-3-formylbenzo[b] furan in moderate yield.  相似文献

The novel polycyclic heterocyclic ring system, naphtho[2,1-b:4,3-g]bisbenzo[b]thiophene was synthesized from 5-[2-(2-bromo-3-thienyl)ethenyl]naphtho[2,1-b][1]benzothiophene. The assignment of its 1H and 13C nmr spectra was also accomplished by utilizing two-dimensional nmr methods.  相似文献

Furo[3,2-b]benzofuran was demonstrated to be an intermediate for the synthesis of dibenzofuran and 1,4-epoxy-1,4-dihydrodibenzofuran ring systems. The synthesis of the title compounds was carried out by cyclization via isocyanate of (E)-4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylideneacetic acid and 4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylacetic acid respectively, which were obtained by the Wadsworth-Emmons modification of the Wittig reaction of 4,5-dihydro-10H-10-oxobenzo[5,6]cyclohepta[1,2-b]thiophene and triethyl phosphonacetate. The syntheses of the K-imine derivatives of carcinogenic benzo[c]phenanthrene and 7,12-dimethylbenz-[a]anthracene are described. Treatment of trans-5-azido-5,6-dihydrobenzo[c]phenanthren-6-ol ( 3 ) and trans-6-azido-5,6-dihydrobenzo[c]phenanthren-5-ol ( 5 ) with thionyl chloride yielded the corresponding β-chloro azides, which in turn, were reacted with lithium aluminium hydride to give 4b,5a-dihydro-5H-benzo[3,4]-phenanthro[1,2-b]azirine ( 2 ). In a similar manner trans-5-azido-5,6-dihydro-7,12-dimethylbenz[a]anthracen-6-ol ( 11 ) and trans-6-azido-5,6-dihydro-7,12-dimethylbenz[a]anthracen-5-ol ( 13 ) were transformed to the respective chloro azides and, converted into 1a,11b-dihydro-6,11-dimethyl-1H-benz[3,4]anthra[1,2-b]azirine ( 10 ).  相似文献

A convenient synthesis of furo[3,2-b]pyridine and its 2- and 3-methyl derivatives from ethyl 3-hydroxypiconate ( 1 ) is described. The hydroxy ester 1 was O-alkylated with ethyl bromoacetate or ethyl 2-bromopropionate to give the diester 2a or 2b . Cyclization of compound 2a afforded ethyl 3-hydroxyfuro[3,2-b]pyridine-2-carboxylate ( 3 ) which in turn was hydrolyzed and decarboxylated to give furo[3,2-b]pyridin-3-(2H)-one ( 4a ). Cyclization of 2b gave the 2-methyl derivative 4b . Reduction of 4a and 4b with sodium borohydride yielded the corresponding hydroxy derivative 5a and 5b respectively, which were dehydrated with phosphoric acid to give furo[3,2-b]pyridine ( 6a ) and its 2-methyl derivative ( 6b ). 2-Acetylpyridin-3-ol ( 8 ) was converted to the ethoxycarbonylmethyl ether ( 9 ) by O-alkylation with ethyl bromoacetate, which was cyclized to give 3-methylfuro[3,2-b]pyridine-2-carboxylic acid ( 10 ). Decarboxylation of 10 afforded 3-methylfuro[3,2-b]pyridine ( 11 ).  相似文献

Substituted 2-aminobenzo[b]pyrans were synthesized by three-component condensation of aromatic aldehydes, cyanoacetic acid derivatives, and cyclic 1,3-diketones. The molecular and crystal structure of 2-amino-3-(2-methoxyethoxycarbonyl)-4-(2-nitrophenyl)-5-oxo-5,6,7,8-tetrahydro-4H-benzo[b]pyran was established by X-ray diffraction analysis.  相似文献

A variety of eleclrophilic substitution reactions have been carried out on 4-methoxybenzo[b]-thiophene (IIIa) and 4-benzoyloxybenzo[b ] thiophene (IVa). Substitution occurs in the 7-position of IIIa and, with the exception of bromination, in the 7-position of IVa. Bromination of IVa occurs in the 3-position. Bromination of 4-hydroxybenzo[b] thiophene (IIa) occurs in the 5-position. The nmr spectra of eleven disubstituted benzo[b] thiophenes have been tabulated.  相似文献

1H-Cyclopropa[b]naphthalene ( 10a ) and 3-methyl or dimethyl derivatives have been synthesized by interception of appropriately substituted o-quinodimethanes 3 with 1-bromo-2-chlorocyclopropene 5 , and subsequent dehydrohalogenation of the adducts. Differences in the characteristics of several common compounds reported in two independent syntheses of the sulfur analog of psilocin required re-examination of our products. Additional evidence supporting the structures previously reported by us is presented. This includes proof of 3-substitution in 4-methoxy-3-β-dimethylaminoethylbenzo[b]thiophene by deuterium exchange of the 2-proton, demonstration that the intermediate 4-methoxy-N, N-dimethyl-3-benzo[b]thienylglyoxamide is different from the 2-isomer, and confirmation of the structure of 4-methoxy-3-methylhenzo[b]thiophene, an intermediate in one of the reported syntheses, by three different synthetic methods.  相似文献

The 1,4-cycloaddition of dichloroketene to N,N-disubstituted 6-aminomethylene-b,7,8,9-tetra-hydro-5H-benzocyclohepten-5-ones afforded N,N-disubstituted 4-amino-3,3-dichloro-3,4,6,7-tetrahydro-5H-benzo[3,4]cyclohepta[2,l-b]pyran-2-ones only in the case of aromatic or strong hindering aliphatic N-substitution. The adducts gave N,N-disubstituted 4-amino-3-chloro-b,7-dihydro-5H-benzo[3,4]cyclohepta[2,l-b]pyran-2-ones by dehydrochlorination with collidine. Upon chromatography on neutral alumina, two products were instead isolated in the case of usual aliphatic N-substitution (diethylamine, piperidine), namely 6-(2,2-dichloroethylidene)-6,7,8,9-tetrahydro-5H-benzocyclohepten-5-one and the dehydrochlorinated 2-pyrone; this latter was the sole product in the case of pyrrolidine substitution. The 1,4-cycloaddition of sulfene occurred readily to give N,N-disubstituted 4-amino-3,4,6,7-tetrahydro-5H-benzo[3,4]cyclohepta-[1,2-e]-1,2-oxathiin 2,2-dioxidesin the case of both aliphatic and partially aromatic N-substitution.  相似文献

Substituted-3-benzylidenebenzo[b]thiophen-2-ones have been synthesized. 5,6-Dimethoxybenzo[b]-thiophen-2-(3H)one with aniline and triethyl orthoformate gives the 3-anilinomethylene compound, which can be hydrolysed to give the corresponding 3-hydroxymethylene derivative.
