This study aimed to investigate the factors affecting molecular weight of poly[(R)-3-hydroxybutyrate] [P(3HB)] when polyhydroxyalkanoate (PHA) synthase (PhaRCBsp) from Bacillus sp. INT005 was used for P(3HB) synthesis in Escherichia coli JM109. It was found that the molecular weight of P(3HB) decreased with time in mid- and late-phase of culture and was strongly affected by culture temperature. At 37 °C culture temperature, the molecular weight of P(3HB) rapidly decreased from 4.4 × 105 to 4.8 × 104 with culture time, whereas it was almost unchanged at 25 °C. Kinetic analysis suggested that the decrease in molecular weight of P(3HB) was due to random scission of the polymer chain. The decrease in molecular weight of P(3HB) was not observed when PHA synthases other than PhaRCBsp were expressed. This study sheds light on the unique behaviour in molecular weight change of P(3HB) that is synthesized by E. coli expressing PhaRCBsp.  相似文献

Poly[(R)-3-hydroxybutyrate] (P(3HB)) fibers with high tensile strength were prepared by stretching the fibers after isothermal crystallization near the glass transition temperature. Two samples with different molecular weights (Mw = 0.7 × 106 and 4.3 × 106) were used to investigate the effect on tensile strength. Increasing the time for isothermal crystallization of P(3HB) fibers resulted in a decrease in the maximum draw ratio. But, the tensile strength of P(3HB) fibers increased remarkably when the isothermal crystallization time was prolonged to more than 24 h. The tensile strength of low-molecular-weight drawn fibers was higher than that of high-molecular-weight fibers. Therefore, it can be concluded that this procedure does not increase the tensile strength of the high-molecular-weight drawn fibers. This is because, in this drawing method, small crystal nuclei grow initially during the isothermal crystallization process. Then, the molecular chains between the small crystal nuclei that acted as the entanglement points are oriented by stretching. In the case of the high-molecular-weight fibers, because the molecular length between the entanglement points of the small crystal nuclei is too long, the molecular chains are not sufficiently oriented by the stretching process. However, in the case of the low-molecular-weight fibers, the molecular length is suitable for generating the extended chains. Based on the result of X-ray analysis of P(3HB) fibers stretched after isothermal crystallization, fibers have the oriented α-form crystal with 21 helix conformation and β-form with planar zigzag conformation. The enzymatic degradation of the stretched P(3HB) fibers was performed by using an extracellular PHB depolymerase purified from Ralstonia pickettii T1. The enzymatic erosion rate of β-form was faster than that of α-form in the P(3HB) fibers stretched after isothermal crystallization.  相似文献

Polyhydroxyalkanoate (PHA) synthase (PhaC) from Wautersia eutropha was expressed in a wide range of production level in Escherichia coli XL1-Blue cells and its effects on PhaC activity, poly[(R)-3-hydroxybutyrate] [P(3HB)] production and its molecular weights were investigated. The production level of PhaC was controlled both by the amount of chemical inducer (isopropyl-β-d-thiogalactopyranoside, IPTG) added into the medium and the use of different copy number of plasmids. In a flask experiment, as PhaC production level in the cells increased, the PhaC activity also increased in the range of low PhaC concentration. However, PhaC activity did not further increase in the range of high PhaC concentration, probably due to the formation of inclusion body in the cells. The molecular weight of P(3HB) was found to decrease with increasing PhaC activity. This trend was also verified in high cell density cultivation using 10-l jar fermentor. Furthermore, we demonstrated that the use of low copy number plasmid and appropriate induction of PhaC expression were effective in achieving both high productivity and high molecular weight of P(3HB).  相似文献

Cupriavidus necator H16CAc, previously constructed for production of poly((R)-3-hydroxybutyrate-co-(R)-3-hydroxyhexanoate) [P(3HB-co-3HHx)] from soybean oil, was further engineered aiming to increase 3HHx composition in the copolyester. PHA synthase gene derived from Aeromonas caviae on the H16CAc chromosome was replaced by a gene encoding the N149S/D171G mutant and this recombination enhanced PHA productivity as well as slightly increased 3HHx composition. Manipulation of phaACn locus partially reduced the amount of 3HB unit concomitantly with relative increase of 3HHx composition, whereas deletion of phaB1Cn resulted in drastic decline of 3HB unit in P(3HB-co-3HHx). Insertion of phaJAc encoding (R)-specific enoyl-CoA hydratase from A. caviae into pha operon significantly enlarged 3HHx fraction without negative effects on the cell growth and polyester accumulation. Consequently, efficient production of P(3HB-co-3HHx) with 3HHx composition of 5.7-9.9 mol% was successfully achieved from soybean oil by the engineered strains.  相似文献

Unlike polyhydroxyalkanoates (PHAs) copolymers, the controlled and efficient synthesis of PHA terpolymers from triglycerides and fatty acids are yet to be established. This study demonstrates the production of P(3HB-co-3HV-co-3HHx) terpolymer with a wide range of 3HV monomer compositions from mixtures of crude palm kernel oil and 3HV precursors using a mutant Cupriavidus necator PHB4 transformant harboring the PHA synthase gene (phaC) of a locally isolated Chromobacterium sp. USM2. The PHA synthase of Chromobacterium has an unusually high affinity towards 3HV monomer. P(3HB-co-3HV-co-3HHx) terpolymers with 3HV monomer composition ranging from 2 to 91 mol% were produced. Generation of 3HHx monomers was affected by the concentration and feeding time of 3HV precursor. P(3HB-co-24 mol% 3HV-co-7 mol% 3HHx) exhibited mechanical properties similar to that of common low-density polyethylene. P(3HB-co-3HV-co-3HHx) terpolymers with a wide range of 3HV molar fraction had been successfully synthesized by adding lower concentrations of 3HV precursors and using a PHA synthase with high affinity towards 3HV monomer.  相似文献

We attempted to synthesize novel polyhydroxyalkanoate (PHA) containing new 3-hydroxy-4-methylvalerate (3H4MV) monomer from the transformed strain of Cupriavidus necator PHB4 harboring the PHA synthase gene of Chromobacterium sp. USM2 (phaCCs). Novel PHA containing SCL and MCL monomers were successfully synthesized from crude palm kernel oil (CPKO) and isocaproic acid. Results showed that P(3HB-co-1 mol% 3HV-co-3 mol% 3H4MV-co-18 mol% 3HHx) possessed higher thermal stability, higher elastomeric behavior at room temperature and higher ductility than the P(3HB-co-5 mol% 3HHx). The novel PHA film was found to possess an interesting rubber-like elasticity and flexibility property which has not been reported. The soil surface degradation study showed that the novel PHA film was degraded faster than the 3HB homopolymer and copolymer with 5 mol% of 3HHx.  相似文献

Poly[(R)-3-hydroxybutyrate-co-4-hydroxybutyrate] (P(3HB-co-4HB)) films were prepared by uniaxial cold-drawing from an amorphous preform at a temperature below, but close to the glass transition temperature. Molecular and highly-ordered structures and physical properties of cold-drawn films were investigated by tensile testing, wide-angle X-ray diffraction and small-angle X-ray scattering. Enzymatic degradation of P(3HB-co-4HB) films was performed using an extracellular polyhydroxybutyrate depolymerase purified from Ralstonia pickettii T1. Tensile strength, elongation to break and Young’s modulus of P(3HB-co-4HB) with cold-drawn ratio 1200% reached 290 MPa, 58% and 2.8 GPa, respectively. X-ray fibre diagrams of cold-drawn P(3HB-co-4HB) films showed a strong reflection on the equatorial line, indicating a planar zigzag conformation (β-form) together with 21 helix conformation (α-form). The β-form seems to contribute to the high tensile strength, and a new mechanism of generation of the β-form is proposed. The enzymatic degradation rate increased with increasing draw ratio, and increased greatly with increasing 4HB content.  相似文献

Atom transfer radical polymerization (ATRP) of tert-butyl methacrylate (tBMA) was investigated using cuprous bromide with different ligands, solvents, deactivators, etc. The polymerization in bulk and diphenyl ether solvent system performed using Cu(I)Br complexed with NNN′, N″, N″-pentamethyldiethylenetriamine (PMDETA) catalyst in conjunction with 2-bromopropionitrile as an initiator at room temperature showed a curvature in the first-order kinetic plot. The controlled polymerization in methanol solution resulted in slower rate of polymerization and lower molecular weights. Well-defined diblock copolymers of PSt-b-PtBMA synthesized by polystyrene bromo macroinitiator (PSt-Br) with Cu(I)Cl/PMDETA catalyst system yielded predetermined molecular weights and lower polydispersities. Otherwise, the Cu(I)Br/PMDETA catalytic system showed an inefficient polymerization of tert-butyl methacrylate with lower molecular weights and higher polydispersities. Subsequent hydrolysis of the homopolymer refluxed in dioxane with addition of HCl afforded well-defined poly(methacrylic acid).  相似文献

The reaction of N9,N9′-(tri or tetramethylene)-bisadenines (Ade2Cx; x = 3 or 4) in HCl 2 M at 50 °C with MCl2 · 2H2O [M = Zn(II), Cd(II)] yields outer sphere compounds like the previously described [(H-Ade)2C3][ZnCl4] · H2O (3) and [(H-Ade)2C3]2[Cd2Cl8(H2O)2] · 4H2O (4) for Ade2C3 and the new {[(H-Ade)2C4][Cd2Cl6(H2O)2] · 2H2O}n (5) for Ade2C4. On the other hand, only in case of Zn(II) complexes by changing [HCl] to 0.1 M, the inner sphere compounds [H-(Ade)2C3(ZnCl3)] (6) and [H-(Ade)2C4(ZnCl3)] · 1.5H2O (7) are obtained. X-ray diffraction study of compound 6, which represents the first inner sphere complex with a N9,N9′-bisadenine, shows a zwitterionic form with one adenine ring protonated at N(1) while the other ring is coordinated via N(7) to a ZnCl3 moiety as in other alkyl-adenine derivatives. In addition, with Ade2C4, is also possible to obtain another inner sphere complex: [(H-Ade)2C4(ZnCl3)2] · 3H2O (8).  相似文献

This paper presents the degradation trends of selected polyhydroxyalkanoate (PHA) films in a tropical mangrove environment. The biodegradability of homopolymer poly(3-hydroxybutyrate) [P(3HB)] and its co-polymers, poly(3-hydroxybutyrate-co-5 mol% 3-hydroxyvalerate) [P(3HB-co-5 mol% 3HV)] and poly(3-hydroxybutyrate-co-5 mol% 3-hydroxyhexanoate) [P(3HB-co-5 mol% 3HHx)], was investigated along with P(3HB) films containing 38 wt% titanium dioxide (TiO2) [P(3HB)-38 wt% TiO2]. The degradation of these formulations was monitored for 8 weeks at three different zones in an intermediate mangrove compartment along Sungai Pinang, adjacent to a famous fishing village on south of Penang Island. The degradation rate was observed both on the surface and in the sediment and was expressed in percentage of weight loss. The microbial enumeration done using sediment from the different zones indicated similar colony-forming unit (CFU) counts even though differences were noticed in the degradation profile of the various films in the respective zones. The results obtained revealed that co-polymers disintegrated at similar or higher rate than the homopolymer, P(3HB). However, the incorporation of TiO2 into PHB films caused the degradation rate of P(3HB)-38 wt% TiO2 composite film to be far slower than all the other PHA films. The overall rate of degradation of all PHA films placed on the sediment surface was slower than those buried in the sediment. Microscopic analyses showed that the surface morphology of P(3HB-co-5 mol% 3HHx) was more porous compared to P(3HB) and P(3HB-co-5 mol% 3HV) films, which may be an important factor for its rapid degradation.  相似文献

Adsorption effects of poly(hydroxybutyric acid) (PHB) depolymerase from Ralstonia pickettii T1 on various polymer single crystals were studied using a catalytically inactive mutant of PHB depolymerase by means of transmission electron microscopy (TEM), atomic force microscopy (AFM), and frictional force microscopy (FFM). Six types of polymer single crystals, poly[(R)-3-hydroxybutyric acid] (P(3HB)), poly[(R)-3-hydroxybutyric acid-co-6 mol% (R)-3-hydroxyvaleric acid] (P(3HB-co-6 mol% 3HV)), poly[(R)-3-hydroxybutyric acid-co-8 mol% (R)-3-hydroxyhexanoic acid] (P(3HB-co-8 mol% 3HH)), poly(l-lactic acid) (PLLA), poly(d-lactic acid) (PDLA), and polyethylene (PE), were prepared to examine the influence of an ester bond and stereoregularity of a polymer on the enzymatic adsorption. The numbers of PHB depolymerase enzymes adsorbed on P(3HB) and P(3HB-co-6 mol% 3HV) single crystals were determined as 171 and 183 enzymes/μm2 by AFM, respectively. AFM observation revealed that the concentration of PHB depolymerase enzymes adsorbed onto PLLA and PDLA single crystals is much higher compared to those on a P(3HB) single crystal, whereas the concentration of enzyme adsorbed onto PE and P(3HB-co-8 mol% 3HH) single crystals is much less. In addition, the single crystals of each polymer were characterized by TEM and FFM before and after enzymatic treatment by mutant for 1 h at 37 °C. The surface properties of P(3HB), P(3HB-co-6 mol% 3HV), and P(3HB-co-8 mol% 3HH) single crystals were changed by the enzymatic adsorption, whereas the internal structures were not affected. On the basis of these results, the properties of the binding domain of PHB depolymerase to polymer chain-folding surfaces have been discussed.  相似文献

Poly(3-hydroxybutyrate) [P(3HB)] homopolymer and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] copolymer was produced by Comamonas sp. EB172 using single and mixture of carbon sources. Poly(3-hydroxyvalerate) P(3HV) incorporation in the copolymer was obtained when propionic and valeric acid was used as precursors. Incorporation of 3HV fractions in the copolymer varied from 45 to 86 mol% when initial pH of the medium was regulated. In fed-batch cultivation, organic acids derived from anaerobically treated palm oil mill effluent (POME) were shown to be suitable carbon sources for polyhydroxyalkanoate (PHA) production by Comamonas sp. EB172. Number average molecular weight (Mn) produced by the strain was in the range of 153-412 kDa with polydispersity index (Mw/Mn) in the range of 2.2-2.6, respectively. Incorporation of higher 3HV units improved the thermal stability of P(3HB-co-3HV) copolymer. Thus the newly isolated bacterium Comamonas sp. EB172 is a suitable candidate for PHA production using POME as renewable and alternative cheap raw materials.  相似文献

The aim of this paper was to demonstrate a fluorescence measurement method for rapid detection of two bacterial count by using water-soluble quantum dots (QDs) as a fluorescence marker, and spectrofluorometer acted as detection apparatus, while Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) were as detection target bacteria. Highly luminescent water-soluble CdSe QDs were first prepared by using thioglycolic acid (TGA) as a ligand, and were then covalently coupled with target bacteria. The bacterial cell images were obtained using fluorescence microscopy. Our results showed that CdSe QDs prepared in water phase were highly luminescent, stable, and successfully conjugated with E. coli and S. aureus. The fluorescence method could detect 102-107 CFU/mL total count of E. coli and S. aureus in 1-2 h and the low detection limit is 102 CFU/mL. A linear relationship of the fluorescence peak intensity and log total count of E. coli and S. aureus have been established using the equation Y = 118.68X − 141.75 (r = 0.9907).  相似文献

Mononuclear, square-planar platinum(II) complexes involving derivatives of aromatic cytokinins as the ligands, and having the general formula cis-[Pt(Ln)2Cl2] (13) and trans-[Pt(Ln)2Cl2] (46), where n = 1–3, L1 = 2-chloro-6-(benzylamino)-9-isopropylpurine, L2 = 2-chloro-6-[(4-methoxybenzyl)amino]-9-isopropylpurine and L3 = 2-chloro-6-[(2-methoxybenzyl)-amino]-9-isopropylpurine, have been synthesized and characterized by elemental analysis, MALDI-TOF mass, FT IR, 1H, 13C, 15N and 195Pt NMR spectral measurements. Dynamic cis-to-trans isomerization process of complex 1 in N,N′-dimethylformamide (DMF) has been investigated by means of multinuclear NMR spectroscopy. The solid-state structures of 1, 4 · (DMF)2, and 5 have been determined by single crystal X-ray analysis. X-ray structures revealed that the heterocyclic ligands are coordinated to platinum via nitrogen atom N(7) in all the complexes studied. In vitro cytotoxicity of the prepared complexes against MCF7, G361, K562, and HOS has been evaluated. Owing to low solubility of the complexes in water, the cytotoxicity has been only tested up to 5 μM concentration. Unfortunately, all complexes have been found to be non-cytotoxic in the accessible concentration range.  相似文献

A new ruthenium complex with benz[f]indenyl ligand, (η5-benz[f]indenyl)Ru(PPh3)2Cl (4), was synthesized and characterized by X-ray diffraction analysis. Complex 4 was examined in comparison with its indenyl analogue (η5-indenyl)Ru(PPh3)2Cl (3) in living radical polymerization of styrene (St) and methyl methacrylate (MMA). Linear increase of molecular weights with conversion and low polydispersities (1.09-1.19 for St, 1.11-1.17 for MMA) were obtained with 4.  相似文献

A novel dichloro zinc complex (L1)ZnCl2, where L1 is N,N′-bis-(2,6-dichloro-benzyl)-(R,R)-1,2-diaminocyclohexane, has been synthesized and characterized. The dimethyl derivatives, generated in situ from the well characterized dichloro zinc complexes (L1)ZnCl2 and (L2)ZnCl2, where L2 is N,N′-bis-(benzyl)-(R,R)-1,2-diaminocyclohexane, were employed as initiators for the ring opening polymerization (ROP) of rac-lactide (rac-LA). The complexes were found to be highly efficient initiators yielding the polylactide (PLA) with a narrow molecular weight distribution. The catalytic activity and heterotactic selectivity of the Zn(II) complexes were affected by the substituents on the phenyl groups of benzyl moieties in (R,R)-1,2-diaminocyclohexane. The dimethyl derivative of (L2)ZnCl2 produced highly stereocontrolled PLA with Pr = 0.75 at −25 °C.  相似文献

Polymer-linked PT (podophyllotoxin) conjugates have been designed to improve the therapeutic efficacy of PT. A new PT-conjugate, 3,6-endo-methylene-1,2,3,6-tetrahydrophthalimido-acetamidoglycinylglycine podophyllotoxin ester (ETPA-gly-gly-PT), was synthesized by covalently coupling its hydroxyl group onto the phthalimido monomer through a glycine-glycine-glycine spacer. Its homo- and copolymer with acrylic acid (AA) were prepared by photopolymerization using 2,2-dimethoxy-2-phenylacetophenone (DMP) as a photoinitiator. ETPA-gly-gly-PT and its polymers were characterized by IR and proton NMR spectra. The ETPA-gly-gly-PT content in the copolymer obtained by elemental analysis was 44 wt%. The number-average molecular weights of the polymers determined by gel permeation chromatography were as follows: Mn = 13,500 for poly(ETPA-gly-gly-PT), Mn = 17,000 for poly(ETPA-gly-gly-PT-co-AA). The in vitro antitumor activity of these conjugates and polymers were determined and used to evaluate the potential applications in antitumor drugs. The IC50 values indicated that the synthesized ETPA-gly-gly-PT and its polymers against cancer cells were much better inhibitors than PT.  相似文献

The synthesis and physico-chemical characterization of Fe(II) and Mn(II) complexes of 4,6-di-tert-butyl-3[(2-hydroxyethyl)sulphanyl]-1,2-dihydroxybenzene (HLI) and 2-amino-4,6-di-tert-butylphenol (HLII) were carried out. Antibacterial activity of the Co(II), Fe(II) and Mn(II) complexes was evaluated in comparison with Cu(II) complexes and three common antibiotics; it was found to follow the order: (1) Сu(LI)2 > Сo(LI)2 > Fe(LI)2 ? Mn(LI)2 > HLI; (2) Сu(LII)2 > Сo(LII)2 > HLII > Fe(LII)2 ? Mn(LII)2; and their reducing ability (determined electrochemically) followed the same order. Spectrophotometric investigation was carried out in order to estimate the rate of the reduction of bovine heart сytochrome c with the ligands and their metal(II) complexes. NADPH:cytochrome P450-reductase was found to increase the rate of сytochrome c reduction with HLI and HLII ligands, while adrenodoxin in couple with NAD(P)H: adrenodoxin reductase had no substantial effect thereon. It was shown that the reduction of сytochrome c with these compounds cannot be related solely to the facility of their oxidation оr ionization.  相似文献

A series of new substituted triphenylamine (TPA) derivatives with alkyl thieno[3,2-b]thiophene and thiophene units were synthesized in a combinatorial manner. Suzuki coupling of a dioxaborolane TPA derivative and 2-bromo-3-nonylthieno[3,2-b]thiophene or Stille coupling of fresh stannyl thieno[3,2-b]thiophene was used. All compounds were characterized by 1H and 13C NMR, HRMS, UV-vis spectrometry and DSC measurements. It was demonstrated that the optical and thermal properties of these materials can be tuned by varying both the conjugation length and thienothiophene and thiophene combination on the TPA branches. Moreover, the measured molar extinction coefficients were increasing from 63,000 (λmax = 354 nm) to 131,000 L mol−1 cm−1 (λmax = 428 nm) for TPA-thienothiophenes and TPA-bithiophene thienothiophenes, respectively. The synthesis of tert-butyl acrylate by atom transfer radical polymerization (ATRP) is reported. This polymer was prepared using FeCl2 · 4H2O(PPh3)2 catalyst system in conjunction with methyl 2-bromopropionate as initiator, in bulk and in solution using acetone as a solvent. The addition of solvent was necessary in order to decrease the polymerization rate and to afford low polydispersity polymers. The number-average molecular weights of the resulting polymers increased in direct proportion to the monomer conversion, and the polydispersities (Mw/Mn) were as low as 1.2. In addition, the preparation of an AB diblock copolymer of poly (n-butyl methacrylate)-block-poly (tert-butyl acrylate) by ATRP is reported. The resulting polymers and copolymers were characterized by means of size exclusion chromatography and 1H-NMR Spectroscopy.  相似文献
