Hypericum perforatum L. (St. John’s Wort) is an important medicinal plant which is widely used in the treatment for depression and irritable bowel syndrome. It is also used as a dietary supplement. Major bioactive phytochemicals of H. perforatum are phenolics and flavonoids. Quality of these phytochemicals is dramatically influenced by environmental and biological factors in the field grown plants. As an alternative, we have developed adventitious root cultures in large-scale bioreactors for the production of useful phytochemicals. Adventitious roots of H. perforatum were cultured in 500 l pilot-scale airlift bioreactors using half-strength Murashige and Skoog medium with an ammonium and nitrate ratio of 5:25 mM and supplemented with 1.0 mg l?1 indole butyric acid, 0.1 mg l?1 kinetin, and 3 % sucrose for the production of bioactive phenolics and flavonoids. Then 4.6 and 6.3 kg dry biomass were realized in the 500 l each of drum-type and balloon-type bioreactors, respectively. Accumulation of 66.9 mg g?1 DW of total phenolics, 48.6 mg g?1 DW of total flavonoids, 1.3 mg g?1 DW of chlorogenic acid, 0.01 mg g?1 DW of hyperin, 0.04 mg g?1 DW of hypericin, and 0.01 mg g?1 DW of quercetin could be achieved with adventitious roots cultured in 500 l balloon-type airlift bioreactors. Our findings demonstrate the possibilities of using H. perforatum adventitious root cultures for the production of useful phytochemicals to meet the demand of pharmaceutical and food industry.  相似文献

Adventitious root cultures of Prunella vulgaris L. were established in shaking flask system for the production of biomass and secondary metabolites. Adventitious root cultures were induced from callus cultures obtained from leaf explants on solid Murashige and Skoog (MS) medium containing combination of 6-benzyladenine (BA; 1.0 mg l?1) and naphthalene acetic acid (NAA; 1.5 mg l?1). Thereafter, 0.49 g inoculum was transferred to liquid MS medium supplemented with different concentrations of NAA (0.5–2.0 mg l?1). Growth kinetics of adventitious roots was recorded with an interval of 7 days for 49 days period. Highest biomass accumulation (2.13 g/l) was observed in liquid medium containing 1.0 mg l?1 NAA after 21 days of inoculation. However, other concentrations of NAA also showed similar accumulation pattern but the biomass gradually decreases after 49 days of inoculation. Adventitious roots were collected and dried for investigation of total phenolics (TP), total flavonoids (TF), and antioxidant activities. Higher TPC (0.995 GAE mg/g-DRB) and TFC (6.615 RE mg/g-DRB) were observed in 0.5 mg l?1 NAA treated cultures. In contrast, higher antioxidant activity (83.53 %) was observed 1.5 mg l?1 NAA treated cultures. These results are helpful in up scaling of root cultures into bioreactor for secondary metabolites production.  相似文献

Adventitious root cultures derived from leaf derived callus of Withania somnifera (L.) Dunal were treated with methyl jasmonate and salicylic acid independently. Biomass accumulation, culture age, elicitation period, and culture duration were optimized for higher withanolides production in the two best-responding varieties collected from Kolli hills (Eastern Ghats) and Cumbum (Western Ghats) of Tamil Nadu, India. Between the two elicitors, salicylic acid (SA) improved the production of major withanolides (withanolide A, withanolide B, withaferin A, and withanone) as well as minor constituents (12-deoxy withastramonolide, withanoside V, and withanoside IV) in the Kolli hills variety. Treatment of root biomass (11.70?g FW) on 30-day-old adventitious root cultures with 150???M SA for 4?h elicitor exposure period resulted in the production of 64.65?mg?g?l dry weight (DW) withanolide A (48-fold), 33.74?mg?g?l DW withanolide B (29-fold), 17.47?mg?g?l DW withaferin A (20-fold), 42.88?mg?g?l DW withanone (37-fold), 5.34?mg?g?l DW 12-deoxy withastramonolide (nine fold), 7.23?mg?g?l DW withanoside V (seven fold), and 9.45?mg?g?l DW withanoside IV (nine fold) after 10?days of elicitation (40th day of culture) when compared to untreated cultures. This is the first report on the use of elicitation strategy on the significant improvement in withanolides production in the adventitious root cultures of W. somnifera.  相似文献

In the study, anti-Candida activity and phenol contents of Lythrum salicaria L. calli and wild species have been evaluated. The seeds of L. salicaria (Lythraceae), collected from Lahidjan City in the north of Iran, were cultured in Murashige and Skoog medium (MSM) with a supplement, gibberellin, to germinate. Callus inductions were performed from segments of seedling on MSM containing different concentrations of plant growth regulators, 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP). The activity of calluses extracts, wild plant, gallic acid, and 3,3′,4′-tri-O-methylellagic acid-4-O-β-d-glucopyranoside (TMEG) as the main phenolic compounds against Candida albicans was assessed using cup plate diffusion method. The total phenols contents of calli and wild plant extracts were analyzed using Folin–Ciocalteu reagent. The callus formation in MSM supplemented with various concentrations of 2,4-D and BAP were 0–100 %. Anti-Candida activity of callus extract which obtained from MSM supplemented with 2,4-D and BAP (1 mg?dm?3) was similar to the wild plant extract. Minimum inhibitory concentration values of gallic acid and TMEG were obtained as 0.312 and 2.5 mg?cm?3, respectively. Gallic acid equivalent values in all treatments were from 0 to 288 μg GAE mg?1. Phenolic contents of plant aerial parts (331?±?3.7 μg GAE mg?1) and the callus, which developed in MSM including 1 mg?dm?3 of both 2,4-D and BAP, showed the same phenolic value and exhibited anti-Candida extract activity.  相似文献

Five naturally bioactive compounds, belonging to the representative flavonoid and pentacyclic triterpenoid types, were separated by liquid chromatography and analyzed on-line with negative ion ESI–MS. Rutin, quercetin, and oleanolic acid in the extracts of Rosa multiflora Thunb. of China were identified by the comparison of their retention times and mass spectrometric data with those of authentic standards, and their contents were 0.22, 3.8, and 45 µg g?1 of dry weight in rose hips, respectively. Five standards, including rutin, quercetin, kaempferol, lysionotin, and oleanolic acid were investigated by MS3, which facilitate further understanding of their structures.  相似文献

Glycosides including triterpenoid saponins and flavonoid glycosides are the main constituents of Glycyrrhiza uralensis Fisch (licorice) and exhibit prominent pharmacological activities. However, conventional methods for the separation of glycosides always cause irreversible adsorption and unavoidable loss of sample due to their high hydrophilicities. The present paper describes a convenient method for the simultaneous separation of triterpenoid saponins and flavonoid glycosides from licorice by pH‐zone‐refining counter‐current chromatography. Ethyl acetate/n‐butanol/water (2:3:5, v/v) with 10 mM TFA in the upper organic stationary phase and 10 mM ammonia in the lower aqueous mobile phase was used as the biphasic solvent system. Three triterpenoid saponins and two flavonoid glycosides including licorice‐saponin A3 (63.3 mg), glycyrrhizic acid (342.2 mg), 3‐O‐[β‐d ‐glucuronopyranosyl‐(1 → 2)‐β‐d ‐galactopyranosyl]glycyrrhetic acid (56.0 mg), liquiritin apioside (232.6 mg), and liquiritin (386.5 mg) were successfully obtained from licorice ethanol extract (2 g) in one step. This method subtly takes advantage of the common acidic properties of triterpenoid saponins and flavonoid glycosides, and obviously is much more efficient and convenient than the previous methods. It is also the first time that the separation of acidic triterpenoid saponins by using pH‐zone‐refining counter‐current chromatography has been reported.  相似文献

The mucilage in Lepidium sativum L. is considered a biologically active compound with diverse medicinal properties. Different explants (hypocotyls and leaf) were transferred to Murashige and Skoog (MS) medium supplemented with twelve different plant growth regulator combinations under two different incubations (light and dark). The best mucilage production from callus (36.76% g g?1 dry weight) was obtained in the MS medium supplemented with 1 mg L?1 of 2, 4-D and 2 mg L?1 of BAP under the light condition. The mucilage produced by callus culture was nearly three times more than the mucilage yield of the seeds. The glucose, arabinose + mannose and galactose were 43.4 (mg g?1 DW), 195.3 (mg g?1 DW) and 86.2 (mg g?1 DW) in the mucilage originated from seed, callus leaf and callus hypocotyl, respectively. The present study proposes an efficient method for producing large scales of mucilage with a favorable sugar aimed at food or pharmaceutical industries.  相似文献

This study aimed to compare phytochemical profiles and antioxidant capacity of various extracts including crude extracts, aqueous- and saponin-enriched butanol fractions prepared from the stems and leaves of Helicteres hirsuta Lour. The results revealed that all the three powdered extracts from the leaves and the stems possessed high levels of phenolics (177.07–241.03 mg GAE g?1), flavonoids (158.03–280.06 mg CE g?1) and saponins (165.77–1035.33 mg ESE g?1) and exhibited strong antioxidant capacity. HPLC analysis identified nine major compounds in the leaf powder crude extract; however, the leaf aqueous fraction had three extra compounds; whereas, the saponin-enriched butanol leaf fraction had seven extra compounds. For the stems, twelve main compounds were evident in either the powdered crude extract or the aqueous fraction, and five new compounds were revealed in the saponin-enriched butanol fraction. The findings revealed that the powdered aqueous fractions and saponin-enriched butanol fractions are potential sources of biologically active compounds for further investigation and industrial utilisation.  相似文献

Licorice is among the most popular herbal medicines and frequently used in traditional medicine, food products, and cosmetics. In China, only Glycyrrhiza uralensis Fisch., Glycyrrhiza inflata Bat. and Glycyrrhiza glabra L. are officially used and are usually processed with honey prior to use. To maintain the quality of commercially available herbal products, a simple, rapid, and reliable ultra high performance liquid chromatography with triple quadrupole mass spectrometry was developed to investigate the major active constituents of commercially available licorice products. Nineteen components were accurately determined, including eight triterpenoid saponins, one triterpene, and ten flavonoids. Subsequently, multivariate statistical analysis methods were employed to further explore and interpret the experimental data. The results indicated that liquiritin apioside may be considered as a candidate index for the quality control of licorice as well as 18β‐glycyrrhizic acid and liquiritin. In addition, both 18β‐glycyrrhizic acid and licorice‐saponin G2 can be used for discrimination between crude and honey‐processed licorice. Furthermore, using 18β‐glycyrrhizic acid and liquiritin as markers, this work revealed that the quality of licorice products may have declined in recent years. This highlights the need for additional effort focused on good agricultural practice during the processing of licorice. In summary, this study provides a valuable reference for the quality assessment of licorice.  相似文献

Methyl jasmonate (MJ)-induced changes of triterpene saponins in Panax ginseng C.A. Meyer adventitious roots, and expression of the genes involved in triterpene biosynthesis, were analyzed. Compared with controls, expression of the squalene epoxidase and dammarenediol synthase genes was clearly upregulated and total saponin content increased in response to MJ. The highest total saponin content was obtained by use of 10 mg L?1 MJ for 24 h, and was 4.76-fold greater than in the control group. Expression of the two genes associated with the cytochrome P450 family was no different from that in controls. The level of ginsenoside in the Rg group thus increased much less than that in the Rb group. This investigation showed that the total saponin content of ginseng adventitious root is related to gene expression in ginsenoside biosynthesis.  相似文献

Dioscorea alata var. purpurae (Indian purple yam) is an important source of diosgenin, a triterpenoid that is used as a raw material in the synthesis of corticosteroid hormones. These drugs are used for the treatment of pharmacological conditions such as arthritis. This paper reports in vitro propagation of explants of various parts of Dioscorea—tuber, leaves, and nodes. Murashige and Skoog media supplemented with hormones and additives was used to get maximum callus initiation and shoot/root induction. All the cultures were maintained at 25?±?2 °C under cool-white fluorescent tubes with 16-h photoperiod. Callus initiation was observed from 8th to 11th day of inoculation, and subsequent root/shoot was initiated in nodal callus after 21 days. Hormones such as kinetin, indole-3-acetic acid, indole-3-butyric acid, α-naphthalene acetic acid, and thiadizuron did not show significant enhancement. Also, there was no need for supplementing additives (silver nitrate, glutamine, l-asparagine monohydrate, polyethylene glycol). Combination of 6-benzylaminopurine (0.2 ppm) and 2,4-dichlorophenoxyacetic acid (2 ppm) hormones gave the best results, and all parts of the plants gave similar callus induction.  相似文献

Batch cultivation of Azadirachta indica hairy roots was carried out in different liquid-phase bioreactor configurations (stirred-tank, bubble column, bubble column with polypropylene basket, and polyurethane foam disc as root supports) to investigate possible scale-up of the A. indica hairy root culture for in vitro production of the biopesticide azadirachtin. The hairy roots failed to grow in the conventional bioreactor designs (stirred tank and bubble column). However, modified bubble column reactor (with polyurethane foam as root support) configuration facilitated high-density culture of A. indica hairy roots with a biomass production of 9.2 g l?1dry weight and azadirachtin yield of 3.2 mg g?1 leading to a volumetric productivity of azadirachtin as 1.14 mg l?1 day?1. The antifeedant activity in the hairy roots was also evaluated by no choice feeding tests with known concentrations of the hairy root powder and its solvent extract separately on the desert locust Schistocerca gregaria. The hairy root powder and its solvent extract demonstrated a high level of antifeedant activity (with an antifeedant index of 97 % at a concentration of 2 % w/v and 83 % at a concentration of 0.05 % (w/v), respectively, in ethanol).  相似文献

The heavy metal content was investigated for six mushroom species native to Jordan. Metal (Cu, Pb, Cd, Fe, Zn, Mn, Ni, and Co) content in soil substrate and their relation to metal concentrations in mushroom and underlying soil were determined by flame and graphite furnace atomic absorption spectrometry. Mushroom species and soil were collected from different places in Jordan. The highest Cu level was 51.84 µg g?1 for the species Lepista nuda; whereas, the lowest Cu level was found to be 18.51 µg g?1 in Calvatia utriformis. Among the wild mushrooms, the highest Pb level was found as 4.81 µg g?1 in Bovista plumbea, whereas the lowest Pb concentration was 2.01 µg g?1 in Calvatia utriformis. The highest Cd level was determined as 1.9 µg g?1 for Lepista nuda, whereas the lowest Cd level was 0.58 µg g?1 for the species of Polyporus frondosus. The highest Zn level was 58.77 µg g?1 for the species of Lepista nuda and the lowest Zn concentration was found 35.98 µg g?1 in Calvatia utriformis. The highest Fe level was found as 317 µg g?1 in Lepista nuda, whereas the lowest Fe concentration was 211.7 µg g?1 in Calvatia utriformis. The highest Mn content was 36.55 µg g?1 for Russula delica, whereas the lowest Mn level was 24.5 µg g?1 for the species Bovista plumbea. The highest Ni content was found as 12.65 µg g?1 for Russula delica, whereas the lowest Ni level was 0.17 µg g?1 for Bovista plumbea. The highest Co content in the tested mushrooms was found as 3.5 µg g?1 for the species of Agaricus bisporus, whereas the lowest Co level was 0.85 µg g?1 for Polyporus frondosus. The results indicated that, in general, heavy metal contents in all mushroom species were lower than the underlying soil substrates except for some mushroom species.  相似文献

Abstract

The objective of this study was to examine the bioactive flavonoids and carotenoids concentration in fifteen south Indian spice and two tree nut species using high performance liquid chromatography (HPLC). Among four flavonoids, catechin concentration was the highest in all spices and nuts and ranged between 97.1 and 1745.4 µg g?1. Quercetin concentration was the greatest in cinnamon, followed by garlic and cumin and ranged from 0.4 to 65 µg g?1 in other spices and nuts. Lutein concentration ranged from 0.1 to 102.8 µg g?1. Of the spices and nuts studied, β-carotene concentration was highest in coriander leaves (74.7 µg g?1), followed by red pepper (12.5 µg g?1) and curry leaves (8.5 µg g?1). This research shows that consumption of south Indian spices and nuts could substantially benefit consumers living in regions experiencing Vitamin A and other micronutrient deficiencies.  相似文献

The present study involved strategies for enhancement in in vitro azadirachtin (commercially used biopesticide) production by hairy root cultivation of Azadirachta indica. Improvement in the azadirachtin production via triggering its biosynthetic pathway in plant cells was carried out by the exogenous addition of precursors and elicitors in the growth medium. Among the different abiotic stress inducers (Ag+, Hg+2, Co+2, Cu+2) and signal molecules (methyl jasmonate and salicylic acid) tested, salicylic acid at 15 mg l?1 of concentration was found to enhance the azadirachtin yield in the hairy roots to the maximum (up to 4.95 mg g?1). Similarly, among the different biotic elicitors tested (filter-sterilized fungal culture filtrates of Phoma herbarium, Alternaria alternata, Myrothecium sp., Fusarium solani, Curvularia lunata, and Sclerotium rolfsii; yeast extract; and yeast extract carbohydrate fraction), addition of filter-sterilized fungal culture filtrate of C. lunata (1 %?v/v) resulted in maximum azadirachtin yield enhancement in hairy root biomass (up to 7.1 mg g?1) with respect to the control (3.3 mg g?1). Among all the biosynthetic precursors studied (sodium acetate, cholesterol, squalene, isopentynyl pyrophosphate, mavalonic acid lactone, and geranyl pyrophosphate), the overall azadirachtin production (70.42 mg l?1 in 25 days) was found to be the highest with cholesterol (50 mg l?1) addition as an indirect precursor in the medium.  相似文献

Linnaea borealis L. (Twinflower)—a dwarf shrub in the Linnaeeae tribe of Caprifoliaceae family—is distributed across the Northern Hemisphere. By means of this study, a reliable protocol for efficient micropropagation of uniform L. borealis L. var. borealis plantlets has been provided for the first time; callus culture was also established. Different initial explants, types of cultures, media systems, and plant growth regulators in Murashige and Skoog (MS) media were tested. Agitated shoot cultures in the liquid media turned out to be the best system for the production of sustainable plant biomass. After stabilization of the callus lines, the highest growth index (c.a. 526%) was gained for callus maintained on MS enriched with picloram. TLC and UHPLC-HESI-HRMS analysis confirmed the presence of phenolic acids and flavonoids, and for the first time, the presence of iridoids and triterpenoid saponins in this species. Multiplication of L. borealis shoot culture provides renewable raw material, allowing for the assessment of the phytochemical profile, and, in the future, for the quantitative analyses and the studies of the biological activity of extracts, fractions, or isolated compounds. This is the first report on in vitro cultures of traditionally used L. borealis rare taxon and its biosynthetic potential.  相似文献

An efficient plant regeneration protocol was established for an endangered ethnomedicinal plant Desmodium gangeticum (Linn.) DC. Morphogenic calli were produced from 96 % of the cultures comprising the immature leaf explants on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxyacetic acid (4.0 mg?l?1) in combination with 6-benzylaminopurine (BA; 0.8 mg?l?1). For callus regeneration, various concentrations of BA (1.0–5.0 mg?l?1) or thidiazuron (TDZ; 1.0–5.0 mg?l?1) alone or in combination with indole-3-acetic acid (IAA; 0.2–1.0 mg?l?1) were used. Highest response of shoot regeneration was observed on MS medium fortified with TDZ (4.0 mg?l?1) and IAA (0.5 mg?l?1) combination. Here, 100 % cultures responded with an average number of 22.3 shoots per gram calli. Inclusion of indole-3-butyric acid in half MS medium favored rooting of recovered shoots. Out of 45 rooted plants transferred to soil, 40 survived. Total DNA was extracted from the leaves of the acclimatized plants of D. gangeticum. Analysis of random amplified polymorphic DNA using 13 arbitrary decanucleotide primers showed the genetic homogeneity in all the ten plants regenerated from callus with parental plant, suggesting that shoot regeneration from callus could be used for the true-to-type multiplication of this plant.  相似文献

Gynostemma pentaphyllum (Thunb.) Makino, a traditional Chinese herb possessing antitumor and antioxidant activities, has been shown to contain several functional components like saponins and flavonoids. However, their identities remain uncertain. The objectives of this study were to develop an appropriate extraction, purification and HPLC-MS method to determine saponins and flavonoids in G. pentaphyllum. Both flavonoids and saponins were extracted with methanol, followed by purification with a C18 cartridge to elute the former with 50% methanol and the latter with 100% methanol. A total of 34 saponins were separated within 40 min by a Gemini C18 column and a gradient mobile phase of acetonitrile and 0.1% formic acid in water, in which 18 saponins were identified by LC-MS with ESI mode and Q-TOF (LC/MS/MS). Similarly, a total of eight flavonoids were separated within 45 min by the same column and a gradient solvent system of methanol and 0.1% formic acid in water, with identification being carried out by a post-column derivatization method and LC-MS with ESI mode. The amounts of flavonoids in G. pentaphyllum ranged from 170.7 to 2416.5 μg g−1, whereas saponins were from 491.0 to 89,888.9 μg g−1.  相似文献

The biotechnological approach of in vitro cultures elicitation offers an alternative strategy for the production of camptothecin (CPT) in Nothapodytes nimmoniana to mitigate indiscriminate harvest of the endangered natural population for the alkaloid. Yeast extract (YE) and vanadyl sulfate (VS) elicitors were used to enhance the biosynthesis of CPT in hypocotyl-derived callus cultures of N. nimmoniana by cultivation using solid and liquid Murashige and Skoog (MS) medium amended with NAA + BAP (2.0 + 1.0 mg L?1). Effects of the two elicitors on biomass and CPT production at 6.25, 12.5, 25, 50 and 75 mg L?1 concentrations using callus cultures from three cell lines were evaluated after 15, 30 and 45 days culture. Yeast extract elicitor treatments showed a linear enhancement effect on biomass and CPT production up to 50 mg L?1 YE and beyond the concentrations, no significant effect was observed. Enhanced biomass and CPT production were achieved with VS elicitor up to 25 mg L?1 concentrations but, 50 and 75 mg L?1 VS had minimal effects on biomass and CPT production in callus sources and incubation duration-dependent manner. The intracellular yield of CPT in liquid media-cultivated cultures at concentrations of the two elicitors was lower when compared to solid media treatments relative control due to the extracellular accumulation but, higher overall production. Accumulation of the biomass showed association with produced CPT in the elicitor treatments and control cultures.  相似文献

High cost becomes the major obstacle for the industrial application of laccase. Many approaches have been applied to enhance the yield and decrease the cost of laccase. Since flavonoids are the natural inducers for laccase production, in this article, flavonoid-rich plants were taken as the sole substrate for the solid-state fermentation of Funalia trogii (Cui 3676). It indicated that flavonoid-rich plants can effectively promote the production of F. trogii laccase without the addition of inducers. The laccase activity was 42.5 IU g?1 substrate when kudzu vine root was used as the substrate, which was enhanced by 4.46 times than that when bran was used as the substrate. Meanwhile, the solid-state fermentation of laccase could enrich flavonoids, benefiting their extraction. The content of flavonoids extracted from fermented kudzu vine root and Ginkgo biloba leaves was enhanced by 56.41 and 24.11 %, respectively, compared to the unfermented substrate, and the relative reductive ability and scavenging ability of hydroxyl radicals of flavonoids in the fermented residues were essentially unchanged. Thus, flavonoid-rich plants will become a kind of potential substrate for laccase fermentation which is beneficial in enhancing the yield and reducing the cost of laccase.  相似文献
