A great number of structurally diverse natural products containing five-membered heterocyclic subunits, such as imidazole, oxazole, thiazole, and their saturated congeners, are abundant in nature. These naturally occurring metabolites often exhibit extensive and pharmacologically important biological activities. This review covers alkaloids classified in the plant families Amaryallidaceae and Sceletium. In addition alkaloids possessing imidazole, oxazole and thiazole structures are listed followed by alkaloids possessing peptide linkages. Finally miscellaneous alkaloids include those compounds found naturally which cannot be classified into known groups; a relative Pandora's box. Human immunodeficiency virus (HIV) especially HIV-1 infection and its progression to acquired immune deficiency syndrome (AIDS) remains a significant global health challenge. The advent of the highly active antiretroviral therapy (HAART) has greatly extended the life expectancy of patients living with HIV, but it has become evident that long-term HAART will not eliminate the HIV reservoir and cure the infection. Moreover, the drug resistance and undesirable side effects hamper efficacious therapy, creating an urgent need to develop novel, more effective and less toxic anti-HIV therapeutics. Imidazole, oxazole and thiazole with two heteroatoms at meta-position of five-membered rings are fascinating structures and constitute an important class of heterocycles in drug discovery. Their derivatives could exert the anti-HIV activity through diverse mechanisms and possess promising antiviral activity against both drug-sensitive and drug-resistant HIV strains. This review summarizes the research progress made regarding the anti-HIV potential of imidazole, oxazole and thiazole hybrids, and the structure–activity relationships (SARs) are also discussed to facilitate further rational design of more effective candidates, covering articles published from 2012 to 2022.  相似文献

The copper(II) coordination chemistry of westiellamide (H(3)L(wa)), as well as of three synthetic analogues with an [18]azacrown-6 macrocyclic structure but with three imidazole (H(3)L(1)), oxazole (H(3)L(2)), and thiazole (H(3)L(3)) rings instead of oxazoline, is reported. As in the larger patellamide rings, the N(heterocycle)-N(peptide)-N(heterocycle) binding site is highly preorganized for copper(II) coordination. In contrast to earlier reports, the macrocyclic peptides have been found to form stable mono- and dinuclear copper(II) complexes. The coordination of copper(II) has been monitored by high-resolution electrospray mass spectrometry (ESI-MS), spectrophotometric and polarimetric titrations, and EPR and IR spectroscopies, and the structural assignments have been supported by time-dependent studies (UV/Vis/NIR, ESI-MS, and EPR) of the complexation reaction of copper(II) with H(3)L(1). Density functional theory (DFT) calculations have been used to model the structures of the copper(II) complexes on the basis of their spectroscopic data. The copper(II) ion has a distorted square-pyramidal geometry with one or two coordinated solvent molecules (CH(3)OH) in the mononuclear copper(II) cyclic peptide complexes, but the coordination sphere in [Cu(H(2)L(wa))(OHCH(3))](+) differs from those in the synthetic analogues, [Cu(H(2)L)(OHCH(3))(2)](+) (L = L(1), L(2), L(3)). Dinuclear copper(II) complexes ([Cu(II) (2)(HL)(mu-X)](+); X = OCH(3), OH; L = L(1), L(2), L(3), L(wa)) are observed in the mass spectra. While a dipole-dipole coupled EPR spectrum is observed for the dinuclear copper(II) complex of H(3)L(3), the corresponding complexes with H(3)L (L = L(1), L(2), L(wa)) are EPR-silent. This may be explained in terms of strong antiferromagnetic coupling (H(3)L(1)) and/or a low concentration of the dicopper(II) complexes (H(3)L(wa), H(3)L(2)), in agreement with the mass spectrometric observations.  相似文献

A variety of computational methods, including the semiempirical techniques AM1, PM3, and MNDO, and the thermochemical basis sets of Benson and Stine, was used to calculate and compare heats of formation (ΔHf°) data for optimized geometries of a variety of aromatic and nonaromatic heterocycles. Detailed analyses, including 6-31G* and MP2/6-31G* ab initio calculations, were performed for the oxazole and thiazole heterocycles. The results indicate a scatter among the methods sensitive to the nature of the heterocycle. This was in particular evident in the oxazole molecule, where AM1 gave a singularly high value of ΔHf° consistent with longer calculated bond lengths, particularly about the oxygen atom. Aromatic stabilization energy appears to be addressed differently among the employed methods. Implications of this contrast applied to calculation of macromolecular systems containing heterocyclic units are discussed.  相似文献

Cyclometaleted rhodium complexes of oxazole and thiazole luminophores [Rh(C??N)2En]Cl [(C??N)?, deprotonated forms of 2,5-diphenyloxazole, 2-phenylbenzothiazole, 2-(biphenyl-4-yl)-6-phenylbenzoxazole, and 2-(biphenyl-4-yl)-5-phenyloxazole; En ?? ethylenediamine] were obtained and characterized by the methods of 1H NMR, IR, and electron absorption and emission spectroscopy. Two cyclometallated ligands in the inner sphere of the complexes are in the cis-C,C positions. Cyclometallating of the luminophores results both in a red shift of intraligand ??-??*-optical transitions (???? ??1.2 kK) as compared to free luminophores and in the appearance of a long-wave band (??max 376?C392 nm) of a mixed nature: metal-ligand charge transfer/intraligand transition. Alongside with the internal conversion to a low-energy state of the metal-ligand charge transfer/ intraligand transition, the emission degradation of photoexcitation energy results in the intraligand ??-??*-fluorescence of the complexes (??max 390?C423 nm) at room temperature.  相似文献

Peptidomimetic-based macrocycles typically have improved pharmacokinetic properties over those observed with peptide analogs. Described are the syntheses of 13 peptidomimetic derivatives that are based on active Sansalvamide A structures, where these analogs incorporate heterocycles (triazoles, oxazoles, thiazoles, or pseudoprolines) along the macrocyclic backbone. The syntheses of these derivatives employ several approaches that can be applied to convert a macrocyclic peptide into its peptidomimetic counterpart. These approaches include peptide modifications to generate the alkyne and azide for click chemistry, a serine conversion into an oxazole, a Hantzsch reaction to generate the thiazole, and protected threonine to generate the pseudoproline derivatives. Furthermore, we show that two different peptidomimetic moieties, triazoles and thiazoles, can be incorporated into the macrocyclic backbone without reducing cytotoxicity: triazole and thiazole.  相似文献

Reactions of benzotriazol-1-yl(1H-pyrrol-2-yl)methanone 10 and benzotriazol-1-yl(1H-indol-2-yl)methanone 11 with diverse ketones, isocyanates, and isothiocyanates in the presence of base afforded pyrrolo[1,2-c]oxazol-1-ones 1, oxazolo[3,4-a]indol-1-ones 2, pyrrolo[1,2-c]imidazoles 3, and imidazo[1,5-a]indoles 4 by a simple one-step procedure.  相似文献
