This work reports a new sensitive multi-residue liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for detection, confirmation and quantification of forty-six pesticides and transformation products belonging to different chemical classes in wines. The proposed method makes use of a solid-phase extraction (SPE) procedure with Oasis HLB cartridges that combines isolation of the pesticides and sample clean-up in a single step. Analysis is performed by liquid chromatography-electrospray ionisation-tandem mass spectrometry (LC-MS/MS) operated in the selected reaction monitoring (SRM) mode, acquiring two specific precursor-product ion transitions per target compound. An investigation of matrix effects has been performed during method validation showing medium to low effects for the majority of the compounds. Limits of detection (LODs) were in the range 0.0003–0.003 mg L−1 and limits of quantification (LOQs) were in the range 0.001–0.01 mg L−1. The average recoveries, measured at two concentration levels (0.010 and 0.050 mg L−1), were in the range 70–110% for most of the compounds tested with % relative standard deviations below 20%, while a value of 0.010 mg L−1 has been established as the method limit of quantification (MLOQ) for all target species. Expanded uncertainty values were in the range 10–40% while the Horrat ratios were below 1. The method has been successfully applied to the analysis of 60 wine samples in the course of an annual monitoring study with carbendazim-benomyl, thiophanate-methyl and carbaryl being the most frequently determined pesticides.  相似文献

A multi-residue method using liquid chromatography coupled to triple quadrupole tandem mass spectrometry (LC-MS/MS), associated with solid-phase extraction (SPE), was developed for the determination of 21 pesticides in water samples. The compounds investigated are used for the maintenance of golf courses and ordinarily measured by gas chromatography-mass spectrometry (GC-MS). Electrospray ionisation (ESI) was applied to all compounds, and LC and MS conditions were optimised to measure them under SRM mode. This method showed excellent linearity ranges for all pesticides, with correlation coefficients greater than 0.996. Two kinds of extraction cartridges, namely, styrene divinylbenzene polymer (Sep-Pak PS-2) and divinylbenzene-N-vinylpyrrolidone copolymer (Oasis HLB), were tested and the extraction conditions were optimised. All the pesticides were determined using acetonitrile and ethyl acetate as eluents in both cartridges, and good recoveries (>77%) and repeatability with low relative standard deviations (RSDs, <12%) were achieved from ultra-pure water. In addition, satisfactory recoveries (>76%) and low intra-day and inter-day RSDs (<15%) of all pesticides were also obtained with the Sep-Pak PS-2 cartridge when using river water. The method limits of detection (LODs) ranged between 0.068 (diazinon) and 3.9 (triclopyrbutoxyethyl)?ng?L?1. The sensitive multiresidual analytical method for simultaneous analysis of 14 most commonly used agricultural pesticides in Serbia was developed and optimized. The selected insecticides, fungicides and herbicides belong to seven chemical classes (organophosphates, neonicotinoids, carbamates, diacylhydrazines, benzimidazoles, triazines and phenylureas). The method was based on solid-phase extraction followed by liquid chromatography-tandem mass spectrometry. The following parameters that may affect the SPE procedure efficiency were optimized: the sorbent type in combination with different elution solvents, the sample pH and the sample volume. For each pesticide, MSn analysis was performed and distinctive ions and transitions were selected for identification and quantification, as well as for confirmation purposes. External matrix-matched calibration method was used to eliminate variable matrix effect and ensure precise quantification. Good recoveries (72-129%), and low limits of detection (0.4-5.5 ng L−1) and quantification (1.1-18.2 ng L−1) were achieved for all selected pesticides. The developed and optimized method was successfully applied in the analysis of several river waters, as well as ground waters in Serbia, influenced by agriculture. The most frequently detected pesticide was carbendazim. Dimethoate, carbofuran and propazine were also found in the investigated samples.  相似文献

Capillary liquid chromatography (cLC) hyphenated with tandem mass spectrometry (MS-MS) was used to separate and quantitate trace concentrations of five estrogens in aqueous samples. New C(18)-based sorption materials bound to the silica support by monomeric and polymeric mechanisms were compared and tested for solid-phase extraction (SPE) of selected analytes with respect to optimization of their preconcentration yield. Application of an endcapped, monomer-bound preconcentration Discovery DSC-18Lt column under the optimized conditions provides yields in the range from 95 to 100% with a high repeatability (n=3, RSD≤7.2%). Using the electrospray ionization in the positive mode (ESI+), the cLC-MS-MS system (the Zorbax SB C18 capillary column and a binary mobile phase of acetonitrile and water containing 0.1% formic acid in both the components) was optimized to attain a sufficient retention of the early eluting estriol, a satisfactory resolution of the analytes and the maximum sensitivity of the determination. Both the isocratic and gradient elution were used and the optimized gradient method permitted analyses of aqueous environmental samples in 14 min within a linearity range from 6.1 to 25.0 (LOQ of analytes) to 500 ng/L and with a very good linearity (r>0.9981) for all the estrogens studied. The detection limits are in the range from 3.0 to 6.8 ng/L (1 μL injection volume). Six environmental water samples were analyzed and the studied estrogens were found in the Vltava river sample collected in Prague (13.2 ng/L for 17β-estradiol) and in the inlet to the wastewater treatment plant in Prague, at an overall concentration of 371.4 ng/L.  相似文献

The detection of ethyl-β-d-6-glucuronide (EtG), a stable phase II metabolite of ethanol, is of interest in both clinical and forensic contexts with the aim of monitoring alcohol abuse. We present a liquid chromatography-electrospray ionisation-tandem mass spectrometry method for the detection and quantification of EtG in hair. Thirty milligrams of washed and cut hair were cleaned up using solid-phase extraction graphite cartridges. Separation was then performed using an Uptisphere-3SI column, and the detection was operated in the negative mode. After validation, the method was applied to hair samples taken from four fatalities (F) with documented excessive drinking habits, 12 heavy drinkers (HD) and seven social drinkers (SD). The method exhibits limits of detection and quantification of 4 and 10 pg/mg, respectively. Intra- and inter-assay standard deviation and relative bias were less than 20% over the calibrating range (10 to 3,000 pg/mg). EtG hair concentrations in SD were <10 pg/mg and >50 pg/mg for F and HD (range, 54 to 497 pg/mg). The present assay appears convenient to carry out owing to the very small quantity of hair samples required to determine an effective heavy alcohol consumption (EtG hair concentration >50 pg/mg).  相似文献

A multiclass method for the analysis of residues, in egg matrices, of 41 antimicrobial agents belonging to seven families (sulfonamides, diaminopyridine derivates, quinolones, tetracyclines, macrolides, penicillins and lincosamides) was developed and validated according to the requirements of European Commission Decision 2002/657. Compounds were extracted with a pressurized liquid extraction (PLE) technique using a 1:1 mixture of acetonitrile and a succinic acid buffer (pH 6.0) at 70 °C. As this resulted in clear extracts, no further clean-up was necessary. Analytes were determined by ultra-high-pressure liquid chromatography–tandem mass spectrometry (UHPL–MS/MS) in a chromatographic run of 13 min. Calibration was carried out with spiked blank samples subjected to the entire analytical procedure. Five compounds, two of them isotopically labelled, were used as internal standards. Most analytes were quantified with errors below 10%. Precision in terms of reproducibility standard deviation was between 10% and 20% in most cases. CCα values were in the range 0.5–3.8 μg kg−1 for the non-authorized compounds. The proposed method would enable an experienced analyst to process about 25 samples per day.  相似文献

Antimicrobials are used in large quantities in human and veterinary medicine. Their environmental occurrence is of particular concern due to the potential spread and maintenance of bacterial resistance. After intake by the organisms, the unchanged drug and its metabolized forms are excreted and enter wastewater treatment plants where they are mostly incompletely eliminated, and are therefore eventually released into the aquatic environment. The reliable detection of several antimicrobials in different environmental aqueous compartments is the result of great improvements achieved in analytical chemistry. This article provides an overview of the more outstanding analytical methods based on liquid chromatography tandem mass spectrometry, developed and applied to determine antimicrobial residues and metabolites present in surface, waste, and ground waters.   相似文献

This paper describes the development and validation of a method for the simultaneous determination of 53 multi-class emerging organic pollutants in water samples using solid-phase extraction (SPE) followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS), using electrospray ionisation (ESI) in both positive and negative modes. Target compounds include acidic herbicides, UV filters, insect repellents, organophosphorous flame retardants, a bactericide, pharmaceuticals and metabolites. A single SPE consisting on the loading of 200-500 mL of sample adjusted to pH 7 on Oasis HLB 200mg cartridges and elution with methanol, permitted obtaining good recoveries: higher than 60% for tap, surface and wastewater in most cases. The 7 isotopically labelled internal standards effectively compensated losses during sample preparation and matrix effects at LC-MS/MS determination. The precision of the method, calculated as relative standard deviation (RSD) was below 15% for all compounds and all tested matrices. Detection limits (LODs) based on the confirmation, less intense, MRM (multiple reaction monitoring) transition and considering blanks varied between 0.3 and 30 ngL(-1). Finally, the developed method was applied to the determination of target analytes in various samples, including tap, surface and waste water. Among the tested emerging pollutants, 31 were found in wastewater in concentrations reaching up to 10 microgL(-1) in the case of ibuprofen. Also, 13 species were detected in tap water with concentrations up to 0.13 microgL(-1) for tri(chloropropyl) phosphate (TCPP).  相似文献

建立了测定植物中痕量脱落酸 (ABA) 的液相色谱-电喷雾串联质谱联用分析方法. 植物提取液先采用固相萃取 (SPE) 富集脱落酸并消除基体干扰, 然后以C18柱为固定相, V(甲醇)∶V(2 g/L甲酸水溶液)=50∶50为流动相分离脱落酸. 电喷雾(ESI)串联质谱采用负离子模式. 选择反应监测 (SRM) 模式用于脱落酸定量, 选择的离子对是263→153, 219. 脱落酸的线性范围为0.005~10 μg/mL, 检测限 (S/N=3) 为0.003 μg/mL, 加标0.02 μg和0.05 μg的回收率分别为98.3% 和103.5%. 该方法用于冬青芽中脱落酸的分析, 结果表明所建立的SPE-LC-MS/MS方法对于分析植物中的痕量脱落酸是有效的.  相似文献

建立了超高效液相色谱-串联质谱测定皮革制品中三丁基锡、三苯基锡、双-三苯基锡氧化物、三环锡等4种三取代有机锡化合物(TOT)的方法。采用盐酸溶液提取皮革制品中的有机锡化合物,使其呈阳离子态,经阳离子固相萃取净化,有机锡化合物在超高效C18色谱柱中完成分离,在正离子模式下采用多反应监测模式进行串联质谱测定。三丁基锡、三苯基锡、双-三苯基锡氧化物(以三苯基锡计)的定量限为10 μg/kg,三环锡的定量限为20 μg/kg;分别添加4种待测物于空白皮革样品中,加标回收率为54.1%~101.4%,相对标准偏差为4.3%~9.8%。实验结果表明,该方法简单、灵敏、准确,各项技术指标均满足国内外法规的要求,可用于皮革制品中4种三取代有机锡化合物残留的确证检测。  相似文献

An automated on-line solid-phase extraction-liquid chromatography-tandem mass spectrometry (SPE-LC-MS/MS) system was developed for the determination of macrolide antibiotics including erythromycin (ETM), roxithromycin (RTM), tylosin (TLS) and tilmicosin (TMC) in environmental water samples. A Capcell Pak MF Ph-1 column packed with restricted access material (RAM) was used as SPE column for the concentration of the analytes and clean-up of the sample. One milliliter water sample was injected into the conditioned SPE column and the matrix was washed out with 3 mL high purity water. By rotation of the switching valve, macrolides (MLs) were eluted in the back-flush mode and transferred to the analytical column by the chromatographic mobile phase. The matrix effect was evaluated by the directly injection LC-MS and on-line SPE-LC-MS methods. The limits of detection (LODs) and limits of quantification (LOQs) obtained are in the range of 2-6 and 7-20 ng L−1, respectively, which means that the proposed method is suitable for trace analysis of MLs at low level concentration. The intra- and inter-day precisions are in the range of 2.9-7.2% and 3.3-8.9%, respectively. In the three fortified levels (20, 200 and 2000 ng L−1), recoveries of MLs ranging from 86.5% to 98.3% are obtained.  相似文献

A new analytical method is proposed to determine more than 40 multiclass pesticides in different kinds of processed (whole, skimmed and powdered) and unprocessed (goat and human) milk samples using solid-phase microextraction (SPME). A comparative study between headspace (HS) and direct immersion (DI) was carried out. The effect of milk dilution and the use of acid to reduce the influence of the matrix in DI-SPME mode were also evaluated. DI of the SPME fiber into previously diluted and acidified milk samples achieved the best sensitivity results. Pesticides were determined using low-pressure gas chromatography-tandem mass spectrometry (LP-GC-MS/MS). Both of the selected techniques have been shown to be effective at reduce fat interference and can determine analytes present at very low concentrations (limits of quantification between 0.02 and 1.00 g L–1). Performance characteristics such as linearity, recovery, precision, and lower limits, together with an estimation of the measurement uncertainty using validation data, are presented for each pesticide. All of the pesticides presented recovery rates of between 81 and 110% and precision values lower than 12% (expressed as the relative standard deviation). The overall uncertainty of the method was estimated at three different concentrations (10, 25 and 50 g L–1) and was lower than 25.5% in all cases. The proposed analytical methodology was applied to the analysis of target pesticides in 35 samples: 15 commercial, 3 human and 17 goat milk samples. The metabolite p,p-DDE was the compound most frequently found in both the breast and goat milk samples, at concentration levels <20 g L–1. However, pesticide residues were not found in any of the other 15 commercial milk samples (skimmed, powdered and whole milk) analyzed.  相似文献

Four different commercial sorbents for solid-phase extraction have been evaluated for the extraction of a group of acidic pharmaceuticals in terms of selectivity and capacity: Oasis hydrophilic-lipophilic balance (HLB), Oasis MAX (strong anion exchange), Oasis WAX (weak anion exchange) and a commercial available molecularly imprinted polymer specific for non-steroidal anti-inflammatory drugs. Among the sorbents studied, molecularly imprinted polymer proved to be very effective in the reduction of matrix interferences and the selective extraction of acidic pharmaceuticals, such as salicylic acid, ibuprofen, fenoprofen, diclofenac and naproxen, among others, from effluent wastewater samples. Moreover, molecularly imprinted solid-phase extraction protocol was applied to liquid chromatography coupled to tandem mass spectrometry (MS/MS) with the purpose of evaluating the clean-up effect on ion suppression/enhancement when the complexity of the samples increases and a reduction of this effect was observed. Molecularly imprinted solid-phase extraction followed by liquid chromatography coupled to ultraviolet detection and liquid chromatography coupled to tandem mass spectrometry validation methodologies with effluent wastewaters were developed, obtaining recoveries between 70 and 85% and limits of detection at low levels of μg/L (0.15-1 μg/L) and ng/L (0.5-2 ng/L), respectively. In this study, a simple, rapid and efficient method has been developed for the extraction and preconcentration of different classes of pesticides, carbofuran (insecticide), clomazone (herbicide) and tebuconazole (fungicide) in aqueous samples by dispersive liquid-liquid microextraction (DLLME) coupled with liquid chromatography-tandem mass spectrometric detection. Some experimental parameters that influence the extraction efficiency, such as the type and volume of the disperser solvents and extraction solvents, extraction time, speed of centrifugation, pH and addition of salt were examined and optimized. Under the optimum conditions, the recoveries of pesticides in water at spiking levels between 0.02 and 2.0 μg L−1 ranged from 62.7% to 120.0%. The relative standard deviations varied between 1.9% and 9.1% (n = 3). The limits of quantification of the method considering a 50-fold preconcentration step were 0.02 μg L−1. The linearity of the method ranged from 1.0 to 1000 μg L−1 for all compounds, with correlation coefficients varying from 0.9982 to 0.9992. Results show that the method we propose can meet the requirements for the determination of pesticides in water samples. The comparison of this method with solid-phase extraction indicates that DLLME is a simple, fast, and low-cost method for the determination of pesticides in natural waters.  相似文献

We developed a sensitive and useful method for the determination of five fluoroquinolones (FQs), enoxacin, ofloxacin, ciprofloxacin, norfloxacin, and lomefloxacin in environmental waters, using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These compounds were analysed within 7 min by high-performance liquid chromatography (HPLC) using a CAPCELL PAK C8 column and aqueous ammonium formate (pH 3.0, 5 mM)/acetonitrile (85/15, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the positive ion mode were optimized for MS/MS detection. In order to optimize the extraction of FQs, several in-tube SPME parameters were examined. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL of sample at a flow-rate of 150 μL/min, using a Carboxen 1010 PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r ≥ 0.997) was obtained in the concentration range from 0.1 to 10 ng/mL for all compounds examined. The limits of detection (S/N = 3) of the five FQs ranged from 7 to 29 pg/mL. The in-tube SPME method showed 60-94-fold higher sensitivity than the direct injection method (5 μL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface waters and wastewaters were collected from the area around Asahi River, and ofloxacin was detected in wastewater samples of a sewage treatment plant and other two hospitals at 17.5-186.2 pg/mL. The recoveries of FQs spiked into river water were above 81% for a 0.1 or 0.2 ng/mL spiking concentration, and the relative standard deviations were below 1.9-8.6%.  相似文献

A simple, rapid and sensitive method for the determination of five estrogens, estrone, 17beta-estradiol, estriol, ethynyl estradiol, and diethylstilbestrol, was developed using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These estrogens were separated within 8 min by HPLC using an XDB-C8 column and 0.01% ammonia/acetonitrile (60/40, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the negative ion mode were optimized for MS/MS detection of the estrogens. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 microL of sample using a Supel-Q PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase, and no carryover was observed. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r > or = 0.9996) was obtained in the concentration range from 10 to 200 pg/mL for all compounds examined. The limits of detection (S/N= 3) of the five estrogens examined ranged from 2.7 to 11.7 pg/mL. The in-tube SPME method showed 34-90-fold higher sensitivity than the direct injection method (5 microL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface water and wastewater samples were collected from the area around Asahi River, and estriol was detected at 35.7 pg/mL in the effluent of a sewage treatment plant. The recoveries of estrogens spiked into river waters were above 86%, except for estriol, and the relative standard deviations were below 0.9-8.8%.  相似文献

A novel procedure for the determination of nine selected fungicides (metalaxyl-M, azoxystrobin, myclobutanil, flusilazole, penconazole, tebuconazole, propiconazole, diniconazole and difenoconazole) in wine samples is presented. Sample enrichment and purification is simultaneously performed using mixed-mode, anion exchange and reversed-phase, OASIS MAX solid-phase extraction (SPE) cartridges. Analytes were determined by liquid chromatography coupled to tandem mass spectrometry using atmospheric pressure electrospray ionization (LC-ESI-MS/MS). Parameters affecting the chromatographic determination and the extraction-purification processes were thoroughly investigated. Under optimized conditions, 10 mL of wine were firstly diluted 1:1 with ultrapure water and then passed through the mixed-mode SPE cartridge at a flow of ca. 5 mLmin(-1). After a washing step with 5 mL of an aqueous NH(4)OH solution (5%, w:v), analytes were recovered with just 1 mL of methanol and injected in the LC-MS/MS system without any additional purification. The selective extraction process avoided significant changes in the ionization efficiency for red and white wine extracts in comparison with pure standards in methanol. Performance of the method was good in terms of precision (RSDs<11%) and accuracy (absolute recoveries>72%, determined against pure standards in methanol) reporting method LOQs in the range of 0.01-0.79 ngmL(-1) for target compounds, which are far below the EU maxima residue levels (MRLs) for fungicides in vinification grapes and wine. Several commercial wines from different geographic areas in Spain were analyzed. In most samples, metalaxyl-M and azoxystrobin were found at concentrations up to several ngmL(-1).  相似文献
