During the last decade, there has been a rapidly growing trend toward the use of cellphone-based devices (CBDs) in bioanalytical sciences. For example, they have been used for digital microscopy, cytometry, read-out of immunoassays and lateral flow tests, electrochemical and surface plasmon resonance based bio-sensing, colorimetric detection and healthcare monitoring, among others. Cellphone can be considered as one of the most prospective devices for the development of next-generation point-of-care (POC) diagnostics platforms, enabling mobile healthcare delivery and personalized medicine. With more than 6.5 billion cellphone subscribers worldwide and approximately 1.6 billion new devices being sold each year, cellphone technology is also creating new business and research opportunities. Many cellphone-based devices, such as those targeted for diabetic management, weight management, monitoring of blood pressure and pulse rate, have already become commercially-available in recent years. In addition to such monitoring platforms, several other CBDs are also being introduced, targeting e.g., microscopic imaging and sensing applications for medical diagnostics using novel computational algorithms and components already embedded on cellphones. This report aims to review these recent developments in CBDs for bioanalytical sciences along with some of the challenges involved and the future opportunities.

A new method for disintegration studies of rapid disintegrating tablet   总被引:4,自引:0,他引:4  
The aim of this study was to develop a simple and suitable disintegration method specific for rapid disintegrating tablets (RDTs). The new disintegration method that we propose employs a rotary shaft to exert mechanical pressure on the RDT. To assess our method, we manufactured several placebo RDTs and exposed them to severe storage conditions (60 degrees C/75%RH for 1 week) in order to obtain RDTs with a wide range of disintegration times. These placebo RDTs were utilized to compare the disintegration times obtained by several methods, including the proposed method. As expected, the disintegration time of the placebo RDTs in human sensory test varied widely. The disintegration times determined by the conventional disintegration test were in good correlation to those in human sensory test, but the slope was far from 1 (0.241). There was no correlation between the disintegration time of RDTs in human sensory test and those determined by the conventional dissolution test. In contrast, we acquired good correlation between the disintegration times obtained with the new method and those in human sensory test, and the slope was very close to 1 (0.858). We attribute this to the use of mechanical stress in the new method, similar to that the RDT is subject to in the oral cavity. We therefore concluded that the proposed method was suitable for the measurement of the disintegration time of RDTs. This new method might provide a valuable approach for the establishment of the official disintegration test for RDTs in the future.  相似文献

Many kinds of rapidly disintegrating or oral disintegrating tablets (RDT) have been developed to improve the ease of tablet administration, especially for elderly and pediatric patients. In these cases, knowledge regarding disintegration behavior appears important with respect to the development of such a novel tablet. Ordinary disintegration testing, such as the Japanese Pharmacopoeia (JP) method, faces limitations with respect to the evaluation of rapid disintegration due to strong agitation. Therefore, we have developed a novel apparatus and method to determine the dissolution of the RDT. The novel device consists of a disintegrating bath and CCD camera interfaced with a personal computer equipped with motion capture and image analysis software. A newly developed RDT containing various types of binder was evaluated with this protocol. In this method, disintegration occurs in a mildly agitated medium, which allows differentiation of minor distinctions among RDTs of different formulations. Simultaneously, we were also able to detect qualitative information, i.e., morphological changes in the tablet during disintegration. This method is useful for the evaluation of the disintegration of RDT during pharmaceutical development, and also for quality control during production.  相似文献

Current contaminant and residue monitoring throughout the food chain is based on sampling, transport, administration, and analysis in specialized control laboratories. This is a highly inefficient and costly process since typically more than 99 % of the samples are found to be compliant. On-site simplified prescreening may provide a scenario in which only samples that are suspect are transported and further processed. Such a prescreening can be performed using a small attachment on a cellphone. To this end, a cellphone-based imaging platform for a microsphere fluorescence immunoassay that detects the presence of anti-recombinant bovine somatotropin (rbST) antibodies in milk extracts was developed. RbST administration to cows increases their milk production, but is illegal in the EU and a public health concern in the USA. The cellphone monitors the presence of anti-rbST antibodies (rbST biomarker), which are endogenously produced upon administration of rbST and excreted in milk. The rbST biomarker present in milk extracts was captured by rbST covalently coupled to paramagnetic microspheres and labeled by quantum dot (QD)-coupled detection antibodies. The emitted fluorescence light from these captured QDs was then imaged using the cellphone camera. Additionally, a dark-field image was taken in which all microspheres present were visible. The fluorescence and dark-field microimages were analyzed using a custom-developed Android application running on the same cellphone. With this setup, the microsphere fluorescence immunoassay and cellphone-based detection were successfully applied to milk sample extracts from rbST-treated and untreated cows. An 80 % true-positive rate and 95 % true-negative rate were achieved using this setup. Next, the cellphone-based detection platform was benchmarked against a newly developed planar imaging array alternative and found to be equally performing versus the much more sophisticated alternative. Using cellphone-based on-site analysis in future residue monitoring can limit the number of samples for laboratory analysis already at an early stage. We have proposed a novel mobile healthcare platform, combining a pocket-sized colorimetric reader (13.5 × 6.5 × 2.5 cm(3)) and commercially available 10-parameter urinalysis paper strips (glucose, protein, glucose, bilirubin, urobilinogen, ketones, nitrite, pH, specific gravity, erythrocytes, and leukocytes), capable of sending data with a smart phone. The reader includes a novel colorimetric multi-detection module, which consists of three-chromatic light-emitting diodes, silicon photodiodes and a novel poly(methylmethacrylate) (PMMA) optical splitter. We employed data reading methods using conversions of the signal data (red, blue, and green) to the hue (H) color map or the Y model data, and used a curve-fitting method for the quantification. The reader is battery-powered, inexpensive, light-weight, and very speedy in analysis. And, it was applied to detection of a thousand of human urine samples and demonstrated reliable quantification of urinary glucose and protein. The features can be used by unskilled people on-site to transfer the analyzed data to experts off-site.  相似文献

Repeated dose toxicity (RDT) is one of the most important hazard endpoints in the risk assessment of chemicals. However, due to the complexity of the endpoints associated with whole body assessment, it is difficult to build up a mechanistically transparent structure–activity model. The category approach, based on mechanism information, is considered to be an effective approach for data gap filling for RDT by read-across. Therefore, a library of toxicological categories was developed using experimental RDT data for 500 chemicals and mechanistic knowledge of the effects of these chemicals on different organs. As a result, 33 categories were defined for 14 types of toxicity, such as hepatotoxicity, hemolytic anemia, etc. This category library was then incorporated in the Hazard Evaluation Support System (HESS) integrated computational platform to provide mechanistically reasonable predictions of RDT values for untested chemicals. This article describes the establishment of a category library and the associated HESS functions used to facilitate the mechanistically reasonable grouping of chemicals and their subsequent read-across.  相似文献

We are describing immunochromatographic test strips with smart phone-based fluorescence readout. They are intended for use in the detection of the foodborne bacterial pathogens Salmonella spp. and Escherichia coli O157. Silica nanoparticles (SiNPs) were doped with FITC and Ru(bpy), conjugated to the respective antibodies, and then used in a conventional lateral flow immunoassay (LFIA). Fluorescence was recorded by inserting the nitrocellulose strip into a smart phone-based fluorimeter consisting of a light weight (40 g) optical module containing an LED light source, a fluorescence filter set and a lens attached to the integrated camera of the cell phone in order to acquire high-resolution fluorescence images. The images were analysed by exploiting the quick image processing application of the cell phone and enable the detection of pathogens within few minutes. This LFIA is capable of detecting pathogens in concentrations as low as 105 cfu mL?1 directly from test samples without pre-enrichment. The detection is one order of magnitude better compared to gold nanoparticle-based LFIAs under similar condition. The successful combination of fluorescent nanoparticle-based pathogen detection by LFIAs with a smart phone-based detection platform has resulted in a portable device with improved diagnosis features and having potential application in diagnostics and environmental monitoring. Smart nanodevices that integrate molecular recognition and signal production hold great promise for the point‐of‐care (POC) diagnostic applications. Herein, the development of a DNA‐mediated proximity assembly of biochemical reactions, which was capable of sensing various bio‐targets and reporting easy‐to‐read signals is reported. The circuit was composed of a DNA hairpin‐locked catalytic cofactor with inhibited activity. Specific molecular inputs can trigger a conformational switch of the DNA locks through the mechanisms of toehold displacement and aptamer switching, exposing an active cofactor. The subsequent assembly of an enzyme/cofactor pair actuated a reaction to produce colorimetric or fluorescence signals for detecting target molecules. The developed system could be potentially applied to smart biosensing in molecular diagnostics and POC tests.  相似文献

Immunological methods are widely applied in medical diagnostics for the detection and quantification of a plethora of analytes. Associated analytical challenges usually require these assays to be performed in a central laboratory. During the last several years, however, the clinical demand for rapid immunodiagnostics to be performed in the immediate proximity of the patient has been constantly increasing. Biosensors constitute one of the key technologies enabling the necessary, yet challenging transition of immunodiagnostic tests from the central laboratory to the point of care. This review is intended to provide insights into the current state of this transition process with a focus on the role of biosensor-based systems. To begin with, an overview on standard immunodiagnostic tests presently employed in the central laboratory and at the point of care is given. The review then moves on to demonstrate how biosensor technologies are reshaping this landscape. Single analyte as well as multiplexed immunosensors applicable to point of care scenarios are presented. A section on the areas of clinical application then creates the bridge to day-to-day diagnostic practice. Finally, the depicted developments are critically weighed and future perspectives discussed in order to give the reader a firm idea on the forthcoming trends to be expected in this diagnostic field.

Magnetorheological Elastomers (MREs) are “smart” materials whose physical properties are altered by the application of magnetic fields. In previous studies the properties of MREs have been evaluated under a variety of conditions, however little attention has been paid to the recording and reporting of the magnetic fields used in these tests [1]. Currently there is no standard accepted method for specifying the magnetic field applied during MRE testing. This study presents a detailed map of a magnetic field applied during MRE tests as well as providing the first comparative results for uniaxial and biaxial testing under high strain fatigue test conditions. Both uniaxial tension tests and equi-biaxial bubble inflation tests were performed on isotropic natural rubber MREs using the same magnetic fields having magnetic flux densities up to 206 mT. The samples were cycled between pre-set strain limits. The magnetic field was switched on for a number of consecutive cycles and off for the same number of following cycles. The resultant change in stress due to the application and removal of the magnetic field was recorded and results are presented.  相似文献

We report cellphone‐based detection of dopamine with attomolar sensitivity in clinical samples with the use of a surface plasmon‐coupled emission (SPCE) platform. To this end, silver‐coated carbon nanotubes were used as spacer and cavity materials on SPCE substrates to obtain up to 100‐fold fluorescence enhancements. The presence of silver on the carbon nanotubes helped to overcome fluorescence quenching arising due to π–π interactions between the carbon nanotube and rhodamine 6G. The competing adsorption of dopamine versus rhodamine 6G on graphene oxide was utilized to develop this sensing platform.  相似文献

This is a novel idea: replacing highly dangerous and complex testing of solid thermally reactive materials through smart technology. We investigated the prediction of the storage lifetime and the thermal impact of thermally reactive material by different thermal analysis models: differential scanning calorimetry (DSC) for non-isothermal tests versus DSC isothermal tests. Two kinds of kinetic models were compared for evaluating appropriate kinetic parameters of thermal decomposition, and then the thermal hazard parameters were estimated by cartridge package simulation, which could result in reliable thermal hazard properties of a thermal reactive material’s thermal decomposition. We also determined the unsafe characteristics of a thermally reactive material stored in a depot under lifetime, so as to prevent runaway reactions that induce incidents by heat attack during storage. We were interested in an effective and smart analysis technology to reduce energy consumption of the dangerous testing. There are also calls for a smart testing technology which is the achieved object here for reducing energy consumption and avoiding runaway reaction disaster of thermally reactive materials.  相似文献

The demand for quick, accurate, and affordable point-of-care (POC) devices increases with the advancement in the dimensions of nanotechnology and digital interfaces (Internet of Things). The future of diagnostic requires the platform which can provide us the following benefits i. e., on-site detection, qualitative as well as quantitative analysis, easy to use, portable, low sample requirement, cost-effective, and have multiplexing proficiency. Multiplex biosensing platforms (MBPs) have the above following advantages so are going to be mostly used in various healthcare applications in near future. MBPs have the potential to fulfill the ‘ASSURED’ criteria specified by the World Health Organization (WHO) for remote-limited settings. This review paper focuses on miniaturized platforms that have multiplexing benefits for the bioanalysis of different clinical samples related to various healthcare applications. In addition to this, screening of pesticides, antibiotics, and hazardous metal ions with these surface-engineered devices has also been accounted in food and environmental samples. Some of the advanced techniques including microfluidics (Lab-on-a-chip), wearable smart devices, and CRISPR/Cas system for multiplexing applications are briefly described here. Furthermore, various needs, challenges, and prospects in commercializing these multiplexed surface-engineered devices have been discussed in this review.  相似文献

Genetic mutations can cause life-threatening diseases such as cancers and sickle cell anemia. Gene detection is thus of importance for disease-risk prediction or early diagnosis and treatment. Apart from genetic defects, gene detection techniques can also be applied to gene-related diseases with high risk to human health such as human papillomavirus (HPV) infection. HPV infection has been strongly linked to cervical cancer. To achieve a high-throughput HPV gene detection platform, the flow-through hybridization system appears to be one of the commercialized diagnostic techniques for this purpose. The flow-through hybridization technique is based on a vacuum-guided flow of DNA fragments which is continuously directed toward the oligoprobes that are immobilized on the testing membrane. However, the conventional colorimetric method and signal read-out approach suffers a problem of low sensitivity. On the contrary, fluorescence approaches allow more sensitive detection and broad sensing ranges. In this work, a fluorescent dye HCAP, which possesses aggregation-induced emission (AIE) properties and is responsive to alkaline phosphatase, was developed and applied to the flow-through hybridization platform to achieve HPV genome diagnosis of clinical samples. Also, an automatic membrane reader was constructed based on the AIE-based diagnosis platform which can identify the diagnostic result of patient DNA with a total concordance rate of 100% in the clinical trial.  相似文献

The possibility of using a microplate reader (MicroReader 4, Hyperion, USA) and 96-well plates for the quantitative photometric analysis of paper tests is demonstrated. In order to decrease the spread of absorbances, it is recommended to use special 96-well plates. The use of microfluidic devices for studying cell biology holds considerable promise when compared to canonical culture systems. However, the lack of compatibility with existing infrastructure hinders the application of microfluidic devices in the life sciences. Here, we present a microchannel cell culture platform having both operational (compatible with plate readers and pipetting) and performance (lower detection limits, controlled microenvironment) advantages. We demonstrated rapid growth assays and immunocytochemistry on mammary epithelial cells (both a cell line and primary cells) in the microchannel arrays. The utilization of ubiquitous pipetting methods and plate reader endpoints lowers the barriers to use. The simplicity and flexibility of the platform, combined with the throughput of automated detection systems, will facilitate the adoption of microfluidic culture systems in biological laboratories.  相似文献

Recently, smart surfaces with switchable wettability have aroused much attention. However, only single surface chemistry or the microstructure can be changed on these surfaces, which significantly limits their wetting performances, controllability, and applications. A new surface with both tunable surface microstructure and chemistry was prepared by grafting poly(N‐isopropylacrylamide) onto the pillar‐structured shape memory polymer on which multiple wetting states from superhydrophilicity to superhydrophobicity can be reversibly and precisely controlled by synergistically regulating the surface microstructure and chemistry. Meanwhile, based on the excellent controllability, we also showed the application of the surface as a rewritable platform, and various gradient wettings can be obtained. This work presents for the first time a surface with controllability in both surface chemistry and microstructure, which starts some new ideas for the design of novel superwetting materials.  相似文献

We report on the development and application of a "smart" activatable peptide-based probe for detection of Ras-related farnesyl protein transferase (FPT). Upon farnesylation by FPT, the probe was brought close to a hydrophobic milieu and as a consequence emitted fluorescent light that could be detected by several media, such as fluorescence microscopy, a plate reader, and an optical imaging system. A FPT activity assay confirmed the specificity of the probe (IC50 = 1.2 muM) for FPT compared to that of the native peptide (IC50 = 0.17 muM). In addition, the probe has remarkable binding constant, Kd = 26 nM. The specificity of enzyme activation was proved in pure enzyme assays as well as in cell-based assays. Furthermore, the fluorescent enhancement of the probe was 30-fold stronger than the control peptide in a live cell assay.  相似文献

Immunodiagnostic tests performed at the point of care (POC) today usually employ antibodies for biorecognition and are read out either visually or with specialized equipment. Availability of alternative biorecognition elements with promising features as well as smartphone-based approaches for signal readout, however, challenge the described established configuration in terms of analytical performance and practicability. Assessing these developments’ clinical relevance and their impact on POC immunodiagnostics is demanding. The first part of this review will therefore give an overview on suitable diagnostic biosensors based on alternative recognition elements (such as nucleic acid-based aptamers or engineered binding proteins) and exemplify advantages and drawbacks of these biomolecules on the base of selected assays. The second part of the review then focuses on smartphone-connected diagnostics and discusses the indispensable considerations required for successful future clinical POCT implementation. Together, the joint depiction of two of the most innovative and exciting developments in the field will enable the reader to cast a glance into the distant future of POC immunodiagnostics.
