Dehydration of 4-hydroxy-4-methyl-3-phenylamino-oxazolidin-2-ones

The dehydration of two 5,5-disubstituted 4-hydroxy-4-methyl-3-phenylaminooxazolidin-2-ones into the corresponding 4-methylene-3-phenylaminooxazolidin-2-ones has been carried out. A convenient synthesis of 3-amino-4-imino(thioxo)-imidazolidin-2-ones

Subsequent treatment of diethylphosphonopropyl α-aminonitriles with 1,1′-carbonyldiimidazole or 1,1′-carbonyl-di-(1,2,4-triazole) and hydrazines afforded substituted 3-amino-4-imino-imidazolidin-2-ones. Their acidic hydrolysis and their reactions with hydrogen sulfide are described.     

Bromination of 5-Substituted 3H-Furan-2-ones and 3H-Pyrrol-2-ones

The bromination of 5-alkyl(aryl)-3H-furan-2-ones and 5-alkyl(aryl)-3H-pyrrol-2-ones and also their derivatives takes place at the ethylene bond with the formation of 4-monobromo derivatives. N-Phenyl-3H-pyrrol-2-ones are brominated simultaneously at the ethylene bond of the heterocycle and at the phenyl substituent at the nitrogen atom.     

Camps reaction for the synthesis of 3-RS-4-arylquinolin-2-ones

The replacement of the chlorine atom in 2-(chloroacetamido)benzophenones on treatment with RSH (R = Alk, Ar, Hetaryl) in the presence of MeONa is accompanied by intramolecular cyclization following the Camps reaction pattern to give 3-RS-4-arylquinolin-2-ones. Cleavage of 4-aryl-3-(benzoxazol-2-ylthio)quinolin-2-ones by morpholine has resulted in the corresponding 4-aryl-3-mercaptoquinolin-2-ones.

Synthesis of Arylidene Derivatives of 1-Aryl-3H-pyrrol-2-ones

The aminolysis of 5-aryl-3-arylidene-3H-furan-2-ones by the action of aromatic amines leads to the formation of substituted amides of 4-oxo acids, the subsequent azacyclization of which in the presence of acetic anhydride leads to the formation of 1,5-diaryl-substituted 3-arylidene-3H-pyrrol-2-ones. The mechanism of the occurring and alternative transformations is discussed.     

Reaction of 2-(2-oxoethylidene)furan-3(2H)-ones with isopropylidenehydrazides of aromatic carboxylic acids: Synthesis of 3-hydroxy-3-(2-oxoethyl)-2,3-dihydropyridazin-4(1H)-ones

3-Hydroxy-3-(2-oxoethyl)-6-phenyl-2,3-dihydropyridazin-4(1H)-ones were obtained by the reaction of methyl 3-oxo-5-phenylfuran-2(3H)-ylideneacetate or 2-[2-(4-chlorophenyl)-2-oxoethylidene]-5-phenylfuran-3(2H)-one with benzoic or p-nitrobenzoic isopropylidenehydrazides. Thermal rearrangement of condensed alkoxy-substituted 2-trifluoromethyl-4H-pyran-4-ones to spiroannelated 3(2H)-furanones

Isomerization of the acetates and benzoates of condensed hydroxy-substituted 2-trifluoromethyl-4H-pyran-4-ones at 300–320°C gave the corresponding derivatives of spiroannelated 3(2H)-furanones. Atom-Economic Route to Densely Functionalized Thiophenes via Base-Catalyzed Rearrangement of 5-Propargyl-2H-thiopyran-4(3H)-ones

An atom-economic route to densely functionalized thiophenes has been developed via base-catalyzed rearrangement of 5-propargyl-2H-thiopyran-4(3H)-ones involving tandem retro-thio Michael addition, propargyl isomerization, and intramolecular thio-addition to allenes.     

Synthesis of 2-Benzylidene and 2-Hetarylmethyl Derivatives of 2H-1,4-Benzoxazin-3-(4H)-ones as Neuroprotecting Agents

A wide variety of the title compounds were synthesized by conventional and microwave methods in which the main step is a condensation of an aldehyde with a 1,4-benzoxazin-3-(4H)-one. In all cases, the Z diasteromers were the major products. Of particular importance is the synthesis of novel 2-(3,5-dibromo-4-hydroxy) and 2-(4-acetoxy-3,5-dibromobenzylidene derivatives of 2H-1,4-benzoxazin-3-(4H)-ones, four of which were shown in a previous publication to exhibit potent neuroprotecting properties. The yields of titled compounds ranged from 25 to 83%.     

Synthesis and structure of 1,3-dialkyl-4-(sulfonylimino)imidazolidin-2-ones

The reactions of 1,3-dialkyl-4,5-dihydroxyimidazolidin-2-ones with alkyl sulfamides have been studied and previously undescribed 1,3-dialkyl-4-(alkylaminosulfonylimino)imidazolidin-2-ones have been obtained. 高温水中2,3-二氢喹唑啉-4(1H)-酮衍生物的合成反应研究

为发展绿色化学,减少化学反应中有毒害溶剂的使用,本文以高温水为溶剂,在未使用任何有机溶剂及催化剂的条件下,以2-氨基苯甲酰胺与醛为底物,成功合成了2,3-二氢喹唑啉-4(1H)-酮衍生物。研究了反应温度、反应时间及芳环上取代基对反应的影响。在摩尔比1∶1、温度为130℃、反应时间1h的优化条件下,收率为32. 4%～85. 7%。     

Synthesis and Anti-HIV-1 Activity of 2-[2-(3,5-Dimethylphenoxy)ethylthio]pyrimidin-4(3H)-ones

New 2-[2-(3,5-dimethylphenoxy)ethyl]thio derivatives of pyrimidin-4(3H)-one containing various substituents at positions 5 and 6 of the pyrimidine ring were synthesized. It was shown that alkylation of 2-thiouracils with 1-bromo-2-(3,5-dimethylphenoxy)ethane in DMF takes place exclusively at the sulfur atom. The obtained 6-benzyl and 6-(2,6-difluorobenzyl) derivatives have clearly defined virus-inhibiting properties with respect to type 1 human immunodeficiency virus in vitro and suppress its reproduction by 50% at concentrations of 1.3 and 11.2 mM respectively.     

One-Pot Synthesis of 4-Aminoquinazolines by Hexamethyldisilazane-Mediated Reaction of Quinazolin-4(3H)-ones with Amines

Hexamethyldisilazane-mediated reaction of quinazolin-4(3H)-ones with primary amines led to facile formation of 4-aminoquinazolines through tandem silylation and substitution in a single pot. Excellent yields of the products (83–97%) and environmental friendliness (avoiding the use of chlorination reagents) are the advantages of this method.      

Unexpected Synthesis of Novel 3-Allyl-5-(arylidene)-2-thioxo-thiazolidin-4-ones in Reactions of 3-Allylrhodanine with 2-Arylidene-4-methyl-5-oxopyrazolidinium Ylides

Abstract

The reaction of 3-allylrhodanine with 2-arylidene-4-methyl-5-oxopyrazolidinium ylides proceeded unexpectedly to form novel 3-allyl-5-(arylidene)-2-thioxo-thiazolidin-4-ones 6a–k in good yields. All structures have been formulated on the basis of their spectral (IR, NMR, MS) data and elemental analyses. In addition, the structure of compound 6a was confirmed by means of x-ray crystallographic analysis.

在高氯酸镁催化作用下,由醛、1,3-二羰基化合物、脲(硫脲)合成了一系列1,2,3,4-四氢嘧啶-2-酮,此合成方法反应温和、产率高,并且高氯酸镁具有能重复使用的优点.     

Synthesis of Some Heterocyclic Compounds from Enamines of 3-Acetylfuran-2(5 H)-ones

The interaction of different 3-acetylfuran-2(5H)-ones (1, 4) with dimethylformamide dimethyl acetal (DMF/DMA) has been investigated, and the condensed furopyridine system 3 was obtained. By means of interaction of compounds 4 and DMF/DMA via the 3-acetylfuran-2(5H)-one enamines, the heterocyclic substituted furanones are obtained. The condensation of 3-acetyl-4,5,5-trimethylfuran-2(5H)-one (1a) with veratraldehyde was realized. Through the interaction of 1a with malononitrile and further intramolecular condensation of the product, 6-amino-dihydroisobenzofuran-5-carbonitrile was also synthesized.     

2-烷硫基-5-苯基亚甲基-4H-咪唑啉-4-酮的合成与表征

用2-硫代-3-正丁基-5-苯基亚甲基4-咪唑啉二酮的-S-烷基化反应合成了2-烷硫基-3-正丁基-5-苯基亚甲基-4H-咪唑啉-4-酮衍生物。探讨了S-烷基化反应的反应条件和所合成化合物的波谱性质。目标产物均为新的化合物，其结构经元素分析，IR，1H NMR和MS确正。     

Reaction of 4-Methylene-1,3-dioxolan-2-ones with Hydrazines

The structure of the products of the reaction of 4-methylene-1,3-dioxolan-2-ones with hydrazines is a function of the structure of the starting hydrazine. 1,3,4-Oxadiazin-2-one derivatives are obtained from hydrazine hydrate. 3-Arylaminooxazolidin-2-ones are obtained from monoarylhydrazines, while mixtures of these derivatives are obtained from aliphatic monoalkylhydrazines.
