The third generation amperometric biosensor for the determination of hydrogen peroxide (H2O2) has been described. For the fabrication of biosensor, o-aminobenzoic acid (oABA) was first electropolymerized on the surface of platinum (Pt) electrode as an electrostatic repulsion layer to reject interferences. Horseradish peroxidase (HRP) absorbed by nano-scaled particulate gold (nano-Au) was immobilized on the electrode modified with polymerized o-aminobenzoic acid (poABA) with L-cysteine as a linker to prepare a biosensor for the detection of H2O2. Amperometric detection of H2O2 was realized at a potential of +20 mV versus SCE. The resulting biosensor exhibited fast response, excellent reproducibility and sensibility, expanded linear range and low interferences. Temperature and pH dependence and stability of the sensor were investigated. The optimal sensor gave a linear response in the range of 2.99×10^-6 to 3.55×10^-3 mol·L^-1 to H2O2 with a sensibility of 0.0177 A·L^-1·mol^-1 and a detection limit (S/N = 3) of 4.3×10^-7 mol·L^-1. The biosensor demonstrated a 95% response within less than 10 s.  相似文献

A novel nanocomposite designed by the assembly of the positively charged poly(diallyldimethylammonium chloride) protected gold nanoparticles (PDDA‐GNPs), and the negatively charged multi‐walled carbon nanotubes (MWCNTs) on ITO electrode via electrostatic interaction, was used as a supporting matrix for immobilizing hemoglobin (Hb) to develop a high‐performance hydrogen peroxide (H2O2) biosensor. The cyclic voltammetrys of immobilized Hb showed a pair of well‐defined and quasi‐reversible redox peaks with the formal potential of ‐0.205V (vs. SCE) and the peak‐to‐peak potential separation of 44 mV at a scan rate of 100 mV×s?1 in 0.1 mol×L?1 pH 7.0 PBS. Under the optimized experimental conditions, a linearity range for determination of H2O2 was from 2.0 × 10?6 to 5.2 × 10?4 mol×L?1 with a correlation coefficient of 0.9994 (n = 37) and a detection limit of 8.4 × 10?7 mol×L?1. The biosensor displayed excellent electrochemical and electrocatalytic response to the reduction of H2O2, high sensitivity, long‐term stability, good bioactivity and selectivity.  相似文献

The highly efficient H2O2 biosensor was fabricated on the basis of the complex films of hemoglobin (Hb), nano ZnO, chitosan (CHIT) dispersed solution and nano Au immobilized on glassy carbon electrode (GCE). Biocompatible ZnO‐CHIT composition provided a suitable microenvironment to keep Hb bioactivity (Michaelis‐Menten constant of 0.075 mmol L?1). The presence of nano Au in matrix could effectively enhance electron transfer between Hb and electrode. The electrochemical behaviors and effects of solution pH values were carefully examined in this paper. The (ZnO‐CHIT)‐Au‐Hb/GCE demonstrated excellently electrocatalytical ability for H2O2. This biosensor had a fast response to H2O2 less than 4 s and excellent linear relationships were obtained in the concentration range from1.94×10?7 to 1.73×10?3 mol L?1 with the detection limit of 9.7×10?8 mol L?1 (S/N=3) under the optimum conditions. Moreover, the stability and reproducibility of this biosensor were evaluated with satisfactory results.  相似文献

Hemoglobin (Hb) was successfully immobilized on a gold electrode modified with gold nanoparticles (AuNPs) via a molecule bridge 1,6-hexanedithiol (HDT). The AFM images suggested that the HDT/gold electrode could adsorb more AuNPs. UV-vis spectra indicated that Hb on AuNPs/HDT film retained its near-native secondary structures. The electrochemical behaviors of the sensor were characterized with cyclic voltammetric techniques. The resultant electrode displayed an excellent electrocatalytical response to the reduction of hydrogen peroxide (H2O2). The linear relationship existed between the catalytic current and the H2O2 concentration ranging from 5.0 × 10?8 to 1.0 × 10?6 mol · L?1. By one‐step co‐electrodeposition CaCO3 nanoparticles‐chitosan composite film on carbon ionic liquid electrode (CILE), and then by spreading the composition of hemoglobin (Hb) and chitosan on the nanoCaCO3‐chi/CILE, a Hb‐chi/nanoCaCO3‐chi/CILE was fabricated and the direct electrochemistry and electrocatalysis of Hb at the electrode was investigated. The electrochemical impedance spectroscopy of the modified electrode showed the electron transfer resistance was 1166 Ω. Investigation results of cyclic voltammetrys showed a pair of well‐defined and quasireversible redox peak of Hb with the formal potentials of ‐0.295 V (vs. SCE) in 0.1 mol·L‐1 pH 7.0 PBS; the response time of the reduction peak currents of Hb was lower than 3s; a linear range for determination of H2O2 was from 5.0 μmol·L‐1 to 1.3 mmol·L‐1 with a detection limit of 1.6 μmol·L‐1 (S/N = 3) and a sensitivity of 0.16 A·M‐1·cm‐2; the electron transfer rate constant and the apparent Michaelis‐Menten constant of Hb were 1.98 s‐1 and 0.81 mmol·L‐1, respectively. As a result, the case of the one‐step co‐electrodeposition and the promising feature of biocomposite could serve as a versatile platform for the fabrication of electrochemical biosensors.  相似文献

A simple and effective glucose biosensor based on immobilization of glucose oxidase (GOD) in graphene (GR)/Nafion film was constructed. The results indicated that the immobilized GOD can maintain its native structure and bioactivity, and the GR/Nafion film provides a favorable microenvironment for GOD immobilization and promotes the direct electron transfer between the electrode substrate and the redox center of GOD. The electrode reaction of the immobilized GOD shows a reversible and surface‐controlled process with the large electron transfer rate constant (ks) of 3.42±0.08 s?1. Based on the oxygen consumption during the oxidation process of glucose catalyzed by the immobilized GOD, the as‐prepared GOD/GR/Nafion/GCE electrode exhibits a linear range from 0.5 to 14 mmol·L?1 with a detection limit of 0.03 mmol·L?1. Moreover, it displays a good reproducibility and long‐term stability.  相似文献

We developed a novel iron‐tetrasulfophthalocyanine‐graphene‐Nafion (FeTSPc‐GR‐Nafion) modified screen‐printed electrode to determine hydrogen peroxide (H2O2) with high sensitivity and selectivity. The nanocomposite film (FeTSPc‐GR‐Nafion) exhibits an excellent electrocatalytic activity towards oxidation of H2O2 at a potential of +0.35 V in the absence of enzyme. A comparative study reveals that the FeTSPc‐GR complexes play a dual amplification role. Amperometric experiment indicates that the sensors possess good sensitivity and selectivity, with a linear range from 2.0×10?7 M to 5.0×10?3 M and a detection limit of 8.0×10?8 M. This sensor has been successfully used to develop the glucose biosensor and has also been applied to determine H2O2 in sterile water.  相似文献

Abstract

A novel approach to assemble an H2O2 amperometric biosensor was introduced. The biosensor was constructed by entrapping horseradish peroxidase (HRP) labeled nano‐scaled particulate gold (nano‐Au) (HRP‐nano‐Au electrostatic composite) in a new silica sol‐gel/alginate hybrid film using glassy carbon electrode as based electrode. This suggested strategy fully merged the merits of sol‐gel derived inorganic‐organic composite film and the nano‐Au intermediator. The silica sol‐gel/alginate hybrid material can improve the properties of conventional sol‐gel material and effectively prevent cracking of film. The entrapment of HRP in the form of HRP‐nano‐Au can not only factually prevent the leaking of enzyme out of the film but also provide a favorable microenvironment for HRP. With hydroquinone as an electron mediator, the proposed HRP electrode exhibited good catalytic activity for the reduction of H2O2. The parameters affecting both the qualities of sol‐gel/alginate hybrid film and the biosensor response were optimized. The biosensor exhibited high sensitivity of 0.40 Al mol?1 cm?2 for H2O2 over a wide linear range of concentration from 1.22×10?5 to 1.46×10?3 mol L?1, rapid response of <5 s and a detection limit of 0.61×10?6 mol L?1. The enzyme electrode has remarkable stability and retained 86% of its initial activity after 45 days of storage in 0.1 mol L?1 Tris‐HCl buffer solutions at pH 7.  相似文献

A new amperometric biosensor for hydrogen peroxide (H2O2) was developed by adsorbing hemoglobin (Hb) on an organic sol‐gel film. The organic sol‐gel was prepared using resorcinol and formaldehyde as monomers. This sol‐gel film shows a biocompatible microenvironment for retaining the native activity of the adsorbed Hb. The direct electron transfer between Hb and electrode is achieved. Hb adsorbed on the film shows an enzyme‐like catalytic activity for the reduction of H2O2. The reduction peak currents are proportional linearly to the concentration of hydrogen peroxide in the range of 6×10?8 to 3.6×10?6 M, with a detection limit of 2.4×10?8 M (S/N=3). This research enlarges the applications of organic sol‐gel materials in biosensor field.  相似文献

In this paper a room temperature ionic liquid 1‐butyl‐3‐methylimidazolium hexafluorophosphate (BMIMPF6) was used as binder for the construction of carbon ionic liquid electrode (CILE) and a new electrochemical biosensor was developed for determination of H2O2 by immobilization of hemoglobin (Hb) in the composite film of Nafion/nano‐CaCO3 on the surface of CILE. The Hb modified electrode showed a pair of well‐defined, quasi‐reversible redox peaks with Epa and Epc as ?0.265 V and ?0.470 V (vs. SCE). The formal potential (E°′) was got by the midpoint of Epa and Epc as ?0.368 V, which was the characteristic of Hb Fe(III)/Fe(II) redox couples. The peak to peak separation was 205 mV in pH 7.0 Britton–Robinson (B–R) buffer solution at the scan rate of 100 mV/s. The direct electrochemistry of Hb in the film was carefully investigated and the electrochemical parameters of Hb on the modified electrode were calculated as α=0.487 and ks=0.128 s?1. The Nafion/nano‐CaCO3/Hb film electrode showed good electrocatalysis to the reduction of H2O2 in the linear range from 8.0 to 240.0 μmol/L and the detection limit as 5.0 μmol/L (3σ). The apparent Michaelis–Menten constant (KMapp) was estimated to be 65.7 μmol/L. UV‐vis absorption spectroscopy and FT‐IR spectroscopy showed that Hb in the Nafion/nano‐CaCO3 composite film could retain its native structure.  相似文献

A simple and efficient electrochemical method is utilized to functionalize aligned carbon nanotubes (ACNTs) by the electrochemical reduction of 4‐carboxyphenyl diazonium salt. Thus hemoglobin (Hb) molecules were covalently immobilized on the diazonium‐ACNTs surface via carbodiimide chemistry. Direct electrochemistry and bioelectrocatalytic activity of the immobilized Hb were then investigated by cyclic voltammetry (CV) and amperometry techniques. It is showed that the Hb film on the diazonium‐ACNTs electrode had well‐defined redox peaks with a formal potential (E°) at ?312 mV (vs. Ag/AgCl), and the Hb‐ACNTs electrode displayed good electrocatalytic activity to H2O2 reduction. Owing to the high Hb covering on the ACNTs surface (Γ*=2.7×10?9 mol cm?2), the catalytic current were significantly improved when compared to the current measured at an Hb‐tangled carbon nanotubes electrode. The Hb‐ACNTs electrode exhibited high sensitivity, long‐term stability and wide concentration range from 40 μM to 3 mM for the amperometric detection of H2O2. A nanobiocomposite film consisted of zirconia, multiwalled carbon nanotubes (MWNTs) and Myoglobin (Mb) was electrochemically deposited on the electrode. Direct electron transfer for the immobilized Mb was realized and high electrocatalytic efficiency toward H2O2 was observed. The proposed biosensor via a simple one‐step electrodeposition method displayed a broader linear range and a lower detection limit for H2O2, as compared with those CNT or ZrO2 based biosensor. The linear range is from 2 × 10?6 M to 1 × 10?3 M with the detection limit of 6 × 10?7 M. The present strategy provides a simple and effective method to assemble CNT, ZrO2 and enzyme nanohybrid on the electrode and expands the scope of CNT‐based electrochemical devices.  相似文献

A biocompatible nanocomposite film was fabricated for hemoglobin (Hb) molecules immobilization. This film consists of multiwalled carbon nanotubes (MWNTs), 1‐pyrenebutanoic acid, succinimidyl ester (PASE), hemoglobin (Hb) and Au nanoparticles (AuNPs). Herein, PASE molecules physically adsorbed onto MWNTs, and its groups then covalently bond with Hb. AuNPs were then linked with Hb/PASE/MWNTs via electrostatic adsorption force. UV‐visible adsorption spectra, Fourier transform infrared spectra, scanning electron microscope and electrochemical impedance spectroscopy have characterized the film. Cyclic voltammetry (CV) scans showed that in the film Hb has well‐defined redox reaction, with the formal potential (E°) at about ?0.27 V (vs. Ag/AgCl). Herein, differential pulse voltammetry (DPV) was employed to electrochemically detect the Hb in the film with a detection limit of 9.3×10?8 M. The proposed method was also succeeded for Hb detection in clinical blood samples. Furthermore, the Hb in the film showed good electrocatalytic activities to the reduction of H2O2, TCA, NaNO2 and O2.  相似文献

Chitosan (CS) was chosen for dispersing multi‐wall carbon nanotubes (MWNTs) to form a stable CS‐MWNTs composite, which was first coated on the surface of a glassy carbon electrode to provide a containing amino groups interface for assembling colloidal gold nanoparticles (GNPs), followed by the adsorption of hemoglobin (Hb). Repeating the assembly step of GNPs and Hb resulted in {Hb/GNPs}n multilayers. The assembly of GNPs onto CS‐MWNTs composites was confirmed by transmission electron microscopy. The consecutive growth of {Hb/GNPs}n multilayers was confirmed by cyclic voltammetry and UV‐vis absorption spectroscopy. The resulting system brings a new platform for electrochemical devices by using the synergistic action of the electrocatalytic activity of GNPs and MWNTs. The resulting biosensor displays an excellent electrocatalytic activity and rapid response for hydrogen peroxide. The linear range for the determination of H2O2 was from 5.0×10?7 to 2.0×10?3 M with a detection limit of 2.1×10?7 M at 3σ and a Michaelis–Menten constant KMapp value of 0.19 mM.  相似文献

When the concentration of dodecyl benzene sulfonic acid sodium salt (SDBS) is 0.7 mmol·L?1, the electrochemical and electrochemiluminescence (ECL) intensity of Ru(bpy)32+‐chlorpheniramine maleate (CPM) system at the Au electrode were studied. The results showed that compared with the absence of SDBS, enhancement of the ECL intensity was 14‐fold at Au electrode. Base on this, an ECL method was established for efficient and simple determination of CPM at Au electrode. Under the optimum experimental condition, the enhanced ECL intensities had good linear relationship with the concentration of CPM in the range of 1.0×10?4–1.0×10?7 mol·L?1, and a linear regression equation was obtained as follows: I (counts)=48.805×106c+394.03 (r=0.9975), the detection limit for CPM was 1.4×10?8 mol·L?1. The RSD for 5 times determinations of 1.0×10?5 mol·L?1 CPM was 3.2%. The results of recovery test were between 96.3%–102.5%, and the RSD of recovery test (n=5) was 2.7%. In addition, eleven kinds of tertiary amines‐Ru(bpy)32+ systems were investigated in the absence and presence of SDBS. The results showed that the enhancement of SDBS on ECL intensity of tertiary amines‐Ru(bpy)32+ systems was universal.  相似文献

A new enzyme‐based amperometric biosensor for hydrogen peroxide was developed relying on the efficient immobilization of horseradish peroxidase (HRP) to a nano‐scaled particulate gold (nano‐Au) film modified glassy carbon electrode (GC). The nano‐Au film was obtained by a chitosan film which was first formed on the surface of GC. The high affinity of chitosan for nano‐Au associated with its amino groups resulted in the formation of nano‐Au film on the surface of GC. The film formed served as an intermediator to retain high efficient and stable immobilization of the enzyme. H2O2 was detected using hydroquinone as an electron mediator to transfer electrons between the electrode and HRP. The HRP immobilized on nano‐Au film maintained excellent electrocatalytical activity to the reduction of H2O2. The experimental parameters such as the operating potential of the working electrode, mediator concentration and pH of background electrolyte were optimized for best analytical performance of amperometry. The linear range of detection for H2O2 is from 6.1×10?6 to 1.8×10?3 mol L?1 with a detection limit of 6.1 μmol L?1 based on signal/noise=3. The proposed HRP enzyme sensor has the features of high sensitivity (0.25 Almol?1cm?2), fast response time (t90%≤10 s) and a long‐term stability (>1 month). As an extension, glucose oxidase (GOD) was chemically bound to HRP‐modified electrode. A GOD/HRP bienzyme‐modified electrode formed in this way can be applied to the determination of glucose with satisfactory performance.  相似文献

A novel method for preparation of hydrogen peroxide biosensor was presented based on immobilization of hemoglobin (Hb) on carbon‐coated iron nanoparticles (CIN). CIN was firstly dispersed in a chitosan solution and cast onto a glassy carbon electrode to form a CIN/chitosan composite film modified electrode. Hb was then immobilized onto the composite film with the cross‐linking of glutaraldehyde. The immobilized Hb displayed a pair of stable and quasireversible redox peaks and excellent electrocatalytic reduction of hydrogen peroxide (H2O2), which leading to an unmediated biosensor for H2O2. The electrocatalytic response exhibited a linear dependence on H2O2 concentration in a wide range from 3.1 μM to 4.0 mM with a detection limit of 1.2 μM (S/N=3). The designed biosensor exhibited acceptable stability, long‐term life and good reproducibility.  相似文献

合成了水溶性硒化镉(CdSe)量子点,利用组装技术和静电吸附作用,将带正电荷的血红蛋白(Hb)和带负电荷的CdSe量子点层层组装到壳聚糖(chit)修饰的玻碳电极(GCE)表面,构建基于{Hb/CdSe}n多层膜的无电子媒介体的电流型生物传感器({Hb/CdSe}3/chit/GCE).运用紫外-可见吸收光谱、电致化学发光、交流阻抗和循环伏安技术来表征修饰膜,并研究传感器的作用机理、性能及分析应用.结果表明:与量子点薄膜法及量子点/血红蛋白复合物法等固载血红蛋白的其他方法相比,层层组装法能显著提高血红蛋白的固定量,保持血红蛋白的生物活性,增强传感器的灵敏度和稳定性.传感器检测H2O2的线性范围为4.0×10-8~4.8×10-6 mol·L-1(r=0.999 1),检测限为2.0×10-8mol·L-l.多层膜的电致化学发光研究,表明修饰电极有望用于电致化学发光传感器的制备.  相似文献

The electrocatalysis oxidation of guanosine‐5′‐monophosphate (GMP) was investigated on Mg‐Al layered double hydroxide (LDH) functionalized with sodium dodecyl sulfate (SDS) and room temperature ionic liquid (RTIL) modified glass carbon electrode (GCE). The cyclic voltammogram of GMP on the modified electrode (RTIL/ LDH‐SDS/GCE) exhibited a well defined anodic peak at 1.091 V in 0.2 mol·L?1 pH 4.4 acetate buffer solution. The GMP oxidation was enhanced in the presence of anionic surfactant in the ?lms. The results suggest that the surfactant molecules intercalate the LDH layers to preconcentrate GMP molecules and the RTIL showed good ionic conductivity. The experimental parameters were optimized, the kinetic parameters were investigated and the probable oxidation mechanism was proposed. Under the optimized conditions, the oxidation peak current was proportional to GMP concentration in the range from 5.0×10?7 to 1.0×10?4 mol·L?1 with the correlation coefficient of 0.9987 and the detection limit was 1.0×10?7 mol·L?1. The RTIL/LDH‐SDS/GCE showed a good electrochemical response to the oxidation of GMP and would be developed into a new biosensor.  相似文献
