The reaction of 4-amino-5-phenyl-3,5-thiaaza-4-pentenoic acid 3 with various nitrilimines 2 led to the formation of substituted carboxymethylthio derivatives of 1,2,4-triazoles (4a–l) rather than the amino derivatives 5 or imidazole derivatives 6. The spectral data of the synthesized compounds are in full agreement with its molecular structure.  相似文献

Reactions of 2,3‐dihydro‐4‐oxo‐thiopyrano[2,3‐b]pyridine with aldehydes and with DMF‐DMA furnished the 3‐benzylidene and 3‐(N,N‐dimethylamino)‐methylene derivatives. The latter products afforded spiro‐pyrazolo‐3,3′‐thiopyrano[2,3‐b]pyridines and new tetra‐ and penta‐heterocyclic ring systems when treated with nitrilimines and aminoazoles, respectively. A number of the products showed high antifungal and antibacterial activities.  相似文献

The 2‐phenylimino‐1,3‐thiazolidin‐4‐one 2 was obtained by thermal cyclization of 4‐amino‐5‐phen‐yl‐3, 5‐thiaaza‐4‐pentenoic acid 1 using DCC as dehydration agent. Treatment of 2‐phenylimino‐1,3‐thiazolidin‐4‐one 2 with various hydrazonoyl halides 3 (nitrilimines 4 precursor) yielded 6‐aryl‐9‐phenyl‐8‐substituted‐1,4,6,7,9‐thiatetrazaspiro‐[4.4]non‐7‐en‐3‐ones 5a‐1. Both analytical and spectroscopic data of all the synthesized compounds are in full agreement with the proposed structures.  相似文献

2‐Acetylbenzothiazole ( 1 ) reacts with dimethylformamide dimethylacetal (DMF‐DMA) to afford the enaminone 2. Compound 2 reacts regioselectively with some nitrilimines 5a–d and nitrile oxides 6b–d to afford the novel pyrazole and isoxazole derivatives 11a–d and 12b–d, respectively, which react with hydrazine hydrate to give the new pyrazolo[3,4‐d]pyridazine and isoxazolo[3,4‐d]pyridazine derivatives 13a–d and 14b–d, respectively. The enaminone 2 reacts with 1H‐benzimidazole‐2‐acetonitrile ( 17 ) to afford the pyrido[1,2‐a]benzimidazole derivatives 19. Compound 2 reacts also with 5‐amino‐3‐phenylpyrazole ( 20 ) and with guanidine to afford the new pyrazolo[1,5‐a]pyrimidine and the 2‐aminopyrimidine derivatives 22 and 24, respectively. © 1999 John Wiley & Sons, Inc. Both substituted 2,4,5,6-tetrahydrocyclopenta[c]pyrazoles and 2,4,5,6-tetrahydropyrrolo[3,4-c]pyrazoles have been synthesized by the 3+2 intramolecular dipolar cycloaddition of nitrilimines to alkynes. This cyclization has been extended to more versatile 3-bromo derivatives by the use of alkynylbromides as dipolarophiles.  相似文献

Compounds 1,3,6,6a‐tetraphenyl‐1,6a‐dihydrocyclopenta[c]pyrazol‐4(3aH)‐ones ( 3 ), 1,3,3a,4‐tetraphenyl‐1,3a‐dihydrocyclopenta[c]pyrazol‐6‐ols ( 4 ) and 6‐hydroxy‐1,3,6,6a‐tetraphenyl‐1,5,6,6a‐tetrahydrocyclopenta[c]pyrazole‐4(aH)‐one ( 5 ) were synthesized by reaction of 3,4‐diphenyl‐4‐hydroxy‐2‐cyclopentenone ( 2a ) with nitrilimines, generated in situ by the action of triethylamine on the corresponding hydrazonyl chlorides 1 .  相似文献

The reactivity of indole derivatives towards nitrilimines has been studied. Substituents at positions 1, 2 and 3 of the indole ring greatly affect the course of the reaction. 1,3-Dipolar cycloaddition products (3a,8a,dihydropyrazole-[3,4-b]-indole derivatives) and non-cyclic addition products (3-indolyl derivatives) were obtained depending on these substituents. The structures reported were assigned on the basis of satisfactory analytical, spectroscopic and chemical data.  相似文献

The latter product undergoes regioselective 1,3‐dipolar cycloaddition with nitrilimines and nitrile oxides to afford the novel 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl)carbonyl‐1‐phenylpyrazole and 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl)carbonyl isoxazole derivatives, respectively. It reacts also with 1H‐benzimidazole‐2‐acetonitrile, 2‐aminobenzimidazole and 3‐amino‐1,2,4‐triazole to afford the novel pyrido[1,2‐a]benzimidazole, pyrimido[1,2‐a]benzimidazole and the triazolo[4,3‐a]pyrimidine derivatives, respectively. The reaction of 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl) carbonyl‐1‐phenylpyrazole derivatives with hydrazine hydrate led to a new pyrazolo[3,4‐d]pyridazine derivatives.  相似文献

The polymer-supported liquid-phase synthesis of small organic molecules has been a subject of intense research activity. 1,2,4-Triazoles are well known for their antifungal[1] and antibacterial[2] activities. This moiety was also found in potent agonist or antagonist receptor lingands. The synthesis of substituted 1,2,4-triazoles via 1,3-dipolar cycloaddition of imines with nitrilimines is well documented.[3] Herein, we would like to report the first liquid-phase synthesis of 1,3,4,5-tetraaryl-4,5-dihydro-1,2,4-triazoles through a 1,3-dipolar cycloaddition of imines with nitrilimines on PEG support.  相似文献

Reaction of nitrilimines 1 with 2-cyanomethylbenzimidazole 2 gave the 3-arylazo-2-methylpyrrolo[1,2-a]benzimidazole 4a rather than the reported 2-arylazo-3-methylpyrrolo[1,2-a]benzimidazole 3a. The correct structure of the product was determined using X-ray crystal structure analysis. The similar reaction of nitrilimines with 2-aminobenzimidazole 5 gave the acyclic nucleophilic addition product 6.  相似文献

Abstract  

Eleven novel dihydropyrrolo[3,4-c]pyrazole derivatives were obtained by the reaction of chiral (1R)-N-(1-phenylethyl)maleimide and C,N-aryl-substituted nitrilimines. The reaction afforded the cycloadducts as a regioisomeric mixture which can be separable in some cases. The structure and stereochemistry of cycloadducts were assigned on the basis of infrared (IR), 1H and 13C nuclear magnetic resonance (NMR), mass and X-ray spectra, optical rotation measurements, and CHN analyses.  相似文献

A new, unusual way to obtain polysubstituted 1H-S-triazole N-imine derivatives is described from reactions of the readily available 1,3,4-oxadiadin-6-one derivatives and nitrilimines. Besides an hydrazide derivative of phenylglyoxylic acid is also formed. Both adducts were characterised by X-ray crystal structure analysis.  相似文献

2‐Thioxo‐1,2,5,6,7,8‐hexahydroquinoline‐3‐carbonitrile ( 2 ) was easily S‐alkylated to produce alkyl mercapto derivatives 3a‐g . The latter compounds were cyclized to afford thienotetrahydroquinolines 4a‐g . Several pyrimidothienotetrahydroquinolines 5a‐d , and 6a‐d were obtained from the condensation of compounds 4c‐f with different reagents. o‐Aminocarbohydrazide derivative 11 was reacted with aromatic aldehydes, acetylacetone, nitrous acid and CS2 to afford compounds 12–15 . Compound 24 was coupled with aryldiazonium chloride to afford arylazo derivatives 25 . Also it condensed with aromatic aldehydes to give arylidene derivatives 26 . Reaction of 1‐(5‐acetyl‐4‐methylthiazol–2‐yl)–3‐phenylthiourea 2 with hydrazonoyl chlorides ( 3a , 3b , 3c , 3d , 3e , 3f ) and 9 yielded the corresponding (thiazolyl)imino–1,3,4‐thiadiazole derivatives ( 6a , 6b , 6c , 6d , 6e , 6f ) and 12 , respectively. Reaction of 2 with ethyl chloroacetate 13 gave (thiazolyl)imino‐1,3‐thiazolidin‐4‐one derivative 15 , which upon condensation with aromatic aldehyde derivatives yielded the 5‐benzylidene derivatives ( 16a , 16b ). In addition, treatment of 2 with 3‐chloropenta‐2,4‐dione 17 afforded the corresponding (thiazolyl)imino‐1,3‐thiazole derivative 19 . The newly synthesized compounds were confirmed from their elemental analyses and spectral data.  相似文献

A variety of 3″,5″‐diaryl‐3″H,4′H‐dispiro[cyclohexane‐1,2′‐chromene‐3′,2″‐[1,3,4]thiadiazol]‐4′‐ones 3a‐c were synthesized regioselectively through the reaction of 4′H,5H‐trispiro[cyclohexane‐1,2′‐chromene‐3′,2″‐[1,3,4]oxadithiino[5,6‐c]chromene‐5″,1″′‐cyclohexan]‐4′‐one ( 1 ) with nitrilimines (generated in situ via triethylamine dehydrohalogenation of the corresponding hydrazonoyl chlorides 2a‐c ) in refluxing dry toluene. Single crystal X‐ray diffraction studies of 3a,b add support for the established structure. Similarly, 3′,5′‐diaryl‐2,2‐dimethyl‐3′H,4H‐spiro[chromene‐3,2′‐[1,3,4]thiadiazol]‐4‐ones 5a‐c were obtained in a regioselective manner through the reaction of 2,2,5′,5′‐tetramethyl‐4H,5′H‐spiro[chromene‐3,2′‐[1,3,4]oxadithiino[5,6‐c]chromen]‐4‐one ( 4a ) with nitrilimines under similar reaction conditions. On the other hand, reaction of 2,5′‐diethyl‐2,5′‐dimethyl‐4H,5′H‐spiro[chromene‐3,2′‐[1,3,4]oxadithiino‐[5,6‐c]chromen]‐4‐one ( 4b ) with nitrilimines in refluxing dry toluene afforded the corresponding 3′,5′‐diaryl‐2‐ethyl‐2‐methyl‐3′H,4H‐spiro[chromene‐3,2′‐[1,3,4]thiadiazol]‐4‐ones 5d‐f as two unisolable diastereoisomeric forms.  相似文献
