4-oxo-4-phenylbutanehydrazide 3 was reacted with aryl or alkyl isothiocyanates to give the corresponding N-substituted-2-(4-oxo-4-phenylbutanoyl) hydrazine-1-carbothioamide 4a-c . Cyclization of thiosemicarbazides 4a-c with sodium hydroxide led to the formation of 3-(4-sub-5-thioxo-1,2,4-triazol-3-yl)-propanone 5a-c . Desulfurization of thiosemicarbazides 4a-c by mercuric oxide afforded 3-(5-(sub-amino)-1,3,4-oxadiazol-2-yl)-propanone 6a-c . The reaction of 4a-c with phosphorus oxychloride gave 3-(5-(sub-amino)-1,3,4-thiadiazol-2-yl)-propanone 7a-c . Treatment of 4a-c with ethyl-bromoacetate or α-bromopropionic acid gave N′-(3-sub-thiazolidin-2-ylidene)-butanehydrazide 8a-c and (N′-(3-sub-oxothiazolidin-2-ylidene)-butanehydrazide 9a-c . Chlorination of oxothiazolidine-hydrazide 9a-c by phosphorus oxychloride afforded N-(3-sub-4-oxothiazolidine)-butane-hydrazonoyl-chloride 10a-c . The reaction of 10a-c with mercaptoacetyl-chloride yielded 2-((4-benzoyl-thiopyrane) hydrazono)-3-sub-thiazolidinone 11a-c . Also, reacted of 10a-c with hydrazine hydrate afforded N″-(3-sub-oxothiazolidine)-butane-hydrazon-hydrazide 12a-c . The 3-sub-2-((pyridazine) hydrazono) thiazolidinone 13a-c was obtained by cyclization of 12a-c via refluxing in DMF. The reaction and cyclized of 9a-c with chloroacetyl-chloride in ethanolic KOH afforded 1-((3-sub-4-oxothiazolidine) amino)-azepine-dione 14a-c . The chemical structures of the new compounds have been confirmed by diverse spectroscopy analyses such as IR, NMR, MS, and elemental analysis. The synthesized compounds were tested for their antimicrobial activity and these compounds were considered (Pyridazin-hydrazono-thiazolidinone 13a-c , oxothiazolidin-azepinedione 14a-c , N-thiazolidin-hydrazon-hydrazide 12a-c , and thiopyran-hydrazono-thiazolidinone 11a-c ) the most effective as antimicrobial activity.  相似文献

Reactions of the iminoesters 3a-c with hydrazine or 1,2-dimethylhydrazine gave 4,5-dihydro- 4a-c or 2,5-dihydro-1,2,4-triazin-6(1H) ones 7a-c , respectively. When methylhydrazine was employed, 1-methyl-4, 5-dihydro- 5a-c and 2-methyl-2,5-dihydro-1,2,4-triazin-6(1H) ones 6a-c were obtained. Compounds 6a-c exist as zwitterions in the solid state and in polar aprotic solvents.  相似文献

4-Oxo-4-phenylbutanehydrazide (1) reacted with many active methylene reagents such as acetylacetone, diethylmalonate, ethylacetoacetate, ethylcyanoacetate, benzoyl-acetonitrile, and malononitrile under neat conditions to afford the corresponding pyrazoles (2–7) , also, treatment of butanehydrazide (1) with electrophilic reagents as triethylorthoformate, dimethylformamide-dimethylacetal, acetic anhydride, and carbon disulfide to give 1,3,4-oxadiazoles (8,10,11) and N′-acetyl-butanehydrazide (9) . Reacted of butanehydrazide (1) with potassium thiocyanate gave 1,2,4-triazoles (12) . Similarly, treatment of (1) with chloroacetamide gave 1,2,4-triazinones (13) . The pyrrolotriazinones (14) was obtained by cyclization of (13) . Also, butanehydrazide ( 1 ) was utilized as a starting material for the synthesized of new Schiff bases as N′-(4-sub-benzylidene)-phenylbutane-hydrazide (15a-c) , which are used as an initiative to prepare new compounds such as 1,2,4-triazepinones (16a-c) , pyrrolotriazepinones (17a-c) , 1,2,4-triazines (18a-c) , and pyrrolotriazines (19a-c) by reacted of (15a-c) with each chloroacetamide or formamide. The chemical structure of the newly prepared compounds was determined through the spectrum data, including IR, NMR, and MS. The prepared compounds were tested for their in vitro antitumor activities. The compounds 17a-c , 16a-c , and 19a-c displayed activity against several types of cancer cell lines.  相似文献

From previous results with lower homologues, dehydroiodination of the three alkenyl-beta-enamino esters 3a-c was expected to provide six-membered N-heterocyclic products. The reactions of 3a-c with triethylamine are found to lead, however, to the unexpected stereoselective synthesis of the trisubstituted cyclopentane derivatives 4a-c, as confirmed by IR and NMR spectroscopy. Cyclopentanes 4a-c bear two chiral centers and a gamma-amino ester moiety, and are therefore conformationally restricted analogues of gamma-amino butyric acid (GABA), which is the major inhibitory neurotransmitter in the central nervous system. Use of electrospray ionization mass (ESI-MS) and tandem mass spectrometry (ESI-MS/MS) allowed the key iminium ion intermediates 5a-c(+), as well as the protonated molecules of both the reactant and final products, [3a-c + H](+) and [4a-c + H](+), to be intercepted and structurally characterized. From these findings a mechanism for this unexpected but synthetically attractive and efficient stereoselective reaction is proposed.  相似文献

A novel D-pi-A system in which tetrathiafulvalene (TTF) and pi-extended TTFs as strong electron donors are covalently connected to a tricarbonyl (eta(6)-arene)chromium complex as the acceptor moiety through a systematically increased conjugated bridge of vinylene units (12a-c, 16a-c) have been synthesized by Wittig-Horner olefination reaction. The electronic spectra as well as the electrochemical data reveal a different behavior of TTF derivatives (12a-c) and of exTTF derivatives (16a-c). Cyclic voltammetry shows the influence of the tricarbonylchromium arene on the oxidation potentials in compounds 12a-c, and no remarkable effect is observed for exTTFs (16a-c). The nonlinear optical properties of 12a-c and 16a-c have been calculated by using the ab initio CPHF/6-31G//B3P86/6-31G model, and the time-dependent density functional theory (TD-DFT) method has been used for the calculation of the electronic transitions. The calculations reveal that an intraligand charge-transfer transition (ILCT) and the metal to ligand charge-transfer transition (MLCT) are responsible for the nonlinear response. In addition, the large angles formed by the ground-state dipole moment and the vectorial hyperpolarizability are responsible for the mubeta values determined experimentally by the EFISH technique.  相似文献

A new versatile synthesis of benzo[c]phenanthridine and protoberberine alkaloids using lithiated toluamide-benzonitrile cycloaddition was carried out. The coupling reaction between benzonitrile 6 with o-toluamides (8a-c) afforded 3-arylisoquinolines (9a-c) that were transformed to the protoberberines (11a-c) or benzo[c]phenanthridines (14a-c). These compounds were synthesized by ring closure of the two-carbon chain on either position 2 or 4 of the 3-arylisoquinolinone (9a-c). Several kinds of substituted benzo[c]phenanthridine alkaloids such as oxysanguinarine, oxyavicine, and oxynitidine as well as protoberberines such as 8-oxocoptisine, 8-oxopseudoberberine, and 8-oxopseudocoptisine were synthesized.  相似文献

Novel C(60)/pi-extended tetrathiafulvalene (exTTF) diads (12a-c) and triads [D(2)A (14a-c) and DA(2) (25, 27a-c)] have been synthesized by the Bingel cyclopropanation reaction of the respective exTTF-containing malonates and [60]fullerene. The reaction of exTTF-bismalonates with C(60) affords the respective C(60)-exTTF diads (26a-c) together with the triad C(60)-exTTF-C(60) (25, 27a-c) and a regioisomeric mixture of bisadducts (28b-c). Theoretical calculations (PM3) predict the favored geometry for triads 14a-c depending upon the orientation (up and down) of the 1,3-dithiole rings in the exTTFs, as well as the more stable regioisomers for the bisadducts 28. Cyclic voltammetry measurements reveal that C(60) and exTTF units do not intereact in the ground state. Compounds 26a-c and 27a-c are not electrochemically stable. A photoinduced electron transfer leading to the formation of the radical pair (C(60)(-)-exTTF(*+)) has been observed for compounds 14a-c.  相似文献

Cyanothioacetamide ( 1 ) reacted with acrylonitrile ( 2 ) to afford the corresponding 6-oxo-2-sulfanylpiperidine-3-carbonitrile ( 6 ), which oxidized to give compounds 7 and 8 under different conditions. Moreover, compound 6 was used as a starting material to synthesize 12a-c , 16a-d , 26a-c , 27a-c , and 30a-c via reactions with aromatic aldehydes 9a-c , diazonium chlorides 13a-d , and 3-arylpropennitrile derivatives 18a-i respectively. Considering the data of IR, 1 H NMR, mass spectra, elemental analyses, and theoretical calculations, all the structures of the newly synthesized heterocyclic compounds were elucidated.  相似文献

Condensations of chiral diamines 11a-c with benzotriazole and formaldehyde gave benzotriazolyl intermediates 12a-c; similar condensations of alpha-amino-amides 10a-c with benzotriazole and paraformaldehyde gave 14a-c. Subsequent treatment of 12a-c and 14a-c with AlCl(3) led to enantiopure tricyclic 1,2,3,5,10,10a-hexahydroimidazo[1,5-b]isoquinolines 1a-c and 2,3,10,10a-tetrahydroimidazo[1,5-b]isoquinolin-1(5H)-ones 15a-c, respectively, via Lewis acid promoted iminium cation cyclizations.  相似文献

The reaction of the hydrazones 5a-c with 2-chloroacrylonitrile produced the 1,2-diazepino[3,4-b]quinoxaline hydrochlorides 6a-c , which were transformed into the 5,6,7,13-tetrahydro-5,14-methano-16-oxo-1,5,6-benzoxadiazonino[3,4-b]quinoxalines 7a-c , respectively. The oxidation of 7a-c with diethyl azodicarboxylate afforded the 7,13-dihydro-5,14-methano-16-oxo-1,5,6-benzoxadiazonino[3,4-b]quinoxalines 8a-c , respectively. Compounds 7a-c and 8a-c were also obtained by a one-pot synthesis from 5a-c and 6a-c , respectively.  相似文献

A new series of tetrahydroquinoline derivatives having pyrazol and hydrazide moieties were synthesized for the purpose of anticancer cell line evaluation. Syntheses of these compounds were firstly achieved by one pot four reactions. The reaction of 3-amino-tetrahydro-1H-pyrazolo [3,4-b]quinolin with aldehydes, indoline-2,3-dione derivatives to give 9a-c , 11a-c , and 13a,b , respectively. In similar manner for biological comparison, the reactions of compound 5 with the same aldehydes and indoline-2,3-dione derivatives to give 19a-c and 20a-c . The newly synthesized compounds were examined in vitro for their cytotoxic activity against HepG-2 and A549 cancer cells. The compounds 11a-c and 20a-c showed promising activity as anticancer agents against HepG-2 and A549 cancer cells.  相似文献

Analogs of 5,7-dihydroxytryptamine (5,7-DHT), namely, 4-fluoro-, 6-fluoro-, and 4,6-difluoro-5,7-DHT's (30a-c) were synthesized starting from 4-fluorophenol (7a), 4-fluorobenzyl alcohol (12) and 2,4-difluorophenol (7b), respectively. Regiospecific hydroxylation and formylation ortho to fluoro groups, both via aryllithium intermediates, were made possible by the blocking effect of tert-butyldimethylsilyloxy functions and allowed the conversion of the starting materials to the key intermediates, namely, 3,5-bis(tert-butyldimethylsilyloxy)-2-fluoro-, 4-fluoro- and 2,4-difluorobenzaldehydes (11a, b and 19, respectively). The latter were converted in one step to the corresponding benzyloxybenzaldehydes, from which indole-2-carboxylates 22a-c were synthesized via azidostyrenes 21a-c, respectively. Decarbonylation of the indole-2-carboxaldehydes (24a-c) produced from 22a-c in two steps gave 2,3-unsubstituted indoles 25a-c, respectively. Introduction of the aminoethyl side chains on C-3 of 25a-c via the corresponding indole-3-acetonitriles, and subsequent debenzylation generated the hydroxytryptamines, which were isolated as their creatinine sulfate salts 30a-c, respectively. Cyclic voltammetric studies indicated that like 5,7-DHT, 30a-c undergo electrochemical oxidation in 1 M H2SO4 via the corresponding p-quinoneimine derivatives 31a-c by an electrochemical-chemical-electrochemical (ECE) process. The voltammetrically detectable products of the ECE process appear to be the corresponding 5-hydroxytryptamine-4,7-dione (6) derivatives 33a-c. The nature of the interaction of dissolved O2 with 30a-c at pH 7.4 appears to be strikingly different from that of 5,7-DHT, which undergoes autoxidation at pH 7.4 via the 4-hydroperoxy derivative 4 to the quinone 6.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Starting from the reaction of thioquinanthrene 1 with sodium methoxide followed by the reaction with α,ω-dihalogenoalkanes, title bis-methoxy oligomers 4a-c with four 3,4-quinolinediyl units were prepared (40–91%). Acid catalysed hydrolysis of methoxy groups in 4a-c gave tetramers 5a-c (46–94%) with two 4(1H)-quinolinone functions. The reactions of bis-quinolinones 5a-c with phosphoryl chloride in DMF run as deoxo-chlorination and afforded tide dichlorotetramers 6a-c (51–66%) with 4-chloroquinolinyl groups. The treatment of bis-methoxy tetramers 4a -c with boiling phosphoryl chloride led to title α,ω-bis(4-chloro-3-quinolinylthio)alkanes 7a-c (52–56%) and thioquinanthrene (65–70%).  相似文献

Adenine tetrachlorocyclodiphospha(V)zane derivatives (III(a-c)) were prepared by the reaction of hexachlorocyclodiphospha(V)zane derivatives (I(a-c)) and adenine (II) as precursors. The synthesized compound's and their structures (III(a-c)) were firmly characterized (based on the presence of an inversion center) using FT-IR (4000-200 cm(-1)), UV-vis. (190-800 nm), (1)H, (13)C NMR and Mass spectral measurements in addition to C, H, N, P elemental analysis. The compounds (III(a-c)) were found to be a 1:2 molar ratio of (I(a-c)) and adenine (II) adducts, respectively. Confident and complete vibrational assignments are proposed for nearly all fundamental vibrations, along with detailed interpretation for all observed signals in both (1)H and (13)C NMR spectra of the investigated phospha(V)zanes (III(a-c)). In addition, unconstrained geometry optimization of III(a-c) were carried out by means of DFT-B3LYP/3-21G(d) calculations to provide new insight into the structural parameters and molecular geometries of compounds III(a-c). The results are reported herein and compared with similar molecules whenever appropriate.  相似文献

Reactions of N,N;-disubstituted hydrazinecarbothioamides 8a-c and substituted thioureidoethylthioureas 9a-c with 2,3,5,6-tetrachloro-1,4-benzoquinone (chloranil, 10a) and 2,3,5,6-tetrabromo-1,4-benzoquinone (bromanil, 10b) to form N,N;-disubstituted [1,3,4]thiadiazole-2,5-diamines 11a-c, 6,7-dichloro-3-substituted amino-1H-benzo[1,3,4]- thiadiazine-5,8-diones 12a-c, 2,3,7,8-tetrahalothianthrene-1,4,6,9-tetraones 13a,b, 5,6,8- trihalo-7-oxo-3,7-dihydro-2H-quinoxaline-1-carbothioic acid substituted amides 14a-c, 15a-c and 7-substituted imino-[1,3,6]thiadiazepane-3-thiones 16a-c are reported. Bishydroxyiminoquinoxalines 3a-b react with ethyl chloroformate 4 to afford the furazano[3,4-b]quinoxalines 5a-b . Bishydroxyiminobenzoxazines 6a-c on treatment with 4 are converted into the fused oxadiazolones 7a-c and 8a-c along with the bisethoxycarbonyloxyimino-derivatives 9a-c . 3-Amino-3-thioxopropanamide (1) reacted with ethyl acetoacetate to form 6-hydroxy-4-methyl-2-thioxo-2,3-dihydropyridine-3-carboxamide (2), which reacted with α-haloketones 3 to produce 2,3-disubstituted-8-hydroxy-6-methyl-2H,5H-pyrido[3,2-f]-[1,4]thiazepin-5-ones 4a-c. Benzoylation of 4c led to the formation of the dibenzoate derivative 9. Compounds 4a-c could be prepared stepwise through the formation of S-alkylated derivatives 10a-c. Compounds 2, 4a-c, 9 and 10a-c were prepared using microwave as a source of heat, and gave better yields in shorter times than those achieved by traditional methods. Coupling of 4a-c with arenediazonium chlorides proceeded unusually to give the 6-hydroxy-4-methyl-2-(arylazo)thieno[2,3-b]pyridin-3(2H)-one ring contraction products 14. Structures of the newly synthesized compounds were proven by spectral and chemical methods.  相似文献

1-Phenyl-3-methyl-4-ylidene-5-pyrazolones ( 1a-c ) produce the respective 1:1 phosphonate adducts ( 4a-i ) upon reaction with the appropriate dialkyl phosphites ( 2a-c ). The reaction of 1a-c with trialkyl phosphites ( 3a-c ) proceeds only in presence of water, also to give phosphonates 4a-c almost exclusively. Hexamethylphosphorus triamide ( 3d ) was successfully utilized to introduce the bis(dimethylamino)-phosphoryl function in the f , g -unsaturated carbonyl moiety in 1a-c , producing phosphoramidates 15a-c . Possible reaction mechanisms were discussed. Compatible elemental and spectroscopic results were recorded for the new products.  相似文献
