We investigated the direct electrochemistry of glucose oxidase (GOx) at gelatin-multiwalled carbon nanotube (GCNT) modified glassy carbon electrode (GCE). GOx was covalently immobilized onto GCNT modified GCE through the well known glutaraldehyde (GAD) chemistry. The immobilized GOx showed a pair of well-defined reversible redox peaks with a formal potential (E0′) of ? 0.40 V and a peak to peak separation (ΔEp) of 47 mV. The surface coverage concentration (Г) of GOx in GCNT/GOx/GAD composite film modified GCE was 3.88 × 10? 9 mol cm? 2 which indicates the high enzyme loading. The electron transfer rate constant (ks) of GOx immobilized onto GCNT was 1.08 s? 1 which validates a rapid electron transfer processes. The composite film shows linear response towards 6.30 to 20.09 mM glucose. We observed a good sensitivity of 2.47 μA mM?1 cm? 2 for glucose at the composite film. The fabricated biosensor displayed two weeks stability. Moreover, it shows no response to 0.5 mM of ascorbic acid (AA), uric acid (UA), acetaminophen (AP), pyruvate (PA) and lactate (LA) which shows its potential application in the determination of glucose from human serum samples. The composite film exhibits excellent recovery for glucose in human serum at physiological pH with good practical applicability.  相似文献

A nanocomposite consisting of cadmium oxide decorated with carbon nanotubes (CdO.CNT NC) was prepared by a wet-chemical technique, and its optical, morphological, and structural properties were characterized by FTIR, UV/Vis, FESEM coupled to XEDS, XPS, and XRD methods. A flat glassy carbon electrode was modified with the nanocomposite to obtain a sensor for L-glutathione (GSH) which displays improved sensitivity, a large dynamic range and good long-term stability. The calibration plot (best acquired at a voltage of 0.5 V) is linear (r 2 = 0.99) in the 0.1 nM to 0.01 M GSH concentration range. The detection limit is as low as 30.0 pM, and the sensitivity is ~9.49 μA?μM?1?cm?2. To the best of our knowledge, this is the first report on the determination of GSH using such a modified glassy carbon electrode (GCE) in combination with I-V method. The GCE was applied to the selective determination of GSH in spiked rabbit serum samples and gave acceptable results.

We report on an electrochemical sensor for the sensitive amperometric determination of ascorbic acid (AA). Aniline containing suspended silicotungstic acid and carbon nanotubes was electropolymerized on the surface of a glassy carbon electrode in a single step which provides a simple and controllable method and greatly improves the electrocatalytic oxidation of AA. The effects of scan rate, solution pH and working potential were studied. A linear relationship exists between the current measured and the concentration of AA in the range from 1 μM to 10 μM and 0.01 mM to 9 mM, with a limit of detection as low as 0.51 μM (S/N?=?3). The sensor is selective, stable and satisfyingly reliable in real sample experiments. In our eyes, it has a large potential for practical applications.

Doped carbon nanotubes are now extremely attractive and important nanomaterials in bioanalytical applications due to their unique physicochemical properties. In this paper, the boron-doped carbon nanotubes (BCNTs) were used in amperometric biosensors. It has been found that the electrocatalytic activity of the BCNTs modified glassy carbon (GC) electrode toward the oxidation of hydrogen peroxide is much higher than that of the un-doped CNTs modified electrode due to the large amount of edge sites and oxygen-rich groups located at the defective sites induced by boron doping. Glucose oxidase (GOD) was selected as the model enzyme and immobilized on the BCNTs modified glassy carbon electrode by entrapping GOD into poly(o-aminophenol) film. The performance of the sensor was investigated by electrochemical methods. At an optimum potential of +0.60 V and pH 7.0, the biosensor exhibits good characteristics, such as high sensitivity (171.2 nA mM(-1)), low detection limit (3.6 microM), short response time (within 6s), satisfactory anti-interference ability and good stability. The apparent Michaelis-Menten constant (K(m)(app)) is 15.19 mM. The applicability to the whole blood analysis of the enzyme electrode was also evaluated.  相似文献

We describe a biosensor for phenolic compounds that is based on a glassy carbon electrode modified with tyrosinase immobilized on multiwalled carbon nanotubes (MWNTs). The MWNTs possess excellent inherent electrical conductivity which enhances the electron transfer rate and results in good electrochemical catalytic activity towards the reduction of benzoquinone produced by enzymatic reaction. The biosensor was characterized by cyclic voltammetry, and the experimental conditions were optimized. The cathodíc current is linearly related to the concentration of the phenols between 0.4???M and 10???M, and the detection limit is 0.2???M. The method was applied to the determination of phenol in water samples.

利用离子液体1-丁基-3-甲基咪唑四氟硼酸盐(BMIMBF4)对玻碳电极(GCE)进行修饰,制备了BMI-MBF4/GCE电极.在0.1mol/L的磷酸盐缓冲溶液中,采用循环伏安法研究了抗坏血酸在BMIMBF4/GCE电极和裸玻碳电极(GCE)上的电化学行为.结果表明,pH=5.7的磷酸盐缓冲溶液为最佳测定底液,最佳富集时间为120s;BMIMBF4/GCE对抗坏血酸的氧化反应有很好的电化学催化作用.抗坏血酸的氧化峰电流与其浓度在2.0×10-4~1.0×10-2 mol/L的范围内呈良好的线性关系,相对标准偏差为4.53%(n=5).  相似文献

We report on a glassy carbon electrode that was modified with a composite made from graphene oxide (GO) and multiwalled carbon nanotubes (MWCNT) that enables highly sensitive determination of L-tyrosine. The sensor was characterized by transmission electron microscopy and electrochemical impedance spectroscopy, and its electrochemical properties by cyclic voltammetry, chronocoulometry and differential pulse voltammetry. The GO/MWCNT hybrid exhibits strong catalytic activity toward the oxidation of L-tyrosine, with a well defined oxidation peak at 761 mV. The respective current serves as the analytical information and is proportional to the L-tyrosine concentration in two ranges of different slope (0.05 to 1.0 μM and 1.0 to 650.0 μM), with limits of detection and quantification as low as 4.4 nM and 14.7 nM, respectively. The method was successfully applied to the analysis of L-tyrosine in human body fluids. The excellent reproducibility, stability, sensitivity and selectivity are believed to be due to the combination of the electrocatalytic properties of both GO and MWCNT. They are making this hybrid electrode a potentially useful electrochemical sensing platform for bioanalysis.

A new approach was attempted to prepare a chemically modified electrode using Cobalt hexacyanoferrate (CoHCF) as the redox mediator and to study its stability and electrocatalytic activity for ascorbic acid (AA) oxidation. The basic principle underlying the electrode modification is the coordination of cobalt ion with the amino nitrogen of aniline adsorbed on the surface of a graphite rod. This surface was subsequently derivatized with ferrocyanide to get CoHCF film on the electrode surface. The CoHCF modified electrode as prepared above was characterized using cyclic voltammetry. The effect of scan rate, supporting electrolyte and pH of the medium on the performance of the modified electrode was investigated. The CoHCF modified electrode exhibited good electrocatalytic activity towards the oxidation of ascorbic acid and gave a linear response from 5.52 x 10(-5) M to 3.23 x 10(-2) M with a correlation coefficient of 0.9929. The detection limit was found to be 3.33 x 10(-5) M. Hydrodynamic voltammetry and chronoamperometry studies for the oxidation of ascorbic acid were also carried out. The electrode was highly stable and exhibited good reproducibility. This modified electrode was also applied for the determination of ascorbic acid in commercial samples.  相似文献

利用水热法在室温条件下合成了一种三维花状氧化锌纳米材料,并将其应用于抗坏血酸(AA)的电催化性能研究。通过滴涂法构筑了三维花状氧化锌修饰的玻碳电极(ZnO/GCE),并用扫描电子显微镜(SEM)对其形貌进行了表征。采用循环伏安法、线性扫描伏安法及差示脉冲伏安法分别考察了AA在该ZnO/GCE上的电化学行为。结果显示,与裸电极相比,三维花状氧化锌纳米结构显著增大了抗坏血酸的电化学响应信号,且在8. 4~171μmol/L的浓度范围内,氧化峰电流与浓度呈现良好的线性关系,(R^2=0. 9987),检出限为0. 67μmol/L(S/N=3)。将该传感器用于维生素C样品中AA含量分析,展现出理想的效果。  相似文献

A nanocomposite consisting of polyaniline and multiwalled carbon nanotubes was tethered with a thiolated thrombin-specific aptamer and placed on a glassy carbon electrode (GCE) to obtain a biosensor for thrombin that has a limit of detection of 80 fM. Tethering was accomplished via a thiol-ene reaction between thiolated thrombin aptamer (TTA) and oxidized polyaniline (PANI) that was chemically synthesized in the presence of solution-dispersed multiwalled carbon nanotubes (MWCNTs). The modified GCE exhibits a pair of well-defined redox peaks (at 50/?25 mV) of self-doped PANI in neutral solution, and the tethered TTA-thrombin interaction gives a decreased electrochemical signal. Cyclic voltammetry, scanning electron microscopy and ultraviolet visible spectroscopy were used to characterize the film properties. This amperometric aptasensor is sensitive, selective and reproducible. It was applied to the determination of thrombin in spiked human serum (0.2 to 4 nM) and gave recoveries that ranged from 95 to 102%.

A novel amperometric sensor for the determination of sulfite was fabricated based on multiwalled carbon nanotubes (MWCNTs)/ferrocene-branched chitosan (CHIT-Fc) composites-covered glassy carbon electrode (GCE). The electrochemical behavior of the sensor was investigated in detail by cyclic voltammetry. The apparent surface electron transfer rate constant (Ks) and charge transfer coefficient (α) of the CHIT-Fc/MWCNTs/GCE were also determined by cyclic voltammetry, which were about 1.93 cm s−1 and 0.42, respectively. The sensor displayed good electrocatalytic activity towards the oxidation of sulfite. The peak potential for the oxidation of sulfite was lowered by at least 330 mV compared with that obtained at CHIT/MWCNTs/GCE. In optimal conditions, linear range spans the concentration of sulfite from 5 μM to 1.5 mM and the detection limit was 2.8 μM at a signal-to-noise ratio of 3. The proposed method was used for the determination of sulfite in boiler water. In addition, the sensor has good stability and reproducibility.  相似文献

We have prepared a nanocomposite consisting of single-walled carbon nanotubes and polylysine. It was characterized by transmission electron microscopy, X-ray photoelectron spectroscopy, and by UV/vis and FTIR spectroscopy. Tyrosinase was covalently immobilized on the nanocomposite, and the resulting bioconjugate deposited on a glassy carbon electrode to form a biosensor for bisphenol A. The biosensor was characterized by scanning electron microscopy and electrochemical impedance spectroscopy. Under optimized experimental conditions, the biosensor gives a linear response to bisphenol A in the 4.00 nM to 11.5 μM concentration range. Its sensitivity is 788 mA M?1 cm?2, and the lower detection limit is 0.97 nM (at an S/N of 3). The biosensor shows good repeatability, reproducibility and long-term stability. In a preliminary practical application, it was successfully applied to the determination of bisphenol A in leachates of plastic spoons.

A novel nitrite sensor was fabricated based on a graphene/polypyrrole/chitosan nanocomposite film modified glassy carbon electrode. The nanocomposite film was characterized by scanning electron microscopy, Fourier transform infrared spectroscopy and Raman spectroscopy. The electron transfer behaviour of the modified electrodes was investigated in [Fe(CN)(6)](3-)/(4-) redox probe using cyclic voltammetry and electrochemical impedance spectroscopy. Differential pulse voltammetry and amperometry were used to study the electrochemical properties of the proposed sensor. Under optimum conditions, the sensor exhibited good reproducibility and stability for nitrite determination. Linear response was obtained in the range of 0.5-722 μM with a detection limit of 0.1 μM (S/N = 3) for nitrite determination.  相似文献

We report on a novel hydrogen peroxide biosensor that was fabricated by the layer-by-layer deposition method. Thionine was first deposited on a glassy carbon electrode by two-step electropolymerization to form a positively charged surface. The negatively charged gold nanoparticles and positively charged horseradish peroxidase were then immobilized onto the electrode via electrostatic adsorption. The sequential deposition process was characterized using electrochemical impedance spectroscopy by monitoring the impedance change of the electrode surface during the construction process. The electrochemical behaviour of the modified electrode and its response to hydrogen peroxide were studied by cyclic voltammetry. The effects of the experimental variables on the amperometric determination of H2O2 such as solution pH and applied potential were investigated for optimum analytical performance. Under the optimized conditions, the biosensor exhibited linear response to H2O2 in the concentration ranges from 0.20 to 1.6?mM and 1.6 to 4.0?mM, with a detection limit of 0.067?mM (at an S/N of 3). In addition, the stability and reproducibility of this biosensor was also evaluated and gave satisfactory results.
