Heterocyclic ketene aminals 1 or 2 reacted with propionyl chloride to give both the mono-N-and C-acylated products 4 and 5 or 6 and 7. While N-methyl heterocyclic ketene aminals 3 reacted with propionyl chloride under the same condition, the reaction took place in complexity. Besides N-and C-acylation, the O-attack and heterocyclic ring cleavage also occurred to give products 8 and 9.  相似文献

A method is provided for the synthesis of the previously unknown spiro heterocyclic ketene aminals, 3,9-bis(substituted-methylene)-2,4,8,10-tetraazaspiro[5.5]undecane 3 in moderate to excellent yields by cyclocondensation reaction of ketene dithioacetals 1 with tetrakis(aminomethyl)methane 2.  相似文献

A scheme for the preparation of novel synthetic reagents. pyrazolyl(ethoxycarbonyl)ketene aminals, from acetyl(ethoxycarbonyl)ketene (N-benzoyl)aminalvia its difluoroboron chelate is suggested. Active methylene β-oxo sulfones add to the C≡N bond of benzoylcyanamide in the presence of catalytic amounts of Ni(acac)2. Debenzoylation of the reaction products under the action of MeONa in MeOH gives N,N′-unsubstituted diaminomethylidene derivatives of β-oxo sulfones (acyl(R-sulfonyl)ketene aminals), which can be used as reagents for heterocyclic synthesis and as chelating ligands. The syntheses of 2-amino-3-arylsulfonylpyridin-4(1H)-ones and 5-sulfonylcytosine derivatives are presented as examples.  相似文献

Abstract

The reaction of 3-oxopropanenitriles with phenyl isothiocyanate in DMF containing KOH afforded the corresponding potassium salts. The latter salts were converted into ketene N,S-acetals upon acidification with hydrogen chloride. The reaction of the ketene N,S-acetals with 2-bromo-1-[5-methyl-1-(p-tolyl)-1H-1,2,3-triazol-4-yl]ethan-1-one or 3-(2-bromoacetyl)-2H-chromen-2-one gave novel thiophenes in good yields. Treatment of the ketene N,S-acetals with hydrazonyl halides afforded 1,3,4-thiadiazoles in good yields. The stereochemistry of the synthesized compounds was studied.  相似文献

Abstract

Two novel methods for synthesis of nicotinic acid derivatives with unsymmetrical substitution patterns were presented via ketene dithioacetals. The ketene N,S-acetals 2 reacted with β-ketoesters or β-cyanoesters to give 4-amino-5-cyano-2-alkyl-6-methylthio-nicotinic acid derivatives 3 or 2,4-diamino-5-cyano-6-methylthio-nicotinic acid ethyl ester 4. However, 6-amino-5-cyano-2-alkyl-4-methylthio-nicotinic acid esters 6 were obtained by the reaction of the ketene dithioacetals 1 and β-amino-crotonates.  相似文献

A concise and efficient route for the synthesis of pyrimido[1,6-a]pyrimidines and imidazo[1,2-c]pyrimidines by simply refluxing a reaction mixture of different heterocyclic ketene aminals and N,N′-bis(arylmethylidene)arylmethane was developed. This protocol provides an alternative method for application in combinatorial and parallel synthesis in drug discovery.  相似文献

Nenitzescu reaction of acetyl ketene aminals (N,N-acetals) was investigated. The interaction of 2-acetyl-1-amino-1-anilinoethene with benzoquinone gave 3-acetyl-2-amino-7a-hydroxy-1-phenyl-5,7a-dihydro-1H-indol-5-one, which was then transformed into 3-acetyl-2-amino-6-chloro-5-hydroxy-1-phenylindole. The reaction of benzoquinone with 2-acetyl-1-amino-1-benzoylaminoethene led to the corresponding hydroquinone-adduct which was oxidized to 4-acetylamino-5-(2,5-dihydroxyphenyl)-2-phenyloxazole. A new representative of hetarylketene aminals, ethyl 3,3-diamino-2-(2-p-tolylpyrimidin-4-yl)acrylate, was synthesized by the reaction ofp-toluamidine with the condensation product of dimethylformamide dimethyl acetal and the difluoroboron chelate of acetyl(ethoxycarbonyl)ketene (N-benzoyl)aminal. A concise approach for the construction of ketene aminals and α-chlorohydrazones has been developed. It involves reactions of the regiodefined gem-dihalo nitrovinyl compound of in situ generated imide chlorides in different media with primary arylamines being dependent on the aryl groups. A range of ketene aminals and α-chlorohydrazones are obtained in good to high yields. In addition, α-aminohydrazones are prepared by using α-chlorohydrazones as the precursors.  相似文献

An efficient one‐pot four‐component protocol for the synthesis of imidazo[1,2‐a]pyridines was developed by condensing ethane‐1,2‐diamine ( 2 ), 1,1‐bis(methylthio)‐2‐nitroethene ( 1 ), aldehydes 3 , and activated methylene compounds in EtOH under reflux conditions (Tables 1–3). The features of this procedure are operational simplicity, good yields of products, in situ preparation of heterocyclic ketene aminals (HKA), and catalyst‐free conditions.  相似文献

The addition reactions of acetylacetone and ethyl acetoacetate with diphenylcarbodiimide and dicyclohexylcarbodiimide in the presence of nickel acetylacetonate afforded N,N"-substituted ,-dioxoketene aminals. Adducts of acetylacetone with carbodiimides are readily deacetylated with MeONa in MeOH to form acetylketene aminals. N,N'-Disubstituted ketene aminals are bioisosteres of thioureas and are useful building blocks in many synthetic operations. A convenient one-pot synthesis of N,N'-disubstituted ketene aminals from activated methylene compounds and isothiocyanates is described. Most of these aminals exist in rotameric equilibrium around the central C=C bonds in solution, and the rotamers are stabilized by intramolecular hydrogen bonding both in solution and in solid states.  相似文献

Treatment of N-(2,2,2-trichloro-1-tosylethyl)dichloroacetamide with excess dimethylamine, piperidine, or morpholine gave substituted aminals of the oxazole series, whose facile acid hydrolysis provided 5-(dimethylamino)-4-tosyl-1,3-oxazol-2-carbaldehyde and its analogs having the piperidino and morpholino group in the 5 position of the oxazole ring. A new strategy for the synthesis of tetrahydroimidazo[1,2-a]pyridines and unusual tetrahydrobenzo[b]imidazo[1,2,3-ij][1,8]naphthyridines has been successfully developed by cascade reactions including Knoevenagel condensation, aza-ene reaction, imine-enamine tautomerization, cyclocondensation/oxidation, and intramolecular SNAr of precursors 2-(2-chloroaroyl)methyleneimidazolidines as new heterocyclic ketene aminals (HKA), which represent a class of polyfunctional scaffolds with four active reaction sites with aromatic aldehydes and malononitrile or ethyl 2-cyanoacetate under mild conditions. In this domino reaction, nine different active sites are involved, and two C-C bonds, two C-N bonds, and two new rings are constructed with all reactants efficiently utilized in the chemical transformation.  相似文献

1-Dialkylaminomethylpyrroles are shown to behave in many respects like aminals. Acylation by an acid chloride, for instance, occurs normally at the amine-type N-atom rather than at the pyrrole ring. Spontaneous cleavage of the resulting quaternary acylammonium salts affords the 5-azoniafulvene ion ( 3 ). This higly reactive iminium ion, and its benzo-annellated analogue ( 4 ) can be trapped by electron rich aromatic compounds such as N-methylpyrrole or N,N-dimethylaniline. More elaborate N-Mannich bases are accessible by addition of indoles to enamines.  相似文献

A facile synthesis of tetracyclo-isocoumarins based on the AcOH-catalyzed cyclocondensation and rearrangement reaction between heterocyclic ketene aminals and 2,2-dihydroxy-2H-indene-1,3-dione is described. This method provides direct access to tetacyclo-isocoumarins, a class of compounds with potential broad spectrum biological activities.  相似文献
