A short and efficient synthesis of 1-(3-tert-butyl-1-phenyl-1H-pyrazol-5-yl)-3-(5-(2-morpholinoethoxy)-2H-chromen-8-yl) urea derivatives (1a–c), a novel type of p38 MAPK inhibitors, is described. The Claisen thermal rearrangement of arylpropargyl ethers was employd as a key step to synthesize the chromene core. The solvent effect on the ratio of the resultant two isomers of Claisen thermal rearrangement, namely 2-methylbenzofuran and 2H-chromen, was also investigated. 相似文献

以金刚烷甲酰氯为起始原料,经亲核取代、吲哚环合及酰化反应制得中间体2-(5-氯-2-金刚烷-1H-吲哚-3-基)-2-氧代乙酰(4); 4与取代胺反应合成了14个新的N-取代2-(5-氯-2-(金刚基-1-基)-1H-吲哚-3-基)-2-氧代乙酰胺衍生物(5a~5n),其结构经1H NMR, 13C NMR和HR-MS(ESI)表征。采用MTT法研究了化合物对人子宫颈癌细胞(Hela)、乳腺癌细胞(MCF-7)和人肝癌细胞(HepG-2)的体外抗肿瘤活性。结果显示:化合物2-(5-氯-2-金刚烷-1H-吲哚-3-基)-N-(3-氯-4-氟苯基)-2-氧代乙酰胺(5e)的体外抑制活性最优,IC50分别为14.10、 10.56和8.55 μmol·L-1。 相似文献

以苯并咪唑-2-甲酸、水合肼、取代苯基异硫氰酸酯和取代苯甲酸等为原料,经多步反应合成了16种2-(苯并咪唑-2-基)-5-取代苯基-1,3,4-噁二唑和2-(苯并咪唑-2-基)-5-取代苯胺基-1,3,4-噁二唑新化合物,并考察了微波辐射对反应的影响.产物结构利用IR,1H NMR,13C NMR和元素分析确证.用生长速率法测试了目标化合物对番茄灰霉病菌和小麦菌核病菌的离体抑制活性,结果表明3种化合物对番茄灰霉病菌有很高的抑制活性,其EC50分别为2.55,6.34和5.12μg/mL,活性高于对照药剂多菌灵(EC50=7.40μg/mL). 相似文献

为寻求高效低毒的农药,设计、合成了一系列新的1-[2-(2,4-二氯苯氧)乙酰基]-5-氨基-1H-吡唑衍生物3a~3j和4a~4d.元素分析,1H NMR,IR和MS证实了它们的结构.初步的生物活性测试结果表明,在10 mg/L浓度下,化合物3a~3j和4a~4d对油菜的根茎均有很好的抑制作用,对油菜的根的抑制率为90.0%~98.7%;于100 mg/L浓度下,对稗草的根抑制率为84.0%~100%. 相似文献

A novel neonicotinoid analogue(C13H19N5O5,3a)had been synthesized,the structure was characterized by elemental analysis,IR and 1H NMR spectra,and the S-(+)-(E)-configuration was confirmed by single-crystal X-ray diffraction.The crystal belongs to monoclinic,space group P21 with a = 8.7076(17),b = 8.3211(17),c = 10.642(2),β = 92.370(3)o,V = 770.4(3)3,Z = 2,Dc = 1.402 g/cm3,μ = 0.110 mm-1,Mr = 325.33,F(000)= 344,S = 1.027,R = 0.0543 and wR = 0.1229 for 3601 unique reflections with 2919 observed ones(I > 2σ(I)).Compound 3a is stabilized by intramolecular hydrogen bonds and intermolecular force.In addition,the structure of compound 3a was optimized by the B3LYP/6-31G(d,p).DFT/B3LYP optimizations were performed based on X-ray geometries applying 6-31G(d,p)basis set.The optimized structure of compound 3a by the B3LYP/6-31G(d,p)method is more bent than in the crystal.IR spectrum of the solid compound is consistent with the X-ray structure.The HOMO-LUMO gap in 3a(5.3 eV)indicates high kinetic stabilities of compound 3a.The preliminary bioassay test showed that 3a exhibited good activities against Nilaparvata legen,Pseudaletia separate Walker and Aphis medicagini at 500 mg/L. 相似文献

Several novel 1-(substituted phenyl)-2-phospholene 1-oxide derivatives, which are analogs of sugars having a phosphorus atom in place of the ring oxygen of normal sugars, were synthesized from the corresponding 2-phospholenes as the starting materials. A number of new1-((aryl)(2-oxoindolin-3-yl)methyl)urea and 1-((aryl)(2-oxoindolin-3-yl)methyl)thiourea derivatives have been prepared by simply combining 2-indolinone, aromatic aldehyde, and urea or thiourea in the presence of p-TsOH in EtOH. This new methodology affords the title compounds in good yields and without the use of chromatography.

The hydrogenation of 2′, 3′-O-isopropylidene-5-methyluridine (1) in water over 5% Rh/Al2O3 gave (5 R)- and (5 S)-5-methyl-5, 6-dihydrouridine (2) , separated as 5′-O-(p-tolylsulfonyl)- (3) and 5′-O-benzoyl- (5) derivatives by preparative TLC. on silica gel and ether/hexane developments. The diastereoisomeric differentiation at the C(5) chiral centre depends upon the reaction media and the nature of the protecting group attached to the ribosyl moiety. The synthesis of iodo derivatives (5 R)- and (5 S)- 4 is also described. There is a serious need for compounds that are effective in the treatment of cancer and virus (HIV. Human Immunodeficiency Virus,etc.). Among the large number of nucleoside analogs which have been synthesized and evaluated(1-2) recently several 5'-alkylthio nucleosides have been reported to exhibit good antitumor and antiviral activities(3-4). Their anticancer mechanism involves blocking cancer cell proliferation and inhibiting the cytoplasmic isozyme(C-TK)(5), one isozyme of thymine kinase, which predominates in human tumor cell lines(6). As we know, many acylic nucleoside analogs have appeared good biological activities(7).
