A Useful Synthesis of 2-Methyl-2,3-dihydrofuro [2,3-b] Quinolines

A convenient, general synthesis of 2-methyl-2,3-dihydrofuro [2,3-b] quinolines is described from o-nitrobenzaldehydes. The benzaldehydes (la-c) on reaction with phosphorane 2 provide (E)-ethyl-α-allylcinnamates (3a-c) in high yields. These esters 3a-c on reduction followed by acid catalysed cyclisation give 2,3-dihydrofuro [2,3-b] quinolines (5a-c).     

Synthesis of 2-Methyl-9-oxo-9H-furo[2,3-c]benzopyrans and 2-Methyl-3H,4H[1]benzopyrano[3,4-b]pyrrol-4-ones

3:4-Fused furocoumarins and pyrrolocoumarins are synthesised from 3-hydroxy and 3-benzamido-substituted coumarins by a novel two step sequence.     

Synthesis of 2-Methyl-3-acetyl-5-oxo-5H-[1]benzopyrano[2,3-b]pyridine

For our synthetic work we needed the title compound. Literature survey revealed that 2-amino-4-oxo-4H-[1] benzopyran-3-carboxaldehyde (2-amino-3-formylchro-mone) can undergo condensation with diethyl malonate (or ethyl cyanoacetate) forming ethyl 2-hydroxy(or amino)-5-oxo-5H-[1]benzopyrano[2,3-b]pyridine-3-carboxylate¹.     

Synthesis of 4-(2-chloroethyl)-2,3-dihydro[1,4]oxazino[2,3-b]quinoline and 4-(2-chloroethyl)2,3-dihydropyrido[2,3-b][1,4]oxazine

The title compounds were synthesized from 3-[bis(2-hydroxyethyl)amino]quinolin-2(1H)-one 11a and 3-[bis(2-hydroxyethyl)amino]pyridin-2(1H)-one 18 respectively. The preparation involved a tandem chlorination/cyclization reaction.     

Synthesis and transformations of 3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridines

Reactions of 2,5-dimethoxytetrahydrofuran with 3-aminothieno[2,3-b]pyridines afford a number of substituted 3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridines. The possibility of the reaction and the yield of the product are determined by the character of a substituent in position 2 of thieno[2,3-b]pyridine. The Curtius rearrangement of 2-acylazido-3(1H-pyrrol-1-yl)thieno[2,3-b]pyridines yields 4,5-dihydropyrido[3",2":4,5]thieno[2,3-e]pyrrolo[1,2-a]pyrazin-4-ones. The molecular and crystal structures of ethyl 4-methoxymethyl-6-methyl-3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridine-2-carboxylate were determined by X-ray diffraction analysis.     

A Facile Synthesis of 2-Benzoyl-6-Hydroxy-3-Methyl-5-(2-Substituted-2,3-Dihydro-1H-1,5-Benzodiazepin-4-YL) Benzo[b]Furans

The synthesis and characterisation of some new 1,5-benzodiazepines prepared from 4,6-diacetylresorcinol is reported.

以1H-吡咯[2,3-b]吡啶-2-甲酸乙酯为起始原料,经5步反应合成了新化合物3-(2-羧基乙基)-1H-吡咯[2,3-b]吡啶-2-甲酸,其结构经1H NMR和HR-MS表征。合成中间(E)-1-Ts-3-(3-乙氧基-3-氧代丙基-1-烯基)-1H-吡咯[2,3-b]吡啶-2-羧酸乙酯的最佳反应(Heck)条件为:无水DMF为溶剂,Pd(OAc)2为催化剂,dppf为配体,三乙胺为碱,收率88%。     

Facile Synthesis of 2-Alkylthio-5,6,7,8-tetrahydrobenzothieno[2,3-d]pyrimidin-4(3H)-ones

Isothiocyanate 2, obtained from aza-Wittig reaction of iminophosphorane 1 with CS₂, reacted with amine to give 2-thioxo-2,3,5,6,7,8-hexahydrobenzothieno[2,3-d]pyrimidin-4(1H)-ones 4 in the presence of sodium ethoxide. S-Alkylation of 4 produced 2-alkylthio-5,6,7,8-tetrahydrobenzothieno[2,3-d]pyrimidin-4(3H)-ones 5 in good yields.     

Synthesis of New Benzopyrano[2,3-c]pyrazoles and 3-triazolonyliminocoumarins From 3-cyano Iminocoumarin Derivatives

Synthesis of benzopyrano[2,3-c]pyrazoles and 3-triazolonyliminocoumarins was successfully performed using heterocyclization of 3-cyano iminocoumarin or their N-ethoxycarbonylated derivatives with semicarbazide reagents. Elemental analysis, infrared, and ¹H NMR spectral data confirmed the molecular structure of the newly synthesized compounds.     

Synthesis of (2S,3S)-[3-(2)H1]-4-methyleneglutamic acid and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid

(2S,3S)-[3-(2)H1]-4-Methyleneglutamic acid 1a and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid 1b have been synthesised for use in biosynthetic and metabolic studies.     

Furopyridines. XV . Synthesis and properties of ethyl 2-(3-furo[2,3-b]-, -[3,2-b]-, -[2,3-c]- and -[3,2-c]pyridyl)acetate

Ethyl 2-(3-furopyridyl)acetates 10a-d were synthesized from furopyridin-3(2H)-ones 4a-d by the Wittig-Horner reaction with diethyl cyanomethylphosphonate, hydrolysis and the subsequent esterification. Reaction of compounds 10a-d with lithium diisopropylamide (IDA) gave the corresponding methylene-lithiated intermediate, and the subsequent reaction with benzaldehyde, acetone and iodomethane afforded the methylene-alkylated product respectively, while N,N-dimethylacetamide did not give any reaction product. The 2-position of 10a, b and d is alkylated by the lithiation with excess of LDA and the successive reaction with an electrophile.     

2-[3-Phenyl-5-(m-Chlorophenyl)-2-Pyrazolin-1-yl]-3-Methyl-4(3H)-Quinazolinone

There are two molecules in the asymmetric unit (I and II). The structure consists of a 2-pyrazoline ring and three aromatic rings two of which are free and the third one is condensed with a 3-methyl-4(3H)-pyrimidine ring (4(3H)-Quinazolinone). While the pyrazoline ring is in the distorted envelope conformation, the free aromatic rings are planar and the pyrimidine ring deviates from the planarity. The crystal structure is stabilized by C–HO inter and intramolecular bonds.     

Studies on the Synthesis of New 3-(3,5-Diamino-1-substituted-pyrazol-4-yl)azo-thieno[2,3-b]pyridines and 3-(2-Amino-5,7-disubstituted-pyrazolo[1,5-a]pyrimidine-3-yl)azothieno[2,3-b]pyridines

Coupling the diazonium salt of 3-amino-2-cyano-4,6-dimethylthieno[2,3-b]pyridine 1 with malononitrile 2 gave 2-cyano-3-(hydrazonomalononitrile)-4,6-dimethylthieno[2,3-b]pyridine 3 which then reacted with hydrazine compounds 4a-4h to yield corresponding 2-cyano-3-(3,5-diamino-1-substituted-pyrazol-4-yl)azo-4,6-dimethylthieno[2,3-b]pyridines 5a-5h. The 2-cyano-3-(2-amino-5,7-disubstituted-pyrazolo-[1,5-a]pyrimidine-3-yl)azo-4,6-dimethylthieno[2,3-b]pyridines 7a-7f were obtained in good yield by the cyclocondensation reaction of 2-cyano-3-(3,5-diamino-pyrazol-4-yl)azo-4,6-dimethylthieno[2,3-b]pyridine 5a with the appropriate 1,3-diketones 6a-6f under acidic condition.     

Derivatives of 5,6-Dimethoxybenzo[b]thiophen-2-(3H)one. Synthesis of 5,6-Dimethoxybenzo[b]thieno[2,3-b]quinoline

Substituted-3-benzylidenebenzo[b]thiophen-2-ones have been synthesized. 5,6-Dimethoxybenzo[b]-thiophen-2-(3H)one with aniline and triethyl orthoformate gives the 3-anilinomethylene compound, which can be hydrolysed to give the corresponding 3-hydroxymethylene derivative. Dimethoxybenzo[b]thieno[2,3-b]-quinoline has been synthesized.
