Methyl (2E,4R)‐4‐hydroxydec‐2‐enoate, methyl (2E,4S)‐4‐hydroxydec‐2‐enoate, and ethyl (±)‐(2E)‐4‐hydroxy[4‐2H]dec‐2‐enoate were chemically synthesized and incubated in the yeast Saccharomyces cerevisiae. Initial C‐chain elongation of these substrates to C12 and, to a lesser extent, C14 fatty acids was observed, followed by γ‐decanolactone formation. Metabolic conversion of methyl (2E,4R)‐4‐hydroxydec‐2‐enoate and methyl (2E,4S)‐4‐hydroxydec‐2‐enoate both led to (4R)‐γ‐decanolactone with >99% ee and 80% ee, respectively. Biotransformation of ethyl (±)‐(2E)‐4‐hydroxy(4‐2H)dec‐2‐enoate yielded (4R)‐γ‐[2H]decanolactone with 61% of the 2H label maintained and in 90% ee indicating a stereoinversion pathway. Electron‐impact mass spectrometry analysis (Fig. 4) of 4‐hydroxydecanoic acid indicated a partial C(4)→C(2) 2H shift. The formation of erythro‐3,4‐dihydroxydecanoic acid and erythro‐3‐hydroxy‐γ‐decanolactone from methyl (2E,4S)‐4‐hydroxydec‐2‐enoate supports a net inversion to (4R)‐γ‐decanolactone via 4‐oxodecanoic acid. As postulated in a previous work, (2E,4S)‐4‐hydroxydec‐2‐enoic acid was shown to be a key intermediate during (4R)‐γ‐decanolactone formation via degradation of (3S,4S)‐dihydroxy fatty acids and precursors by Saccharomyces cerevisiae.  相似文献

Acylation of 2H-thiopyran-3,5(4H,6H)-dione and 2-methyl-2H-thiopyran-3,5(4H,6H)-dione with acetyl chloride or propionyl chloride afforded the corresponding 4-acyl-2H-thiopyran-3,5(4H,6H)-diones. Oxidation of 4-acetyl-2H-thiopyran-3,5(4H,6H)-dione with m-chloroperoxybenzoic acid gave 4-acetyl-2H-thiopyran-3,5(4H,6H)-dione 1-oxide. 4-Acyl-2H-thiopyran-3,5(4H,6H)-diones and 4-acetyl-2H-thiopyran-3,5(4H,6H)-dione 1-oxide reacted with pyrrolidine, allylamine, and p-anisidine, resulting in formation of the corresponding 4-aminomethylene derivatives.  相似文献

(+)- -threo-4-Fluoroglutamic acid [(+)-(2S, 4S)-fluoroglutamic acid] has been synthesizedstarting with the natural (−)-4-trans-hydroxy- -proline. Its acetylation at nitrogen followedby esterification with diazomethane afforded methyl 1-acetyl-trans-4-hydroxy- -prolinatewhich was converted to methyl 1-acetyl-cis-4-fluoro- -prolinate by means of diethylaminosulfurtrifluoride (DAST) or 2-chloro-1,1,2-trifluorotriethylamine. The mixture wasoxidized by ruthenium tetroxide to methyl 1-acetyl-cis-4-fluoro- -pyrrolidin-5-one-2-carboxylate,whose acid hydrolysis yielded the title compound. A similar sequence of reactionsconverted cis-4-hydroxy- -proline to (−)- -erythro-4-fluoroglutamic acid [(−)(2R, 4S)-fluoroglutamic acid]. (−)- -threo-4-Fluoroglutamic acid [(−)-(2R, 4R)-floroglutamicacid] was prepared analogously from trans-4-hydroxy- -proline, obtained from its diastereomerby inversion of configuration at carbon 4 of the pyrrolidine ring using thediethyl azodicarboxylate-triphenylphosphine procedure. cis-4-Hydroxy- -proline, necessaryfor the synthesis of (+)- -erythro-4-fluoroglutamic acid [(+)-(2S, 4R)-fluoroglutamicacid], was prepared from trans-4-hydroxy- -proline by benzyloxycarbonylation at thenitrogen, oxidation of the 1-benzyloxycarbonyl-trans-4-hydroxy- -proline to 1-benzyloxy-carbonyl-4-oxo- -proline, its reduction to 1-benzyloxycarbonyl-cis-4-hydroxy- -proline anddeprotection of the latter at the nitrogen. (−)-cis-4-Fluoro- -proline and (+)-trans-4-fluoro- -proline were isolated after the hydrolysis of incompletely oxidized methyl 1-acetyl-cis-4-fluoro- -prolinate and methyl 1-acetyl-trans-4-fluoro- -prolinate, respectively.  相似文献

Nucleophilic substitution of the halogen atom in dimethyl (S)-4-bromoglutamate followed by removal of the protecting groups and closure of a lactam ring afforded (2S,4S)-4-(indolin-1-yl)-5-oxoproline. The indoline fragment was oxidized into the indole fragment to give (2S,4S)-4-(indol-1-yl)-5-oxoproline; reduction of the carbonyl groups with BH3 yielded (2S,4S)-4-(indol-1-yl)prolines and (2S,4S)-2-hydroxymethyl-4-(indol-1-yl)pyrrolidines. Reduction of (2S,4S)-4-arylamino-5-oxoprolines with BH3 to the corresponding (2S,4S)-4-arylaminoprolines and (2S,4S)-4-arylamino-2-hydroxymethylpyrrolidines was studied.  相似文献

By treating 2e-methyl-4-oxo-trans-decahydroquinoline with lithium acetylide a mixture of stereoisomeric 4-cis-hydroxy-2e-methyl- and 4-trans-hydroxy-2e-methyl-4-ethynyl-trans-decahydroquinolines was obtained in 3:2 ratio. Their reaction with acetonitrile in the presence of sulfuric acid (Ritter's reaction) results in a mixture of stereoisomeric 4-cis-acetylamino-2e-methyl- and 4-trans-acetylamino-2e-methyl-4-ethynyl-trans-decahydroquinolines in the same ratio. The ethynyl group of alcohols synthesized is not hydrated under conditions of Kuchrerov's reaction. The boiling of the alcohols with formic acid furnished a mixture of 4-acetyl-2e-methyl and 2e-methyl-4-ethynyl-1,2,3,6,7,8,9,10-octahydroquinolines in 1:7 ratio. The former of these compounds under conditions of Ritter's reaction yielded a mixture (1:1.4) of stereoisomeric 4-acetyl-4-trans-acetylamino- and 4-acetyl-4-cis-acetylamino-2e-methyl-trans-decahydroquinolines. From 2e-methyl-4-ethynyl-1,2,3,6,7,8,9,10-octahydroquinoline under the same conditions were obtained both already mentioned stereoisomeric 4-acetylamino-2e-methyl-4-ethynyl-decahydroquinolines (53% of cis-isomer and 35% of trans-isomer in the mixture) and 4-acetyl-4-acetylamino-2e-methyldecahydroquinolines (7% of cis-isomer and 5% of trans-isomer).  相似文献

A series of novel 2‐(aryl)‐3‐[5‐(2‐oxo‐2H‐3‐chromenyl)‐1,3‐oxazol‐2‐yl]‐1,3‐thiazolan‐4‐ones 4a , 4b , 4c , 4e , 4f , 4g , 4h , 4i , 4j have been synthesized and assayed for their antibacterial activity against Gram‐positive bacteria viz. Bacillus subtilis (ATCC 6633), Staphylococcus aureus (ATCC 6538p), Micrococcus luteus (IFC 12708), and Gram‐negative bacteria viz. Proteus vulgaris (ATCC 3851), Salmonella typhimurium (ATCC 14028), Escherichia coli (ATCC 25922), and also antifungal activity against Candida albicans (ATCC 10231), Aspergillus fumigatus (HIC 6094), Trichophyton rubrum (IFO 9185), and Trichophyton mentagrophytes (IFO 40996). Among the screened compounds, 4d , 4e , 4f , 4g , and 4j exhibited potent inhibitory activity compared with the standard drug at the tested concentrations. The results reveal that, the presence of difluorophenyl in 4f and pipernyl ring in 4j at 2‐position of thiazolidine‐4‐one ring show significant inhibitory activity. The other compounds also showed appreciable activity against the test bacteria and fungi and emerged as potential molecules for further development. J. Heterocyclic Chem., 2011.  相似文献

Synthesis of N-Methyl- and N,N-Dimethylmerucathine and of N-Methyl- and N,N--Dimethylpseudomerucathine Starting from L -Alanine Starting form L -alanine, N-methylmerucathine (= (3R,4S)-4-(methylamino)1-phenyl-1-penten-3-ol; (3R,4S,)- 6 ), N,N-dimethylmerucathine (= (3R,4S)-4-(dimethylamino)-1-phenyl-1-penten-3-ol; (3R,4S)- 9 ), N-methylpseudomerucathine (= (3S,4S)-4-(methylamino)-1-phenyl-1-penten-3-01; (3S,4S)-6), and N,N-dimethylpseudomerucathine (= (3S,4S)-4-(dimethylamino)-1-phenyl-1-penten-3-ol; (3S,4S)- 9 ) were synthesized. A synthesis is reported for (4R,5R)-and (4S,5S)-4-hydroxymethyl-5-(4-nitrophenyl)oxazolidin-2-ones and (1′R,4R)-and (1′S,4S)-4-[hydroxy(4-nitrophenyl)methyl]oxazolidin-2-ones from (1R,2R)-and (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediols. Five new stilbene glucosides, gnemonosides F, G, H, I, and J were isolated from the stem lianas of Gnetum gnemonoides Brongn and Gnetum africanum Welw along with nine known stilbenoids. The structures of the new compounds were elucidated as gnetin E 4a,4b,4c‐Oβ‐triglucopyranoside ( 2 ), gnetin E 4a,4c‐Oβ‐diglucopyranoside ( 3 ), gnetin C 4a,4b,11a‐Oβ‐triglucopyranoside ( 4 ), gnetin D 4a,4b‐Oβ‐diglucopyranoside ( 5 ), and gnetuhainin A 4a,4b‐Oβ‐diglucopyranoside ( 6 ) on the basis of spectroscopic evidence.  相似文献

We report herein DABCO mediated one pot synthesis of 2-(3-benzyl-2, 6-dioxo-3,6-dihydropyrimidin-1[2H]-yl)-N-(4-(1,3-dioxo-1H-benzo[de]isoquinolin-2[3H]-yl) aryl) acetamides ( 4a-j ). The silent features of this new one pot synthesis include the shorter reaction time, high yields, simple workup, and simultaneous formation of N-Amide and N-benzyl bonds in the one pot. The newly synthesized compounds ( 4a-j ) were characterized by different spectral techniques such as IR, 1H-NMR, 13C-NMR, HRMS. All the synthesized compounds were evaluated for their anti-bacterial and anti-fungal activities. The anti-bacterial activities results reveal that the compounds 4a , 4g , 4i , and 4j are most active against S. aureus. In the case of B. subtilis the compounds 4a , 4i , and 4j are found to be most active. The compounds 4c , 4e , 4i , and 4j are most active against E. coli. In the case of P. aeruginosa 4a , 4i & 4j are found to be more active. On the other hand, the anti-fungal activity result shows that the compounds 4d , 4f , 4i , and 4j are more active against A. niger. The compounds 4a , 4d , 4i , and 4j are found to be more active against C. albicans.  相似文献

Reactions of 4-arylidene-3-methyl-5(4H)-isoxazolones with nucleophiles lead to 4-substituted-benzyl-3-methyl-5(2H)-isoxazolones. Formation of 5(4H)- and 5(2H)-isoxazolone derivatives have been observed when 4-substituted-arylidene-3-rriethyl-5(4H)-isoxazolones are reacted with methyl magnesium iodide. Oxidation of 4-substituted-benzyl-3-methyl-5(2H)-isoxazolones with manganic acetate and pyridinium chlorochromate, gives 4,4′-bis[4-substituted-benzyl-3-methyl-5(4H)-isoxazolone]. Oxidation of 4-substituted-benzyl-3-methyl-5(2H)-isoxazolones with metachloroperbenzoic acid gives 4-hydroxy-4-substituted-benzyl-3-methyl-5(4H)-isoxazolones and reactions of the same substrates with N-bromosuccinimide furnish 4-bromo-4-substitutedbenzyl-3-methyl-5(4H)isoxazolones.  相似文献

Summary N-Acylanthranilamides react with dibromotriphenylphosphorane in the presence of triethylamine as HBr captor to give 4-imino-4H-3,1-benzoxazines in good yields. If the reaction is carried out without acid acceptor, N-acetylanthranilamides yield 2-methyl-4-quinazolones, whereas N-benzoylanthranilamides give 2-phenyl-4-imino-4H-3,1-benzoxazines. It has also been found that 2-methyl-4-imino-4H-3,1-benzoxazines rearrange under the influence of HCl or HBr into the respective 2-methyl-4-quinazolones; 2-phenyl-4-imino-4H-3,1-benzoxazines, however, do not undergo such a rearrangement.

Zusammenfassung Die Umsetzung von N-Acyl-anthranilsäure-amiden mit Triphenyldibromphosphoran in Gegenwart von Triethylamin als HBr-Akzeptor führt mit guten Ausbeuten zu 4-Imino-4H-3,1-benzoxazinen. Wird die Reaktion ohne säurebindendes Mittel durchgeführt, dann entstehen aus N-Acetyl-anthranilsäure-amiden 2-Methylchinazolone-4, jedoch erhält man aus N-Benzoylanthranilsäure-amiden 2-Phenyl-4-imino-4H-3,1-benzoxazine. 2-Methyl-4-imino-4H-3,1-benzoxazine erleiden unter dem Einfluß von HBr oder HCl eine Umlagerung in entsprechende 2-Methylchinazolone-4, während 2-Phenyl-4-imino-4H-3,1-benzoxazine zu einer solchen Umlagerung nicht befähigt sind.

Reaction of 4-arylsulfonylimino-2,3,5,6,6-pentachloro-2-cyclohexen-l-ones with tosylhydrazine gives rise toN-arylsulfonyl-N-p-tolylsulfonyl-2,5,6,-tetrachloroaminophenols. With 4-arylsulfonylimino-2,2,3-trichloro-1,2,3,4-tetrahydronaphthalen-1-ones and 4-arylsulfonylimino-2-dihalo-1,2,3,4-tetrahydronaphthalen-1-ones reaction products are 4-arylsulfonylamido-2,3-dichloro-4-p-tolylsulfonylhydrazido-l,4-dihydronaphthalen-1-ones and 2-p-toIylsulfonyl-l,4-naphthoquinone-4-diazide respectively. The trans and cis form of 4-(4-methoxystyryl)pyridine N-oxide were studied. The spectral characteristics of cis-4-(4-methoxystyryl)pyridine N-oxide were determined in acetonitrile. The melting and thermal decomposition processes of the trans and cisforms of 4-(4-methoxystyryl)pyridine N-oxide were studied by thermochemical methods. It was establish that the thermal decomposition of 4-(4-methoxystyryl)pyridine N-oxide begins with the cleavage of the bond between the pyridine and benzene rings.

rel-(4R,5R)-4-Benzoylamino-5-phenyl-3-pyrazolidinone ( 1 ) was alkylated at position 1 with carbonyl compounds 2a-g . The corresponding rel-(4R,5R)-4-benzoylamino-5-phenyl-3-pyrazolidinone-1-azomethine imines 3a-g , were treated with sodium borohydride to give rel-(4R,5R)-1-alkyl-4-benzoylamino-5-phenyl-3-pyrazolidinones 4a-g . Reduction of pyrazolidinones 4a-g with Raney-nickel alloy in methanolic potassium hydroxide furnished rel-(4R,5R)-N-benzoyl-3-alkylamino-3-phenylalanine amides 5a-f .  相似文献

Five new aromatics bearing a 4‐O‐methylglucose unit, namely 3‐methoxy‐1,4‐hydroquinone 1‐(4′‐O‐methyl‐β‐glucopyranoside) (=4‐hydroxy‐3‐methoxyphenyl 4‐O‐methyl‐β‐glucopyranoside; 1 ), 3‐methoxy‐1,4‐hydroquinone 4‐(4′‐O‐methyl‐β‐glucopyranoside) (=4‐hydroxy‐2‐methoxyphenyl 4‐O‐methyl‐β‐glucopyranoside; 2 ), vanillic acid 4‐(4′‐O‐methyl‐β‐glucopyranoside) (=3‐methoxy‐4‐[(O‐methyl‐β‐glucopyranosyl)oxy]benzoic acid; 3 ), 5‐methoxycinnamic acid 3‐O‐(4′‐O‐methyl‐β‐glucopyranoside) (=(2E)‐3‐{3‐methoxy‐5‐[(4‐O‐methyl‐β‐glucopyranosyl)oxy]phenyl}prop‐2‐enoic acid; 4 ), and naphthalene‐1,8‐diol 1,8‐bis(4′‐O‐methyl‐β‐glucopyranoside) (=naphthalene‐1,8‐diyl bis(4‐O‐methyl‐β‐glucopyranoside; 5 ), were isolated from the cultivated Cordyceps cicadae mycelia, together with thirteen known compounds. Their structures were determined by spectroscopic methods. The absolute configurations of the sugar units were not determined.  相似文献

A series of novel 1‐methyl‐3‐(4‐phenyl‐4H‐1,2,4‐triazol‐3‐yl)‐1H‐indazoles was synthesized in three steps from 5‐(1‐methyl‐1H‐indazol‐3‐yl)‐4‐phenyl‐2H‐1,2,4‐triazole‐3(4H)‐thiones. 5‐(1‐Methyl‐1H‐indazol‐3‐yl)‐4‐phenyl‐2H‐1,2,4‐triazole‐3(4H)‐thiones were converted into 1‐methyl‐3‐(5‐(methylsulfonyl)‐4‐phenyl‐4H‐1,2,4‐triazol‐3‐yl)‐1H‐indazoles upon methylation followed by treatment with aq. KMnO4. The reaction of 1‐methyl‐3‐(5‐(methylsulfonyl)‐4‐phenyl‐4H‐1,2,4‐triazol‐3‐yl)‐1H‐indazoles with Raney nickel resulted in desulphonylation to afford corresponding 1‐methyl‐3‐(4‐phenyl‐4H‐1,2,4‐triazol‐3‐yl)‐1H‐indazoles. All the new synthesized compounds were characterized by spectral techniques.  相似文献

New ferroelectric liquid crystals containing two chiral centers, 4-(4′-n-alkyloxyphenyl)phenyl 4-{2(S)-[2(S)-methylbutyloxy]propoxy}benzoate ( 4a-4f ) and 4 -(n-alkyloxy)phenyl 4-{4′-[2(S)-(2(S)-methylbutyloxy)propoxy]phenyl}benzoate ( 5a-5f ) were synthesized and their physical properties studied. A phase-transition sequence of C-Sc*-N*-I was observed in most cases. Some homologues of them, 4a-4d , possess monotropic Sc * phase. Not only the Sc* phase-transition temperature of 5a-5f is lower than that of the corresponding 4a-4f , but their Sc * phase-transition temperature range is also wider than the corresponding 4a-4f . The Sc * phase temperature range can be up to 48 °C. The spontaneous polarization of 8-28 nC/cm2 and the electric rise time of 240-420 μs were measured in FLCs 4a-5f .  相似文献
