ABSTRACT

3-Prop-2′-ynyloxybenzothiapyran-4-one derivatives (3a–d) were prepared from 3-hydroxy benzothiapyran-4-one (1) and prop-2-ynylic halides (2) in 70–90% yield. The ethers (3a–d) were then heated in refluxing chlorobenzene to give furo[3,2-b]benzothiapyran-9-one derivatives (4a–d) in 87–95% yields. 2-Chloro-2-methylbut-3-yne (2e) on reaction with 3-hydroxybenzothiapyran-4-one (1a) directly afforded 2-isopropyl furo[3,2-b]benzothiapyran-9-one (4e) in 70% yield.  相似文献

A series of benzyloxybenzaldehyde derivatives (13) were prepared by the reactions of 4-nitrobenzyl bromide with 4-hydroxy-3-methoxybenzaldehyde (vanillin), 2-hydroxy-3-methoxybenzaldehyde (o-vanillin) and 2-hydroxy-4-methoxybenzaldehyde. When the reaction time is quite long, benzofuran derivatives (4 and 5) were obtained by the reactions of ortho-hydroxyaldehydes with the 4-nitrobenzyl bromide. Condensation reactions among the three benzyloxybenzaldehyde derivatives (13) with 2-aminomethylfuran (furfurylamine) yielded the new imine compounds (68). The structures of these aldehydes (13), benzofuran derivatives (4 and 5) and imine compounds (68) were confirmed on the basis of elemental analyses, IR, 1H NMR and 13C NMR and mass spectroscopy. The solid-state structures of compounds 46 were determined by single-crystal X-ray crystallography.  相似文献

In this study, we synthesized a new series of substituted aliphatic 1,3,4-thiadiazol-2(3H)-one derivatives (6-24) in yields ranging from 42 to 70% with an interesting mechanism that involves internal nucleophilic substitution followed by an SN2-type nucleophilic substitution. First, 1-(4-chlorophenyl)-2-((5-methyl-1,3,4-thiadiazol-2-yl)thio)ethanone (3) was synthesized from the reaction of 5-methyl-1,3,4-thiadiazole-2-thiol (1) with 2-bromo-1-(4-chlorophenyl)ethanone (2) in the presence of potassium hydroxide. Then, 1-(4-chlorophenyl)-2-((5-methyl-1,3,4-thiadiazol-2-yl)thio)ethanol (4) was synthesized by a reduction reaction of this compound using NaBH4. Finally, 5-methyl-3-alkyl-1,3,4-thiadiazol-2(3H)-one derivatives (6-24), which are the target compounds, were synthesized from the reaction of this compound (4), which is a secondary alcohol with various alkyl halides (5a-n) in the presence of sodium hydride (NaH). This study presents an interesting reaction mechanism related to the synthesis of aliphatic 1,3,4-thiadiazol-2(3H)-one derivatives that is not recorded in the literature.  相似文献

A series of novel helicid derivatives containing 3,4-dihydropyrimidin-2(1H)-one and 3,4-dihydropyrimidine2(1H)-thione moiety (3a–3f and 4a–4f) were synthesized starting from helicid. The structure of the new compounds were characterized by 1H NMR, IR and HR-MS spectra. The sedative-hypnotic activities of the target compounds were evaluated using the test of spontaneous locomotor activity in mice. All of the derivatives produced moderate to high activities; in particular, compound 4a presented the most potent sedative-hypnotic effect in comparison to the other derivatives, and derivatives 3a, 3c, 3d, 3e and 3f also showed potent activities.  相似文献

Summary The syntheses of 3-chloro derivatives of 2-alkyl-pyrimido[1,6-a]benzimidazol-1(2H)-ones2a, b as well as of 4,4-dichloro and 4,4-dibromo derivatives of 2-alkylpyrimido[1,6-a]benzimidazole-1,3(2H,4H)-diones3 a, b and4 are reported. Methods for converting some of the chloro compounds to azido (5, 6), amino (8), morpholino (9 a,10,11), piperidino (9 b), cyano (12), and methoxy (13) derivatives of the adopted tricyclic system are also described.

Diese Verbindungen lassen sich zu den entsprechenden Azido- (5, 6), Amino- (8), Morpholino- (9 a, 10, 11), Piperidino- (9 b), Cyano- (12) und Methoxy- (13) Derivaten umwandeln.

Treatment of 2-aminopyridine ( 1 ) with chloroacetyl chloride in dry benzene gave 2-chloro-N-(pyridin-2-yl)acetamide ( 3 ), which on further reaction with potassium thiocyanate gave 2-imino-3-(pyridin-2-yl)thiazolidin-4-one ( 4 ) as an intermediate compound for the synthesis of pyridin-2-yl substituted 2-imino-thiazolidine-4-one derivatives. Cyclocondensation reaction of ( 4 ) with a series of aromatic aldehydes gave 5-arylidene derivatives of pyridin-2-yl substituted 2-imino-thiazolidine-4-ones 5a–j . 1 H and 13C NMR spectroscopy, as well as elemental analyses, were used for the identification of these new compounds.  相似文献

The 3(2H)-oxo-, 3(2H)-thioxo- or 3-amino-pyridazine derivatives 4, 6 and 7 were coupled with N-phthalyl- or N-tosyl-amino acids in one-step using N,N′-dicyclohexyl-carbodiimide as the condensing agent to furnish the corresponding 3-(N-phthalyl- or N-tosyl-aminoacyl)pyridazine derivatives 8–10 respectively. Hydrazinolysis of the N-phthalyl derivatives in methanol yielded the corresponding unprotected derivatives 11–13. The antibacterial activities of the prepared compounds were tested.

Abstract

Regioselective benzoylations of 2-deoxy-2-[(2, 2-diethoxycarbonylvinyl) amino] -α-D-glucopyranose (1) yielded 6-mono-O- (2) 3, 6-di-O- (3), 4, 6-di-O- (4) and 1, 3, 6-tri-O-benzoyl- (5) derivatives. The fully benzoylated compound 6 was the major product when excess of benzoyl chloride was used. The di-O-acetyl derivatives (7 and 8) of 3 and 4 respectively were also prepared. The structures of 2-8 are based on analytical and/or spectroscopic data. Some data on MS fragmentation pathways of the partially protected compounds 2 and 4 are also reported.  相似文献

5-Benzylidene-3-(p-chlorophenyl)-2-aminothiocarbonyl-1,2,4-triazine (2) was prepared via condensation of oxazolinone (1) with thiosemicarbazide. Fused 1,2,4-triazine derivatives (3, 4 and 9) were synthesized from the reaction of compound 2 with ω-bromomethyl aryl ketones, ethyl chloroacetate, and acetic anhydride. Treatment of 4 with acetic anhydride and aromatic aldehydes yielded the corresponding acetyl, diacetyl derivatives (6 and 7) and 7-benzylidene-5-(p-chlorophenyl)-4-thioxo-3-arylidene-1,2,4-triazino [2,1-a]-1,2,4-triazine-1,8-diones (8). The electron impact mass spectra of both the above series of compounds have also been recorded and their fragmentation pattern is discussed. All synthesized fused 1,2,4-triazine derivatives were primary in vitro screened for their antimicrobial and antitumor activity.

A series of thidiazole derivatives (4, 7) from pyrazole-3-carboxylic acid chloride (2) and pyrazole-3,4-dicarboxylic acid chloride derivatives (6) were synthesized and characterized. The structures of the new compounds were confirmed byelemental analysis, NMR (1H and 13C) and IR spectra. The molecular and crystal structure of 4-benzoyl-N-[5-(methylthio)-1,3,4-thiadiazol-2-yl]-1,5-di-phenyl-1H-pyrazole-3-carboxamide(4d)was determined by single crystal X-ray diffraction method.  相似文献

Ricinine (1), a known major alkaloid in Ricinus communis plant, was used as a starting compound for the synthesis of six ricinine derivatives; two new and four known compounds. The new derivatives; 3-amino-5-methyl-1H-pyrazolo[4,3-c]pyridin-4(5H)-one (2), and 3-amino-5-methyl-1-(phenylsulfonyl)-1H-pyrazolo[4,3-c]pyridin-4(5H)-one (3), as well as the previously prepared derivatives (47) were subjected for antimicrobial and antiquorum-sensing evaluation in comparison to different R. communis extracts. Acetyl ricininic acid derivative (5) showed the highest antimicrobial activity among all tested derivatives against Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeuroginosa and Candida albicans. However, compound 7 (4-methoxy-1-methyl-2-oxo-1,2-dihydropyridine-3-carboxamide) showed the highest antiquorum-sensing activity among all tested compounds and extracts. These findings proved the usefulness of ricinine as a good scaffold for the synthesis of new antimicrobial and antiquorum-sensing derivatives in spite of its poor contribution to the antimicrobial activity of the plant extracts.  相似文献

Chalcone-like compounds 3a–l, 2-(benzylidene)-3,4-dihydronaphthalen-1(2H)-one, were synthesized from the addition of different benzaldehyde derivatives (2a–l) to 1,2,3,4-tetrahydro-1-napthalone (1) in basic medium. Mn(OAc)3-mediated addition of dimedone (4) to chalcone-like compounds gave the spirobenzofuran derivatives (5a-l), 6,6-dimethyl-3-aryl-3′,4′,6,7-tetrahydro-1′H,3H-spiro[benzofuran-2,2′-naphthalene]-1′,4 (5H)-dione, in good yields. The structures of synthesized compounds 5a–l were elucidated on basis of spectral data (NMR, IR) and elemental analysis. In addition, their antibacterial activities were screened against some human pathogenic microorganisms.  相似文献

Herein the synthesis and extraction abilities of new d-/l-phenylalaninol substituted p-tert-butylcalix[4]arene triamide derivatives (3 and 4) towards amino acids are reported. These compounds (3 and 4) have been easily synthesized via aminolysis of p-tert-butylcalix[4]arene trimethylester (2) with d-/l-phenylalaninol in methanol-toluen solvent system at one step. The extraction properties of the prepared chiral calix[4]arene triamide derivatives (3 and 4) towards some selected amino acid methylesters are studied by liquid–liquid extraction. Results show that these chiral calix[4]arene triamide derivatives (3 and 4) exhibited a good affinity towards all amino acid species without any remarkably discrimination.  相似文献

Abstract

Suitably protected 1-deoxynojirimycin (l, 5-dideoxy-l, 5-imino-D-glucitol; DNJ) and its 2-acetamido derivative, i.e., 2, 3, 6-tri-O-benzyl-.N-benzyloxycarbonyl-l, 5-dideoxy-1, 5-imino-D-glucitol (6) and 2-acetamido-3, 6-di-O-benzyl-N-benzyloxycarbonyl-1, 2, 5-trideoxy-l, 5-imino-D-glucitol (14) were each coupled with methyl 2, 3, 4, 6-tetra-O-acetyl-1-thio-β-D-galactopyranoside (15) in the presence of dimethyl(methylthio)-sulfonium triflate (DMTST) as a promoter, to give 16 and 18, which were converted to the novel disaccharides (20, 21) related to lactose and lactosamine. Coupling of 14with methyl 3, 4, 6-tri-O-acetyl-2-deoxy-2-phthalimido-l-thio-β-D-glucopyranoside (22) gave achitobiose analog (25). O-(β-D-Galactopyranosyl)-(l→3)-DNJ derivatives (38, 39) and O-(β-D-glucopyranosyl)-(l→3)-DNJ (45) were also synthesized. Conformational analysis of a variety of DNJ derivatives, based on the 1H NMR data, is also discussed.  相似文献

Three new thiacalix[4]arene derivatives, 5,11,17,23-tetra-tert-butyl-25,27-di(2-hydroxyethoxy)-26,28-dihydroxythiacalix-{}[4]arene (2), 5,11,17,23-tetra-tert-25, 26,27,28-tetrakis[(methylcarboxyl)methoxy]thiacalix[4]arene (3),5,11,17,23-tetra-tert-butyl-25,26,27,28-tetrakis(2-hydroxy-1-propanoxy)thiacalix[4]arene (4), were synthesized for the first time. The coordination properties of thiacalix[4]arene(1) and its derivatives (2 and 4) were investigated by detecting the interactions betweenthese compounds and two palladium complexes, cis-[Pd(en)(H2O)2]2+ and cis-[Pd(dtco-3-OH)(H2O)2]2+, by means of electrospray ionization mass spectrometry (ESI-MS) technique.  相似文献

The isolation and structural characterisation of two ionic complexes (S)-3 and(S)-4 based on fluoride ion-mediated deprotonation of 1,1′-bi-2-naphthol (BINOL) derivatives (S)-1 and (S)-2 have been carried out for the first time. X-ray crystallographic study showed that the deprotonated forms (S)-3 and (S)-4 adopt remarkably different molecular geometries, bond parameters as well as molecular packing modes from their neutral analogs, in agreement with their significant fluorescence changes upon the addition of fluoride ion, giving insights into the actual mechanism of fluoride ion fluorescence sensing. The deprotonation–protonation processes in two BINOL derivatives were also investigated by both fluorescence measurements and X-ray structural analyses. Such chiral basic compounds can be promising organocatalysts for asymmetric reactions.  相似文献

The synthesized series of new thiosemicarbazide derivatives ( 1 , 6–10 ) in reactions with carbon disulphide produced, according to the reaction conditions, the dithioacids ( 4 , 30 ) or the 5-substituted 1,3,4-thiadiazolo-2-thiol derivatives ( 2 , 27 ). The dithioacids were cyclized, in the reaction with hydrazine, into the 4-ami-no-1,2,4-triazolo-2-thiol derivatives ( 5 , 31 ). One of these compounds ( 31 ) was transformed into the 1,2,4-triazolo-1,3,4-thiadiazine derivative ( 33 ). The compo-unds 6–9 were also exposed to the condensation with aldehydes. 4-phenylpipera-zinocarbothiohydrazide ( 6 ) was exposed to the action of isothiocyanates, which gave the compounds 16–20 , and these cyclized to the 1,3,4-thiadiazoloamino derivatives ( 21–23 ).

The susceptibility of aerobic and anaerobic bacteria to some of the new derivatives were tested. The anaerobes were the most susceptible at concentrations in ranges less than 6.2 to 100 μg/mL to derivative: 9 (64% were susceptible), 1 , 13 (for 60%), and 7 (for 56%).  相似文献

The synthesis of a number of C-4 and C-9 substituted derivatives of KDN2en methyl ester 2 is reported. 9-Deoxy-9-iodo, 9-azido-9-deoxy and 9-O-methyl derivatives of 2(compounds 5, 7and 9) were prepared from the corresponding 9-O-tosylate, methyl 2,6-anhydro-3-deoxy-9-O-p-toluenesulfonyl-D-glycero-D-galacto-non-2-enonate (3). These compounds have been fully characterised as the peracetates 6, 8 and 10. Treatment of 3 with KSAc gave the 9-thioacetyl derivative which was isolated as the peracetate 11. 4-C-Ethenyl-4-deoxy (14), 4-C-phenyl-4-deoxy (15) and 4-C-[1-(methoxycarbonyl)ethenyl]-4-deoxy (16) derivatives of 2were prepared via the palladium-catalysed coupling of the 4-epi-chloride, methyl 5,7,8,9-tetra-O-acetyl-2,6-anhydro-4-chloro-3,4-dideoxy-D-glycero-D-talo-non-2-enonate (12) with the appropriate organostannanes.  相似文献

Summary By condensation of 2,2-difluoro-4-methyl-benzo[d]-1,3,2-2H-dioxaborines (12) with cyano acetic acid derivatives in presence of weak bases, 3-cyano-4-methyl-benzo[b]pyran-2-ones (13) or their 3-cyano-4-methyl-benzo[b]pyran-2-imine precursors (14) are available in satisfactory yields.

Zusammenfassung Dihydronorthevinon (2b) wurde aus Dihydrothevinon (2a) mit Azodicarbonsäurediethylester (DEAD) hergestellt und zu einigen neuen N-substituierten Dihydronorthevinon-Derivaten (2c–2g) umgesetzt. Es wurdenGrignard-Reaktionen dieser Verbindungen mit Methylmagnesiumiodid bzw.tert-Butylmagnesiumchlorid durchgeführt. O-Demethylierungen von3a–3j ergaben die entsprechenden N-substituierten Buprenorphin- bzw. Diprenorphin-Analoga4a–4j.

Summary Dihydronorthevinone (2b) was prepared from dihydrothevinone (2a) with diethyl azodicarboxylate (DEAD) and transformed into a number of new N-substituted dihydronorthevinone derivatives (2c–2g).Grignard reactions of the new compounds with methylmagnesium iodide andtert-butyl-magnesium chloride were studied. O-Demethylation of3a–3j resulted in the corresponding N-substituted buprenorphine and diprenorphine analogs4a–4j.
