Stereoselective syntheses on a gram scale of (3Z,6E,8E)-, (3Z,6E,8Z)-and (3Z,6Z,8Z)-3,6,8-dodecatrien-1-ol, 8, 9 and 10, respectively, are described. A key step of the synthesis of 8 consisted of a copper-mediated coupling reaction between 4-(2-tetrahydropyranyloxy)-1-butynylmagnesium bromide (15) and the mesyl ester of (2E,4E)-2,4-octadien-1-ol (14). A similar copper-mediated reaction between 15 and the mesyl ester of (E)-2-octen-4-yn-1-ol (19) was used to construct the C-12 carbon skeleton of 9. On the other hand, the synthesis of 10 was based on a palladium-promoted reaction between (Z)-1-bromo-1-pentene (23) and the organozinc bromide derived from 3,6-heptadiyn-1-yl acetate (27).  相似文献

The convenient synthesis of both enantiomers of the piperidine alkaloids such as dumetorine and epidihydropinidine is described. Pure enantiomers of 2-(2-hydroxy-ethyl)-piperidine-1-carboxylic acid tert-butyl ester are used as a common starting material. The syntheses are based on a RCM reaction and on methylation of the piperidine ring according to Beak–Lee methodology, respectively.  相似文献

A facile synthesis of 2,3,6,11-tetrahydro-1H,5H-indolizino[8,7-b]indole-11b-carboxylic acid methyl ester, a versatile intermediate utilized in the synthesis of indole alkaloids, was achieved in two steps. Condensation of tryptamine with dimethyl α-ketoglutarate led to the formation of the corresponding indolizino[8,7-b]indolone ester, which subsequently underwent an efficient lactam reduction with 9-BBN to generate the tertiary amine ester in good yield.  相似文献

The first enantiospecific total synthesis of the indole alkaloid (+)-dehydrovoachalotine (1) has been achieved from d-(+)-tryptophan methyl ester in 28% overall yield. The formation of the prochiral quaternary carbon center at C-16 in the key intermediate (12) was realized via a Tollens reaction from Na-methylvellosimine (13) in 95% yield. This approach could also be applied to the synthesis of many other indole alkaloids that contain a quaternary carbon center at C(16).  相似文献

The so-called Securinega alkaloids constitute a class of tetracyclic biologically active specialised metabolites isolated principally from subtropical plants belonging to the Phyllanthaceae family. Following a strategy based on alternative hypotheses for their biosynthesis, an easy and time-efficient divergent synthesis enabled access to twelve of those alkaloids featuring (neo)(nor)securinane skeletons. A highly stereoselective and efficient total synthesis of trans‐dihydronarciclasine from a readily available chiral starting material was developed. The synthesis defines two of the five stereogenic centers of the natural product by an amino acid ester–enolate Claisen rearrangement. The other three stereogenic centers are created in a highly stereocontrolled fashion via a six‐ring vinylogous ester intermediate, which is generated from the γ,δ‐unsaturated ester functional group of the Claisen rearrangement product in an efficient three‐step sequence. This concise total synthesis exemplifies the use of a highly regioselective Friedel–Crafts‐type cyclization to form the B ring via an isocyanate intermediate derived from an N‐Boc group, which is superior to the conventional method using an imino triflate intermediate. This same N‐Boc group is employed to give high selectivity in the Claisen rearrangement earlier in the sequence.  相似文献

A novel synthesis for rac-pilosinine ( 6 rac) and transformation of (+)-pilosinine ( 6 ) into the pilocarpus alkaloids (+)-isopilosine ( 9 ) and (+)-pilocarpine ( 18 ) are described. By correlation via 5 with 18 the absolute configuration of (+)-isopilosine ( 9 ) is established.  相似文献

A new synthesis of (+)-pinitol 9 has been developed starting from the 7-oxanorbornenic sulfone (+)-5, prepared in enantiomerically pure form by resolution of the sulfonyl-7-oxanorbornanol 4. 6-Cyanobenzo[b]furan-2-boronic acid pinacol ester (10) is a potentially useful two-point scaffold for the construction of specific compounds or compound libraries with benzofuran cores. Using a per-iodination/de-iodination strategy coupled with Sonogashira alkynylation and Cu-catalyzed heteroannulation, we have developed a procedure that allows the preparation of benzo[b]furan-6-carbonitrile (9) and 6-cyanobenzo[b]furan-2-boronic acid pinacol ester (10) in gram quantities.

We report the first catalytic asymmetric approach to octahydroindolones and a divergent enantioselective synthesis of perhydroindole alkaloids, as exemplified by lycorine‐type Amaryllidaceae alkaloids (+)‐α‐lycorane and (+)‐lycorine, from a common intermediate by using a highly concise route. The assembly of octahydroindolones employs a catalytic enantioselective 1,4‐conjugate addition of nitro dienynes, followed by a TsOH‐catalyzed cascade synthesis of highly functionalized enones, and a diastereoselective intramolecular Michael addition.  相似文献

(+)-Lentiginosine (14) and (7R)-7-hydroxylentiginosine (26), powerful inhibitors of amyloglucosidases, and their enantiomers were obtained in high overall yields by a multistep synthesis involving 1,3-dipolar cycloaddition of enantiopure tartaric acid derived pyrroline N-oxides. Structurally related (S,S)-3,4-dihydroxypyrrolidines 29-33 were synthesized as simpler models and tested towards 24 glycosidases.  相似文献

The synthesis of N-(2-hydroxy-1-phenoxyacetyl)prolylproline 2 from 2-acetoxyphen-oxyacetic acid is described. A firstsynthesis led toN-(2-acetoxy-1-phenoxyacetyl)prolylproline methylester 8b that fragmented upon attempted esterhydrolysis with 1N NaOH. A second synthesis gave thecorresponding benzyl ester 13, which wasconverted to 2 by deacylation of the phenolicacetoxy group with pyrrolidine followed byhydrogenolysis of the ester. The first enantiospecific total synthesis of the antibacterial natural product (+)‐pleuromutilin has been achieved. The approach includes the synthesis of a non‐racemic cyclisation substrate from (+)‐trans‐dihydrocarvone, a highly selective SmI2‐mediated cyclisation cascade, an electron transfer reduction of a hindered ester, and the first efficient conversion of (+)‐mutilin to the target.  相似文献

The utility of a D-glucose-derived aziridine carboxylate was demonstrated for the synthesis of polyhydroxylated quinolizidine and indolizidine alkaloids. The chemoselective reduction of 1 followed by two-carbon homologation by the Wittig reaction afforded gamma,delta-aziridino-alpha,beta-unsaturated ester 9, which on regioselective nucleophilic aziridine ring opening either by using water as a nucleophile or hydrogenation afforded delta-lactams 11/16--true synthons for the synthesis of four structurally different iminosugars, namely quinolizidine alkaloids 5b/5c, swainsonine 6b and lentiginosine 7b analogues. Glycosidase inhibitory activities of these iminosugars were investigated.  相似文献

The synthesis of the naturally occurring and biologically active alkaloids 1 and 2, first isolated from the red ascidian Botryllus leachi by Duran et al. [1], is described and the structure proposed for Botryllazine B (1) is confirmed. The analytical data for 2-(p-hydroxybenzoyl)-4-(p-hydroxyphenyl)imidazole (2) are discussed and compared with the literature. A total synthesis of the cembranoid (+)‐sarcophytin is presented, featuring a Diels–Alder cycloaddition of an enone as the dienophile with an ester‐derived dienoate. The study highlights a peculiar geometric preference for the Z dienoate to furnish the cycloadduct. The endgame involves a reaction cascade, including lactone opening, alcohol oxidation, and ketone epimerization to complete an efficient synthesis. A salient feature of the synthesis is the resulting reassignment of the absolute configuration, which corrects the previously reported nominal structure.  相似文献

Asymmetric transfer hydrogenation was used as a key step in the synthesis of two guanidine-derived alkaloids (+)-trypargine and (+)-crispine E. The enantiomeric compositions of key intermediates were established on the basis of 1H NMR spectra with chiral solvating agents. Abstract

Total synthesis of two arabidopyrones, iso-arabidopyl alcohol (1) and iso-arabidopic acid (2) isolated from Arabidopsis thaliana was achieved for the first time using Claisen condensation and Wittig reaction as the key steps. In addition, arabidopic acid (4) was synthesized from the methyl ester of arabidopyl alcohol (3). Thus, chemical synthesis of the unique natural α-pyrones 14 was accomplished with a short synthetic route by a systematic approach from readily available substances.  相似文献
