分子内1,3-偶极环加成反应的研究

近几年来,人们对于1,3-偶极环加成反应的理论研究及其在合成上的应用已给予广泛的兴趣和重视。我们曾详细地研究了4-取代苯基(N-4-戊烯基)硝酮的热化学反应性能和其区域选择性以及呋哺偶极体系的热和光化学反应。本文进一步探讨了取代基效应对环加成反应的影响,利用色谱法定量地研究了取代基的电子效应与区域选择性间的关系,并对肉桂醛肟的构型及有关的反应作了研究。     

Synthesis of Novel Spiro Pyrrolidine and Pyrrolizine Derivatives by 1,3-Dipolar Cycloaddition

The 1,3‐dipolar cycloaddition of azomethine ylides derived from isatin and amino acids viz. sarcosine and proline to 2‐arylmethylidene‐5,6‐dihydroimidazo[2,1‐b]thiazol‐3(2H)‐ones afforded novel spiro pyrrolidines and pyrrolizidines regio‐ and stereoselectively in moderate yields. The products were characterized thoroughly by IR, MS, NMR together with elementary analysis.

利用肌氨酸与对位含不同取代基的苯甲醛、共轭烯烃醛类及酮类反应生成的含氮Ylide对C60的加成反应，合成并分离、纯化制得了一系列含不同取代基的C60吡咯环纯衍生物，并用1HNMR、13CNMR、FDMS、FT－IR、UV－Vis等光谱手段确定了它们的分子结构．     

Expedient Approach for the Synthesis of Trispiropyrrolidine Bisoxindoles Through 1,3-Dipolar Cycloaddition Reactions

A series of trispiropyrrolidine bisoxindoles has been achieved via a three-component 1,3-dipolar cycloaddition reaction of 3-aryl-5-arylmethylenespiro[indole-3′,2-[1,3]thiazolane]-2′(1H),4-dione, isatin, and sarcosine in refluxing toluene, which produced the corresponding cycloadducts in good yields (79–88%). Their structures were determined by infrared, ¹H and ¹³C NMR, elementary analysis, and single-crystal x-ray diffraction analysis, and the cycloaddtion reaction was found to be highly regio- and stereoselective.     

1,3-偶极环加成合成双-1,2,4-噁二唑啉衍生物及单晶结构

2-苯基-1,2,3-三唑基-4-甲醛与1,3-丙二胺或对苯二胺缩合, 形成新的双席夫碱3和4, 然后分别与氯代肟在三乙胺条件下发生1,3-偶极环加成, 得到新型双-1,2,4-噁二唑衍生物6a～6f和7a～7f. 结构经元素分析, IR, 1H NMR和MS确证, 并用X射线衍射法测定了化合物6e的晶体结构. 化合物6e属于单斜晶系, C2/c空间群, 晶胞参数 a＝3.5134(8) nm, b＝0.50575(11) nm, c＝2.0598(5) nm, α＝90°, β＝102.02(8)°, γ＝90°, Mr＝756.81, V＝3.5798(14) nm3, Dc＝1.404 g/cm3, Z＝4, F(000)＝1576, μ＝0.095 mm－1, R＝0.0787, wR＝0.2093.     

1,3-Dipolar Cycloaddition of Nitrones with 5-Methylenehydantoins: Synthesis and Transformation of New Spirohydantoin Derivatives

1,3-Dipolar cycloaddition of various acyclic nitrones with 5-methylenehydantoin derivatives afforded new chiral spiroadducts in good yields. All the spirohydantoins were obtained through a regio- and stereospecific pathway, and the spirocarbon atom was linked to the isoxazolidine oxygen atom. A representative example of the reduction of the spirohydantoin 8 with Zn/AcOH led to the substituted 1,3-aminoalcohol hydantoin 20.     

甲亚胺叶立德的1,3-偶极环加成反应合成螺吲哚里西啶类化合物

通过7-芳亚甲基-6,7-二氢中氮茚-8(5H)-酮与甲亚胺叶立德(经苊醌或靛红与肌氨酸反应原位生成)进行的l,3-偶极环加成反应,合成了具有良好产率的新的1’-甲基-4’-芳基-5",6"-二氢-2H,8"H-二螺[苊烯-1,2’-吡咯烷-3’,7"-中氮茚]-2,8"-二酮类化合物和1’-甲基-4’-芳基-5",6"-二氢-1H,8"H-二螺[吲哚-3,2’-吡咯烷-3’,7"-中氮茚]-2,8"-二酮类化合物.采用NMR,IR,质谱,元素分析以及X射线单晶衍射等多种谱学技术对产物进行了结构表征.     

微波促进1,3-偶极环加成反应合成氮杂糖苷衍生物

利用微波促进氮杂糖硝酮(2)与丙烯酸类衍生物(3)发生1,3-偶极环加成反应,立体选择性地得到了一系列新的含异噁唑烷的氮杂糖衍生物(4),反应效率显著提高,反应时间由95h缩短为5～15min,收率由67%提高到78%～88%.利用NMR和HRMS等方法结合化合物(4d-1)的单晶结构确定了产物的结构和相对构型.     

Regioselective Synthesis of Substituted Triazolium Salts via 1,3-Dipolar Cycloaddition Reactions

The regioselectivity of 1,3-dipolar cycloaddition reactions of 1-aza-2-azoniaallene salts with ,-unsaturated nitriles such as acrylonitrile or cyclohexylidene acetonitrile afforded only 1,2,4-triazolium salts via addition to the nitrile group, while the other expected pyrazolium salts were not observed. Moreover, 1-aza-2-azonia-allene salts reacted with other competitive systems such as -iminonitrile derivatives yielding only triazolium salts via addition to the nitrile and not to the imino group. Treatment of cumulene with 3-pyridylnitrile afforded the pyridinium salt. However, 2,3-dimethyl-5-(2,6-dimethoxyphenyl)-[1,2,4]triazole could be prepared from cumulene and 2,6-dimethoxybenzonitrile. Some reactions of nitriles with 1-aza-2-azonia-allene salts prepared from 1,2,3-indantrione and 9-acetylphenanthrene are discussed.     

Fluorous Synthesis of Biaryl-Substituted Proline Analogs by 1,3-Dipolar Cycloaddition and Suzuki Coupling Reactions

A solution-phase synthesis of bicyclic prolines containing four points of diversity has been developed by a two-step synthesis involving 1,3-dipolar cycloaddition of perfluoroalkylsulfonyl-protected hydroxybenzaldehydes followed by Pd-catalyzed Suzuki coupling reaction of fluorous sulfonates with boronic acids. Both reactions are conducted under microwave irradiation and reaction mixtures are purified by solid-phase extractions without performing chromatography.     

Synthesis of 1-(2-Fluorophenyl)pyrazoles by 1,3-Dipolar Cycloaddition of the Corresponding Sydnones

3-Arylsydnones bearing fluorine and bromine atoms on the benzene ring were synthesized from N-nitroso-2-fluorophenylglycines and characterized by NMR spectroscopy. These were employed further in synthesis of the corresponding 1-(2-fluorophenyl)pyrazoles by 1,3-dipolar cycloaddition reaction with dimethyl acetylenedicarboxylate (DMAD) as activated dipolarophile. The sydnones as reaction intermediates were characterized by single crystal X-ray diffraction analysis showing interesting features such as halogen bonding as an important interaction in modeling the crystal structure.

Schiff碱3和肟或氯代肟在三乙胺作用下通过1,3-偶极环加成生成一系列含2-苯基-1,2,3-三唑基的1,2,4-(口恶)二唑啉类化合物4a～4g, 5a～5g和6a～6g. 21个新化合物的结构经元素分析,1H NMR, IR 和MS谱确证.     

1,3-偶极环加成合成新型4,5-二氢-2′,5′-二氧杂吡咯啉并[4,5-c]异噁唑衍生物

N-取代苯基马来酰亚胺与α-氯代-1,2,3-三唑-4-甲醛肟和α-氯代-喹喔啉-2-甲醛肟在三乙胺作用下通过1,3-偶极环加成合成了一系列4,5-二氢-2′,5′-二氧杂吡咯啉并[4,5-c]异(口恶)唑衍生物.化合物结构经元素分析,IR, 1H NMR及MS得到确证.     

An Approach to New Water-soluble Oligo（ethylene glycol） Camptothecin Analogues by 1,3-Dipolar Cycloaddition

C-甲基硝酮与硝基乙烯1,3-偶极环加成反应及其溶剂效应的理论研究

用密度泛函B3LYP方法研究了硝基乙烯与C-甲基硝酮的1,3-偶极环加成反应, 并利用自洽反应场方法(SCRF)计算了环已烷与乙腈这两种溶剂分别对反应所产生的 影响。该反应进行时反应物的接近有四种方式,对气相情况下和上述两种溶剂中的 反应物、四种可能产物及其相应过渡态和反应前期复合物构型分别进行了优化并计 算了振动频率。四个反应均为放热反应,且均具有很低的反应势垒,四种产物均容 易生成。但在气相反应和以环已烷为溶剂的情况下,生成的产物中endo-4型产物略 占优势;而在乙腈溶剂中得到endo-5型略占优势的产物。     

Chemo- and Regioselective 1,3-Dipolar Cycloaddition of 2-Diazopropane over 1,4-Benzoquinone: Synthesis of New Pyrazoloquinones

Abstract

1,3-Dipolar cycloaddition reaction of 2-diazopropane 1 with 1,4-benzoquinone 2 carried out at ?20 °C led to a minor mono-cycloadduct 4 and mixture of bis-cycloadducts 6 and 7. The same addition realized with 3H-pyrazole 7 at ?60 °C for 2 h yields a mixture of compounds 8 and 9 and results in O-alkylation. The reaction of 3H-pyrazoles 4 and 7 with dimethylsulfoxide and oxalyl chloride under Swern conditions led to pyrazolenines 11 and 12.     

Heterocyclic Derivatives of Fullerene C60. 1. Synthesis of New Fulleropyrazolines by the 1,3-Dipolar Cycloaddition of Nitrile Imines

New stable [6,6]-closed cycloadducts, fulleropyrazolines containing aryl, hetaryl, trifluoromethyl, and other substituents in the five-membered ring, have been obtained by the 1,3-dipolar cycloaddition of nitrile imines to C₆₀. Two methods of generating nitrile imines in situ have been used, the dehydrohalogenation of the corresponding hydrazonoyl halides by the action of triethylamine and the thermal decomposition of 2,5-diaryltetrazoles.
