An improved enantioselective synthesis of protected (3S,4S)-4-amino-3,5-dihydroxypentanoic acid (ADPA) from L-serine has been developed. Enantioselectivity is improved by replacing the methyl ester with the tert-butyl ester and using neutral magnesium salt of esters to give β-keto ester.  相似文献

本文报道了以(E)-1-(2-对甲苯磺酰胺基)-3-芳基丙-2-烯-1-酮(1)为底物与N-硫代丁二酰亚胺(2)通过亲电环化反应合成2-芳基-3-硫代-2,3-二氢喹啉-4(1H)-酮类化合物。以三氟化硼乙醚为催化剂(20 mol %),1,2-二氯乙烷为溶剂,反应温度为60 oC,可以60-92%的收率得到一系列2-芳基-3-硫代-4(1H)-喹啉酮衍生物,化合物3a-k均未见文献报道,其结构均经过1H NMR, 13C NMR以及高分辨质谱进行确定。  相似文献

L -Aspartic acid by successive N-tosylation, anhydride formation, and reduction was converted into (3S)-3-(tosylamino)butano-4-lactone ( 4 ). Electrophilic methylation of 4 , subsequent iodo-esterification and nucleophilic methylation, followed by saponification and deprotection, gave (2S, 3R)-3-amino-2-methylpentanoic acid ( 2 ) with an ee of > 99% in seven steps and in an overall yield of 34%.  相似文献

Treatment of (R) methionine sulfoxide with NaOD led to exchange of the C-4 methylene and C-5 methyl protons; exchange of the chiral C-2 proton did not occur. Reducation with mercaptoacetic acid gave (R)-[4-2H2, 5-2H3] methionine. The latter was converted into its carbobenzyloxy methyl ester sulfoxide, pyrolysis of which followed by deprotection yielded (R)-[4-2H2] vinylglcine as the hydrochloride.  相似文献

A new tripeptide (dimer), bis[(L -cysteine-S-acetyl)-L -hemicystinyl(S2S2)-D -valine] ( 6 ) was synthesized by coupling N-(tert-butoxycarbonyl)-S-carboxymethyl-L -cysteine benzyl ester ( 1 ) with S-trityl-L -cysteinyl-D -valine benzyl ester and subsequent removal of the protecting groups. 3-Ethylamino-2-(4-nitro-benzoyl)-but-2-enoic acid phenylamide is synthesized, isolated, spectroscopical and structural characterized by means of linear-polarized IR-spectroscopy (IR-LD) of oriented solids as a colloidal suspension in nematic liquid crystal. The experimental IR-band assignment and structural information are supported by quantum chemical calculations at MP2 and B3LYP level of theory and 6-311++G** basis set. The geometry is characterized by two inramolecular hydrogen bonds of NH...O=C with bond lengths of 2.589 and 3.449 angstroms. The NHO angles are 142.1(3) degrees and 125.8(4) degrees , respectively. A transoide configuration of amide fragments is predicted with a torsion NH-C=O angle of 150.6 degrees . Phenyl fragments are mutually perpendicular oriented closing an angle of 92.6 degrees .  相似文献

A general synthetic scheme for the preparation of 2-amino-4,(4-imidazolyl)pyridines, potential histamine H2 antagonists, is described. The synthesis is based on the Neber rearrangement of l,(4-pyridyl)l-alkanone oxime 0-tosylates to the appropriate α-aminoketones or α-aminoketals, which are then converted to the corresponding imidazoles. The reaction of Grignard reagents with 2-chloroisonicotinonitrile, as well as nucleophilic displacement of chloride by amines on 2-chloroisonicotinonitrile and derivatives, are discussed in relation to the preparation of the ketone intermediates.  相似文献

以1-磺丁基-3-甲基咪唑硫酸氢盐离子液体(b)为反应介质,7-氨基头孢烷酸(7-ACA,2)为原料,与4-甲基-5-甲酰基噻唑经缩合反应制得(6R,7R)-7-氨基-3-[2-(4-甲基-5-噻唑)乙烯基]-8-氧代-5-硫-1-氮杂双环[4.2.0]辛-2-烯-2-羧酸(7-ATCA,1),其结构经1H NMR,13C NMR和IR表征。考察了离子液体及其用量,原料摩尔比r[n(4-甲基-5-甲酰基噻唑)∶n(2)],反应温度和反应时间对1收率的影响。在最佳反应条件[b为反应介质,b用量为20 m L,r=1.3,于65℃反应5 h]下,1收率可达90%以上。  相似文献
