Treatment of 3-acetonyl-5-cyano-1,2,4-thiadiazole (1) with 4-methyl or 4-methoxyphenylhydrazine hydrochloride provided 5-cyano-3-(2,5-dimethylindol-3-yl)-1,2,4-thiadiazole (2) or 5-cyano-3-(5-methoxy-2-methylindol-3-yl)-1,2,4-thiadiazole (3) as the sole product, respectively. In contrast, treatment of 1 with phenylhydrazine hydrochloride resulted in the formation of 5-cyano-3-(2-methylindol-3-yl)-1,2,4-thiadiazole (4) and the unexpected 5-cyano-3-(3,5-dimethyl-1-phenylpyrazol-4-yl)-1,2,4-thiadiazole (5). In a similar manner, when 1 was treated with 4-chlorophenylhydrazine hydrochloride, indolization was suppressed by phenylpyrazolation giving rise to 5-cyano-3-(5-chloro-2-methylindol-3-yl)-1,2,4-thiadiazole (6) and 5-cyano-3-[1-(4-chlorophenyl)-3,5-dimethylpyrazol-4-yl]-1,2,4-thia diazole (7). The reaction mechanism is discussed. Compounds 4, 5 and 6 exhibited weak antimicrobial activity against Helicobacter pylori.  相似文献

By adding diethyl hydrogen phosphite to 5-trifluoromethyl-2-furaldehyde (5-trifluoromethylfur-2-yl)(diethoxyphosphoryl)methanol was synthesized. It was oxidized with DMSO-acetic anhydride mixture to diethyl 5-trifluoromethyl-2-furoyl phosphonate. The reaction of the latter with ethoxymethylenetriphenylphosphorane gives ethyl (2E)-3-(diethoxyphosphoryl)-3-(5-trifluoromethylfur-2-yl)propenoate. Analogous reaction of (diethoxyphosphoryl)(5-trifluoromethylfur-2-yl)acetic aldehyde yields ethyl (4E)-4-(diethoxyphosphoryl)-4-(5-trifluoromethylfur-2-yl)-but-3-enoate. The addition of nitromethane to these esters of unsaturated acids in the presence of potassium fluoride gives a mixture of diastereomers of phosphorylated esters of 2-nitromethyl-3-(5-trifluoromethylfur-2-yl)propanoic and 3-nitromethyl-4-(5-trifluoromethylfur-2-yl)butanoic acids respectively. By the reduction of ethyl nitropropanoate with zinc and formic acid in dioxane ethyl 2-aminomethyl-3-(diethoxyphosphoryl)-3-(5-trifluoromethylfur-2-yl)propanoate was prepared in a low yield. It may be considered as the derivative of β-alanine containing additional pharmacophore fragments.  相似文献

以阿魏酸甲酯为原料,通过氧化偶联构建2-芳基苯并二氢呋喃骨架,再经傅克酰基化和酯缩合反应依次制得(E)-3-[2-(4-羟基-3-甲氧基-5-乙酰基)苯基-3-甲氧羰基-7-甲氧基-2,3-二氢苯[b]并呋喃-5-基]丙烯酸甲酯(3)和(E)-3-[2-(4-羟基-3-甲氧基-5-甲氧羰基乙酰基)苯基-3-甲氧羰基-7-甲氧基-2,3二氢苯并[b]呋喃-5-基]丙烯酸甲酯(4); 4经水解反应合成3-【2-羟基-3-甲氧基-5-{5-[2-(甲氧基羰基)乙烯基]-7-甲氧基-3-甲氧羰基-2,3-二氢苯并[b]呋喃-2-}基】苯基-3-氧丙酸(5),化合物3~5未见文献报道,其结构经1H NMR, 13C NMR和MS(ESI)表征。采用分子对接软件Autodock vina对化合物2~5与HIV-1整合酶核心部位高度同源的PFV IN(PDB: 3L2V)进行对接,计算结果显示该类化合物能与整合酶形成稳定的复合物,具有1,3-二酮基团的化合物3, 4和5能与整合酶中金属离子产生螯合作用,其中化合物5的结合作用最强。  相似文献

The reaction of 3,4,4-trichloro-1-(2-thienyl)but-3-en-1-one with hydroxylamine gave 3-hydroxyiminomethyl-5-(2-thienyl)isoxazole which was converted into 5-(2-thienyl)isoxazole-3-carbonitrile by the action of acetic anhydride in pyridine. 5-(2-Thienyl)isoxazole-3-carbonitrile reacted with hydroxylamine to produce the corresponding amide oxime. Heterocyclization of its O-acyl derivatives in acetic acid afforded 5-substituted 3-[5-(2-thienyl)isoxazol-3-yl]-1,2,4-oxadiazoles.  相似文献

以2,3-二氯吡啶为起始原料,通过水合肼亲核取代、马来酸二乙酯环合制得2-(3-氯-2-吡啶基)-5-氧-3-吡唑烷甲酸乙酯(5);5经苯磺酰氯酯化、溴化氢溴化制得3-溴-1-(3-氯-2-吡啶基)-4,5-二氢-1H-吡唑-5-甲酸乙酯(7);7经脱氢、水解合成了用于制备氯虫酰胺的关键中间体--3-溴-1-(3-氯-2-吡啶基)-1H-吡唑-5-甲酸,总收率39.5%,其结构经~1H NMR和MS确证.  相似文献

Successive treatment of 5α-cholestan-3-one ( 1 ) with O2 under basic conditions and then NaBH4 led to 5α-3-oxa-cholestan-2-one ( 5 ). Analogous reactions with 5β-cholestan-3-one ( 6 ) yielded 5α-4-oxa-cholestan-3-one ( 7 ) and 5 ξ-3-oxa-cholestan-4-one ( 8 ). 4-Cholesten-2-one ( 10 ), which was prepared starting from 4-cholesten-3-one, was isomerized by methanolic KOH to give a mixture of 5α-cholest-3-en-2-one ( 11 ) and 5β-cholest-3-en-2-one ( 12 ). 5β-Cholestane-2,3-dione ( 17 ) was synthesized from 4β-bromo-5β-cholestan-3-one ( 13 ). Ozonolysis of the dione 17 and subsequent NaBH4 reduction of the oxidation product gave both 5β-2-oxa-cholestan-3-one ( 18 ) and 5β-3-oxa-cholestan-2-one ( 19 ).  相似文献

3, 5, 6-哌嗪三酮-2-甲酰胺(5)经与PCl5-POCl3反应制得3, 5, 6-三氯-2-氰基吡嗪(6)。6经转化为甲酯(7)再水解为3, 5, 6-三氯-2-吡嗪甲酸(8)。由8出发合成了两个2, 3-二氯-5-取代-5, 8-二氢-8-氧-吡啶并[2, 3-b]吡嗪-7-羧酸乙酯(11a~b)。其中5, 7~11均为新化合物, 其组成和结构经元素分析、IR、^1H NMR、MS证实。  相似文献

三(三甲硅基)环戊二烯基三羰基钼负离子锂盐[{η^5-(Me~3Si)~3C~5H~2}Mo(CO)~3]^-Li^+(1), 分别与MeI、phCH~2Cl及ClCH~2COOC~2H~5反应生成相应的烃基化钼衍生物[{η^5-(Me~3Si)~3C~5H~2}Mo(CO)~3R,] (R=-CH~3, 2; -CH~2ph, 3;-CH~2COOC~2H~5, 4)。1与PCl~3反应除得到预期的钼氯化物[{η^5-(Me~3Si)~3C~5H~2}Mo(CO)~3Cl](5)外, 主要得到钼磷氯化物[{η^5-(Me~3Si)~3C~5H~2}Mo(CO)~3PCl~2] 6; 1与碘反应得到钼碘化物[{η^5-(Me~3Si)~3C~5H~2}Mo(CO)~3I] 7; 1与HOAc作用后分别和CCl~4、NBS室温反应, 仅分离到脱去一个Me~3Si的钼卤化物[{η^5-(Me~3Si)~2C~5H~2}Mo(CO)~3X], (X:Cl, 8; Br, 9)。  相似文献

The synthesis of two new acyclic nucleoside analogs, 2-(2′,3′-dihydroxypropyl)-5-amino-2H-1,2,4-thiadiazol-3-one (1) and 3-(2′,3′-dihydroxypropyl)-5-amino-3H-1,3,4-thiadiazol-2-one (2), is reported. The first compound, 1, was obtained by reaction of 3-chloro-1,2-propanediol with the sodium salt of 5-amino-2H-1,2,4-thiadiazol-3-one (3) in anhydrous dimethylformamide. Similarly, 5-amino-3H-1,3,4-thiadiazol-2-one (4) reacted with 3-chloro-1,2-propanediol to give 2. The thiadiazole 4 was prepared by condensation-cyclization of hydrazothiodicarbonamide (9).  相似文献

4-Toluenesulfonyl isothiocyanate reacted with 1,2-epoxy-3-phenoxypropane and 2,3-epoxypropyl 4-methoxyphenyl ether to give, respectively, 3-(4-toluenesulfonyl)-5-phenoxymethylene-2-oxazolidmethione ( I ) and 3-(4-toluenesulfonyl)-5-(4-methoxyphenoxymethylene)-2-oxazolidinethione ( II ) in high yields. The sulfonyl isothiocyanate reacted further with styrene oxide to give a mixture of oxazolidinethiones from which a solid III was isolated. The structure of III is either the 4- or 5-phenyl derivative of 3-(4-toluenesulfonyl)-2-oxazolidinethione. Reactions of the isothiocyanate with 3-chloro-1,2-epoxypropane and 1,2-epoxybutane afforded, respectively, 3-(4-toluenesulfonyl)-5-chloromethyl-2-oxazolidinethione ( IV ) and 3-(4-toluenesulfonyl)-4-ethyl-2-oxazolidinethione ( V ). 以(1S,5S)-(-)-α-蒎烯为原料合成了系列新型(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物. (-)-α-蒎烯经硼氢化氧化、重铬酸吡啶盐(PDC)氧化得到(1S,2S,5R)-(-)-3-蒎酮; 在BF3•(C2H5)2O催化下(1S,2S,5R)-(-)-3-蒎酮与伯胺化合物反应生成Schiff碱, 再经KBH4或NaBH4还原得到(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物. 采用FT-IR, 1H NMR, 13C NMR和GC-MS等分析手段对合成所得(1S,2S,5R)-N-烷基-3-蒎烷亚胺和(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物的结构进行了表征. 考察了(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物对大肠杆菌(E. coli)、金黄色葡萄球菌(S. aureus)、枯草芽胞杆菌(B. subtilis)、荧光假单胞菌(P. fluorescens)、白色念珠菌(C. albicans)、黑曲霉(A. niger)和米根霉(R. oryzae)等细菌和真菌的抑菌和杀菌活性. 结果表明(1S,2S,3S,5R)-N-正庚基-3-蒎胺对真菌和细菌均表现出良好的杀菌和抑菌活性.  相似文献

Oxidation of 2′-hydroxy-8-methylspiro[4H-benz-1,3-oxazin-2-one-6,1′-cyclopentane] or N-mesyl-2-(cyclopent-1-en-1-yl)-6-methylaniline provided the corresponding ketones. The rearrangement of these ketones oximes under treatment with thionyl chloride gave rise to nitriles of 5-(2-amino-3-methylphenyl)-5-oxopentanoic or 5-(2-methanesulfamido-3-methylphenyl)-5-hydroxypentanoic acids. By heating 5-(2-acetylamido-3-methylphenyl)-5-oxopentanoic acid with LiH in THF3-(2,8-dimethylquinol-4-on-3-yl)propanoic acid was obtained.  相似文献

3-Chloro- and 3-bromo-4-methoxycoumarins 1a,b were readily transformed into 4-halo-5-(2-hydroxyphenyl)-3-oxo-2,3-dihydropyrazoles 2a,b with hydrazines. In the reaction of 1a,b with excess hydrazine and phenylhydrazine in boiling ethanol, unexpected 4-hydrazono-3-(2-hydroxyphenyl)-2-pyrazolin-5-ones 3 , 5 were obtained. The structure of 3 was determined by X-ray diffraction analysis.  相似文献

A reaction of 2-(2-nitrobenzoylmethyl)-1,3-dioxolane ( 3 ) with hydroxylamine, followed by acid catalyzed cyclization, produced 5-(2-nitrophenyl)isoxazole ( 5 ) as the only isolable product, whereas 2-(benzoylmethyl)-1,3-dioxolane ( 9 ) under identical conditions produced a 2.5:1 mixture of 3-phenyl and 5-phenylisoxazoles 10 and 11 . These findings contradict the literature report that β-keto ethyleneacetals on treatment with hydroxylamine produce exclusively 3-substituted isoxazoles. As an additional proof, 3-(2-nitrophenyl)isoxazole ( 8 ) was prepared by an unambiguous method via the nitrile oxide route for comparison. The intermediate obtained on treatment of 2-(2-nitrobenzoylmethyl)-1,3-dioxolane ( 3 ) with hydroxylamine was found to be an isomeric mixture of 5-hydroxy-5-(2-nitrophenyl)-2-isoxazoline ( 4 ) and the syn and anti mono-oximes 19 (at least in solution), either of which could give 5-(2-nitrophenyl)isoxazole ( 5 ) on acid treatment. A mechanistic rationale is provided to explain the anomalous results.  相似文献

Microbial enantioselective ester hydrolysis for the preparation of optically active (3R,5S)-(-)-5-phenyl-4,1-benzoxazepine-3-acetic acid derivatives as potent squalene synthase inhibitors was investigated. Pseudomonas diminuta and Pseudomonas taetrolens hydrolyzed the racemic ethyl ester of the 5-(2-chlorophenyl) analogue to yield the (-)-carboxylic acid with excellent enantiomeric excess (>99% ee). We found that the (-)-enantiomer was an active inhibitor. Bulkiness of the ester moiety did not affect the enantioselectivity but did affect reactivity. The racemic ethyl ester of the 5-(2-methoxyphenyl) analogue, 5-(2,3-dimethoxyphenyl) analogue and 5-(2,4-dimethoxyphenyl) analogue were also hydrolyzed with Pseudomonas taetrolens to afford enantiomerically pure (-)-carboxylic acids in large scale. As another route to (3R,5S)-(-)-7-chloro-5-(2,3-dimethoxyphenyl)-1-neopentyl-2-oxo-1,2,3,5-tetrahydro-4,1-benzoxazepine-3-acetic acid [(-)-1c], the earlier intermediate (-)-2-amino-5-chloro-alpha-(2,3-dimethoxyphenyl)benzyl alcohol [(-)-12] was successfully obtained by asymmetric hydrolysis of (+/-)-5-chloro-alpha-(2,3-dimethoxyphenyl)-2-pivaloylaminobenzyl acetate with Pseudomonas sp. S-13 with >99% ee in kilogram scale followed by alkaline treatment. The product (-)-12 was converted to (-)-1c without racemization.  相似文献

The effects of native cyclodextrins (alpha, beta or gamma), hydroxypropyl-beta-cyclodextrin, beta-cyclodextrin solubilized in urea, soluble starch and glucose in water solution on the fluorescence behaviour of melatonin (N-acetyl-5-methoxytryptamine) (M) and 5-methoxytryptamine [5-methoxy-3-(2-aminoethyl)indole] (5M) were determined. In addition, the effect of methanol and propanol with and without beta-cyclodextrin or hydroxypropyl-beta-cyclodextrin was assessed. From the fluorescence changes with pH, the values of the pKa for the ground (9.9 +/- 0.2) and the excited state (7.7 +/- 0.2) for 5M were determined. From the fluorescence changes with beta-cyclodextrin or hydroxypropyl-beta-cyclodextrin, the association constants of M, 5MH [5-methoxy-3-(2-ammoniumethyl)indole] and 5M with the two hosts were determined. The values with beta-cyclodextrin were KAssoc5MH = (1.4 +/- 0.4) x 10(2) mol-1 dm3, KAssoc5M = (1.6 +/- 0.1) x 10(2) mol-1 dm3 and KAssocM = (1.1 +/- 0.2) x 10(2) mol-1 dm3, and with hydroxypropyl-beta-cyclodextrin KAssoc5MH = (1.1 +/- 0.3) x 10(2) mol-1 dm3, KAssoc5M = (2.5 +/- 0.1) x 10(2) mol-1 dm3 and KAssocM = (1.51 +/- 0.07) x 10(2) mol-1 dm3. The ratios of the fluorescence quantum yields for the bound and free substrate (phi b/phi f) were in the range 1.15-1.48. The detection limits under the optimum conditions were 0.381 +/- 0.001 ng cm-3 for the complex 5MH-hydroxypropyl-beta-cyclodextrin in water and 0.290 +/- 0.001 ng cm-3 for the complex M-hydroxypropyl-beta-cyclodextrin in water with 5% of methanol. The recovery of melatonin from pharmaceutical preparations was 98-103% with an RSD of 2%. The recovery from rat pineals was also good. The method is direct, simple and accurate.  相似文献

文中深入研究了5-(l-孟氧基)-3-溴-2(5H)-呋喃酮新手性源(5a)的合成方法及其不对称合成反应。5a不仅制备方法简便,光学纯度单一,而且它作为稳定的Michael受体,可与碳、氧、氮、硫等不同的亲核试剂发生串联的Michael加成/分子内亲核取代反应,制备得到含有多个手性中心的双环[3.3.0^3^,^7]-5-辛烯类化合物、双环[3.1.0^3^,^5]-己烷类化合物和螺-环丙烷类化合物。本文报道了该手性呋喃酮5a的合成和构型测定。  相似文献
