Mono-, di-, and tetraiodothiacalix[4]arenes 13-16 have been successfully synthesized for the first time by the Griess reaction of diazonium salts of the corresponding aminothiacalix[4]arenes 4-7. X-ray crystallography reveals that monoiodinated compound 13 adopts a distorted pinched cone conformation, in which the three hydroxy groups and the iodine atom form a pseudo-cyclic hydrogen bonding. On the other hand, tetraiodinated compound 16 adopts a 1,3-alternate conformation presumably due to the steric hindrance and dipole repulsion between the iodine atoms.  相似文献

Tetranitroazacalix[4]arenes have been synthesized by the nucleophilic aromatic substitution of 1,5-difluoro-2,4-dinitrobenzene with 1,3-diaminobenzenes. An X-ray crystal structure analysis revealed that the azacalixarenes adopt a non-symmetrical 1,3-alternate conformation, and the dinitrobenzene rings strongly conjugate with the bridging nitrogen atoms. In the 1H NMR spectrum (CDCl3, 30 °C), the tetraisopropyl derivative 3b displays a pair of diastereotopic methyl signals of the isopropyl groups in agreement with the frozen 1,3-alternate conformation on the NMR time scale. The free energy of activation () for the macrocyclic inversion was determined to be 87.5 kJ mol−1 by temperature-dependent NMR spectroscopy.  相似文献

Enolizable ketones react with m-nitroaniline in the presence of strong base such as t-BuOK to give 4- and 6-substituted nitroindoles. The reaction proceeds via oxidative nucleophilic substitution of hydrogen in m-nitroaniline with enolate anions in positions ortho to the amino group giving anionic σH adducts that are additionally stabilized by intramolecular interaction between the amino and the carbonyl group. Spontaneous oxidation of the σH adducts followed by the Bayer type condensation of the produced ortho-aminonitrobenzyl ketones gives 4- and 6-substituted nitroindoles. The scope of this reaction and its basic mechanistic features are discussed.  相似文献

A convergent synthetic strategy towards novel 1,2,4-triazole dendrimers, in which 3,5-dichloro-4-(4-methoxyphenyl)-4H-1,2,4-triazole was used as the heterocyclic building block, was successfully explored. Nucleophilic aromatic substitution at this novel AB2-monomer was used as the key step in the propagation of the heterocyclic dendrons and these dendrons were attached to both a 1,3,5-triazine and a methylene core. The peripheral 1,2,4-triazole could be varied not only by nucleophilic aromatic substitution but also by Suzuki cross-coupling. The presented dendrimers are promising candidates to be used in applications where the large number of heteroatoms can be exploited or a better resistance to the applied conditions is required.  相似文献

The first uncatalyzed preparation of tetraaza[1.1.1.1]m,p,m,p-cyclophanes (symmetrical and unsymmetrical) gave access to previously unknown N(H)-bridged derivatives that could be further substituted. NMR studies and theoretical calculations show that these macrocycles adopt an 1,3-alternated conformation.  相似文献

We describe an efficient synthesis of enantiopure tetrahydro-1,4-benzodiazepine-3-ones derived from l-alanine. Diverse substitution at N1, N4, and C10 can be achieved by coupling various N-alkyl derivatives of l-alanine and N-allyl-(2-fluoro-5-nitro)benzylamine. Cyclization of this intermediate proceeds in high yield and without racemization, providing diversity at N1. The NO2 group was easily transformed into other functional groups or removed, providing diversity at C10. Finally, oxidative deallylation allows diverse substitution to be installed at N4.  相似文献

A synthetic route to a 2,4-disubstituted pyridine as a potential β-strand mimetic has been developed and applied in the synthesis of a tripeptidomimetic of Leu-Gly-Gly. The pyridine scaffold replaces the central glycine, and is substituted with analogues of leucine and glycine in positions 4 and 2, respectively. 2-Fluoro-4-iodopyridine was chosen as the functionalized scaffold and was substituted with protected leucinal in position 4 via a Grignard exchange reaction using iso-propyl magnesium chloride. The glycine moiety was introduced in position 2 via a nucleophilic aromatic substitution reaction (SNAr) facilitated by microwave irradiation. The synthetic sequence involved 12 steps with an overall yield of 7%.  相似文献

The effects on response times of representative commercially available dielectrically negative (−LC) and positive (+LC) type liquid crystal mixtures doped with small quantities of various tetrafluorinated cycloalkoxyphenyl ether derivatives were investigated. The synthesis of various polyfluoroaryl systems by nucleophilic aromatic substitution processes is described and electro-optical measurements concerning the rise and decay times and voltage holding ratios of each doped LC formulation are reported. The response time of the dielectrically negative LC host was improved upon doping with each of the tetrafluorinated cycloalkoxyphenyl ether derivatives, although no improvement in the performance of the dielectrically positive LC host was achieved.  相似文献

A short, divergent route to G1 oligo(aryl sulfone)s and a G2 oligo(aryl sulfone) dendrimer using nucleophilic aromatic substitution reactions is described. A range of tetrasubstituted pentasulfones are proposed for applications as homogeneous supports for synthesis. Key to achieving selectivity in the syntheses is the activation of leaving groups by sulfide to sulfone oxidation. Preparation of the G2 oligo(aryl sulfone) is low-yielding due to competition from SET processes that are interesting from a mechanistic point of view. The utility of the supports is exemplified with a four step synthesis of a dipeptide and by ‘react and release’ synthesis of amides.  相似文献

The synthesis of 3-cyano-2-fluoropyridines from readily available precursors was achieved via nucleophilic substitution of a leaving group in the 2-postion with KF or Bu4NF in polar aprotic solvents such as DMF and DMSO. Ionic tetrahydrothiophenium fragment is the most effective leaving group, the methanesulfonyl moiety is a somewhat less effective, and Br- and Cl- are the least effective. Relatively mild conditions of the reaction between (2-pyridyl)-tetrahydrothiophenium salts and KF, as well as the convenience of one-step synthesis of these salts from 2(1H)-pyridinethiones, make these salts the compounds of choice for the preparation of ring-fluorinated pyridines.  相似文献

Vinylic halides having alcohol, sulfonamide, active methine, and thiol moieties as nucleophiles cyclize to hetero- and carbocycles by intramolecular nucleophilic substitution at the sp2 carbon centers. The density functional theory calculations suggest that the cyclization proceeds through SN2-type substitution (the in-plane vinylic nucleophilic substitution, SNVσ), when vinyl halides are substituted with oxygen, nitrogen, and carbon nucleophiles. The substitution with sulfur nucleophiles, in contrast, proceeds through both routes of SNVσ and out-of-plane vinylic nucleophilic substitution (SNVπ).  相似文献

Photodynamic therapy (PDT) is a developing modality for the treatment of certain tumorous and other diseases. Considerable progress has been made in recent years in the search for new photosensitizers, in particular elucidating the role of localization of the photosensitizer. Known successful photosensitizers of the tetrapyrrole type are amphiphilic molecules, preferably localizing in cellular membrane structures. Thus, the quest for new photosensitizers requires the synthesis of unsymmetrically substituted (amphiphilic) tetrapyrroles. In this article, we describe strategies for the de novo synthesis of amphiphilic tetrapyrroles using a 3-hydroxyphenyl substituted tetrapyrrolic system (Temoporfin) as the lead structure. From an applied science-oriented approach, such a set of amphiphilic porphyrins is best synthesized by combining well-developed condensation methods with subsequent functionalization via organolithium compounds or transition metal catalyzed coupling protocols. Starting from simple A2- or AB-porphyrins, the synthesis of A2B-, A3-, A3B-, and A2BC-porphyrins with a mixed hydrophilic/hydrophobic substitution pattern is described. Because of the versatility of this approach to unsymmetrically substituted porphyrins it is also applicable to other areas where porphyryns with a tailor-made substitution patterns are needed, for example, catalysts or molecular electronic devices based on tetrapyrroles.  相似文献

A phenylazo group was used for selective activation of ortho fluorine and chlorine atoms towards nucleophilic aromatic substitution with the propanethiolate anion. This enabled a regioselective synthesis of three substituted 4-alkoxyanilines. The regioselectivity of substitution was confirmed by comparison of experimental NMR chemical shifts with empirically predicted values. The observed reactivity of the substrates is discussed in the context of the substituent effect.  相似文献

New aromatic diamine monomers prepared from condensation reactions of aniline, p-chloroaniline, p-nitro aniline, p-chloro-m-nitro aniline with 4-amino-N,N-dimethylaniline (2:1) (aniline:reagent) in the presence of potassium dichromate in acidic media yielded new monomers of a highly colored violet and reddish-violet. Mechanism of the reaction of aniline with 4-amino-N,N-dimethylaniline in the presence of potassium dichromate as an oxidant is expected to proceed through nucleophilic substitution reaction, and the mechanism proceeds facilitated a nucleophilic attack of the substituted aniline ring on the –NH2 group of the reagent; through partial protonation of their –NH2 group, forming in diamine dye and their identification was confirmed by IR, 1H NMR, and CHN analyses.  相似文献
