2-[3′-(Trifluoromethyl)anilino]-5-hydroxynicotinic acid (2) was synthesized by two routes: a) by direct hydroxylation of 2-[3′-(trifluoromethyl)anilino]nicotinie acid (1) ; and b) by the following sequence starting from 2-chloro-3-methyl-5-nitropyridine (3) via 5-amino-2-chloro-3-methylpyridine (4) , 2-ehloro-5-hydroxy-3-methylpyridine (6) , 5-acetoxy-2-chloro-3-methylpyridine (7) , 5-acetoxy-2-chloronicotinie acid (8) , and 2-chloro-5-hydroxynicotinic acid (9). The correlation of 2 with one of the metabolites of 1 has been accomplished, and the identities of both compounds have been proven.  相似文献

A new variant of the synthesis of 7-amino-4-methyl-coumarin from m-aminophenol via a three-step scheme is proposed. Acylation of m-aminophenyl with methoxycarbonyl chloride gave m-(N-methoxycarbonylamino)phenol, which was converted to 7-(N-methoxycarbonyl-amino)-4-methylcoumarin by condensation with acetoacetic ester in sulfuric acid. Heating of the coumarin with concentrated alkali leads to an intermediate, which, after acidification, is converted to 7-amino-4-methylcoumarin in high yield.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 3, pp. 312–314, March, 1990.  相似文献

Two closely structurally related coumarins, 4-hydroxy-8-isopropyl-5-methylcoumarin and 4-hydroxy-6-chloro-7-methylcoumarin were acylated at C-3 and further converted to the respective chalcones and two series of eighteen new compounds, which were evaluated for possible antiviral activity.  相似文献

Fragmentation of (S)-endo-5-norbornenyl-2-oxychlorocarbene [(S)-8] in cyclohexane-d12 gives approximately 20% (S)-endo-2-chloro-5-norbornene [(S)-7] with approximately 50% ee, 65-70% (R)-exo-2-chloro-5-norbornene [(R)-4] with >95% ee, and approximately 12% (R)-3-nortricyclyl chloride [(R)-5] with approximately 22% ee. (Analogous stereochemical results were also obtained starting with the enantiomeric carbene (R)-8.) The (S)-8 to (S)-7 and (S)-8 to (R)-4 conversions are ascribed mainly to retention and inversion S(N)i transition states, respectively. These have been located by computational methods and are nearly isoenergetic. In more polar solvents (CDCl3 and CD3CN), the fragmentation of (S)-8 increasingly occurs via competitive ion pair pathways in which steroselectivity is diminished, and escape to the norbornenyl-nortricyclyl cation directs the products away from endo-2-chloro-5-norbornene toward exo-chloride 4 and nortricyclyl chloride 5.  相似文献

以2,3-二氯吡啶为起始原料,通过水合肼亲核取代、马来酸二乙酯环合制得2-(3-氯-2-吡啶基)-5-氧-3-吡唑烷甲酸乙酯(5);5经苯磺酰氯酯化、溴化氢溴化制得3-溴-1-(3-氯-2-吡啶基)-4,5-二氢-1H-吡唑-5-甲酸乙酯(7);7经脱氢、水解合成了用于制备氯虫酰胺的关键中间体--3-溴-1-(3-氯-2-吡啶基)-1H-吡唑-5-甲酸,总收率39.5%,其结构经~1H NMR和MS确证.  相似文献

A series of new N1-(coumarin-7-yl)amidrazones incorporating N-piperazines and related congeners were synthesized by reacting the hydrazonoyl chloride derived from 7-amino-4-methylcoumarin with the appropriate piperazines. The chemical structures of the newly prepared compounds were supported by elemental analyses, 1H-NMR, 13C-NMR, and ESI-HRMS spectral data. The antitumor activity of the newly synthesized compounds was evaluated. Among all the compounds tested, 7-{2-[1-(4-(1-benzyl-2-ethyl-4-nitro-1H-imidazol-5-yl)piperazin-1-yl)-2-oxopropylidene]hydrazinyl}-4-methyl-2H-chromen-2-one (3n) was the most potent against MCF-7 and K562 cells, with IC?? values of 20.2 and 9.3 μM, respectively.  相似文献

A new method was developed for the synthesis of some 7-substituted 3-chloro-3,4-dihydro-l-hydroxycarbostyrils 3c-g in which α-chloro-β-(4-substituted-2-nitrophenyl)propionic acids 2c-g were reductively cyclized by catalytic hydrogenation over platinum-on-carbon sulfided catalyst. In particular, this method was applied to α-chloro-β-(2-nitrophenyl)propionic acids bearing 4-methyl 2c , 4-ethyl 2d , 4-ethoxy 2e , 4-(n-butyl 2f and 4-phenyl 2g substituents to afford good yields of the corresponding 7-methyl 3c , 7-ethyl 3d , 7-ethoxy 3e , 7-(n-butyl) 3f , and 7-phenyl 3g substituted 3-chloro-3,4-dihydro-l-hydroxycarbostyrils. The various 4-substituted α-chloro-β-(2-nitrophenyl)propionic acids 2c-q were synthesized by reacting the in situ diazotized salts of the appropriate 4-substituted-2-nitroanilines in aqueous acetone with acrylic acid in the presence of cuprous chloride and hydrochloric acid. Intramolecular cyclization of 2-(allylthio)propionyl chloride ( 3a ) effected by aluminum chloride gave two dihydrothiopyran-3-ones ( 5a,6a ) and 5-chloro-2-methyltetrahydrothiopyran-3-one ( 7a ). Treatment of propargylthioacetyl chloride ( 4a ) with aluminum chloride afforded 5-chloro-3,6-dihydro-2H-thiopyran-3-one ( 10a ). 4-Chloro-3-(trifluoroacetyl)- and 4-chloro-3-(methoxalyl)coumarins react with electron-rich aminoheterocycles, dimethyl 1,3-acetonedicarboxylate, hydrazines, alkyl thioglycolates, and methyl sarcosinate to give a variety of 3,4-heteroannulated coumarins. The starting materials were prepared by the reaction of 4-hydroxycoumarin with trifluoroacetic anhydride and methyl 2-chloro-2-oxoacetate in the presence of trimethylsilylchloride. An NMR study and X-ray crystallographic analysis are reported.  相似文献

Resynthesis of NSC 341,964, which had been assigned structure 1 (1-[[3-(7-chloro-4-oxo-4H-3,1-benzoxazin-2-yl)phenyl]methyl]pyridinium chloride) was approached via 7-chloro-2-(3-methylphenyl)-4H-3,1-benzoxazin-4-one ( 5 ) obtained from 3-methylbenzoyl chloride ( 2 ) and 2-amino-4-chlorobenzoic acid ( 3 ) followed by dehydration in acetic anhydride. Radical bromination provided 6 which with pyridine afforded the bromide analog 7 of 1 . Ion exchange, however, gave ring-opened benzoic acid 8 rather than 1 . The original sample of NSC 341,964 also proved to be ring-opened material. However, 7 upon standing exhibited slow hydrolysis to 8 so that the structure of the original NSC 341,964 remains uncertain. A more direct route to compound 8 is also described.  相似文献

6-Chloro-2-chloromethylpyridine is prepared from 6-chloro-2-methylpyridine by a route in which the 2-Me substituent was successively converted to 2-acetoxymethyl, 2-hydroxymethyl and finally to the required 2-chloromethyl substituent. Attempts to simultaneously monochlorinate the Me group and reduce the N-oxide function of 6-chloro-2-methylpyridine-N-oxide with methanesulphonyl chloride and p-toluenesulphonyl chloride gave only very small yields of 6-chloro-2-chloromethyl-pyridine. 4-Chloro-2-chloromethylpyridine is prepared from 2-methylpyridine-N-oxide by nitration, followed by substitution of the 4-nitro group by chloro using conc HCl; side chain chlorination of the 2-Me group using p-toluenesulphonyl chloride yields 4-chloro-2-chloromethylpyridine. Phosphoryl chloride reacts with 2-chloromethylpyridine-N-oxide hydrochloride to yield only 14.4% of 4-chloro-2- chloromethylpyridine, together with 2-chloromethylpyridine (2.2%), 2-dichloromethylpyridine (41.6%) and 6-chloro-2- chloromethylpyridine (41.8%). Attempts to N-oxidise 2-chloromethylpyridine with peracids led to either 2-hydroxymethylpyridine (peracetic, m-chloroperbenzoic and performic acid) or no reaction (pertrifluoroacetic acid); none of the peracids led to any detectable N-oxidation.  相似文献

Cyclization of 2-chloro-6-[(3-chlorophenyl)thio]benzoic acid ( 2 ) gave a mixture of 1,8-, 3 , and 1,6-dichloro-9H-thioxanthen-9-ones 4 . The mixture was converted to 1,8-diamino- 7 , and 1-amino-6-chloro-9H-thioxanthen-9-ones 8 , from which 3 and 4 were prepared separately, respectively. From a mixture of 4 and 3,6-dichloro-9H-thioxanthen-9-one ( 11 ) obtained by cyclizing 4-chloro-2-[(3-chlorophenyl)thio]benzoic acid ( 10 ) was separated 11 by conversion of 4 to 8 .  相似文献

The reaction of 6-chloro-2-(1-methylhydrazino)quinoxaline 4-oxide 8 with acetic anhydride resulted in the intramolecular cyclization to give 8-chloro-2,4-dimethyl-4H-1,3,4-oxadiazino[5,6-b]quinoxaline 7a , while the reaction of compound 8 with acetic anhydride/pyridine or acetic anhydride/acetic acid afforded 3-(2,2-diacetyl-1-memymydrazmo)-7-chloro-2-oxo-1,2-dihydroquinoxaline 9 , effecting no intramolecular cyclization. The reaction of 2-(2-acetyl-1-methylhydrazino)-6-chloroquinoxaline 4-oxide 10a or 6-chloro-2-(1-methyl-2-trifluoroacetylhydrazino)quinoxaline 4-oxide 10b with phosphoryl chloride provided compound 7a or 8-chloro-4-memyl-2-trifluoromethyl-4H-1,3,4-oxadiazino[5,6-b]quinoxaline 7b , respectively. The reaction of compound 7b with phosphorus pentasulfide gave 7-chloro-3-(1-methyl-2-trifluoroacetylhydrazino)-2-thioxo-1,2-dihydroquinoxaline 11 , whose dehydration with sulfuric acid in acetic acid afforded 8-chloro-4-methyl-2-trifluoromemyl-4H-1,3,4-thiadiazino[5,6-b]quinoxaline 12 .  相似文献

We have obtained 2-(4-arylthiazol-2-yl)-4-chloro-3-oxobutyronitriles by acylation of 4-aryl-2-cyanomethylthiazoles with -chloroacetyl chloride. We have studied their reaction with secondary aliphatic amines, leading to formation of 4-dialkylamino-2-(4-arylthiazol-2-yl)-3-oxobutyronitriles, and also intramolecular alkylation with formation of 3-aryl-7-cyano-6(5H)-oxopyrrolo[2,1-b]thiazoles. We have determined some aspects of the tautomerism of the synthesized compounds.  相似文献

In the reaction of 2-(2-pyridylcarbonyl)benzoyl chloride, which exists in the form of 6,11-dioxo-6,11dihydrobenzo[b]quinolizinium chloride, with p-nitroaniline, 2-(4-nitrophenylimino)-6, 11-dihydro-2H-benzo-[b]quinolizine-6, I I -dione is unexpectedly formed. When it reacts with water or methanol there is an opening of the quinolizine ring and aromatization of the quinoid fragment with the formation of 2-[4-(4nitrophenylamino)-2-pyridylcarbonyl]benzoic acid or its methyl ester. Under the action of antimony pentachloride, 2- 2-quinolylcarbonyl)benzoylchloride-3 (2-quinolyl)-3-chloro- 1, 3-dihydrobenzo[c]furan-1 -one -is converted to 3-(2-quinolyl)-1,3-dihydrobenzo[c]furan-1-on-3-ylium hexachlorantimonate, which undergoes isomerizing recyclization upon heating to 7,12-dioxo-7,12-dihydrobenzo[b/ hexachloroantimonate. The latter enters into an analogous reaction with p-nitroaniline, thereby forming 5-(4-nitrophenylimino)-7,12-dihydro-5H-dibenzo[b,f]quinolizine-7,2-dione.Riga Technical University, Riga LV-1048. ABSTRACT

Selective substitution of the primary sulfonate group in 2, 3, 4, 5-tetra-0-methanesulfonyl-D-arabinose dimethyl acetal (1) gives 5-S-acetyl-2, 3, 4-tri-0-methanesulfonyl--5-thio-D-arabinose dimethyl acetal (2) which is further converted into the title compound (3). Reductive desul-furization of 3 afforded deoxy dimethyl acetal derivatives 5 and 6 in a low yield. Unsaturated monosaccharide derivative 7 was obtained as the only reaction product from 3 with triphenylphosphine. Catalytic hydrogenation of 7 gave dideoxy-sugar 6 in a satisfactory yield. Finally, epi-sulfide 3 with acetyl chloride afforded 4-chloro-4-deoxy derivative 4, which can be recycled into the starting 3.  相似文献

A facile method for the synthesis of a new series of R,R-2-(4-(2-(4-(5-chloro-3-halopyridin-2-yloxy)-phenoxy)-propionyloxy)-phenoxy) -propionic acid ester derivatives containing a halo-substituted pyridine carrying two R,R chiral centers from(R)-2-(4- hydroxyphenoxy)propionic acid,halopyridines and alcohols using base/cyanuric chloride/catalyst system is reported.Their herbicidal activities against grass weeds and crops selectivity were evaluated.  相似文献

Chloromethylation of 4- and 5-chlorobenzo-2,1,3-thiadiazoles with dichlorodimethyl ether in the presence of anhydrous aluminum chloride gives 4-chloro-7-chloromethyl- and 5-chloro-4-chloromethylbenzo-2, 1,3-thiadiazoles respectively. Reductive scission followed by treatment with thionyl chloride converts them to 4-chloro-7-methyl- and 5-chloro-4-methylbenzo-2,1,3-thiadiazoles; Chlorination of the latter gives 4-methyl-5,7-dichlorobenzo-2,1,3-thiadiazole. Replacement of the chlorine in the chloromethyl groups gives 4-chloro-7-hydrosymethyl-,5-chloro-4-hydroxymethyl-, 4-chloro-7-cyanomethyl-,4-chloro-7-carboxymethyl-,5-chloro-4-carboxymethylbenzo-2,1,3-thiadiazoles. Reductive scission of 4-chlorobenzo-2,1,3-thidiazole followed by treatment with sodium selenite gives 4-chlorobenzo-2,1,3-selenadiazole.Part XLIII see [1].  相似文献

Reaction of 2-[(organylthio)methyl]oxiranes with acetic anhydride gives a mixture of 3-(organylthio)-1,2-diacetoxypropane and 2-(organylthio)-1,3-diacetoxypropane, and with acetyl chloride a mixture of 2-chloro-3-(organylthio)-1-acetoxypropane and 3-chloro-2-(organylthio)-1-acetoxypropane.
