Direct electrochemistry and electrocatalysis of heme proteins, such as hemoglobin (Hb), myoglobin (Mb), and horseradish peroxidase (HRP), incorporated in gluten biopolymer films cast on pyrolytic graphite (PG) electrodes, were studied by voltammetry and amperometry. All the three protein-gluten films exhibited a pair of well-defined, quasi-reversible cyclic voltammetric peaks at about −0.28 V versus saturated calomel electrode (SCE) in pH 5.5 buffers, respectively, characteristic of the heme Fe(III)/Fe(II) redox couples, indicating enhanced electron transfer between the proteins and PG electrodes in a gluten film environment. The protein-gluten hydrogel films showed excellent stability. Positions of Soret absorption band of protein-gluten films suggested that the heme proteins kept their secondary structure similar to their native state in the films in the medium pH range. The heme proteins in gluten films were act as a biologic catalyst to catalyze reduction of oxygen or hydrogen peroxide. The voltammetric or amperometric responses of H2O2 at the protein-gluten film electrodes could be used to determine the concentration of H2O2 in solution.  相似文献

Stable heme proteins entrapped in dimethylformamide (DMF)–chitosan organohydrogel films modified electrodes were operated in neat hydrophilic room-temperature ionic liquid (IL) [bmim][BF4] for the first time. The modified electrodes possess outstanding electrochemical response in [bmim][BF4] without adding water. The morphology studies of films were demonstrated by atomic force microscopy (AFM). UV–Vis and FTIR spectroscopy showed that the heme proteins retained their native structure in organohydrogel films. Direct electrochemistry and bioelectrocatalysis of heme protein–organohydrogel films were investigated. Several electrochemical parameters such as the charge transfer coefficients (α) and the apparent electron transfer rate constant (ks) of these processes were calculated by performing nonlinear regression analysis of square wave voltammetry (SWV) experimental dates. Furthermore, high electrocatalytic activity to hydrogen peroxide (H2O2) was observed, indicating that heme proteins entrapped in organohydrogel films retained their bioelectrocatalytic activities in [bmim][BF4]. Kinetic analysis of the cyclic voltammetry dates shows that heme protein–organohydrogel films operated in IL bring up to an enhancement of the biosensor sensitivity and a high affinity for H2O2.  相似文献

Amperometric biosensors were prepared by immobilizing heme proteins in single-wall carbon nanotubes (SWCNTs)-cetyltrimethyl ammonium bromide (CTAB) surfactant films based on 1-butyl-3-methylimidazolium hexafluorophosphate ([bmim][PF6]) as nonaqueous electrolyte. The electrochemistry and electrocatalysis of protein-SWCNTs-CTAB films were investigated. A pair of well-defined and nearly reversible redox peaks was observed when the heme protein films were immersed into [bmim][PF6]. Moreover, the electrocatalytic activity of heme proteins for two model peroxides compounds (hydrogen peroxide (H2O2) and tert-butyl hydroperoxide (t-BuOOH)) was observed. Some electrocatalytic parameters for heme proteins were calculated. Compared with the detection results of t-BuOOH in other organic solvents, kinetic analysis in this experiment indicates that heme protein-SWCNTs-CTAB films operated in ionic liquid provide a higher affinity and sensitivity.  相似文献

The adsorption processes and electrochemical behavior of 4-nitroaniline (4-NA) adsorbed onto glassy carbon electrodes (GCE) have been investigated in aqueous 0.1 M nitric acid (HNO3) electrolyte solutions using cyclic voltammetry (CV). 4-NA adsorbs onto GCE surfaces, and upon potential cycling past −0.2 V, is transformed into the arylhydroxylamine (ArHA) derivative which exhibits a well-behaved pH dependent redox couple centered at 0.32 V at pH 1.5. It is noted as arylhydroxylamine modified glassy carbon electrodes (HAGCE). This modified electrode can be readily used as an immobilization matrix to entrap proteins and enzymes. In our studies, myoglobin (Mb) was used as a model protein for investigation. A pair of well-defined reversible redox peaks of Mb (Fe(III)-Fe(II)) was obtained at the Mb/arylhydroxylamine modified glassy carbon electrode (Mb/HAGC) by direct electron transfer between the protein and the GCE. The formal potential (E0), the apparent coverage (Γ*) and the electron-transfer rate constant (ks) were calculated as −0.317 V, 8.26 × 10−12 mol/cm2 and 51 ± 5 s−1, respectively. Dramatically enhanced biocatalytic activity was exemplified at the Mb/HAGC electrode by the reduction of hydrogen peroxide (H2O2), trichloroacetic acid (TCA) and oxygen (O2). The Mb/arylhydroxylamine film was also characterized by UV-visible spectroscopy (UV-vis), scanning electron microscope (SEM) indicating excellent stability and good biocompatibility of the protein in the arylhydroxylamine modified electrode. This new Mb/HAGC electrode exhibited rapid electrochemical response (2 s) for H2O2 and had good stability in physiological condition, showing the potential applicability of the films in the preparation of third generation biosensors or bioreactors based on direct electrochemistry of the proteins.  相似文献

The pre-grafted screen-printed gold electrode modified with phenyl-amino monolayer was investigated for covalent immobilization of phenyl-amine functionalized single-walled carbon nanotubes (PA-SWCNT) and metal tetra-amino phthalocyanine (MTAPc) using Schiff-base reactions with benzene-1,4-dicarbaldehyde (BDCA) as cross-linker. The PA-SWCNT and MTAPc modified electrodes were applied as hybrids for electrochemical sensing of H2O2. The step-by-step fabrication of the electrode was followed using electrochemistry, impedance spectroscopy, scanning electron microscopy and Raman spectroscopy and all these techniques confirmed the fabrication and the immobilization of PA-SWCNT, MnTAPc and CoTAPc onto gold surfaces. The apparent electron transfer constant (kapp) showed that the carbon nanotubes and metallo-phthalocyanines hybrids possess good electron transfer properties compared to the bare, pre-grafted and the MTAPc modified gold electrode surfaces without PA-SWCNT. The electrochemical sensing of hydrogen peroxide was successful with PA-SWCNT-MTAPc hybrid systems showing higher electrocatalytic currents compared to the other electrodes. The analytical parameters obtained using chronoamperometry gave good linearity at H2O2 concentrations ranging from 1.0 to 30.0 μmol L−1. The values for the limit of detection (LoD) were found to be of the orders of 10−7 M using the 3δ for all the electrodes. The PA-SWCNT-MTAPc modified SPAuEs were much more sensitive compared to PA-MTAPc modified SPAuEs.  相似文献

This paper describes the direct electrochemistry and electrocatalysis of myoglobin immobilized on graphene‐cetylramethylammonium bromide (CTAB)‐ionic liquid nanocomposite film on a glassy carbon electrode. The nanocomposite was characterized by transmission electron microscopy, scanning electron microscopy, X‐ray photoelectron spectroscopy, and electrochemistry. It was found that the high surface area of graphene was helpful for immobilizing more proteins and the nanocomposite film could provide a favorable microenvironment for MB to retain its native structure and activity and to achieve reversible direct electron transfer reaction at an electrode. The ionic liquid may play dual roles here: it keeps the protein's activity and improves stability of the nanocomposite film; it also serves as a binder between protein and electrode, therefore, enhancing the electron transfer between the protein and the electrode. The nanocomposite films also exhibit good stability and catalytic activities for the electrocatalytic reduction of H2O2.  相似文献

A novel nanocomposite integrating the good biocompatibility of polyacrylic resin nanoparticles (PAR) and the good conductivity of colloidal gold nanoparticles was proposed to construct the matrix for the immobilization of hemoglobin (Hb) on the surface of a glassy carbon electrode (GCE). UV‐vis spectra demonstrated that Hb preserved its native structure after being entrapped into the composite film. The direct electrochemistry of hemoglobin (Hb) in this nanocomposite films showed a pair of well‐defined and quasi‐reversible cyclic voltammetric peaks with a formal potential of ?0.307 mV and a constant electron transfer rate of 2.51±0.2 s?1. The resultant amperometric biosensor showed fast responses to the analytes with excellent detection limits of 0.2 µM for H2O2 and 0.89 µM for TCA (S/N=3), and high sensitivity of 1108.6 for H2O2 and 77.14 mA cm?2 M?1 for TCA, respectively. The linear current response was found in the range from 0.59 to 7.3 µM (R2=0.9996) for H2O2 and from 5 to 85 µM (R2=0.9996) for TCA, while the superior apparent Michaelis–Menten constant was 0.012 mM for H2O2 and 0.536 mM for TCA, respectively. Therefore, the PAR‐Au‐Hb nanocomposite as a novel matrix opens up a possibility for further study on the direct electrochemistry of other proteins.  相似文献

A novel multi-function Metal-Organic Framework composite Ag@Zn-TSA (zinc thiosalicylate, Zn(C7H4O2S), Zn-TSA) was synthesized as highly efficient immobilization matrixes of myoglobin (Mb)/glucose oxidase (GOx) for electrochemical biosensing. The electrochemical biosensors based on Ag@Zn-TSA composite and ionic liquid (IL) modified carbon paste electrode (CPE) were fabricated successfully. Furthermore, the properties of the sensors were discussed by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and amperometric current-time curve, respectively. The results showed the proposed biosensors had wide linear response to hydrogen peroxide (H2O2) in the range of 0.3–20,000 μM, to nitrite (NO2) for 1.3 μM–1660 μM and 2262 μM–1,33,000 μM, to glucose for 2.0–1022 μM, with a low detection limit of 0.08 μM for H2O2, 0.5 μM for NO2, 0.8 μM for glucose. The values of the apparent heterogeneous electron transfer rate constant (ks) for Mb and GOx were estimated as 2.05 s−1 and 2.45 s−1, respectively. Thus, Ag@Zn-TSA was a kind of ideal material as highly efficient immobilization matrixes for sensitive electrochemical biosensing. In addition, this work indicated that MOF nanocomposite had a great potential for constructing wide range of sensing interface.  相似文献

A novel matrix, gold nanoparticles-bacterial cellulose nanofibers (Au-BC) nanocomposite was developed for enzyme immobilization and biosensor fabrication due to its unique properties such as satisfying biocompatibility, good conductivity and extensive surface area, which were inherited from both gold nanoparticles (AuNPs) and bacterial cellulose nanofibers (BC). Heme proteins such as horseradish peroxidase (HRP), hemoglobin (Hb) and myoglobin (Mb) were successfully immobilized on the surface of Au-BC nanocomposite modified glassy carbon electrode (GCE). The immobilized heme proteins showed electrocatalytic activities to the reduction of H2O2 in the presence of the mediator hydroquinone (HQ), which might be due to the fact that heme proteins retained the near-native secondary structures in the Au-BC nanocomposite which was proved by UV-vis and IR spectra. The response of the developed biosensor to H2O2 was related to the amount of AuNPs in Au-BC nanocomposite, indicating that the AuNPs in BC network played an important role in the biosensor performance. Under the optimum conditions, the biosensor based on HRP exhibited a fast amperometric response (within 1 s) to H2O2, a good linear response over a wide range of concentration from 0.3 μM to 1.00 mM, and a low detection limit of 0.1 μM based on S/N = 3. The high performance of the biosensor made Au-BC nanocomposite superior to other materials as immobilization matrix.  相似文献

Polymerized ionic liquid-wrapped carbon nanotubes (PIL-CNTs) were firstly designed for direct electrochemistry and biosensing of redox proteins. The CNTs were coated successfully with polymerized ionic liquid (PIL) layer, as verified by transmission electron microscopy (TEM), thermogravimetric analysis (TGA) and Fourier transform infrared (FT-IR) spectroscopy. The PIL-CNTs were dispersed better in water and showed superior electrocatalysis toward O2 and H2O2 comparing to pristine CNTs and the mixture of IL monomer and CNTs. With glucose oxidase (GOD) as a protein model, the direct electrochemistry of the redox protein was investigated on the PIL-CNTs modified glassy carbon (GC) electrode and excellent direct electrochemical performance of GOD molecules was observed. The proposed biosensor (GOD/PIL-CNTs/GC electrode) displayed good analytical performance for glucose with linear response up to 6 mM, response sensitivity of 0.853 μA mM−1, good stability and selectivity.  相似文献

Direct electrochemistry of hemoglobin (Hb) is observed at carbon nanotube (CNT) interface. The adsorbing Hb can transfer electron directly at CNT interface compared with common carbon material. The heterogeneous electron transfer rate constant k of Hb can be calculated as 0.062 s−1, the transfer coefficient α is 0.21 and the average surface coverage of Hb on CNT surface is 3.58 × 10−9 ± 2.7 × 10−10 mol/cm2. It is found that the adsorbing Hb still keeps its catalytic activity to H2O2. This sensor was used to detect H2O2. The apparent Michaelis-Menten constant is calculated as 6.75 × 10−4 mol L−1.  相似文献

TiO2-graphene nanocomposite was prepared by hydrolysis of titanium isopropoxide in colloidal suspension of graphene oxide and in situ hydrothermal treatment. The direct electrochemistry and electrocatalysis of hemoglobin in room temperature ionic liquid 1-Butyl-3-methylimidazolium hexafluorophosphate, chitosan and TiO2-graphene nanocomposite modified glassy carbon electrode were investigated. The biosensor was examined by using UV-vis spectroscopy, scanning electron microscopy and electrochemical methods. The results indicated that hemoglobin remained its bioactivity on the modified electrode, showing a couple of well-defined and quasi-reversible redox peaks, corresponding to hemoglobin FeIII/FeII couple. The kinetic parameters for the electrode reaction, such as the formal potential (Eo'), the electron transfer rate constant (ks), the apparent coverage (Γ), and Michaelis–Menten constant (Km) were evaluated. The biosensor showed good electrochemical responses to the reduction of H2O2 in the ranges of 1–1170 μM. The detection limit was 0.3 μM (S/N = 3). The properties of this composite film, together with the bioelectrochemical catalytic activity, could make them useful in the development of bioelectronic devices, and investigation of electrochemistry of other heme proteins at functional interface.  相似文献

A novel cobalt-tetraphenylporphyrin/reduced graphene oxide (CoTPP/RGO) nanocomposite was prepared by a π–π stacking interaction and characterized by ultraviolet–visible absorption spectroscopy (UV–vis), Fourier transform infrared spectroscopy (FTIR) and electrochemical impedance spectroscopy (EIS). The CoTPP/RGO nanocomposite exhibited high electrocatalytic activity both for oxidation and reduction of H2O2. The current response was linear to H2O2 concentration with the concentration range from 1.0 × 10−7 to 2.4 × 10−3 mol L−1 (R = 0.998) at the reductive potential of −0.20 V and from 1.0 × 10−7 to 4.6 × 10−4 mol L−1 (R = 0.996) at the oxidative potential of +0.50 V. The H2O2 biosensor showed good anti-interfering ability towards oxidative interferences at the oxidative potential of +0.50 V and good anti-interfering ability towards reductive interferences at the reductive potential of −0.20 V.  相似文献

The transition metal-substituted heteropolyoxoanion, Cd4(H2O)2(As2W15O56)212− (As4W30Cd4), is one of the trivacant Dawson derivatives. Its redox electrochemistry has been studied in acid buffer solutions using cyclic voltammetry. It exhibited three steps of four-electron redox waves attributed to redox processes of the tungsten-oxo framework. Through layer-by-layer assembly, the compound was first successfully immobilized on a 4-aminobenzoic acid modified glassy carbon electrode surface by alternate deposition with a quaternized poly(4-vinylpyridine) partially complexed with [Os(bpy)2Cl]2+/+ (denoted as QPVP-Os). Thus, prepared multilayer films have been characterized by cyclic voltammetry (CV), X-ray photoelectron spectroscopy (XPS) and UV-vis spectroscopy (UV-vis). The electrocatalytic activities of the multilayer films containing As4W30Cd4 have been investigated on the reduction of three substrates of important analytical interests, NO2, BrO3 and IO3. And with the increase of the number of As4W30Cd4 layers, the catalytic current towards the reduction of BrO3 was enhanced and the catalytic potential shifted positively.  相似文献

In this report, a highly sensitive amperometric sensor based on MnO2-modified vertically aligned multiwalled carbon nanotubes (MnO2/VACNTs) for determination of hydrogen peroxide (H2O2) was fabricated by electrodeposition. The morphology of the nanocomposite was characterized by scanning electron microscopy, energy-dispersive X-ray spectrometer and X-ray diffraction. Cyclic voltammetry, chronoamperometry and electrochemical impedance spectroscopy were applied to investigate the electrochemical properties of the MnO2/VACNTs nanocomposite electrode. The mechanism for the electrochemical reaction of H2O2 at the MnO2/VACNTs nanocomposite electrode was also discussed. In borate buffer (pH 7.8, 0.20 M), the MnO2/VACNTs nanocomposite electrode exhibits a linear dependence (R = 0.998) on the concentration of H2O2 from 1.2 × 10−6 M to 1.8 × 10−3 M, a high sensitivity of 1.08 × 106 μA M−1 cm−2 and a detection limit of 8.0 × 10−7 M (signal/noise = 3). Meanwhile, the MnO2/VACNTs nanocomposite electrode is also highly resistant towards typical inorganic salts and some biomolecules such as acetic acid, citric acid, uric acid and d-(+)-glucose, etc. In addition, the sensor based on the MnO2/VACNTs nanocomposite electrode was applied for the determination of trace of H2O2 in milk with high accuracy, demonstrating its potential for practical application.  相似文献

Electrocatalytic activities of graphene nanosheets/Nile blue nanocomposite, synthesized and adsorbed simultaneously on the glassy carbon (GC?GNs?NB) electrode, are investigated. The nanocomposite was characterized by ATR?FTIR, FESEM and voltammetry. Activity of the electrode toward reduction of H2O2 and oxidation of NO2? was studied electrochemically. Values of 1.95 and 0.730 mM are found for the Michaelis?Menten constant of the electrode toward H2O2 and NO2?, respectively. Wide dynamic response ranges were observed for the electrode, with DLs of 0.22 μM H2O2 and 1.1 μM NO2?. Effect of interferences was studied. The sensor was successfully tested for H2O2 and NO2? contents in real samples, respectively.  相似文献

A highly sensitive NOx sensor was designed and developed by electrochemical incorporation of copper nanoparticles (CuNP) on single-walled carbon nanotubes (SWCNT)-polypyrrole (PPy) nanocomposite modified Pt electrode. The modified electrodes were characterized by scanning electron microscopy and energy dispersive X-ray analysis. Further, the electrochemical behavior of the CuNP-SWCNT-PPy-Pt electrode was investigated by cyclic voltammetry. It exhibited the characteristic CuNP reversible redox peaks at −0.15 V and −0.3 V vs. Ag/AgCl respectively. The electrocatalytic activity of the CuNP-SWCNT-PPy-Pt electrode towards NOx is four-fold than the CuNP-PPy-Pt electrode. These results clearly revealed that the SWCNT-PPy nanocomposite facilitated the electron transfer from CuNP to Pt electrode and provided an electrochemical approach for the determination of NOx. A linear dependence (r2 = 0.9946) on the NOx concentrations ranging from 0.7 to 2000 μM, with a sensitivity of 0.22 ± 0.002 μA μM−1 cm−2 and detection limit of 0.7 μM was observed for the CuNP-SWCNT-PPy-Pt electrode. In addition, the sensor exhibited good reproducibility and retained stability over a period of one month.  相似文献

A recently developed atmospheric pressure ionization source, a distributed plasma ionization source (DPIS), was characterized and compared to commonly used atmospheric pressure ionization sources with both mass spectrometry (MS) and ion mobility spectrometry (IMS). The source consisted of two electrodes of different sizes separated by a thin dielectric. Application of a high RF voltage across the electrodes generated plasma in air yielding both positive and negative ions. These reactant ions subsequently ionized the analyte vapors. The reactant ions generated were similar to those created in a conventional point-to-plane corona discharge ion source. The positive reactant ions generated by the source were mass identified as being solvated protons of general formula (H2O)nH+ with (H2O)2H+ as the most abundant reactant ion. The negative reactant ions produced were mass identified primarily as CO3, NO3, NO2, O3 and O2 of various relative intensities. The predominant ion and relative ion ratios varied depending upon source construction and supporting gas flow rates. A few compounds including drugs, explosives and amines were selected to evaluate the new ionization source. The source was operated continuously for 3 months and although surface deterioration was observed visually, the source continued to produce ions at a rate similar that of the initial conditions.  相似文献

Graphene/Fe3O4 nanocomposite was prepared for the immobilization of hemoglobin (Hb) to improve the electron transfer between Hb and glass carbon electrode (GCE). The characterization of nanocomposites was described by transmission electron microscopy, Fourier transform infrared, Raman spectroscopy, and X-ray photoelectron spectroscopy, respectively. The electrochemistry of Hb on the graphene/Fe3O4-based GCE was investigated by cyclic voltammetry and amperometric measurement. The modified electrode showed a wide linear range from 0.25 μmol/L to 1.7 mmol/L with a correlation coefficient of 0.9967. The detection limit of the H2O2 biosensor was estimated at 6.0?×?10?6?mol/L at a signal-to-noise ratio of 3.  相似文献

Phosphomolybdate, H3PMo12O40, (PMo12)-doped-poly(3,4-ethylenedioxythiophene) (PEDOT) coated gold nanoparticles have been synthesized in aqueous solution by reduction of AuCl4 using hydroxymethyl EDOT as a reducing agent in the presence of polystyrene sulfonate and PMo12. The resulting PMo12-doped-PEDOT stabilized Au nanoparticles are water soluble and have been characterized by UV–visible spectroscopy, scanning electron microscopy and electrochemistry. Glassy carbon electrodes modified with these Au nanoparticles show excellent stability and catalytic activity towards the reduction of bromate in an aqueous electrolyte solution containing 10 mM H2SO4 and 0.1 M Na2SO4.  相似文献
