In China, Semen Cassiae has long been used to protect liver, brighten eyes, and relieve constipation. Prepared Semen Cassiae is produced from raw Semen Cassiae by processing, the two forms of Semen Cassiae have different clinical applications. Pathological state is an important factor affecting the efficacy of drugs, the pharmacokinetic behavior of drugs could be significantly changed when people or animal were under different pathological state. To clarify the effect of processing mechanism and pathological state for pharmacokinetic behavior, the pharmacokinetics of nine components of raw and prepared Semen Cassiae under normal and acute liver injury rats were examined. The results showed that the bimodal phenomenon appeared on the plasma concentration‐time profiles of obtusin, emodin, chrysophanol, aloe emodin and rhein. The Tmax of aurantio‐obtusin, obtusin, chrysoobtusin, emodin, chrysophanol, aloe emodin, physcion in normal groups administrated prepared Semen Cassiae were shorter than those administrated raw Semen Cassiae. For the AUC0–t, aurantio‐obtusin, obtusin, chrysoobtusin, chrysophanol, aloe emodin and physcione in model groups administrated prepared Semen Cassiae were significantly higher than other groups, unlike above components, rhein had poor absorption in model groups. The study would be useful for further studies on pharmacokinetics and clinical application of raw and prepared Semen Cassiae.  相似文献

Aloe‐emodin, a natural polyphenolic anthraquinone, has shown various beneficial bioactivities in vitro. The aim of this study was to investigate the pharmacokinetics and metabolism of aloe‐emodin. Aloe‐emodin was intravenously and orally administered to rats. The concentrations of aloe‐emodin and rhein, a metabolite of aloe‐emodin, were determined by HPLC method prior to and after hydrolysis with β‐glucuronidase and sulfatase/β‐glucuronidase. The results showed that the systemic exposures of aloe‐emodin and its metabolites were ranked as aloe‐emodin glucuronides (G) > rhein sulfates (S) > aloe‐emodin > rhein and rhein G when aloe‐emodin was given intravenously. In contrast, when aloe‐emodin was administered orally, the parent form of aloe‐emodin was not absorbed per se, and the systemic exposures of its metabolites were ranked as aloe‐emodin G > rhein G > rhein. Prepared rhubarb, as one of the main processed products of rhubarb, has a good effect on promoting blood circulation. In this paper we describe a rapid, sensitive, and selective ultra‐fast liquid chromatography with tandem mass spectrometry method for simultaneous quantification of five anthraquinones (rhein, aloe‐emodin, chrysophanol, emodin, and physcion) and gallic acid in plasma. Chromatographic separation was performed on an Extend C18 column at the temperature of 30°C using a mobile phase that consisted of 0.1% aqueous formic acid and acetonitrile. Satisfactory linearity, precision, accuracy, extraction recovery, and matrix effect have been achieved. Then, the validated method was successfully applied to a comparative pharmacokinetic study. The results might be helpful for guiding clinical application of prepared rhubarb in the future.  相似文献

This work concentrates on the in vitro metabolism of anthraquinone derivatives from rhubarb. Different metabolic reactions for anthraquinone derivatives were dependent on the substituent group in the skeleton. Monohydroxylation in the exocyclic methyl group was the major metabolic modification for emodin. Aloe‐emodin containing a hydroxymethyl group was mainly metabolized through a methylation reaction. For rhein, both hydrogenation and methyl substitution on the benzene ring were the major metabolic reactions. Hydroxyl substitution on the benzene ring was the predominant metabolic reaction for chrysophanol. Dahuang‐mudan decoction (DMD) has been widely used for disease treatment in China for 1700 years. The formula consists of Rhubarb, moutan bark, Prunus persica, wax gourd kernel and mirabilite, which have been well studied by multidisciplinary approaches. However, the role of the mineral mirabilite in DMD is unclear. The objective of this study was to investigate the effects of mirabilite on the absorption and pharmacokinetics of the ingredients in DMD. The constituents were identified in DMD extract and the plasma of mirabilite–DMD (MDMD, 50 g kg?1) treated rats and nonmirabilite–DMD (NMDMD, 50 g kg?1) treated rats. The plasma was also used to investigate the effects of mirabilite on the pharmacokinetics of active ingredients in DMD using a new validated UPLC–MS/MS method. The results showed that 63 compounds were identified in the extract of DMD, 27 and 22 of which were found in the plasmas of MDMD‐ and NMDMD‐treated rats, respectively. Furthermore, the results of a pharmacokinetic study suggested that mirabilite influenced the absorption of the five constituents by decreasing the absorption of emodin and rhein while increasing the absorption of aloe‐emodin, paeoniflorin and amygdalin; the pharmacokinetic parameters, including the Tmax, Cmax, AUC0–t, MRT0–t, CLz and t1/2 of five constituents, significantly changed in MDMD‐treated rats compared with the NMDMD. The method validation for selectivity, precision, accuracy, matrix effect, recovery and stability met the acceptance criteria. These findings uncover the roles of mirabilite in DMD and demonstrate the application of scientific principles to the study of DMD in human health care.  相似文献

The structural and electronic properties of hydroxyanthraquinone derivatives in rhubarb, namely, chrysophanol, emodin, physcion, aloe‐emodin, rhein, and their radicals were investigated at density functional level. The bifurcated hydrogen bond property of the studied structures was investigated using the atoms in molecules theory. It turned out that the presence of the dihydroxy functionality increases the radical stability through hydrogen bonds formation and favors hydrogen atom abstraction. Bond dissociation energy and ionization potential were also determined to know if the radical scavenging activity of these compounds proceeds via an H‐atom or an electron‐transfer mechanism. © 2010 Wiley Periodicals, Inc. A highly sensitive method using reduced graphene oxide with iron oxide (rGO/Fe3O4) as the sorbent in magnetic SPE has been developed for the purification of five anthraquinones (emodin, rhein, aloeemodin, physcion, and chrysophanol) in rhubarb and rat urine by ultra‐HPLC coupled with quadrupole TOF/MS. The extraction was accomplished by adding trace amount rGO/Fe3O4 suspension to 200 mL of aqueous mixture, and the excellent adsorption capacity of the nanoparticles was fully demonstrated in this procedure. Under the optimized conditions, the calibration curves were linear in the concentration range of 0.05–27.77 ng/mL with correlation coefficients varying from 0.9902 to 0.9978. The LODs ranged from 0.28 to 58.99 pg/mL. The experimental results indicated that the proposed method was feasible for the analysis of anthraquinones in rhubarb and urine samples.  相似文献

This study presents an efficient strategy based on liquid–liquid extraction, high‐speed counter‐current chromatography, and preparative HPLC for the rapid enrichment, separation, and purification of four anthraquinones from Rheum tanguticum. A new solvent system composed of petroleum ether/ethyl acetate/water (4:2:1, v/v/v) was developed for the liquid–liquid extraction of the crude extract from R. tanguticum. As a result, emodin, aloe‐emodin, physcion, and chrysophanol were greatly enriched in the organic layer. In addition, an efficient method was successfully established to separate and purify the above anthraquinones by high‐speed counter‐current chromatography and preparative HPLC. This study supplies a new alternative method for the rapid enrichment, separation, and purification of emodin, aloe‐emodin, physcione, and chrysophanol.  相似文献

A rapid and sensitive high‐performance liquid chromatography–mass spectrometric (HPLC‐MS) method was developed and validated for simultaneous determination of benzoylhypaconine (BHA), benzoylmesaconine (BMA), benzoylaconine (BAC) and hypaconitine (HA) in rat plasma for the first time. The analytes were separated on a Kromasil C18 column with a total running time of 11 min. The validation data demonstrated a sound feasibility for the newly developed method and it was then applied to the pharmacokinetic study of these analytes in rats. Pharmacokinetic behaviors of BHA, BMA, BAC and HA in rats were studied after oral administration of Radix Aconiti Lateralis Praeparata extract (FZ) and Dahuang Fuzi Decoction (DFD). The main parameters for the two groups of subjects were compared, and significant differences between Radix Aconiti Lateralis Praeparata extract group and Dahuang Fuzi Decoction group in calculated parameters, such as the area under the plasma concentration–time from zero to the last quantifiable time‐point (AUC0–t), the area under the plasma concentration–time curve from zero to infinity (AUC0–∞), peak plasma concentration (Cmax), half‐life of elimination (T1/2), mean retention time (MRT0–t), plasma clearance (CL), volume of distribution (Vd) and time to reach Cmax (Tmax), were found. After oral administration of DFD, the AUC0–t, AUC0–∞ and Cmax of BHA, BMA, BAC and HA decreased remarkably (p < 0.05) compared with those of the FZ extract group. Vd and CL values of BHA, BMA, BAC and HA increased, two of which showed significant difference (p < 0.05). T1/2 and MRT0–t values of BHA, BMA and BAC in the DFD group were significantly delayed compared with those of FZ extract group. Only the Tmax of HA, the toxic ingredient in FZ, delayed significantly in DFD group compared with the value of FZ group. Docetaxel, frequently used for the treatment of breast cancer, is mainly metabolized via hepatic cytochrome P450 (CYP) 3A in humans and is also a substrate of P‐glycoprotein (P‐gp). Wogonin has been shown to be able to modulate the activities of CYPs and P‐gp, and it could serve as an adjuvant chemotherapeutic agent. However, the impacts of co‐administration of wogonin and docetaxel on their pharmacokinetics have not been studied because of a lack of an analytical method for their simultaneous measurement. In the present study, we established an HPLC–MS/MS method for simultaneous measurement of wogonin and docetaxel in rat plasma, and it was then utilized to explore the pharmacokinetics of wogonin and the herb–drug interactions between wogonin and docetaxel after their combined administration in rats with mammary tumors. The rats received 10, 20 and 40 mg/kg wogonin via oral administration, with or without docetaxel intravenously administered at 10 mg/kg, and the plasma concentrations of wogonin and docetaxel were measured using the established and validated HPLC–MS/MS method. The Cmax and AUC0–t of wogonin were proportionally increased in the dose range from 10 to 40 mg/kg, suggesting a linear pharmacokinetics of wogonin. Moreover, the Cmax and AUC0–t of docetaxel and the AUC0–t of wogonin were increased after co‐administration (p < 0.05), indicating increased in vivo exposures of both wogonin and docetaxel, which might lead to an increase in not only therapeutic but also toxic effects. Thus the alterations of pharmacokinetics should be taken into consideration when wogonin and docetaxel are co‐administered.  相似文献

A sensitive high-performance liquid chromatographic method with fluorescence detection (excitation 435 and emission 515 nm) was established and validated for quantification of five anthraquinones (aloe-emodin, rhein, emodin, chrysophanol and physcion) in rat plasma. Following a single-step liquid-liquid extraction, the analytes and internal standard (1,8-dihydroxyanthraquinone) were separated on a reversed-phase C(18) column with water-phosphoric acid-methanol as mobile phase at a flow rate of 1 mL/min. The linear ranges of the calibration curves were 6.5-1300 ng/mL for aloe-emodin, 20-4000 ng/mL for rhein, 40-8000 ng/mL for emodin, 15-3000 ng/mL for chrysophanol and 13-2600 ng/mL for physcion. The lower limit of quantification was 6.5 ng/mL for aloe-emodin, 20 ng/mL for rhein, 40 ng/mL for emodin, 15 ng/mL for chrysophanol and 13 ng/mL for physcion. The mean accuracy was 94.3-105.1% for aloe-emodin, 90.3-108.8% for rhein, 92.6-106.7% for emodin, 95.8-103.8% for chrysophanol and 98.7-101.2% for physcion. The within-batch and between-batch precisions were < or = 5.5% and < or = 13.4%, respectively. This method is suitable for determining the five anthraquinones in plasma simultaneously and thus investigating the pharmacokinetics of anthraquinones from Xiexin decoction in rats.  相似文献

Lizhong decoction (LZD), a classic formula, has been used to treat ulcerative colitis (UC) for thousands of years in clinical practice. However, the pharmacokinetic characteristics of its major bioactive components in rats under different physiological and pathological states are not clear. Thus, in this study, a rapid and sensitive analytical method, ultra‐performance liquid chromatography coupled with mass spectrometry (UPLC–MS/MS) method, was developed and applied to simultaneously determine glycyrrhizic acid, liquiritin, isoliquiritin, glycyrrhizin, isoliquiritigenin, 6‐gingerol, ginsenoside Rg1, ginsenoside Rb1 and ginsenoside Re in normal and UC rats after oral administration of LZD extract. A Waters BEH C18 UPLC column was used for chromatographic separation, while acetonitrile and 0.1% formic acid were selected as mobile phase. The linearity of nine analytes was >0.9920. Inter‐ and intra‐day accuracy was ≤ 11.4% and precision was from 1.1 to 12.7%. Additionally, stable and suitable extraction recoveries were also obtained. The established method was validated and found to be specific, accurate and precise for nine analytes. Furthermore, it was successfully applied to the pharmacokinetic investigation of nine major components after oral administration of LZD extracts to normal and model rats, respectively. The results showed that the pharmacokinetic parameters (Cmax, Tmax, AUC0–t, AUC0–∞) in the plasma of UC rats were significantly different from those of normal rats, which could provide a reference for the clinical application of LZD.  相似文献

A simple, fast and reliable high‐performance liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous quantification and pharmacokinetic study of three flavonoids (liquiritigenin, isoliquiritigenin and formononetin) and three anthraquinones (emodin, rhein and aloe‐emodin), which are the bioactive ingredients of Wei‐Chang‐Shu tablet found in rat plasma. After extraction by liquid–liquid extraction with ethyl acetate, chromatographic separation was achieved on an Agilent Zorbax SB‐C18 column (4.6 × 150 mm, 5 μm) at a flow rate of 1 mL/min by gradient elution using 0.1% aqueous acetic acid and acetonitrile. The detection was performed using a triple quadrupole mass spectrometer equipped with electrospray ionization source in the negative ionization and selected reaction monitoring mode. Method validation was performed in terms of specificity, carryover, linearity (r > 0.99), intra−/inter‐day precision (1.0–10.1%), accuracy (relative error, <7.6%), stability (0.6–13.2%), extract recovery (74.9–91.9%) and matrix effect (89.1–109%). The lower limits of quantification of the six analytes varied from 0.92 to 10.4 ng/mL. The validated method was successfully applied to compare the pharmacokinetic properties of Wei‐Chang‐Shu tablet in normal rats and in rats with gastrointestinal motility disorders. The results indicated that there were obvious differences in the pharmacokinetic behavior between normal and model rats. This study will be helpful in the clinical application of Wei‐Chang‐Shu tablet.  相似文献

An ultra‐performance liquid chromatography (UPLC) method coupled with photodiode array (PDA) detection has been developed and validated for simultaneous quantification of 12 active components (berberine, palmatine, jatrorrhizine, aloe‐emodin, rhein, emodin, chrysophanol, physcion, baicalin, baicalein, wogonoside and wogonin) in Yiqing granule. Optimum separation were achieved on a C18 column (50 × 2.1 mm i.d., 1.7 µm particle) through a 7.5 min gradient delivery of a mixture of A (acetonitrile) and B (0.1% aqueous phosphoric acid containing 1.8 mmol/L sodium dodecyl sulfonate and 10% acetonitrile, v/v) at a flow rate of 0.3 mL/min at 30°C. Because of the different UV characteristics of these components, three detection wavelengths were used for quantitative analysis. All of the analytes showed good linearity (r of >0.999). The method was validated for repeatability, precision, stability, accuracy and selectivity. The validated method was applied to quality control of Yiqing granule from different production batches. A simple and rapid HPLC–MS/MS method was developed and validated for simultaneous measurement of phosphocreatine and its metabolites creatine and creatinine in children's plasma. A 50 μL aliquot of plasma was prepared by protein precipitation with acetonitrile–water (1000 μL, 1:1, v/v) followed by separation on a Hypersil Gold C18 column (35°C) with gradient mobile phase consisting of 2 mm ammonium acetate aqueous solution (pH 10) and methanol at a flow rate of 0.3 mL/min and analyzed by mass spectrometry in both positive (phosphocreatine) and negative (creatine and creatinine) ion multiple reaction monitoring mode. Good linearity (r > 0.99) was obtained for the three analytes. The intra‐day and inter‐day values of CV were <5.46% (?13.09% ≤ RE ≤ 2.57%). The average recoveries of the three analytes were 70.9–97.5%. No obvious impact was found for the quantitation of three analytes in normal, hemolyzed and hyperlipemic plasma. In the end, this method was successfully applied to a pharmacokinetic study of phosphocreatine in children (six cases) with viral myocarditis of children after intravenous infusion of 2 g of the test drug. The pharmacokinetc parameters of phosphocreatine/creatine were as follows: t1/2 0.24/0.83 h, Tmax 0.49/0.55 h, Cmax 47.34/59.29 μg/mL, AUClast 17.07/59.63 h μg/mL, AUCinf 17.16/79.01 h μg/mL and MRT 0.29/0.67 h.  相似文献

Five hydroxyanthraquinone derivatives physcion, emodin, fallacinal, teloschistin, and 1,3-dimethoxy-8-hydroxy-6-methyl-9,10-anthraquinone were isolated from the lichen Caloplaca cerina growing in Serbia. Three anthraquinone derivatives, namely physcion, emodin, and rhein were isolated and identified from the Thai medicinal plant Cassia tora. In this study, we used a self‐contrast method, which excluded the individual difference, to evaluate the inhibitory effect of chrysosplentin (CHR) in the presence or absence of artemisinin (ART) on the P‐glycoprotein (P‐gp) transport activity. A sensitive and rapid UHPLC–MS/MS method was applied for quantification of digoxin, a P‐gp‐specific substrate, in rat plasma. A pharmacokinetic study was carried out: first after an oral administration of digoxin at a dose of 0.09 mg/kg (first period), followed by a 20‐day wash‐out, then after another administration of digoxin (second period). During the second period, test compounds were orally given three times per day for seven consecutive days. Results showed that the t1/2 of digoxin in all the groups had no significant difference between the first and second periods. The AUC0–24, Cmax, tmax, and Clz/F of the negative control and ART alone groups showed no difference. However, the AUC0–24 and Cmax in the CHR alone, CHR–ART (1:2) and verapamil (positive control) groups showed 2.34‐, 3.04‐, 1.79‐, and 1.81‐, 1.99‐, 2.06‐fold increases along with 3.50‐, 3.84‐ and 4.76‐fold decreases for CLz/F, respectively. The tmax in the CHR–ART (1:2) group increased 3.73‐fold. In conclusion, our self‐contrast study suggested that CHR, especially when combined with ART in a ratio of 1:2, inhibited P‐gp activity while ART alone has no effect. The thermal behavior of five free anthraquinones (chrysophanol, emodin, physcion, aloe-emodin, and rhein) from rhubarb had been investigated using TG, DTG and DTA technique. The results show that all the free anthraquinones have the similar TG and DTG curve shapes, however, due to the substituted groups attached on the skeleton of 1,8-dihydroxy anthraquinone are different, every anthraquinone has different mass loss features. Moreover, all the DTA curves of these free anthraquinones have two obviously characteristic peaks, but with special curvilinear types, peak location and peak values. Therefore, thermal analysis (TA) characteristics of anthraquinones above mentioned could be established, and it is possible to easily distinguish these anthraquinones by using TA technique.  相似文献

A sensitive and specific liquid chromatography–electrospray ionization–tandem mass spectrometric (LC‐ESI‐MS/MS) method was developed and validated to simultaneously quantify 11 active compounds (coptisine, jatrorrhizine, berberine, palmatine, baicalin, baicalein, wogonoside, wogonin, rhein, emodin and aloeemodin) from Xiexin decoction (XXD) in rat plasma. Plasma samples extracted by a single‐step protein precipitation procedure were separated using the gradient mode on a Dikma ODS‐C18 column. Selected reaction monitoring scanning was employed for quantification with switching electrospray ion source polarity between positive and negative modes in a single run. Calibration curves offered satisfactory linearity (r > 0.995) at linear range of 0.47–60 ng/mL for coptisine, jatrorrhizine, berberine and palmatine, 15–1930 ng/mL for baicalin, 20–2560 ng/mL for baicalein, 14–1790 ng/mL for wogonoside, 0.57–72.8 ng/mL for wogonin, 10–1280 ng/mL for rhein, 0.6–76.8 ng/mL for emodin and 3.0–384 ng/mL for aloeemodin. The intra‐ and interday precisions were less than 10.2% in terms of relative standard deviation (RSD), and the accuracies were within ±10.84% in terms of relative error (RE).
