[reaction: see text] Photolysis of hydroxyacetophenone and thermolysis of the corresponding dioxetane afford spiroiminodihydantoin rather than 4,8-dihydro-4-hydroxy-8-oxo-2'-deoxyguanosine (4-HO-8-oxodG) through the oxidation of 2'-deoxyguanosine (dG) by triplet-excited hydroxyacetophenone and the peroxyl radicals derived thereof by alpha cleavage and subsequent oxygen trapping. The structure of the spiroiminodihydantoin is assigned by the SELINQUATE NMR technique, which unequivocally establishes the spirocyclic connectivity.  相似文献

On thermolysis of the methoxy (MeO-TMD), tert-butoxy (tBuO-TMD), and hydroxy (HO-TMD) derivatives of 3,3,4,4-tetramethyl-1,2-dioxetane (TMD) in the presence of dG and calf-thymus DNA, the guanine is oxidized considerably more efficiently than the parent TMD. The same trend in the oxidative reactivity is observed for the photolysis of the corresponding oxy-substituted ketones versus acetone. The oxidative reactivity order in the dioxetane thermolysis, as well as in the ketone photolysis, parallels the ability of the excited ketones to release radicals (determined by spin trapping with DMPO and EPR spectroscopy) upon alpha cleavage (Norrish-type-I reaction). In the presence of molecular oxygen, the carbon-centered radicals are scavenged to produce peroxyl radicals, which are proposed as the reactive species in the oxidation of the guanine in dG and calf-thymus DNA.  相似文献

The photolysis of acetophenone, benzophenone, 4-carboxybenzophenone and benzil was studied in air-saturated aqueous solution in the presence of alcohols. The overall reaction is an oxidation of 2-propanol to acetone. The quantum yield of oxygen uptake (Phi(-O(2))) increases with increasing 2-propanol concentration up to 0.9. The photoreaction can also be initiated by quenching of the ketone triplet state by ascorbic acid, formate or an amine e.g. triethylamine. Subsequent reactions of the involved radicals with oxygen yield the superoxide radical and eventually hydrogen peroxide. For the ketones in the presence of 3-30 mM ascorbic acid or triethylamine Phi(-O(2)) = 0.3-0.9. The specific properties of ketones, including 4-methoxyacetophenone and 2-acetonaphthone, the radicals involved and the pH and concentration dependences of Phi(-O(2)) are discussed.  相似文献

On irradiation of N-hydroxythiazole-2(3H)-thione 3 at 300 nm, the photoproducts disulfide 4, bisthiazole 5 and thiazole 6 are formed. During this photolysis, hydroxyl radicals are released, which have been detected by spin trapping with 5,5-dimethyl-1-pyrroline N-oxide (DMPO), coupled with electron paramagnetic resonance spectroscopy. In the presence of supercoiled pBR322 DNA, irradiation of thiazolethione 3 induces strand breaks through the photogenerated hydroxyl-radicals, as confirmed by control experiment with the hydroxyl-radical scavenger isopropanol. Singlet oxygen appears not to be involved, as attested by the lack of a D2O isotope effect. During the photoreaction of thiazolethione 3 in the presence of 2'-deoxyguanosine (dG), the latter is photooxidized (ca 10% conversion after 2 h of irradiation) to the 7,8-dihydro-8-oxo-2'-deoxyguanosine as the main oxidation product. The dG conversion levels off after complete consumption of thiazolethione 3 and is suppressed by the addition of the hydroxyl-radical scavenger 2,6-di-tert-butylcresol or DMPO. Since the photoproducts 4-6 are ineffective as sensitizers for the photooxidation of dG and DNA, the hydroxyl radicals released in the photolysis of thiazolethione 3 are the oxidizing species of DNA and dG. These results suggest that the thiazolethione 3 may serve as a novel and effective photochemical hydroxyl-radical source for photobiological studies.  相似文献

Abstract— The photochemical reactions of benzophenone and acetophenone with purine and pyrimidine derivatives in aqueous solutions have been investigated by flash photolysis and steady-state experiments. Upon excitation of these two ketones in aqueous solutions, two transient species are observed: molecules in their triplet state and ketyl radicals. The triplet state lifetimes are 65 μsec for benzophenone and 125 μsec for acetophenone. The ketyl radicals disappear by a second order reaction, controlled by diffusion. In the presence of pyrimidine derivatives, the triplet state is quenched and the ketyl radical concentration is decreased without any change in its kinetics of disappearance. Ketone molecules in their triplet state react with purine derivatives leading to an increase in the yield of ketyl radicals due to H-atom abstraction from the purines. Steady-state experiments show that benzophenone and acetophenone irradiated in aqueous solution at wavelengths longer than 290 nm undergo photochemical reactions. The rate of these photochemical reactions is increased in the presence of pyrimidine derivatives and even more in the presence of purine derivatives. Following energy transfer from the triplet state of benzophenone to diketopyrimidines, cyclobutane dimers are formed. The energy transfer rate decreases in the order orotic acid > thymine > uracil. Benzophenone molecules in their triplet state can also react chemically with pyrimidine derivatives to give addition photoproducts. All these results are discussed with respect to photosensitized reactions in nucleic acids involving ketones as sensitizers.  相似文献

Abstract The photooxidative DNA damage by iV-hydroxy-2-pyri-done (1) is caused by hydroxyl radicals, as confirmed by electron paramagnetic resonance studies with the spin trap 5,5-dimethylpyrroline JV-oxide. Irradiation of the pyridone 1 at 300 nm induced strand breaks in super-coiled pBR322 DNA, while in calf-thymus DNA and 2'-deoxyguanosine (dG), respectively, 8-oxoguanine and 8-oxo-7,8-dihydro-2'-deoxyguanosine were formed. Time-dependent control experiments disclosed that photoprod-ucts of pyridone 1, e.g. 2-pyridone (3), are not responsible for the modification of DNA. Also the photosensitization by the pyridine-2-one chromophore was excluded, because JV-methylpyridine-2-one (2), which cannot generate hydroxyl radicals, was ineffective in the photooxidation of DNA and dG. Thus, the photolysis of pyridone 1 serves as a specific source of hydroxyl radicals for DNA damage, both strand breaks and base modifications.  相似文献

Irradiation of acetophenone or benzophenone in the presence of norbornadiene or quadricyclene gives 1: 1-adducts. These arise via reaction of the ketone triplet with the quadricyclene. Similar reactivity is exhibited in an intramolecular sense by 7-phenacylnorbornadiene and the corresponding quadricyclene. Addition of triplet benzophenone to 5-methylene norbornene is regiospecific in contrast to expectations based on a comparison of the reactivities of 2-methylenenorbornane and norbornene. The possibility is raised that, in contrast to the simple olefins and the quadricyclenes, exciplexes formed by reaction of the rigid homoconjugated dienes with the triplet ketone are endo orientated with participation of both double bounds.  相似文献

The phosphorescence characteristics (excitation and emission spectra and lifetimes) of acetophenone (AP), benzophenone (BP), p-aminobenzophenone (PABP) and Michler's ketone (MK) adsorbed on Whatman No. 1 filter paper were measured at various temperatures, and compared with the phosphorescence characteristics in different solvent glasses at 77 K. Both AP and BP phosphoresce on filter paper only at low temperature (208 K). The phosphorescence lifetimes of AP and BP are < 1 msec, indicating a (3)(n,pi(*)) lower triplet level for paper substrates. With PABP, the low lying triplet state in polar solvents is (3)(CT) and in non-polar solvents is (3)(n, pi(*)); PABP on filter paper results in spectral characteristics similar to those of PABP in polar solvents at 77 K. The lifetime of PABP is longer than that of BP, indicating a (3)(CT) low-lying triplet state. MK, like PABP, has strongly environment-dependent photophysical properties. MK, when adsorbed on filter paper, has an intense long-lived luminescence at room temperature, resulting in a limit of detection of 3 ng ml or 3 pg, and a linear dynamic range of over 3 orders of magnitude. MK appears to be strongly hydrogen-bonded to the filter paper. In studies in ethanol and other solvents, MK adsorbed on filter paper shows a dramatic change in its phosphorescence spectrum when the temperature is lowered from 298 K to 208 K; the phosphorescence peak moves to longer wavelengths and the intensity decreases. The temperature effect could arise from the presence of several conformers of MK or be due to different environmental sites or E-type delayed fluorescence. The low-lying triplet state of MK on filter paper is most likely a (3)(CT) state. Lowering the temperature appears to increase the phosphorescence intensity for ketones which phosphoresce in the (3)(n,pi(*)) triplet state, but affects it only slightly for analytes which phosphoresce in the (3)(pi,pi(*)) triplet state. Room-temperature phosphorescence seems to arise for aromatic ketones and aldehydes with low-lying (3)(pi, pi(*)) or (3)(CT) triplet states.  相似文献

Laser flash photolyses have been carried out on solutions of the 2-naphthylammonium ion (RNH+3?) benzophenone (BP) [or acetophenone (AP)] system. It is found that the hydrogen-atom transfer reaction from 3RNH+3 (produced by triplet sensitization of the ketones) to the ground BP (or AP) occurs effectively to give RNH+3 and > COH.  相似文献

The 2,6-diamino-4-hydroxy-5-formamidopyrimidine of 2'-deoxyguanosine (FaPydG) is one of the major DNA lesions found after oxidative stress in cells. To clarify the base pairing and coding potential of this major DNA lesion with the aim to estimate its mutagenic effect, we prepared oligonucleotides containing a cyclopentane based analogue of the DNA lesion (cFaPydG). In addition, oligonucleotides containing the cyclopentane analogue of 2'-deoxyguanosine (cdG), and oligonucleotides containing 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) were synthesized. The thermodynamic stability of duplexes containing these building blocks and all canonical counterbases were determined by concentration dependent melting-point measurements (van't Hoff plots). The data reveal that cFaPydG greatly destabilizes a DNA duplex (DeltaDeltaG degrees (298K) approximately 2-4 kcal mol(-1)). The optimal base pairing partner for the cFaPydG lesion is dC. Investigation of duplexes containing dG and cdG shows that the effect of substituting the deoxyribose by a cyclopentane moiety is marginal. The data also provide strong evidence that the FaPydG lesion is unable to form a base pair with dA. Our computational studies indicate that the syn-conformation required for base pairing with dA is energetically unfavorable. This is in contrast to 8-oxodG for which the syn-conformation represents the energetic minimum. Kinetic primer extension studies using S. cerevisiae Pol eta reveal that cFaPydG is replicated in an error-free fashion. dC is inserted 2-3 orders of magnitude more efficiently than dT or dA, showing that FaPydG is a lesion which retains the coding potential of dG. This is also in contrast to 8-oxodG, for which base pairing with dC and dA was established.  相似文献

Abstract— Quenching of the triplet states of the aromatic ketones (KCO), benzophenone, acetophenone and xanthone, by indole and 3-methyl indole gives rise to the neutral radicals resulting from hydrogenatom transfer with variable efficiency (40–100%). Thus in addition to the reaction,
3KCO*+ R H →KCOH + R .
some other quenching path or paths occur. There is no evidence for any triplet energy transfer even when this is energetically favourable, and it is suggested therefore that quenching to give ground state species following favourable charge-transfer interactions accounts for the proportion of quenching without reaction.
The spectra of the indole radicals, R ., were determined and the kinetics of their decay in aerated and deaerated solution were investigated and reaction schemes proposed to explain these observations.  相似文献

The C-O bond cleavage from benzophenone substituted with 4-CH2OR (p-BPCH2OR, 1-3), such as p-phenoxymethylbenzophenone (1, R= C6H5) and p-methoxymethylbenzophenone (2, R= CH3), occurred by a stepwise two-photon excitation during two-color, two-laser flash photolysis. On the other hand, no C-O bond cleavage occurred from p-hydroxymethylbenzophenone (3, R = H). The first 355-nm laser excitation of 1-3 generates p-BPCH2OR in the lowest triplet excited state (T1) which has an absorption at 532 nm. When p-BPCH2OR(T1) is excited with the second 532-nm laser to p-BPCH2OR in the higher triplet excited state (T(n)), the C-O bond cleavage occurred within the laser flash duration of 5 ns. The quantum yields of the C-O bond cleavage during the second 532-nm laser irradiation were found to be 0.015 +/- 0.007 and 0.007 +/- 0.003 for 1 and 2, respectively. Although these values are low, the diminishing 1(T1) or 2(T1) was found to convert, in almost 100% yield, to phenoxyl (C6H5O*) and p-benzoylbenzyl (BPCH2*) radicals or methoxyl (CH3O*) and BPCH2* radicals, respectively. The T(n) excitation energy, the energy barrier along the potential surface between the T(n) states and product radicals, and delocalization of the T(n) state molecular orbital including BP and CH2OR (R = C6H5, CH3, H) moieties are important factors for the occurrence of the C-O bond cleavage. It is found that the C-O bond cleavage and production of free radicals, such as BPCH2*, C6H5O*, and CH3O*, can be performed by a stepwise two-photon excitation. The present study is an example in which the chemical reactions can be selectively initiated from the T(n) state but not from the S1 and T1 states.  相似文献

The dihydroxo(tetraphenylporphyrinato)antimony(V) complex (SbTPP) demonstrates bactericidal activity under visible-light irradiation. This phototoxic effect could be caused by photodamage to biomolecules, but the mechanism has not been well understood. In this study, to clarify the mechanism of phototoxicity by SbTPP, DNA damage photosensitized by SbTPP was examined using [(32)P]-5'-end-labeled DNA fragments. SbTPP induced markedly severe photodamage to single-stranded rather than to double-stranded DNA. Photo-irradiated SbTPP frequently caused DNA cleavage at the guanine residue of single-stranded DNA after Escherichia coli formamidopyrimidine-DNA glycosylase or piperidine treatment. HPLC measurement confirmed the formation of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG), an oxidation product of 2'-deoxyguanosine, and showed that the content of 8-oxodG in single-stranded DNA is larger than that in double-stranded DNA. The effects of scavengers of reactive oxygen species on DNA damage suggested the involvement of singlet oxygen. These results have shown that the mechanism via singlet oxygen formation mainly contributes to the phototoxicity of SbTPP. On the other hand, SbTPP induced DNA damage specifically at the underlined G of 5'-GG, 5'-GGG, and 5'-GGGG in double-stranded DNA. The sequence-specificity of DNA damage is quite similar to that induced by the type I photosensitizers, suggesting that photo-induced electron transfer slightly participates in the phototoxicity of SbTPP. In conclusion, SbTPP induces DNA photodamage via singlet oxygen formation and photo-induced electron transfer. A similar mechanism can damage other biomacromolecules, such as protein and the phospholipid membrane. The damage to biomacromolecules via these mechanisms may participate in the phototoxicity of SbTPP.  相似文献

Illumination of air-free aqueous solutions containing sulfonated poly(ether ether ketone) and poly(vinyl alcohol) with 350 nm light results in benzophenone ketyl radicals of the polyketone. The polymer radicals form with a quantum yield 0.02 and decay with a second-order rate constant 6 orders of magnitude lower than that of typical alpha-hydroxy radicals. Evidence is presented that the polymeric benzophenone ketyl radicals reduce Ag+, Cu2+, and AuCl4- to metal particles of nanometer dimensions. Decreases in the reduction rates with increasing Ag(I), Cu(II), and Au(III) concentrations are explained using a kinetic model in which the metal ions quench the excited state of the polymeric benzophenone groups, which forms the macromolecular radicals. Quenching is fastest for Ag+, whereas Cu2+ and AuCl4- exhibit similar rate constants. Particle formation becomes more complex as the number of equivalents needed to reduce the metal ions increases; the Au(III) system is an extreme case where the radical reactions operate in parallel with secondary light-initiated and thermal reduction channels. For each metal ion, the polymer-initiated photoreactions produce crystallites possessing distinct properties, such as a very strong plasmon in the Ag case or the narrow size distribution exhibited by Au particles.  相似文献

Photosensitized hydrogen abstraction from 2-alkyl-1,3-dioxolanes by triplet benzophenone gives the corresponding 1,3-dioxolan-2-yl radicals and these are trapped by ,β-unsatured ketones yielding monoprotected 1,4-diketones. With open chain ketones (3-buten-2-one and 4-penten-3-one) the yields are low and competitive pathways in part consume the radicals. With cyclic enones however, yields are good as tested with cyclopentenone, cyclohexenone and 4-hydroxy-cyclopentenone. More generally, this is a viable alternative for the synthesis of 1,4-diketones via radicals while the thermal initiation gives only low yield. The reaction cannot be extended to strongly stabilized radicals, such as the 2-phenyl-1,3-dioxolanyl radical.  相似文献

Photochemical processes of benzophenone (BP) and xanthone (XT) with tryptophan (TrpH) and tyrosine (TyrOH) were studied using the laser flash photolysis technique.It has been observed that the triplet state of BP and XT reacted with TrpH and TyrOH by hydrogen transfer with the formation of BP and XT ketyl radicals and oxidized radicals of TrpY and TyrOY.The related rate constants of these reactions were determined in this paper.The free energy changes (G) of these reactions suggested that the proposed hydrogen transfer mechanism was thermodynamically feasible.These results provide theoretical foundation for further studying structural effects on the photochemical behaviors of proteins with triplet state BP and XT.  相似文献

Unsubstituted or alkyl-substituted cyclic ketones react with PdCl2 in methanol under a CO atmosphere to give mainly acyclic diesters along with some acyclic chloro-substituted monoesters. The monosubstituted cyclic ketones, 2-hydroxy- and 2-methoxycyclohexanone, do not give ring cleavage but rather produce 2-(carbomethoxy)cyclohex-2-en-1-one. 13CO labeling experiments indicate one CO is inserted in forming the diester product so the second ester group must arise from the original ketone group. Two mechanisms are possible for the diester reaction. One involves initial Pd(II)-CO2CH3 insertion across the double bond of the enol form of the ketone while the second involves initial addition of Pd(II)-OCH3 followed by CO insertion into the new Pd(II)-carbon bond formed. Pd(II) elimination and acid-catalyzed ring cleavage produce the second methyl ester group in both routes. The chloro-substituted monoester is formed by initial Pd(II)-Cl insertion across the double bond followed by the acid-catalyzed ring cleavage. The 2-(carbomethoxy)cyclohex-2-en-1-one must result from elimination of water or methanol from the alpha-ketoester product formed by the initial methoxycarbonylation of the enol form of the ketone. As expected, the acyclic ketone, 2-decanone, formed methyl acetate and a mixture of methyl nonanoate and 1-chlorooctane as products.  相似文献

A series of benzophenone (BP) and naphthalene (NA) labeled poly(aryl ether) dendrimers (BP-Gn-NA), generations 1-4, were synthesized, and their photophysical properties were examined. Flash photolysis demonstrates that the triplet energy in BP-Gn-NA can be transferred from the peripheral BP chromophores to the core NA group with the efficiencies of ca. 0.97, 0.96, 0.88, and 0.54 and with the rate constants of 1.4x10(8), 1.2x10(8), 9.5x10(7), and 1.3x10(7) s-1 at room temperature for generations 1-4, respectively. The transient absorption spectra of BP-Gn-NA show clearly the formation of the triplet NA absorption along with the decay of the triplet BP one with an isosbestic point at 475 nm, which gives direct evidence of the triplet energy transfer from the periphery BP chromphores to the core NA group. The phosphorescence of the NA group attached to the focal point was observed when the periphery BP chromophores were selectively irradiated in BP-G1-NA at 77 K. The triplet energy transfer occurs at 77 K with the efficiencies of 1.0, 0.16, 0.17, and 0.21 for generations 1-4, respectively. The intramolecular triplet energy transfer is proposed to proceed mainly via a through space mechanism.  相似文献

UVA-irradiation of the photo-Fenton reagents N-isopropoxypyridone 2b and N-isopropoxythiazole-2(3H)-thione 3b releases radicals which induce strand breaks. Transient spectroscopy establishes N-O bond scission [Phi(N)(-)(O) = (75 +/- 8)% for 2b and (65 +/- 7)% 3b] as the dominating primary photochemical process to afford the DNA-damaging radicals. Product studies and laser-flash experiments reveal that the thiazolethione 3b leads primarily to the disulfide 5, from which through C-S bond breakage, the bithiazyl 6, the thiazole 7, and the isothiocyanate 8 are derived. Upon irradiation of pyridone 2b (300 nm) in aqueous media, a mixture of isopropoxyl and 2-hydroxyprop-2-yl radicals is formed, as confirmed by trapping with 5,5-dimethyl-1-pyrroline N-oxide (DMPO) and EPR spectroscopy. In contrast, the photolysis of the thiazolethione 3b (350 nm) affords exclusively the DMPO adducts of the isopropoxyl radicals. Control experiments disclose that the thiazolethione-derived photoproduct disulfide 5, or the intermediary thiyl radicals B, scavenge the carbon-centered 2-hydroxyprop-2-yl radicals, which are generated from the isopropoxyl radicals by hydrogen shift. With supercoiled pBR 322 DNA in a 60:40 mixture of H(2)O-MeCN, the pyridone 2b and the thiazolethione 3b display moderate strand-break activity (17% open-circular DNA for 2b and 12% for 3b). In pure water, however, the pyridone 2b photoinduces substantially more DNA cleavage (32% open-circular DNA), which is attributed to the peroxyl radicals generated from the 2-hydroxyprop-2-yl radicals by oxygen trapping. The lower strand-break activity of the thiazolethione 3b derives presumably from isopropoxyl radicals, because only these are detected in the photolysis of this photo-Fenton reagent.  相似文献

Collision-induced dissociation (CID) of 8-(4'-hydroxyphenyl)-2'-deoxyguanosine and 8-(2'-hydroxyphenyl)-2'-deoxyguanosine was investigated using sequential tandem mass spectrometry. These adducts represent biomarkers of DNA damage linked to phenolic radicals and were investigated to gain insight into the effects of chemical structure of a C-8 modification on fragmentation pathways of modified 2'-deoxyguanosine (dG). CID in MS(2) of the deprotonated molecules of both the isomers generated the same product ion having the same m/z values. CID in MS(3) of the product ion at m/z 242 and CID in MS(4) experiments carried out on the selected product ions at m/z 225 and m/z 218 afford distinct fragmentation patterns. The conformational properties of isomeric product ions from CID showed that the ortho-isomers possess the unique ability to tautomerize through an intramolecular proton transfer between the phenolic OH group and the imine nitrogen (N7). Tautomerization of ortho-isomers to their keto-tautomers led to differences in their system of conjugated double bonds compared with either their enol-tautomer or the para-isomer. The charge redistribution through the N-7 site on the imidazole ring is a critical step in guanosine adduct fragmentation which is disrupted by the formation of the keto-tautomer. For this reason, different reaction pathways are observed for 8-(4'-hydroxyphenyl)-2'-deoxyguanosine and 8-(2'-hydroxyphenyl)-2'-deoxyguanosine. We present herein the dissociation and the gas-phase ion-molecule reactions for highly conjugated ions involved in the CID ion chemistry of the investigated adducts. These will be useful for those using tandem mass spectrometry for structural elucidation of C-8 modified dG adducts. This study demonstrates that the modification at the C-8 site of dG has the potential to significantly alter the reactivity of adducts. We also show the ability of tandem mass spectrometry to completely differentiate between the isomeric dG adducts investigated.  相似文献
