Bis(di-isobutyl octadecylsiloxy)silicon 2,3-naphthalocyanine (isoBOSINC) is a synthetic potential photosensitizer for tumor therapy. A new method, which combines solvent extraction and several purification steps, has been developed to determine its presence in tissues. Separation and quantitation of isoBOSINC is done by high-performance liquid chromatography on a silica column with toluene as a mobile phase and using fluorescence detection (lambda ex = 365 nm, lambda em = 750 nm). For recovery studies, isoBOSINC was added to muscles at levels of 0.067 and 0.67 micrograms/g; the mean recoveries were 100%, with coefficients of variation of 6.1 and 6.4%, respectively. For liver samples, the amounts added were 0.67 and 6.7 micrograms/g and for serum 0.67 and 6.70 micrograms/ml. The mean recoveries for liver were 86 and 93%, with coefficients of variation of 7.7 and 4.4%, respectively. For serum, the mean recoveries were 99 and 96%, with coefficients of variation of 2.6 and 6.9%, respectively. Due to its low detection limit and selectivity, the method is appropriate for pharmacokinetic as well as tumor uptake studies following in vivo exposure to isoBOSINC. Preliminary data on tissue distribution of the photosensitizer in normal rats are also presented.  相似文献

A thin-layer chromatographic (TLC) method with densitometric quantitation using the absorption reflectance mode at 280 nm was developed and validated for the determination of chloramphenicol residues in controlling pharmaceutical equipment cleanliness. Simulated samples at residue levels 0.5, 1, and 1.2 mg/m2 were prepared by spreading the calculated amount of chloramphenicol solution on a 10 dm2 stainless steel surface. After evaporation of the solvent, the residue was removed by 2 methanol-wetted cotton swabs, which were then extracted with methanol. The extract was applied on a high-performance TLC (HPTLC) silica gel F254 plate together with standards ranging from 10 to 60 ng. Plates were developed in a horizontal developing chamber from both sides (36 applications per plate) by using n-hexane-ethyl acetate (35 + 65, v/v) as developing solvent. The mean recovery (n=6) at 1 mg/m2 was 95.8%, and the coefficient of variation was 5.8%. The absolute detection limit was 3 ng, and the quantitation limit 10 ng. The method detection limit was 0.3 mg/m2 by swabbing 2.5 dm2 and 0.075 mg/m2 by swabbing 10 dm2. Chloramphenicol was stable on the plate 2 h before and 24 h after development. Additionally, it was stable during 7 days storage on the cotton swabs in the solvent at room temperature and in diluted standard solution stored in darkness at 4 degrees C. The method can be applied to routine control of pharmaceutical equipment cleanliness by sampling from the stainless steel surface areas of 2.5 to 10 dm2, and an acceptable residue limit of 1 mg/m2.  相似文献

This work describes the analytical performance of the enzyme-linked immunosorbent assay (ELISA) for the fungicide chlorothalonil to effectively exploit as a simple and rapid detection system for pesticide residue on the scenes of the agricultural production and distribution. This ELISA represents the satisfactory analytical characteristics (I50 value, 0.34 ng/g; limit of detection, 0.052 ng/g) to detect chlorothalonil at the regulatory values or thereabout in a sample. Noticeable cross-reactivities were shown with two fungicides, fthalide (58.8%) and pentachloronitrobenzene (quintozene) (20.0%), and some non-agrochemicals such as tetrachloroterephthalonitrile (96.8%) and tetrachlorophthalonitrile (68.3%). The influence of three organic solvents (methanol, acetone, and acetonitrile) used as extractants for chlorothalonil residue was evaluated, with the result that methanol was the most suitable solvent for the ELISA, and the final concentration in the well could be up to 5% (v/v) without any negative influence on the ELISA. It has been possible to directly analyze chlorothalonil residue only by giving dilution of each sample extract with water prior to the ELISA analysis. The average recovery values from the spiked samples by the ELISA were between 101.7 and 113.6% with the average coefficients of variation between 2.6 and 5.9%. Although the results obtained from the ELISA correlated well with those from the reference GC/MS methods for all agricultural samples (r>0.98), the linear function inclined to the ELISA results because of loss during complex sample preparations for GC/MS analysis. Nevertheless, the results demonstrated that the proposed ELISA is a reliable, cost-effective, and rapid quantitative method for chlorothalonil residue.  相似文献

The Willems badge, a diffusive sampler for nitrogen dioxide, has previously been validated for ambient air measurements. This paper describes the laboratory and field validation of the Willems badge for personal sampling under working environment conditions. The mean sampling rate in the laboratory tests was 46 ml min(-1), with an RSD of 12%. No statistically significant effects on sampling rate of the sampling time, concentration of NO2 or relative humidity were found. A slightly decreased sampling rate was observed at low wind velocity. This was also confirmed during static sampling, which makes the sampler less appropriate for static sampling indoors. No back diffusion was observed. Storage of the samplers for two weeks before or after exposure did not affect the sampling rate. Our analysis is based on a modified colorimetric method, performed by FIA (flow injection analysis). This technique was compared to ion chromatography analysis. The use of ion chromatography lowered the detection limit from 11 to 2 microg m(-3) for an 8 h sample, and furthermore enabled the detection of other anions. In conclusion, the diffusive sampler was found to perform well for personal measurements in industrial environments.  相似文献

An HPTLC method with densitometric quantification using fluorescence at 313 nm was developed and validated for the determination of ofloxacin residue in controlling pharmaceutical equipment cleanliness. Simulated samples at a residue level of 1 mg/m2 were prepared by spreading the calculated amount of ofloxacin solution on 1, 5, and 10 dm2 stainless steel surfaces. After evaporation of the solvent, the residue was removed by two ethanol wetted cotton swabs, which were thereafter extracted with the mixture of ethanol and Na2EDTA-water solution at pH 11 for 15 min with sonication. The extract and standards were applied on HPTLC silica gel 60 plates and then developed in a horizontal developing chamber from both sides using ethanol-conc. ammonia (4+1, v/v) as the mobile phase. The mean recovery (n=6) at 1 mg/m2 from 1, 5, and 10 dm2 was 95.3, 88.6, and 89.7% with the CV values 3.78, 4.41, and 4.97%, respectively. The absolute detection limit was 0.6 ng and the quantitation limit was 2 ng, but it was shown that these can be improved by immersion of the developed plate into a solution of liquid paraffin-n-hexane (1+2, v/v) to approximately 0.25 and 0.9 ng, respectively. The LOD of the method using detection without paraffin-n-hexane was 3, 0.6, and 0.3 microg/m2 by swabbing 1, 5, and 10 dm2, respectively. The method can be applied to routine control of pharmaceutical equipment cleanliness by sampling from stainless steel surface areas of 1 to 10 dm2 with acceptable residue limit/surface of 1 mg/m2.  相似文献

A highly sensitive gas chromatographic-mass spectrometric (GC-MS) analytical method for the determination of the opioid narcotics fentanyl, alfentanil, and sufentanil in industrial hygiene personal air samples and surface contamination wipes was developed and comprehensively validated. Sample preparation involved a single step extraction of the samples with methanol, fortified with a fixed amount of the penta-deuterated analogues of the opioid narcotics as internal standard. The GC-MS analytical procedure using selected ion monitoring (SIM) was shown to be highly selective. Linearity was shown for levels of extracted wipe and air samples corresponding to at least 0.1-2 times their surface contamination limit (SCL) and accordingly to 0.1-2 times their time weighted average occupational exposure limit (OEL-TWA) based on a full shift 9601 air sample. Extraction recoveries were determined for spiked air samples and surface wipes and were found to be quantitative for both sampling media in the entire range studied. The air sampling method's limit of detection (LOD) was determined to be 0.4 ng per sample for fentanyl and sufentanil and 1.6 ng per sample for alfentanil, corresponding to less than 1% of their individual OEL for a full shift air sample (9601). The limit of quantification (LOQ) was found to be 1.4, 1.2, and 5.0 ng per filter for fentanyl, sufentanil, and alfentanil, respectively. The wipe sampling method had LODs of 4 ng per wipe for fentanyl and sufentanil and 16 ng per wipe for alfentanil and LOQs of respectively, 14, 12, and 50 ng per wipe. The analytical intra-assay precision of the air sampling and wipe sampling method, defined as the coefficient of variation on the analytical result of six replicate spiked media was below 10 and 5%, respectively, for all opioids at all spike levels. Accuracy expressed as relative error was determined to be below 10%, except for alfentanil at the lowest spike level (-13.1%). The stability of the opioids during simulated air sampling was investigated. For fentanyl and sufentanil a quantitative recovery was observed at all spike levels, while for alfentanil recoveries ranged from 60.3 to 85.4%. When spiked air samples were stored at ambient temperature and at -15 degrees C quantitative recovery was found for fentanyl and sufentanil after 7 and 14 days. For alfentanil a slight loss seemed to occur upon storage during 7 days, being more explicit after 14 days. Ambient storage of spiked wipes seemed to lead to significant losses of all opioids studied, yielding recoveries of 37.7-88.3%. Upon storage of similar wipes at -15 degrees C a significantly higher recovery was found ranging from 77.3 to 88.3%. The developed analytical and sampling procedures have been recently applied in an explorative field study of which the results of surface contamination wipe sampling are presented in this paper. To our knowledge, this is the first study addressing the development and validation of analytical procedures for the assessment of external occupational exposure to potent opioid narcotics.  相似文献

The presence of both pharmaceuticals and pesticides in the aquatic environment has become a well-known environmental issue during the last decade. An increasing demand however still exists for sensitive and reliable monitoring tools for these rather polar contaminants in the marine environment. In recent years, the great potential of passive samplers or equilibrium based sampling techniques for evaluation of the fate of these contaminants has been shown in literature. Therefore, we developed a new analytical method for the quantification of a high number of pharmaceuticals and pesticides in passive sampling devices. The analytical procedure consisted of extraction using 1:1 methanol/acetonitrile followed by detection with ultra-high performance liquid chromatography coupled to high resolution and high mass accuracy Orbitrap mass spectrometry. Validation of the analytical method resulted in limits of quantification and recoveries ranging between 0.2 and 20 ng per sampler sheet and between 87.9 and 105.2%, respectively. Determination of the sampler-water partition coefficients of all compounds demonstrated that several pharmaceuticals and most pesticides exert a high affinity for the polydimethylsiloxane passive samplers. Finally, the developed analytical methods were used to measure the time-weighted average (TWA) concentrations of the targeted pollutants in passive samplers, deployed at eight stations in the Belgian coastal zone. Propranolol, carbamazepine and seven pesticides were found to be very abundant in the passive samplers. These obtained long-term and large-scale TWA concentrations will contribute in assessing the environmental and human health risk of these emerging pollutants.  相似文献

Two new methods for the determination of dithiocarbamate pesticides in occupational hygiene sampling devices are described. Dithiocarbamate spiked occupational hygiene sampling devices, consisting of glass fibre (GF/A) filters, cotton pads, cotton gloves and disposable overalls, were reduced under acidic conditions and the CS2 evolved as a decomposition product was extracted into isooctane. The isooctane was then analysed using gas chromatography with mass spectrometry, for CS2, which provided a quantitative result for dithiocarbamates. Recoveries obtained were generally within a 70-110% range and reproducibilities better than 15% RSD were typically achieved. The method has been successfully applied to samples collected during occupational exposure surveys. A second method employing automatic thermal desorption-gas chromatography-mass spectrometry (ATD-GC-MS) has also been developed and applied to the direct analysis of GF/A (airborne) samples. The method relies on the thermal degradation of dithiocarbamates to release CS2, which is used to quantify the analytes. Thiram spiked GF/A filters gave an average recovery of 107% with an RSD of 4%. The performance of the two analytical methods were directly compared by analysing sub-portions of GF/A filters collected during a survey to evaluate occupational exposures to thiram during seed treatment operations. Both methods performed well for the analysis of airborne (GF/A) samples and produced results in good agreement. ATD-GC-MS is the preferred method for studies involving GF/A (airborne) samples only. Because of the wider applicability of the isooctane method for other sampling devices, it is the preferred choice when carrying out surveys which require a dermal as well as respirable exposure assessment.  相似文献

Winter is the key period for the control of apple diseases, and fungicides are needed to protect the trunk or main branches. Fungicide residue in apple tree bark is an important basis for the action of the pesticide, but there are no reports on analytical methods or dissipation patterns. In this work, thiophanate-methyl, carbendazim, tebuconazole and pyraclostrobin were selected as typical fungicides and a new QuEChERS–HPLC–VWD(QuEChERS extraction followed by high-performance liquid chromatography detection with a variable wavelength detector) analytical method was developed to estimate their residue kinetics in apple tree bark during the winter months. In the pretreatment step, the sorbent for the clean-up of extracts was optimized as 60 mg/ml primary secondary amine and a gradient-elution model followed by a variable wavelength detection was developed for instrumental analysis. Then this method was validated and applied to the analysis of apple tree bark samples with the linearity range of 0.010–50.00 mg/L, quantification limit range of 0.028–0.080 mg/kg and recovery range of 86.1–101.4%. The dissipation kinetics of thiophanate-methyl and pyraclostrobin could be described by the first-order and two-phase kinetics models, respectively. For carbendazim and tebuconazole, two new models were developed to describe their residue kinetics.  相似文献

A liquid chromatographic-tandem mass spectrometric multiresidue method for the simultaneous quantitative determination of the tetrahydroimidazole, levamisole and the benzimidazoles thiabendazole, oxfendazole, oxibendazole, albendazole, fenbendazole, febantel and triclabendazole in milk has been developed and validated. The anthelmintic residues were extracted with ethyl acetate. The liquid chromatographic separation was performed on a reversed-phase C18 column with gradient elution. The analytes were detected by tandem quadrupole mass spectrometry after positive electrospray ionisation by multiple reaction monitoring. The confirmatory method is very sensitive and each component can be detected at a residue level lower than 1 microgram/l. The method is validated according to the revised European Union requirements and all parameters were found conform the criteria. The evaluated parameters were linearity, specificity, stability, recovery, precision (repeatability and within-laboratory reproducibility) and analytical limits (detection limit, decision limit and detection capability). This analytical method is applied in the Belgian monitoring programme for classical anthelmintic veterinary drugs in raw farm cow's milk.  相似文献

A new analytical procedure using solid-state Raman spectroscopy within the THz-region for the quantitative determination of mixtures of different conformations of trifloxystrobin (EE, EZ, ZE and ZZ), tebuconazole (1), and propiconazole (2) as an effective method for the fungicide product quality monitoring programmes and control has been developed and validated. The obtained quantities were controlled independently by the validated hybrid HPLC electrospray ionization (ESI) tandem mass spectrometric (MS) and matrix-assisted laser desorption/ionization (MALDI) MS methods in the condensed phase. The quantitative dependences were obtained on the twenty binary mixtures of the analytes and were further tested on the three trade fungicide products, containing mixtures of trifloxystrobin-tebuconazole and trifloxystrobin-propiconazole, as an emissive concentrate or water soluble granules of the active ingredients. The present methods provided sufficient sensitivity as reflected by the metrologic quantities, evaluating the concentration limit of detection (LOD) and quantification (LOQ), linear limit (LL), measurement accuracy and precision, true quantity value, trueness of measurement and more.  相似文献

In order to assess exposure levels of hospital personnel involved in the preparation and administration of antineoplastic drugs, environmental monitoring should be carried out. Wipe samples, pads, gloves and air samples should be collected at the end of each work shift, properly treated and then analysed using instrumental techniques which are sufficiently sensitive and specific to detect even trace amounts of drug. In this study, a method using high performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS), incorporating solid phase extraction (SPE), was validated for determination of methotrexate (MTX) in wipe and air samples. Each step of the method was first developed and optimised using ultraviolet detection (UV), and afterwards tandem mass spectrometry was used to obtain a lower limit of quantitation when the expected drug level was less than the analytical UV detection limit. SPE enabled a 20-fold preconcentration of the analyte when using HPLC/UV and a further 30-fold preconcentration was obtained when analysing samples by HPLC/MS/MS. For example, the limit of quantitation (LLQ) was lowered from 3000 ng on wipe (direct injection onto an HPLC/UV system) to 5 ng on wipe (SPE plus HPLC/MS/MS). 7-hydroxymethotrexate was used as internal standard to assess precision and accuracy.  相似文献

The use of a small precolumn instead of an injection loop for the determination of a new phytochemical drug, fellavine, and its metabolites is described. The method combines the direct injection of plasma and urine into the reversed-phase precolumn with separation on a Spheri-5 RP-18 analytical column. Different sorbents in the precolumn were compared. A recovery of fellavine and its metabolites from biological fluids except rat plasma of almost 100% was achieved on Chrompack RP (30-40 microns) and LiChrosorb RP-18 (7 microns). For rat plasma only the last sorbent gave 80% fellavine recovery. The influence of the protein binding on the fellavine recovery was examined. The limit of detection was equal to 0.05 micrograms/ml fellavine for plasma and 0.02 micrograms/ml for urine. To enhance the limit of detection longer precolumns were perferred.  相似文献

A rapid multiresidue method for the simultaneous determination of 14 fungicides in soil was developed. Fungicides were exacted from soil, placed in small columns, by sonication-assisted extraction with ethyl acetate. The effect of residue residence time and soil moisture content on the fungicide recovery was studied. Residue levels in soil were determined by gas chromatography with electron-capture and nitrogen-phosphorus detection. Residue identities were confirmed by gas chromatography coupled with mass spectrometry, in the selected ion monitoring mode. Recovery studies were carried out at 0.5, 0.1 and 0.05 microg/g fortification levels for each fungicide, and average recoveries obtained for these compounds ranged from 80 to 104% with relative standard deviations between 1 and 8%. The method is linear over the range assayed, 0.5-0.05 microg/g, and the detection limit for the fungicides studied varied from 2 to 10 microg/kg.  相似文献

Summary A diffusive sampler for the large scale routine determination of airborne formaldehyde was developed. Formaldehyde is sampled in a badge-type passive sampler containing a 2,4-dinitrophenylhydrazine-coated filter paper as sampling layer. Formaldehyde is immediately converted to the corresponding hydrazone, which, after desorption with acetonitrile, is separated and quantified by gradient HPLC using UV detection at 345 nm. Calibration was done via an active sampling method and showed an excellent, time- and concentration-independent linear performance of the diffusive samplers. A detection limit of about 0.05 ml/m3·h (ppm·h) and a relative standard deviation of about 10% ensured a good analytical reliability. By testing the influence of air movements at the sampler surface, a minimum air velocity of 10 cm/s was found necessary to ensure representative sampling.  相似文献

A diffusive sampling method is presented for the determination of atmospheric levels of ammonia based on the collection of ammonia at the phosphoric acid coated bottom of small glass vials and subsequent flow injection analysis of the trapped ammonium. In order to avoid effects of varying wind velocities on the sampling rate, the tube-type samplers are covered with a hydrophobic microporous membrane. The experimentally obtained uptake rate of the diffusive samplers (i.e. 7.8 ml min–1) is in excellent agreement with the theory. The analytical characteristics of the method with respect to sensitivity, selectivity, dynamic range, precision and practicability are evaluated in detail. As the detectability of the proposed method is related to both the mass collected during exposure and the mass sensitivity of the detection step, the role of microanalytical procedures in general, and significance of meticulous control of contamination in particular are discussed. Under optimum conditions the gas-phase detection limit achievable (in terms of time independent exposure dose value) is about 2.3×103 g m–3 s–1, which permits measurements at low g m–3-levels with a time resolution of a few hours. The present study was carried out to develop an analytical method for the detection and quantification of bistrifluron, a benzoylphenylurea compound, in pear using high‐performance liquid chromatography with UV detection. Samples were extracted using conventional, AOAC and EN quick, easy, cheap, effective, rugged and safe ‘QuEChERS’ methods. As expected, conventional and EN‐QuEChERS methods gave higher recoveries than AOAC. In addition, interference around the analyte retention time was observed in the conventional method. Thus, the EN‐QuEChERS method was selected and validated by studying various parameters, including linearity, limit of detection, limit of quantification (LOQ), recovery and precision. Linearity was excellent, with a correlation coefficient of 0.9998. Recovery rates at three spiking levels (0.05, 0.2 and 1 mg/kg) ranged from 73.76 to 98.66%. Intra‐ and inter‐day precisions, expressed as relative standard deviations, were <6%. The LOQ of 0.05 mg/kg was considerably lower than the maximum residue limit (1 mg/kg) set by the Korean Ministry of Food and Drug Safety. The developed method was successfully applied to open‐field pear samples, in which the target analyte was slowly dissipated (55% decline) over 14 days with a half‐life of 10.19 days. Notably, the residue levels throughout the period of sample collection (14 days) were lower than the maximum residue limit, indicating that the residue was not hazardous for consumers. A method for the co-extraction and simultaneous chromatographic determination of fenthion and its five oxidation products (or metabolites) fenoxon, fenoxon-sulfoxide, fenoxon-sulfone, fenthion-sulfoxide, and fenthion-sulfone in personal protection equipment (PPE) of pesticide applicators was developed and validated. Capillary gas chromatography-nitrogen-phosphorus detection was used for the analytical determination of all the aforementioned compounds over the concentration range of 0.1-0.5 microg/mL. All necessary validation criteria of the method were met. The method was found to be highly selective, accurate, and precise, gave satisfactory recovery (>70%) and RSD values (<20%) for fenthion and its metabolites in all tested specimens of personal protection equipment and in air samplers. The limit of quantification (LOQ) and linearity were determined for the most relevant PPE (i.e. inner coverall) for the parent compound and for the metabolites. The LOQ values ranged from 0.05 to 0.1 ppb while the linearity in the tested range of 0.1-0.5 ppm had r(2)>0.994 for all analytes.  相似文献

A simple high-performance liquid chromatographic method is presented for the determination of trace amounts of sulphadimidine (SDD), its hydroxylated metabolites and N4-acetyl-SDD in blood plasma, urine, hepatocyte culture media and microsomal incubations. The synthesis of 5-hydroxy-SDD and an improved method for the isolation of 4-methylhydroxy-SDD from urine are described and their respective specific absorption coefficients at 265 nm are calculated by on-line radiochemical and ultraviolet detection. The limit of detection of the analytical method is 0.05 micrograms/ml for SDD and its hydroxy metabolites and 0.2 micrograms/ml for N4-acetyl-SDD. Linear calibration graphs for SDD and its metabolites were constructed from 0.2 to 50 micrograms/ml. The method has been applied to biotransformation studies in vivo and in vitro.  相似文献

A new analytical method was developed using liquid chromatography with tandem mass spectrometry for the routine analysis of 31 multi-class pesticide residues and applied to approximately 50 fresh fruit and vegetable samples (green bean, cucumber, pepper, tomato, eggplant, watermelon, melon and zucchini). Extraction of the pesticides with ethyl acetate was carried out. The optimal ionisation conditions were selected for each pesticide in the same run. The procedure was validated and the values of some merit figures, such as recovery, precision, linear range, detection limit and quantification limit for each pesticide were calculated together with its calculated expanded uncertainty (U). The average recoveries in cucumber obtained for each pesticide ranged between 74 and 105% at two different fortification levels (n = 10 each) that ranged between 9 and 250 ng g(-1) (depending on the pesticide). The uncertainty associated to the analytical method was lower than 23% for all compounds tested. The calculated limits of detection and quantitation were typically <1 ng g(-1) that were much lower than the maximum residue levels established by European legislation.  相似文献
