The interaction between lanthanide cationic porphyri
n and bovine serum albumin (BSA) was studied by fluorescence and UV-Vis spectrum. The static quenching of BSA was observed in the presence of YbTMPyP. According to the thermodynamic parameters, this binding was regarded as "enthalpy-driven" reaction. Furthermore, YbTMPyP is so close to the residues of BSA that molecular resonance energy transfer occurs between them. Besides, the red drift and hypochromicity of absorption spectrum of YbTMPyP were accompanied with the binding reaction. 相似文献

以光谱技术与微量热技术相结合的方法研究水溶液中金霉素与牛血清白蛋白分子间结合作用的热力学性质.荧光猝灭法测得该反应的结合常数K=2.09×105L/mol,结合位点数n=1.75,微量法测得反应的焓变△rHm= -17.50 kJ/mol; 依据Forster非辐射能量转移机制,得到授体-受体间的结合距离(r1=1.67 nm, r2=1.46 nm)和能量转移效率(E1=0.41, E2=0.66). 金霉素与牛血清白蛋白分子间有较强的结合作用, 且结合力以疏水作用为主. 相似文献

在模拟生理条件下,采用荧光光谱法、圆二色光谱法和红外光谱法研究了花椒油素(XT)与牛血清白蛋白(BSA)的相互作用。结果表明花椒油素与牛血清白蛋白之间发生动态和静态联合猝灭,二者间的的猝灭常数(K)在286, 298和310 K分别为3.31 × 105, 到2.03 × 105 和 0.94 × 105 L∙mol-1. 热力学参数表明, 花椒油素与牛血清白蛋白间以疏水作用力为主。圆二色光谱和红外光谱法表明加入花椒油素后,牛血清白蛋白的二级结构发生了变化,其中α-螺旋减少了3.9%。另外,我们还研究了共存离子对两者结合的影响。 相似文献

利用傅立叶变换红外(FT-IR)和拉曼(FT-Raman)光谱研究了高浓度磷脂酰胆碱(PC)与牛血清白蛋白(BSA)的相互作用,及Eu3+对该作用的影响.FT-IR结果显示,PC/BSA混合体系中二者的相互作用主要发生在PC头部极性基团,且这一作用随BSA含量的增加而增强,作用后蛋白质二级结构中α螺旋的比例有所增加. FT-Raman光谱说明PC与BSA的相互作用影响磷脂CH链的排列有序程度. PC/BSA/Eu3+体系的红外光谱显示, Eu3+与PC的磷氧键发生了强相互作用,并使蛋白α螺旋的比例进一步增加. 相似文献

在模拟生理条件下,用光谱法研究了硝酰基(HNO)探针(4-[2-(diphenylphosphino)benzoate]-N-butyl-1,8-naphtalimide,NIM)与牛血清白蛋白(BSA)的相互作用。结果表明,探针与BSA之间主要是静态猝灭方式。药物代替实验表明,探针与BSA的色氨酸残基相结合。由热力学数据确定了二者之间的作用力类型为静电引力。二者之间的结合距离为3.73nm。利用同步荧光、CD光谱、红外光谱以及三维荧光光谱考察了探针对BSA构象的影响。 相似文献

采用电化学方法并结合紫外、红外光谱分析, 研究了近生理pH 实验条件下新型抗血脂紊乱药物阿托伐他汀钙(AC)与牛血清白蛋白(BSA)的相互作用, 探讨了BSA对AC峰电流的增敏和减敏效应. 实验发现, 在pH为7.17的NaH2PO4-Na2HPO4缓冲溶液中, AC在静汞电极上产生一个还原峰, 加入BSA后峰电位发生移动且峰电流发生变化. 电化学研究结果表明, 部分疏水性AC小分子的芳香基团通过疏水作用镶嵌到BSA疏水微区内部, 使AC与BSA之间通过疏水作用力形成一种1:1的结合物, 结合常数为1.67×105. 紫外吸收光谱结果表明,AC的加入使BSA的紫外吸收峰发生红移且有减色效应, 导致BSA 构象改变、α-螺旋含量减小. 红外光谱结果显示, AC与BSA分子中氨基酸残基的硫及氮原子形成键合作用. 相似文献

运用荧光光谱和紫外-可见吸收光谱研究了在缓冲溶液中不同温度下孔雀石绿(MG)与牛血清白蛋白(BSA)之间的相互作用. 实验结果表明, MG对BSA的内源荧光猝灭为静态猝灭过程. 测定了该反应在不同温度下的结合常数KA, KA分别为7.69×104 L·mol-1(10 ℃)、5.31×104 L·mol-1(20 ℃)和4.85×104 L·mol-1(37 ℃), MG与BSA以摩尔比1:1结合. 根据Forster非辐射能量转移理论, 求出了37 ℃时给体(MG)和受体(BSA)之间能量转移效率和结合距离分别为E=0.1635 和r=2.30 nm. 计算出的热力学参数表明, MG 和BSA之间的作用力主要是通过氢键和范德华力相互作用. 相似文献

Abstract The binding of trazodone hydrochloride (TZH), an antidepressant drug, to bovine serum albumin (BSA) has been investigated by fluorescence spectroscopic analysis. The fluorescence emission of BSA (λem=350 nm) was quenched by TZH while that of this ligand was enhanced (λem=443 nm). The spectral behavior was consistent with the static quenching mechanism, and the apparent binding constant, K a (1.05×104 l mol?1) as well as binding site number, n (~1), were estimated. Thermodynamic parameters obtained from the measured data at different temperatures showed that the binding of TZH to BSA involved predominantly hydrophobic interactions as well as smaller contributions from electrostatic forces. Phenylbutazone and ibuprofen were utilized as competitive markers for sites I and II, respectively, in the interaction of TZH with BSA. This competitive displacement procedure indicated that the likely binding was site I, i.e., subdomain IIA, and this was supported by the observation that up to 50% of this site marker, phenylbutazone, could be exchanged with TZH whilst only a few percent of ibuprofen were so affected. 相似文献
