A method using sequential supercritical fluid extraction (SFE) and enzymatic transesterification has been developed for the rapid determination of total nutritional fat content in meat samples. SFE conditions of 12.16 MPa and 50°C were utilized to extract lipid species from the sample matrix. The enzymatic transesterification of the lipids by methanol was catalyzed by an immobilized lipase isolated from Candida antarctica. Conversion of the triglycerides to fatty acid methyl esters was monitored by supercritical fluid chromatography, while the fatty acid content of the extract was determined by capillary gas chromatography (GC). Total fat, saturated fat and monounsaturated fat contents were calculated from the GC data and compared to values from traditional extraction and lipid determination methods. Both off-line SFE and automated SFE followed by on-line GC analysis using two different instruments were utilized in this study. The enzymatic-based SFE method gave comparable results to the organic solvent extraction-based method followed by conventional BF3-catalyzed esterification.  相似文献

Analysis of low concentration polymer additives has been a challenging problem. The commonly used methods of analysis involve the initial extraction of polymer additives with solvents, often in a Soxhlet apparatus, followed by liquid, size exclusion, or gas chromatography. This paper describes the on-line super-critical fluid extraction (SFE)-supercritical fluid chromatographic (SFC) determination of different additives from low density polyethylene. Cryogenic collection was used as an interface between SFE and SFC to focus the extraction eluate before transfer to an analytical SFC column for quantitative analysis.  相似文献

The use of supercritical fluid extraction (SFE) is growing, with an expanding range of applications in many different fields as a consequence of its advantages compared with traditional extraction methods. In order to develop an analytical method to determine dibutyl phthalate (DBP) and dioctyl phthalate (DOP) traces (<20 ppm) in flexible poly(vinyl chloride) (PVC) formulations, a maximum efficiency in the extractive process and an adequate separative system are needed to avoid interferences between these two plasticizers and other additives that could be present at high concentrations in flexible PVC formulations. In order to determine the optimum SFE conditions, the extraction time, temperature and pressure were controlled. The separation and quantitation of individual components in the PVC extracts were carried out off-line by using a semicapillary column in gas chromatography (GC). Samples with different DOP content (41.18%, 33.33% and 23.08%) and DBP content (41.18%), as well as samples with both plasticizers (20.59% DOP and 20.59% DBP) were prepared. Some other samples were also prepared to study detection limits for these two PVC additives. Recoveries and reproducibilities were studied in every sample. Finally, this method was compared with Soxhlet liquid extraction. Determination by gravimetric analysis of the total extracted material was found to be particularly suitable for PVC. This study demonstrates the potential of SFE to shorten extraction times with similar or even better extraction efficiencies compared with traditional liquid methods.  相似文献

Supercritical fluid extraction (SFE) of the fluoroquinolones norfloxacin and ofloxacin from chicken breast muscles was examined. A liquid chromatography with fluorescence detection was used for the determination of the fluoroquinolones. Extraction conditions of the SFE were optimized by determining the extraction parameters to achieve a sufficiently high recovery of each fluoroquinolone in fortified-muscle samples. Recovery values for the extraction of the fluoroquinolones using the SFE ranged from 70 to 87%. Chickens were treated orally with each fluoroquinolone and their muscles were extracted at set time intervals for time-course determination of the fluoroquinolones in chickens. The SFE combined with liquid chromatographic analysis showed that the concentrations of the fluoroquinolones decreased gradually with time in the chicken muscles after oral treatment, giving a concentration less than 5 ng/ml in 120 h. No further sample cleanup procedures were required after the SFE. These results suggest that SFE method is an extraction method for the determination of norfloxacin and ofloxacin in chicken muscle.  相似文献

An analytical method based on CO2 supercritical fluid extraction (SFE) followed by gas chromatography (GC) was evaluated, compared to Soxhlet extraction, and found to determine accurately and precisely diesel fuel contamination of standard soil samples at a total petroleum hydrocarbon level of 100 μg/g in soil. While both extraction methods have the same 3% relative repeatability standard deviation for determination of total hydrocarbon contamination at this level, SFE requires much less time, uses less organic solvent and provides better recovery of the more volatile n-C10 to n-C12 hydrocarbons.  相似文献

An automated method using supercritical CO2 and clean-up by solid-phase extraction (SPE) using graphitized carbon black, has been developed for the quantitative determination of organophosphorus pesticide (OPP) residues in wheat and maize. Recoveries were as good as, or better than, those obtained using liquid extraction (LE) and gel permeation chromatography (GPC) for 10 OPP's spiked at levels equivalent to 0.05 and 0.50 microg/g. Lower limits of detection were possible using supercritical fluid extraction (SFE). Incurred residues were found in wheat and maize samples, and good agreement was obtained using SFE+SPE and LE+GPC. The SFE+SPE method required less analyst time and organic solvent, and hazardous waste was reduced.  相似文献

Supercritical fluid extraction (SFE) of essential oil from Pimpinella anisum, using carbon dioxide as a solvent is presented in this work. An orthogonal array design OA9 (3(4)) was applied to select the optimum extraction condition. The effects of pressure, temperature, dynamic extraction time and methanol volume on the extraction efficiency were investigated by the three-level orthogonal array design. Results show that pressure has a significant effect on the extraction efficiency. The extract obtained from P. anisum by using supercritical fluid extraction was compared with the essential oil obtained by hydrodistillation, considering both quantity and quality of the product. SFE products were found to be of markedly different composition, compared with the corresponding hydrodistilated oil. The total amount of extractable substances obtained in SFE (7.5%) is higher than that obtained by hydrodistillation (3.1%) and SFE is faster than hydrodistillation method.  相似文献

Supercritical fluid extraction (SFE) with CO(2), a clean and rapid alternative to conventional Soxhlet extraction, was investigated for the extraction of nitro compounds from soil samples. Quantitative extraction by SFE was accomplished at a pressure of 25 MPa and an extraction temperature of 60 degrees C, for 30 min in dynamic mode and using acetonitrile as modifier, and the results were comparable with those obtained by acetonitrile Soxhlet extraction (3 h) for all soil samples. Extracts from these two procedures were analyzed by gas chromatography coupled with mass spectrometry. Quantitative reproducibility for SFE extracts was acceptable (RSD 2-10%), and the quantity of solvent was reduced from 160 mL for Soxhlet extraction to 5 mL in the case of SFE.  相似文献

A two-step supercritical fluid extraction (SFE) method has been developed for the analysis of oxygenated and nitrated polycyclic aromatic hydrocarbons (oxy- and nitro-PAHs, respectively) present in urban aerosol samples. The proposed SFE procedure first involves an extraction step using pure CO2 in order to remove the less polar compounds from the matrix and a second consecutive step using toluene-modified CO2. The oxy- and nitro-PAHs are obtained in the second step. Parameters affecting both collection efficiencies and the selective extraction of oxy- and nitro-PAHs in the second SFE step were optimised. Analysis of the extracts was performed using gas chromatography with electron-capture detection and coupled to mass spectrometry. The proposed SFE method was compared with a conventional extraction technique such as sonication and good agreement in the results was obtained. Nevertheless, clean up of sonication extracts was needed, whereas no purification was necessary for SFE extracts. The SFE method was applied to the analysis of oxy- and nitro-PAHs in urban aerosol samples and 9-fluorenone, 9,10-anthraquinone, 2-methyl-9,10-anthraquinone, benzanthrone, benz[a]anthracene-7,12-dione and 1-nitropyrene were identified at concentrations ranging between 15 and 364 pg m(-3).  相似文献

Summary The determination of 2,4-D (2,4-dichlorophenoxyacetic acid) and Dicamba (2-methoxy-3,6-dichlorobenzoic acid) residues in sugar cane, rice and corn was performed by a supercritical fluid extraction (SFE) method using CO2/acetone as extraction mix and an SFE apparatus developed in our laboratory. The extracts were cleaned up after extraction by both liquid-liquid partition and a Florisil column. Micellar electrokinetic capillary chromatography (MEKC) coupled with ultraviolet on-column detection was used for the analysis of these pesticides. The detection limits were improved by the preparation of a special detection cell with an increased pathlength that gave detection limits of ca. 0.6 pg for 2,4-D and Dicamba. Our results demonstrated that capillary electrophoresis can be a powerful new analytical tool for pesticide residue analysis.  相似文献

Abstract

Application of supercritical fluid extraction (SFE) utilizing pure carbon dioxide for selective isolation of organophosphates from contaminated cereals has been tested.

At the beginning of the experiments the extractability of added standards from an empty extraction vessel (thimble) and from various materials such as filter paper, sand, Celite and anhydrous sodium sulfate was tested to estimate the behavior of organophosphates. Further method development was carried out using a spiked sample of flour, which was analyzed within the proficiency testing for organophosphorus pesticides analysis (round 7) organized by Food Analysis Performance Assessment Scheme (FAPAS, MAFF-UK).

Comparison of the SFE method with a classical method currently employed for sample preparation (i.e. extraction with acetone/methanol mixture followed by gel permeation chromatographic clean up) showed advantages of the SFE technique such as simplification of the sample preparation step and thereby significant speeding up of the determination of organophosphates in cereals.  相似文献

Supercritical fluid extraction (SFE) has been evaluated as an extraction technique for the isotope-dilution quantification of polychlorinated biphenyls (PCBs) in a sediment sample. A high-resolution mass-spectrometric system equipped with a gas chromatograph was employed for the determination of seven target PCB congeners. The effect of the operation parameters on the SFE efficiency was investigated, in which the analytical values of five target PCB congeners significantly increased with increases in the extraction temperature and pressure, and that of 4,4'-dichlorobiphenyl further increased by applying static extraction. The following conditions were found to be optimal: extraction temperature, 140 degrees C; pressure, 30 MPa; time and mode, static for 15 min then dynamic for 30 min. Under these conditions, the addition of modifiers influenced the extraction of polar compounds, but did not affect the analytical values of the PCB congeners. The optimized method was suitable for high-throughput analysis as well as for providing accurate analytical results, which were comparable to or better than the analytical results obtained by Soxhlet extraction.  相似文献

A method combining the techniques of liquid – solid disk extraction (LSDE) and supercritical fluid elution (SFE) has been developed for the phenols regulated by the Clean Water Act. LSDE uses a disk or membrane made of polytetrafluoroethylene (PTFE) fibrils impregnated with small particles, e.g. styrene divinylbenzene (SDB) resin, to extract phenols from water. After disk extraction the retained analytes are eluted from the disk using SFE. SFE is used as an alternative to liquid solvent elution with an organic solvent. Analytes are separated, identified, and quantified using gas chromatography – ion trap detector mass spectrometry (GC-ITDMS). The method is capable of sub parts per billion detection limits, and precision of 5–28% RSD. Evaluation of various disks or membranes, such as C18-silica disks, SDB disks, and ion exchange membranes, has also been performed for the extraction of phenols from water. The results obtained from the in-situ aqueous acetylation of phenols and extraction of their acetates are quantitative. The utilization of LSDE and SFE techniques has proven to be a more effective approach than liquid – liquid extraction in minimizing air pollution and solvent waste.  相似文献

The efficiencies of extraction of vapor-deposited pyrene from a high-carbon coal stack ash by Soxhlet extraction with methanol, ultrasonic extraction with toluene, acid pretreatment and subsequent ultrasonic extraction with toluene, batch extraction with toluene, and supercritical fluid extraction (SFE) are compared. SFE using CO(2) or isobutane yielded extraction recoveries virtually identical with those obtained using ultrasonic or Soxhlet extraction processes. Collection of the SFE extract was performed by expansion into a solvent or onto the head of a gas chromatography (GC) column. No loss of extracted pyrene was observed upon collection of methanol-modified CO(2) SFE by expansion into methanol. Also, no loss of pure CO(2) SFE extract was observed upon collection on the head of a GC column. However, use of a methanol or toluene modifier for CO(2) SFE directly coupled to GC effected complete loss of extracted pyrene.  相似文献

The volatile components of Cnidium monnieri were obtained by supercritical fluid extraction (SFE) and analyzed by GC‐MS (identification and determination of metabolites). The compounds were identified according to their retention times and mass spectra. The effects of different parameters, such as extraction pressure, temperature, dynamic extraction time, flow rate of CO2, on the SFE of C. monnieri extracts were investigated. A total of 14 compounds of SFE extracts were identified. Osthole (69.52%), bornyl acetate (10.03%), α‐pinene (4.71%), and imperatorin (2.42%) were the major compounds identified in C. monnieri SFE extracts. The quantitation of osthole and imperatorin were then accomplished. The linear calibration ranges were all 5–1000 μg/mL for osthole and imperatorin by GC‐MS analysis. The recovery of osthole and imperatorin were in the range 96.5–101.8%. The LODs for osthole and imperatorin were 1.0 and 0.6 μg/mL, respectively.  相似文献

A novel integral restrictor–collector has been designed for use with a conventional supercritical fluid extraction (SFE) apparatus. The assembly reduces the path length between a micrometering valve and collector (a solid phase extraction (SPE) column), obviating the need for the complicated tubing and connectors usually associated with such devices. Also described is a heating-block assembly which encases the micrometering valve and provides uniform heating of the valve during extraction. The valve–SPE column assembly was part of a system used to perform the first reported SFE multi-residue drug recovery from fortified liver. Extractions used carbon dioxide pressurized to 690 bar as the supercritical fluid. Flow rates of expanded gas through the SPE columns were 3–4 L/min with concomitant quantitative trapping of the analytes on the sorbent bed. After SFE the three nitrobenzamide antimicrobial drug residues from the liver were eluted from the SPE columns by off-line analysis. The results demonstrated that losses of trace level analytes in tissue may be significantly reduced by including an integral metering valve-collector assembly as part of the SFE apparatus.  相似文献

Two different relatively simple, commercially available supercritical fluid extractors (SFE), Leco and Foss-Tecator, were tested for the determination of total fat content in meat and meat products. The fatty acid composition in meat and meat products was also determined after the Foss-Tecator extraction in an aliquot of the extract. Total fat was determined by weighing after the different extraction procedures and the fatty acid composition by gas chromatography after hydrolysis and methylation of the extract. The results for total fat content agreed well with results from a standard method of Schmid, Bondzynski, and Ratzlaff, which uses conventional solvent extraction. Fatty acid composition was compared with the Bligh and Dyer extraction, and showed good agreement. The average relative difference between SFE and Bligh and Dyer of all fatty acids in the sample was <3% for acids exceeding 0.5% of total fatty acid amount. The advantages of SFE over traditional methods are a much lower consumption of hazardous organic solvents and shorter extraction times. To obtain quantitative recoveries by SFE, ethanol was added to the extraction cells before extraction.  相似文献
