Substitution reactions of the complexes [Pt(terpy)(H(2)O)](2+), [Pt(terpy)(cyst-S)](2+) and [Pt(terpy)(guo-N(7))](2+), where terpy = 2,2':6',2"-terpyridine, cyst = L-cysteine and guo = guanosine, with some biologically relevant ligands such as inosine (INO), inosine-5'-monophosphate (5'-IMP), guanosine-5'-monophosphate (5'-GMP), l-cysteine, glutathione, thiourea, thiosulfate and diethyldithiocarbamate (DEDTC), were studied in aqueous 0.10 M NaClO(4) at pH 2.5 and 6.0 using variable-temperature and -pressure stopped-flow spectrophotometry. The reactions of [Pt(terpy)(H(2)O)](2+) with INO, 5'-IMP and 5'-GMP showed that these ligands are very good nucleophiles. The second order rate constants varied between 4 x 10(2) and 6 x 10(2) M(-1) s(-1) at 25 degree C. The [Pt(terpy)(cyst-S)](2+) complex is unreactive towards nitrogen donor nucleophiles, and cysteine cannot be replaced by N(7) from INO, 5'-IMP and 5'-GMP. However, sulfur donor nucleophiles such as thiourea, thiosulfate and diethyldithiocarbamate could displace the Pt-cysteine bond. Diethyldithiocarbamate is the best nucleophile and the order of reactivity is: thiourea < thiosulfate < DEDTC with rate constants of 0.936 +/- 0.002, 5.99 +/- 0.02 and 8.88 +/- 0.07 M(-1) s(-1) at 25 degree C, respectively. The reactions of [Pt(terpy)(guo-N(7))](2+) with sulfur donor ligands showed that these nucleophiles could substitute guanosine from the Pt(ii) complex, of which diethyldithiocarbamate and thiosulfate are the strongest nucleophiles. The tripeptide glutathione is also a very efficient nucleophile. Activation parameters (Delta H(++), Delta S(++) and Delta V(++)) were determined for all reactions. The crystal structures of [Pt(terpy)(cyst-S)](ClO(4))(2).0.5H(2)O and [Pt(terpy)(guo-N(7))](ClO(4))(2).0.5guo.1.5H(2)O were determined by X-ray diffraction. Crystals of [Pt(terpy)(cyst-S)](ClO(4))(2).0.5H(2)O are orthorhombic with the space group P2(1)2(1)2(1), whereas [Pt(terpy)(guo-N(7))](ClO(4))(2).0.5guo.1.5H(2)O crystallizes in the orthorhombic space group P2(1)2(1)2. A typical feature of terpyridine complexes can be found in both molecular structures: the Pt-N (central) bond distance, 1.982(7) and 1.92(2) A, respectively, is shorter than the other two Pt-N distances, being 2.043(7) and 2.034(7) A in [Pt(terpy)(cyst-S)](ClO(4))(2).0.5H(2)O and 2.03(2) and 2.04(2) A in [Pt(terpy)(guo-N(7))](ClO(4))(2).0.5guo.1.5H(2)O, respectively. In both crystal structures two symmetrically independent cations representing different conformers are present in the asymmetric unit. The results are analysed in reference to the antitumour activity of Pt(II) complexes, and the importance of the rescue agents are discussed.  相似文献

The novel dinuclear Pt(II) complexes [{trans-Pt(NH(3))(2)Cl}(2)(μ-pyrazine)](ClO(4))(2) (Pt1), [{trans-Pt(NH(3))(2)Cl}(2)(μ-4,4'-bipyridyl)](ClO(4))(2)·DMF (Pt2), and [{trans-Pt(NH(3))(2)Cl}(2)(μ-1,2-bis(4-pyridyl)ethane)](ClO(4))(2) (Pt3), were synthesized. Acid-base titrations, and temperature and concentration dependent kinetic measurements of the reactions with biologically relevant ligands such as thiourea (Tu), glutathione (GSH) and guanosine-5'-monophosphate (5'-GMP) were studied at pH 2.5 and 7.2. The reactions were followed under pseudo-first-order conditions by stopped-flow and UV-vis spectrophotometry. (1)H NMR spectroscopy was used to follow the substitution of chloride in the complex [{trans-Pt(NH(3))(2)Cl}(2)(μ-4,4'-bipyridyl)](ClO(4))(2)·DMF by guanosine-5'-monophosphate (5'-GMP) under second-order conditions. The results indicate that the bridging ligand has an influence on the reactivity of the complexes towards nucleophiles. The order of reactivity of the investigated complexes is Pt1 > Pt2 > Pt3.  相似文献

A dinuclear platinum(II) complex that was recently investigated in our group was tested for its cytostatic activity and found to be active against HeLa S3 cells. The complex consists of a bidentate N,N-donor chelating ligand system in which the two platinum centers are connected by an aliphatic chain of 10 methylene groups. The complex [Pt(2)(N(1),N(10)-bis(2-pyridylmethyl)-1,10-decanediamine)(OH(2))(4)](4+) (10NNpy) is of further special interest, since only little is known about the substitution behavior of such dinuclear platinum complexes that contain a bidentate coordination sphere. The complex was investigated using different biologically relevant nucleophiles, such as thiourea (tu), L-methionine (L-Met), glutathione (GSH), and guanine-5'-monophosphate (5'-GMP), at two different pH values (2 and 7.4). The substitution of coordinated water by these nucleophiles was studied under pseudo-first-order conditions as a function of nucleophile concentration, temperature, and pressure, using stopped-flow techniques and UV-vis spectroscopy. The reactivity of 10NNpy with the selected nucleophiles was found to be tu ? 5'-GMP > L-Met > GSH at pH 2 and GSH > tu > L-Met at pH 7.4. The results for the dinuclear 10NNpy complex were compared to those for the corresponding mononuclear reference complex [Pt(aminomethylpyridine)(OH(2))(2)](2+), Pt(amp), studied before in our group, by which the effect of the addition of an aliphatic chain, an increase in the overall charge, and a shift in the pK(a) values of the coordinated water ligands could be investigated. The kinetics of the substitution reactions between the mono-functional Au(III) complexes, [Au(dien)Cl](2+) and [Au(terpy)Cl](2+) (dien = 3-azapentane-1,5-diamine, terpy = 2,2';6',2'-terpyridine) and bi-functional Au(III) complexes, [Au(bipy)Cl(2)](+) and [Au(dach)Cl(2)](+) (bipy = 2.2'-bipyridine, dach = (1R,2R)-1,2-diaminocyclohexane) and biologically relevant ligands such as l-histidine (l-His), inosine (Ino), inosine-5'-monophosphate (5'-IMP) and guanosine-5'-monophosphate (5'-GMP), were studied in detail. All kinetic studies were performed in 25 mM Hepes buffer (pH = 7.2) in the presence of NaCl to prevent the spontaneous hydrolysis of the chloride complexes. The reactions were followed under pseudo-first order conditions as a function of ligand concentration and temperature using stopped-flow UV-vis spectrophotometry. The results showed that the mono-functional complexes react faster than the bi-functional complexes in all studied reactions. The [Au(terpy)Cl](2+) complex is more reactive than the [Au(dien)Cl](2+) complex, which was confirmed by quantum chemical (DFT) calculations. A more than 50% lower activation energy for the terpy than for the dien based complex was found. The bi-functional [Au(bipy)Cl(2)](+) complex is more reactive than the [Au(dach)Cl(2)](+) complex. The reactivity of the studied nucleophiles follows the same order for all studied systems, viz. l-His > 5'-GMP > 5'-IMP > Ino. According to the measured activation parameters, all studied reactions follow an associative substitution mechanism. Quantum chemical calculations (B3LYP/LANL2DZp) suggest that ligand substitution in [Au(terpy)Cl](2+) and [Au(dien)Cl](2+) by imidazole follows an interchange mechanism with a significant degree of associative character. The results demonstrate the strong connection between the reactivity of the complexes toward biologically relevant ligands and their structural and electronic characteristics. Therefore, the binding of gold(III) complexes to 5'-GMP, constituent of DNA, is of particular interest since this interaction is thought to be responsible for their anti-tumour activity.  相似文献

The formation equilibria of the [Pt(SMC)(H2O)2]+ complex with some biologically relevant ligands such as L-methionine (L-met) and glutathione (GSH) were studied. The stoichiometry and stability constants of the formed complexes are reported, and the concentration distribution of the various complex species has been evaluated as a function of pH. The reaction between [PtCl2(SMC)] and guanosine-5′-monophosphate (5′-GMP) was studied by 1H NMR spectroscopy. The NMR spectra indicated that first step is the hydrolysis of the [PtCl2(SMC)] complex and second step is the substitution of an aqua ligand, either in the cis or trans position with guanosine-5′-monophosphate in molar ratio 1:1. The values of rate constant showed faster substitution of coordinated H2O in the trans position to the S donor atom of S-methyl-L-cysteine, whereas the slower reaction was assigned to the displacement of the cis coordinated aqua molecule. This is due to the strong trans labilization effect of coordinated sulfur. Complexes of the type [Pt2(N,N,N',N'-tetrakis(2-pyridylmethyl)diamine(HO)2]4+ and [Pt2(N,N,N',N'-tetrakis(2-pyridylmethyl)diamine(Cl)2]2+ were used to study their reactions with a series of bio-relevant nucleophiles, viz. thiourea, L-methionine and guanosine-5'-monophosphate (5'-GMP2-) as a function of nucleophile concentration and temperature. The reactions with the sulfur containing nucleophiles (thiourea and L-methionine) were followed under pseudo-first-order conditions by stopped-flow and UV-Vis spectrophotometry. The reaction with 5'-GMP2- was carried out under second order conditions and studied by NMR spectroscopy. The results indicate that the bridged dinuclear complexes remain intact after coordination of the studied nucleophiles for an extended period of time, which differs significantly from that reported for other multinuclear platinum complexes in the literature.  相似文献

The kinetics and the mechanism of the substitution reactions of the complex [(TL(tBu))PdCl](+), where TL(tBu) is 2,6-bis[(1,3-di-tert-butylimidazolin-2-imino)methyl]pyridine, with nucleophiles (guanosine-5'-monophosphate (5'-GMP), l-Methionine (l-Met) and l-Histidine (l-His)) were studied using variable-temperature stopped-flow techniques in aqueous 0.1 M NaClO(4) with 10 mM NaCl at 298 K. The order of reactivity is: l-Met > 5'-GMP > l-His. The formation equilibria of [(BL(iPr))Pd(H(2)O)(2)](2+), where BL(iPr) is 1,2-bis(1,3diisopropyl-4,5-dimethylimidazolin-2-imino)ethane, and [(TL(tBu))Pd(H(2)O)](2+) with some biologically relevant ligands (l-Met, 5'-GMP and l-His) were also studied. The stoichiometry and stability constants of the newly formed complexes are reported, and the concentration distribution of the various complex species has been evaluated as a function of pH. Comparing the values of logβ(1,1,0) for 5'-GMP, l-His and l-Met complexes, the most stable complex is with 5'-GMP followed by l-His and l-Met for both complexes, [(BL(iPr))Pd(H(2)O)(2)](2+) and [(TL(tBu))Pd(H(2)O)](2+). The crystal structures of [(TL(tBu))PdCl]ClO(4) and [(BL(iPr))PdCl(2)] were determined by X-ray diffraction. The coordination geometries around the palladium atoms are distorted square-planar, with the Pd-N1 distance to the central nitrogen atom of the TL(tBu) ligand, 1.944(2) ?, being shorter than those to the other two nitrogen atoms of TL(tBu), viz. 2.034(3) and 2.038(2) ?. The BL(iPr) complex displays similar Pd-N distances of 2.031(2) and 2.047(2) ?.  相似文献

A dimer-of-dimers model compound for the oxygen-evolving complex of photosystem II, [[(H(2)O)(terpy)Mn(IV)(micro-O)(2)Mn(IV)(terpy)](2)(micro-O)](ClO(4))(6) (terpy = 2,2':6',2' '-terpyridine), has been prepared and characterized by X-ray crystallography and ESI-MS. Low pH was found to promote the disproportionation of [Mn(III/IV)(2)O(2)(terpy)(2)(OH(2))(2)](3+) to Mn(2+) and a Mn(IV/IV)(2)O(2)(terpy)(2) species; the latter complex slowly dimerizes to form the title complex. Protonation of a micro-oxo bridge is proposed to initiate the disproportionation, based on analogy with the [Mn(III/)(IV)(2)O(2)(bpy)(4)](3+) system.  相似文献

Compounds [RuII(bipy)(terpy)L](PF6)2 with bipy = 2,2'-bipyridine, terpy = 2,2':6',2"-terpyridine, L = H2O, imidazole (imi), 4-methylimidazole, 2-methylimidazole, benzimidazole, 4,5-diphenylimidazole, indazole, pyrazole, 3-methylpyrazole have been synthesized and characterized by 1H NMR, ESI-MS and UV/Vis (in CH3CN and H2O). For L = H2O, imidazole, 4,5-diphenylimidazole and indazole the X-ray structures of the complexes have been determined with the crystal packing featuring only few intermolecular C-H...pi or pi-pi interactions due to the separating action of the PF6-anions. Complexes with L = imidazole and 4-methylimidazole exhibit a fluorescence emission with a maximum at 662 and 667 nm, respectively (lambdaexc= 475 nm, solvent CH3CN or H2O). The substitution of the aqua ligand in [Ru(bipy)(terpy)(H2O)]2+ in aqueous solution by imidazole to give [Ru(bipy)(terpy)(imi)]2+ is fastest at a pH of 8.5 (as followed by the increase in emission intensity). Coupling of the [Ru(bipy)(terpy)]2+ fragment to cytochrome c(Yeast iso-1) starting from the Ru-aqua complex was successful at 35 degrees C and pH 7.0 after 5 d under argon in the dark. The [Ru(bipy)(terpy)(cyt c)]-product was characterized by UV/Vis, emission and mass spectrometry. The location where the [Ru(bipy)(terpy)] complex was coupled to the protein was identified as His44 (corresponding to His39 in other numbering schemes) using digestion of the Ru-coupled protein by trypsin and analysis of the tryptic peptides by HPLC-high resolution MS.  相似文献

We report that the ability of disodium 5'-deoxy-5'-thioguanosine-5'-monophosphate, Na(2)(5'-GSMP), to self-associate into a helical G-quadruplex structure in aqueous solution at pH 8 is significantly higher than that of disodium guanosine-5'-monophosphate, Na(2)(5'-GMP), which supports our earlier hypothesis regarding the importance of cation bridging.  相似文献

The kinetics and mechanism of substitution reactions of novel monofunctional [Pt(tpdm)Cl](+) and [Pd(tpdm)Cl](+) complexes (where tpdm = tripyridinedimethane) and their aqua analogues with thiourea (tu), L-methionine (L-met), glutathione (GSH), and guanosine-5'-monophosphate (5'-GMP) were studied in 0.1 M NaClO(4) at pH = 2.5 (in the presence of 10 mM NaCl for reactions of the chlorido complexes). The reactivity of the investigated nucleophiles follows the order tu > l-met > GSH > 5'-GMP. The reported rate constants showed the higher reactivity of the Pd(II) complexes as well as the higher reactivity of the aqua complex than the corresponding chlorido complex. The negative values reported for the activation entropy as well as the activation volume confirmed an associative substitution mode. In addition, the molecular and crystal structure of [Pt(tpdm)Cl]Cl was determined by X-ray crystallography. The compound crystallizes in a monoclinic space group C2/c with two independent molecules of the complex and unit cell dimensions of a = 38.303(2) ?, b = 9.2555(5) ?, c = 27.586(2) ?, β = 133.573(1)°, and V = 7058.3(8) ?(3). The cationic complex [Pt(tpdm)Cl](+) exhibits square-planar coordination around the Pt(II) center. The lability of the [Pt(tpdm)Cl](+) complex is orders of magnitude lower than that of [Pt(terpyridine)Cl](+). Quantum chemical calculations were performed on the [Pt(tpdm)Cl](+) and [Pt(terpyridine)Cl](+) complexes and their reactions with thiourea. Theoretical computations for the corresponding Ni(II) complexes clearly demonstrated that π-back-bonding properties of the terpyridine chelate can account for acceleration of the nucleophilic substitution process as compared to the tpdm chelate, where introduction of two methylene groups prevents such an effective π-back bonding.  相似文献

We report a combined NMR and dynamic light scattering (DLS) study on the size of supramolecular structures formed by disodium guanosine 5'-monophosphate, Na(2)(5'-GMP), at pH 8. In general, two distinct types of aggregate species are present in an aqueous solution of Na(2)(5'-GMP). One type consists of stacking 5'-GMP monomers, and the other contains stacking G-quartets. Both types of aggregates can be modeled as rodlike cylinders. The cylinder diameter is 10 and 26 A for monomer aggregates and quartet aggregates, respectively. For Na(2)(5'-GMP) concentrations between 18 and 34 wt %, the cylinders formed by stacking G-quartets have an average length between 8 and 30 nm, corresponding to a stack of approximately 24-87 G-quartets. These nanoscale aggregates are significantly larger than what had previously been believed for Na(2)(5'-GMP) self-association at pH 8. The length of both types of 5'-GMP aggregates was found to increase with Na(2)(5'-GMP) concentration but was insensitive to the added NaCl in solution. While the aggregate size for monomer aggregates increases with a decrease in temperature, the size of G-quartet aggregates is essentially independent of temperature. We found that the size of G-quartet aggregates is slightly larger in D(2)O than in H(2)O, whereas the size of monomer aggregates remains the same in D(2)O and in H(2)O. We observed a linear relationship between the axial ratio of the 5'-GMP cylinders and the Na(2)(5'-GMP) concentration for both types of 5'-GMP aggregates, which suggests a common stacking mechanism for monomers and G-quartets.  相似文献

This paper reports on the chemistry of platinum complexes containing bidentate pyridine-carboxylate (pyAc = pyridin-2-yl-acetate and picEt = pyridine-2-ethylcarboxylate, ethylpicolinate) (N,O) ligands. The pyridine-2-acetate and ethylpicolinate ligands form six- and five-membered chelates, respectively, upon formation of the Pt-carboxylate bond. In all reactions with picEt with various platinum complex starting materials, spontaneous de-esterification of the pendant carboxylate ester occurs to give directly the chelates K[PtCl(2)(pic-N,O)]-trans-[Pt(pic-N,O)(2)] and SP-4,2-[PtCl(pic-N,O)(NH(3))] without any evidence of intermediates. The de-esterification is solvent dependent, and molecular modeling was used to explain this reaction. The reactions of the geometric isomers of [PtCl(pyAc-N,O)(NH(3))] with 5'-guanosine monophosphate, 5'-GMP, and N-acetyl-l-methionine, AcMet, were investigated by NMR spectroscopy. The objective was to ascertain by model chemistry the feasibility of formation of ternary DNA-Pt-protein adducts in biology. Model nucleotide and peptide compounds were formed in situ by chloride displacement giving [PtL(pyAc-N,O)(NH(3))](+) (L = 5'-GMP or AcMet). Competitive reactions were then examined by addition of the complementary ligand L. Sulfur displacement of coordinated 5'-GMP was slow. For SP-4,3-[Pt(AcMet)(NH(3))(PyAc-N,O)](+), a rapid displacement of the sulfur ligand by 5'-GMP was observed, giving SP-4,2-[Pt(5'-GMP-N7)(pyAc-N,O)(NH(3))](+).  相似文献

Reaction of the novel thiopropyl-closo-1,2-carborane ligand bearing a pendant glycerol group HS(CH(2))(3)CB(10)H(10)CCH(2)OCH(CH(2)OH)(2)(L) with the labile platinum(ii) precursor [Pt(MeCN)(terpy)](OTf)(2)(terpy = 2,2':6',2'-terpyridine; OTf = trifluoromethanesulfonate) affords the highly water-soluble platinum(ii) complex [PtL(terpy)]OTf, the first example of a metal-carborane complex functionalised with a water-solubilising glycerol group.  相似文献

The kinetics of complex-formation reactions of six Pd(dach) complexes, dach = 1,2-trans-R,R-diaminocyclohexane, viz. [Pd(dach)Cl2], [Pd(dach)(H2O)2]2+, and four complexes with different chelating leaving groups X-Y, viz. [Pd(dach)(O,O-cyclobutane-1,1-dicarboxylate)], [Pd(dach)(N,O-glycine)](+), [Pd(dach)(N,S-methionine)]+ and [Pd(dach)(O,O-oxalate)], were studied. The effect of the leaving group on the lability of the resulting Pd(ii) complexes was studied for the nucleophiles inosine, inosine-5'-monophosphate and guanosine-5'-monophosphate under pseudo-first-order conditions as a function of nucleophile concentration, temperature and pressure using stopped-flow techniques. Two consecutive reaction steps, which both depend on the nucleophile concentration, were observed. The rate constants for all reactions indicate a direct substitution of the X-Y chelate by the selected nucleophiles, thereby showing that the nature of the chelate, viz. O-O (cbdca), (ox), N-O (gly) or S-N (l-met), plays an important role in the kinetic and mechanistic behavior of the Pd(ii) complexes. The mechanism of the substitution reactions is associative in nature as supported by the large and negative values of DeltaS(double dagger) and DeltaV(double dagger).  相似文献

DFT calculations using the B3LYP functional support the suggestion that the [(terpy)(H(2)O)Mn(IV)(micro-O)(2)Mn(III)(H(2)O)(terpy)](3+) (terpy=2,2':6,2' '-terpyridine) complex functions as a synthetic O(2) catalyst. The calculated barrier for O-O bond formation with water is 23 kcal/mol. In this complex, as well as in models of the oxygen evolving complex in PSII, the active species is a Mn(IV)-oxyl radical. From comparisons with inactive Mn(V)-oxo complexes, it is proposed that radical formation is actually a requirement for O(2) formation activity in Mn-complexes.  相似文献

Insight into the N7/O6 equatorial binding interactions of the antitumor active complex Rh(2)(OAc)(4)(H(2)O)(2) (OAc(-) = CH(3)CO(2)(-)) with the nucleotide 5'-GMP and the DNA fragment d(pGpG) has been obtained by one- (1D) and two-dimensional (2D) NMR spectroscopy. The lack of N7 protonation at low pH values and the significant increase in the acidity of N1-H (pK(a) approximately 5.6 as compared to 8.5 for N7 only bound platinum adducts), indicated by the pH dependence study of the H8 (1)H NMR resonance for the HT (head-to-tail) isomer of Rh(2)(OAc)(2)(5'-GMP)(2), are consistent with bidentate N7/O6 binding of the guanine. The H8 (1)H NMR resonance of the HH (head-to-head) Rh(2)(OAc)(2)(5'-GMP)(2) isomer, as well as the 5'-G and 3'-G H8 resonances of the Rh(2)(OAc)(2) [d(pGpG)] adduct exhibit pH-independent titration curves, attributable to the added effect of the 5'-phosphate group deprotonation at a pH value similar to that of the N1 site. The enhancement in the acidity of N1-H, with respect to N7 only bound metal adducts, afforded by the O6 binding of the bases to the rhodium centers, has been corroborated by monitoring the pH dependence of the purine C6 and C2 (13)C NMR resonances for Rh(2)(OAc)(2)(5'-GMP)(2) and Rh(2)(OAc)(2) [d(pGpG)]. The latter studies resulted in pK(a) values in good agreement with those derived from the pH-dependent (1)H NMR titrations of the H8 resonances. Comparison of the (13)C NMR resonances of C6 and C2 for the dirhodium adducts Rh(2)(OAc)(2)(5'-GMP)(2) and Rh(2)(OAc)(2) [d(pGpG)] with the corresponding resonances of the unbound ligands at pH 8.0, showed substantial downfield shifts of Deltadelta approximately 11.0 and 6.0 ppm, respectively. The HH arrangement of the bases in the Rh(2)(OAc)(2) [d(pGpG)] adduct is evidenced by intense H8/H8 ROE cross-peaks in the 2D ROESY NMR spectrum. The presence of the terminal 5'-phosphate group in d(pGpG) results in stabilization of one left-handed Rh(2)(OAc)(2) [d(pGpG)] HH1 L conformer, due to the steric effect of the 5'-group, favoring left canting in cisplatin-DNA adducts. Complete characterization of the Rh(2)(OAc)(2[d(pGpG)] adduct revealed notable structural features that resemble those of cis-[Pt(NH(3))(2) [d(pGpG)]]; the latter involve repuckering of the 5'-G sugar ring to C3'-endo (N-type) conformation, retention of C2'-endo (S-type) 3'-G sugar ring conformation, and anti orientation with respect to the glycosyl bonds. The superposition of the low energy Rh(2)(OAc)(2) [d(pGpG)] conformers, generated by simulated annealing calculations, with the crystal structure of cis-[Pt(NH(3))(2) [d(pGpG)]], reveals remarkable similarities between the adducts; not only are the bases almost completely destacked upon coordination to the metal in both cases, but they are favorably poised to accommodate the bidentate N7/O6 binding to the dirhodium unit. Unexpectedly, the two metal-metal bonded rhodium centers are capable of engaging in cis binding to GG intrastrand sites by establishing N7/O6 bridges that span the Rh-Rh bond.  相似文献

Capillary electrophoresis-electrospray ionization-mass spectrometry (CE-ESI-MS) has been used for investigating the influence of the sulfur containing amino acid L-methionine (L-Met) on the binding behavior of oxaliplatin (trans-R,R-diaminocyclohexane-(oxalato)platinum(II)) to 5'-GMP. L-Methionine competes with 5'-GMP for the platinum binding site and forms as well as 5'-GMP adducts with oxaliplatin. The formation of the prognosed complexes [Pt(DACH)(L-Met-S,N)]+ and [Pt(DACH)(5'-GMP)2]2- (DACH = 1,2-diaminocyclohexane) could be proved directly by using CE-ESI-MS. Furthermore, we could now bring forward proofs, that the coordination of 5'-GMP with oxaliplatin is inhibited by L-methionine and could show, that the 5'-GMP ligands of the [Pt(DACH) (5'-GMP)2]2- complex can be replaced slowly by L-methionine whereas methionine can not be replaced by GMP.  相似文献
