Summary. Ethyl 2-(N-morpholinyl)cyclopent-1-ene-1-carboxylate reacted smoothly with cyanothioacetamide to give morpholinium 4-cyano-1-oxo-2,5,6,7-tetrahydro-1H-cyclopenta[c]pyridine-3-thiolate; the former when treated with N-benzyl-α-chloroacetamide gave either a S-alkyl derivative or cyclopenta[d]thieno[2,3-b]pyridine, depending on the reaction conditions. Under Mannich-type aminomethylation with primary amines and formaldehyde the above thiolate afforded derivatives of the previously unknown heterocyclic system, cyclopenta[g]pyrido[2,1-b][1,3,5]thiadiazine in 81–90% yields. 以4-取代苯乙炔化亚铜与3-甲氧基-4-羟基-5-溴肉桂酸甲酯为原料进行缩合反应得到2-取代芳基并[b]呋喃, 并将其衍生化, 得到14个2-芳基-5-烷基-7-甲氧基苯并[b]呋喃化合物, 并确证了其结构.  相似文献

以溴代丁二酰亚胺为溴化试剂, 对1-氰基-2-甲基薁进行了溴代反应研究, 将溴甲基化产物与水杨醛类衍生物在通常的加热反应及微波辐射条件下进行缩合, 以较高的收率得到2-薁基取代苯并[b]呋喃, 其结构经1H NMR, IR及元素分析等得以证实.  相似文献

New imidazo[4,5-b]pyridine derivatives with various substituents in the 2-position (,-unsaturated ketones, imines, 2-pyrazolines, pyrimidines, 1,2,3,4-tetrahydropyrimidines) and derivatives of the new pyrido[3',2':4,5]imidazo[1,2-d][1,2,4]triazine ring system were synthesized. The reaction of 3-amino-4,6-dimethyl-2-thieno[2,3-b]pyridine carbohydrazide ( 1 ) with appropriate chalcones 2a-2d in the presence of acid catalyst produced the corresponding 3-amino-2-[(3,5-disubstituted-pyrazolin-1-yl)carbonyl]-4,6-dimethylthieno[2,3-b]pyridines 3a-3d . 3-Amino-2-[(3-substituted-pyrazolin-1-yl)carbonyl]-4,6-dimethylthieno[2,3-b]pyridines 7a, 7b were also obtained by the cyclization reaction of carbohydrazide 1 with Mannich base derivatives 6a, 6b under basic condition.  相似文献

A method was developed for the synthesis of pyrano[4,3-b]thieno[3,2-e]pyridine derivatives based on the reaction of 3-amino-7-ethyl-7-methyl-7,8-dihydro-5H-pyrano[4,3-b]thieno[3,2-e]pyridine-2-carboxylic acid ethyl ester with chloroacetic acid chloride, triethyl orthoformate, hydrazine hydrate, and also phenyl chloroformate. The synthesis of various new representatives of pyrano[2″,3″:5′,6′]pyrido[3′,2′:4,5]thieno[3,2-dl-pyrimidine series was carried out.

Total syntheses of (−)-plectrodorine [(−)-1] and (+)-oxerine [(+)-3] possessing the cyclopenta[c]pyridine ring system have been accomplished through a route starting from the chiral γ-butyrolactone 7 and exploiting the intramolecular oxazole–olefin Diels–Alder reaction. The sign of specific rotation for the synthetic (+)-3 was in disagreement with that reported for natural oxerine, leaving the absolute configuration of this monoterpene alkaloid incomplete.  相似文献

The 4-nitro derivatives and an oxidation by-product, 9-hydroxy-2-methyl-3-oxo-9-phenyl-2,3-dihydro-9H-indeno[2,1-c]pyridine were obtained by the nitration of N-alkyl-9-phenyl-2,3-dihydro-1H-indeno-[2,1-c]pyridines with sodium nitrite in acetic acid. Their molecular structures were studied by X-ray structural analysis. Monoterpene pyridine alkaloids (MTPAs) are alkaloids derived from iridoid glycosides (IGs). The common molecular structure of MTPAs is the pyridine ring, while some of them have a cyclopenta[c]pyridine skeleton. Some compounds containing this structure are potentially bioactive medicinal agents. In this paper, seven drug candidates (A–G), ninety natural source products (1–90), thirty-seven synthesized compounds (91–127), as well as twenty-six key intermediates (S1–S26) were summarized. We categorized five types of MTPAs and one type of cyclopenta[c]pyridine alkaloids in all. Additionally, their possible genetic pathways were proposed. Then, the chemical transformation, biotransformation, chemical synthesis, as well as the bioactivity of MTPAs and cyclopenta[c]pyridine derivatives were analyzed and summarized. Cyclopenta[c]pyridine derivatives can be concisely and chirally synthesized, and they have shown potentials with antibacterial, insecticidal, antiviral, anti-inflammatory, and neuropharmacological activities.  相似文献

用邻氨基噻吩腈与不同结构的二羰基化合物反应,合成了新的多取代噻吩并吡啶类衍生物4-氨基-5-取代羰基噻吩并[2,3-b]吡啶-6-酮。用元素分析、红外光谱、核磁共振氢谱和质谱等测试技术对所合成化合物的结构进行了表征。初步生物活性测试表明,该类化合物有较好的杀菌活性和一定的除草活性。  相似文献

Ethyl [4-oxo-3-(2-oxo-2-arylethyl)thiazolidin-2-ylidene]acetates and [4-oxo-3-(2-oxo-2-arylethyl)thiazolidin-2-ylidene]acetonitriles were shown to react with substituted benzaldehydes at the endocyclic methylene group leading to the corresponding 5-arylmethylidene derivatives. Their treatment with DMF · POCl3 complex yielded 3-oxo-5-aroyl-2-arylmethylidene-2,3-dihydropyrrolo[2,1-b]thiazole-7-carboxylic acids ethyl esters and -7-carbonitriles. The structures of the pyrrolothiazoles were confirmed by an X-ray crystallographic study, which indicated the (Z)-configuration at the arylmethylidene moiety.  相似文献

以廉价易得的2,6-二氯吡啶为原料,经过硝化、叠氮化、热解环化步骤得到中间体[1,2,5]噁二唑并[3,4-e]四唑并[1,5-a]吡啶-3-氧化物(4b),再与浓硫酸/硝酸钾、甲醇钠和甲胺水溶液反应分别得到5-取代的氧化呋咱并[3,4-b]吡啶衍生物5~7。 研究了化合物4b结构的稳定性,发现其中的氧化呋咱环在强酸性、强碱性和弱碱性条件下较稳定,而吡啶环与叠氮基形成的四唑环结构则不太稳定。  相似文献

Reduction of 1-substituted [1,2,3]triazolo[4,5-c]pyridines with nickel-aluminum alloy in aqueous alkali gave 2-azaspinaceamines. Reduction of imidazo[4,5-c]pyridine and [1,2,3]triazolo[4,5-c]pyridine derivatives with formic acid in the presence of triethylamine resulted in formation of 5-formylspinaceamines and 2-azaspinaceamines. The 5-formyl group in the latter can be removed by acid hydrolysis. Unsubstituted 2-azaspinaceamine, an aza analog of natural spinaceamine, was synthesized for the first time.  相似文献
