A method for the determination of 16 polycyclic aromatic hydrocarbons in water has been developed. First, we made a solid‐phase extraction column. After this, the parameters affecting the efficacy of the experimental method were optimized, including appropriate selection of a solid‐phase extraction column and cleanup conditions on columns. The separation was achieved by gas chromatography and detection with triple quadrupole tandem mass spectrometry. The method showed satisfactory linearity (R> 0.999) over the range assayed (0.01–1 μg/mL), and limits of quantification ranging from 0.0011 to 0.0199 μg/L. The recoveries ranged from 83 to 113%. The relative standard deviation is in the range 0.86–3.1%. The results indicated that this method had high selectivity and precision that was suitable for the simultaneous determination of 16 polycyclic aromatic hydrocarbons in water.  相似文献

Biogenic amines have been reported in many foods such as fish, meat, and soy sauce. The consumption of foods containing high concentrations of biogenic amines has been associated with health hazards. In this study, a green and efficient method using supercritical fluid chromatography coupled with single quadrupole mass spectrometry was developed for determination of biogenic amines in soy sauce. The chromatographic and mass spectrometry conditions were systematically optimized in terms of selectivity and peak shape. Nine biogenic amines were well separated within 25 min on a Cosmosil 5HP column using 5% (v/v) water and 0.2% (v/v) ammonia solution in methanol as mobile phase additives at a backpressure of 120 bar and temperature of 40°C. The established method was fully validated regarding the linearity, sensitivity, precision, and accuracy. The limits of detection and limits of quantification ranged from 0.03 to 10.50 μg/mL and 0.10 to 23.1 μg/mL, respectively. The relative standard deviations for intra‐ and interday precisions were all lower than 9.36% and the recoveries ranged from 75.82 to 99.63% and 80.10 to 99.89% for two levels of standards spiked in soy sauce, respectively. Finally, the established method was successfully applied to the quantitative analysis of biogenic amines in soy sauce.  相似文献

An on‐line supercritical fluid extraction coupled with supercritical fluid chromatography method was developed for the determination of four major aromatic constituents in vanilla. The parameters of supercritical fluid extraction were systematically investigated using single factor optimization experiments and response surface methodology by a Box–Behnken design. The modifier ratio, split ratio, and the extraction temperature and pressure were the major parameters which have significant effects on the extraction. While the static extraction time, dynamic extraction time, and recycle time had little influence on the compounds with low polarity. Under the optimized conditions, the relative extraction efficiencies of all the constituents reached 89.0–95.1%. The limits of quantification were in the range of 1.123–4.747 μg. The limits of detection were in the range of 0.3368–1.424 μg. The recoveries of the four analytes were in the range of 76.1–88.9%. The relative standard deviations of intra‐ and interday precision ranged from 4.2 to 7.6%. Compared with other off‐line methods, the present method obtained higher extraction yields for all four aromatic constituents. Finally, this method has been applied to the analysis of vanilla from different sources. On the basis of the results, the on‐line supercritical fluid extraction‐supercritical fluid chromatography method shows great promise in the analysis of aromatic constituents in natural products.  相似文献

建立了超高效液相色谱-三重四极杆质谱法(UPLC-MS/MS)同时测定水粉画颜料、油画颜料、丙烯画颜料等美术颜料中33种初级芳香胺(PAAs)的检测方法。样品中的初级芳香胺用乙腈提取,经离心分离、氮吹浓缩后,以甲醇-水(1:9, v/v)定容至2 mL, 0.22 μm膜过滤后上机检测。采用BEH Phenyl柱(100 mm×2.1 mm, 1.7 μm),以含0.07%(v/v)甲酸的甲醇溶液-水(1:9, v/v)为流动相,梯度洗脱分离,UPLC-MS/MS多反应监测模式(MRM)检测,同位素内标法定量。方法优化了色谱分离条件、质谱碎裂电压、碰撞能量等,并考察了提取时间、提取溶剂、浓缩方式等对回收率的影响。33种初级芳香胺的方法检出限为5~50 μg/kg,定量限为15~150 μg/kg, 3种不同基质样品在3个添加水平的平均回收率为70.1%~115.8%,相对标准偏差(RSD)为2.1%~15%。本方法操作简便、快速、准确、灵敏度高,能满足相关测定的要求。  相似文献

建立了检测油脂及油炸食品中7种杂环胺类物质的超高效液相色谱-三重四极杆质谱(UPLC-MS/MS)的检测方法。以含1%(体积分数)氨水的乙腈溶液为提取液,采用乙腈饱和的正己烷脱脂,以PCX固相萃取柱为净化柱,10 mmol/L甲酸铵溶液(pH 6.8)和乙腈为流动相,采用Waters ACQUITY UPLC BEH C18反相色谱柱(50 mm×2.1 mm, 1.7μm)梯度洗脱,流速为0.3 mL/min,在正离子扫描、多反应监测模式下测定,内标法定量。结果表明,杂环胺类物质在各自范围内,线性关系良好,相关系数大于0.999;在3个加标水平下,杂环胺类物质在油脂和油炸食品中的平均回收率为64.31%~113.8%,相对标准偏差为0.18%~9.26%,检出限(S/N=3)和定量限(S/N=10)分别为0.01~0.14 ng/g和0.09~0.38 ng/g。该方法具有灵敏、准确等优点,适用于油脂和油炸食品中杂环胺的确证检测。  相似文献

An efficient on‐site extraction technique to determine carcinogenic heterocyclic aromatic amines in seawater has been reported. A micro‐solid‐phase extraction device placed inside a portable battery‐operated pump was used for the on‐site extraction of seawater samples. Before on‐site applications, parameters that influence the extraction efficiency (extraction time, type of sorbent materials, suitable desorption solvent, desorption time, and sample volume) were investigated and optimized in the laboratory. The developed method was then used for the on‐site sampling of heterocyclic aromatic amines determination in seawater samples close to distillation plant. Once the on‐site extraction completed, the small extraction device with the analytes was brought back to the laboratory for analysis using high‐performance liquid chromatography with fluorescence detection. Based on the optimized conditions, the calibration curves were linear over the concentration range of 0.05–20 μg/L with correlation coefficients up to 0.996. The limits of detection were 0.004–0.026 μg/L, and the reproducibility values were between 1.3 and 7.5%. To evaluate the extraction efficiency, a comparison was made with conventional solid‐phase extraction and it was applied to various fortified real seawater samples. The average relative recoveries obtained from the spiked seawater samples varied in the range 79.9–95.2%.  相似文献

建立了液相色谱-离子阱-飞行时间串联质谱法(LC-IT-TOF MS)同时测定葡萄酒中14种杂环胺(HAAs)残留量的分析方法。样品于乙酸乙酯碱性条件下提取,内标法定量,Phenomenex Kinetex C18 100A色谱柱(100 mm×2.1 mm, 2.6 μm)分离,以乙腈和30 mmol/L甲酸铵为流动相进行梯度洗脱。实验结果表明,2-氨基-3,7,8-三甲基咪唑[4,5-f]喹喔啉在1~500 μg/L、2-氨基-3,8-二甲基咪唑[4,5-f]喹喔啉在10~500 μg/L、其他12种杂环胺在5~500 μg/L质量浓度范围内线性良好,相关系数均不小于0.9945,检出限(以信噪比为3计) 0.33~1.77 μg/L。14种杂环胺在加标水平为10、50、100 μg/L时的回收率分别为71.6%~96.4%、72.9%~101.9%、74.5%~103.3%,相对标准偏差(RSD, n=6)分别为2.9%~7.9%、1.7%~5.3%、1.8%~4.8%。结果显示该方法线性范围宽,精密度和准确度较高,分析时间短,净化效果好,可满足葡萄酒中多种杂环胺残留同时检测的要求。  相似文献

Thirteen polycyclic aromatic hydrocarbons have been determined in soy‐based nutraceutical products. First, an optimization of extraction procedure was performed, and a solid–liquid extraction assisted by sonication and a dilute and shoot procedure were compared, selecting the dilute and shoot approach for the extraction of target compounds, utilizing a mixture of acetone/n‐hexane (1:1 v/v) as extractant solvent. After this, a clean‐up step was needed bearing in mind the complexity of these matrices. Dispersive solid‐phase extraction, using a mixture of C18 and Zr‐Sep+ (25 mg/mL each) was used. The separation was achieved by gas chromatography and detection with triple quadrupole tandem mass spectrometry. For quantification purposes, matrix‐matched calibration was used. The validation was applied at three concentration levels (20, 100 and 250 μg/kg), obtaining recoveries between 70 and 120% and precision values equal to or lower than 23%. Limits of detection and quantification were below 8 and 20 μg/kg, respectively. The method was applied in 11 samples, detecting five polycyclic aromatic hydrocarbons at concentrations ranging from 4.1 to 18.5 μg/kg.  相似文献

A lyophilization?supercritical fluid extraction coupled with supercritical fluid chromatography?quadrupole tandem mass spectrometry online method was developed for the determination of lipid mediators in breast cancer cells. Supercritical fluid extraction was applied to the cell samples for the first time due to the use of lyophilization. The conditions of supercritical fluid extraction and supercritical fluid chromatography?quadrupole tandem mass spectrometry were investigated systematically. Under the optimized conditions, all the calibration curves for the lipid mediators showed good linearity (correlation coefficient > 0.99). The limits of detection and the limits of quantification were in the range of 0.190?5.36 pg and 0.560?16.2 pg, respectively. The recoveries were in the range of 70.3?125%. The relative standard deviations of the precision ranged from 1.49?18.7% and the accuracies were higher than 84%. Compared with liquid?liquid extraction coupled with liquid chromatography and tandem mass spectrometry method, the present approach reduced the manual labor and obtained higher sensitivity as well as higher extraction recoveries for all 15 lipid mediators. Finally, the online method was applied to the quantification of lipid mediators in breast cancer cells and normal mammary epithelial cells. On the basis of the results, this lyophilization?supercritical fluid extraction online coupled with supercritical fluid chromatography?quadrupole tandem mass spectrometry method showed great promise in the analysis of lipid mediators in complex biological samples.  相似文献

The fragmentation of heterocyclic amines (HAs) in an ion trap was studied by means of the infusion of methanolic solutions containing the compounds under assay, and using an atmospheric pressure chemical ionization (APCI) as ion source. The MS(n) spectra obtained for compounds included in the same family, either aminoimidazoazaarenes (AIAs) or carbolines, were compared in order to propose fragmentation pathways for each HA. Moreover, labelled AIAs were used to establish the mechanisms. The protonated molecule was always obtained, but subsequent fragmentation was different for both families. In the case of AIAs, major product ions came from the fragmentation of the aminoimidazole moiety, thus the base peak in MS(2) corresponded to the loss of the methyl group, and losses of C(2)NH(3) and CN(2)H(2) were also observed. Further fragmentation occurred in the heterocyclic rings, mainly with losses of HCN and CH(3)CN. For carbolines, the most important product ions came from the loss of ammonia, except for harman and norharman, the loss of a methyl group for methylated carbolines or the loss of diverse fragments from the heterocyclic rings. In some cases, ion-molecule reactions into the ion trap were observed. For instance, for AalphaC or MeAalphaC one ion originating from these reactions corresponded to the base peak.  相似文献

A multiresidue method for determining 22 polychlorinated biphenyls (PCBs) in air has been developed and validated by gas chromatography (GC) coupled to tandem mass spectrometry (MS/MS) using a triple quadrupole analyzer (QqQ). The method was validated in terms of both steps of sampling and analysis. The sampling method, which is based on active sampling using polyurethane foam (PUF) as adsorbent, was validated by generating standard atmospheres. The retention capacity of this sampling sorbent allows up to 5 m3 of air to be sampled without any breakthrough for most compounds. Two solvent extraction methods were compared: sonication and Soxhlet extraction with a mixture of n-hexane:diethyl ether (95:5 v/v). Both extraction methods yielded similar results, but the first one required less solvent and time. The method exhibited good accuracy (80.3–99.8%), precision (2.2–15.2%) and lower limits that allowed quantification and confirmation at levels as low as 0.008 ng/m3. Finally, the method was applied to the analysis of PCBs in the air in areas near to a municipal solid-waste landfill and directly above the refuse in the landfill, where it indicatedd the presence of some of the target compounds. A multiclass method has been developed for screening, quantification and confirmation of organic micro-pollutants in water by gas chromatography coupled to mass spectrometry with a triple quadrupole analyzer. The work has been focused on the determination of more than 50 compounds belonging to different chemical families: 19 organochlorine and organophosphorus insecticides, 6 herbicides, 7 polychlorinated biphenyls, 16 polycyclic aromatics hydrocarbons, 2 brominated diphenyl ethers, and 3 octyl/nonyl phenols and pentachlorobenzene. Most of these analytes are included in the list of priority substances in the framework on European Water Policy.Analyte extraction was performed by solid phase extraction using C18 cartridges, and five isotopically labeled standards were added before extraction as surrogates. Analyses were performed by gas chromatography with tandem mass spectrometry (MS/MS) in electron impact mode. Accuracy and precision were evaluated by means of recovery experiments using water samples fortified at two concentration levels (25 and 250 ng L−1), with satisfactory results for most of analytes. The excellent selectivity and sensitivity reached in selected reaction monitoring mode allowed us satisfactory quantification and confirmation at levels as low as 25 ng L−1. Two MS/MS transitions were acquired for each analyte, using the Q/q intensity ratio as a confirmatory parameter. The method developed was applied to the analysis of surface, ground and wastewater samples collected from the Valencia Region (Spain).Analytical methodology using negative chemical ionization mode was also validated for the organochlorine compounds selected, showing a superior sensitivity and lower detection limits.  相似文献

In this work, a facile and environmentally friendly solid‐phase microextraction assay based on on‐fiber derivatization coupled with gas chromatography and mass spectrometry was developed for determining four nonvolatile index biogenic amines (putrescine, cadaverine, histamine, and tyramine) in fish samples. In the assay, the fiber was firstly dipped into a solution with isobutyl chloroformate as derivatization reagent and isooctane as extraction solvent. Thus, a thin organic liquid membrane coating was developed. Then the modified fiber was immersed into sample solution to extract four important bioamines. Afterwards, the fiber was directly inserted into gas chromatography injection port for thermal desorption. 1,7‐Diaminoheptane was employed as internal standard reagent for quantification of the targets. The limits of detection of the method were 2.98–45.3 μg/kg. The proposed method was successfully applied to the detection of bioamines in several fish samples with recoveries ranging 78.9–110%. The organic reagent used for extraction was as few as microliter that can greatly reduce the harm to manipulator and environment. Moreover, the extraction procedures were very simple without concentration and elution procedures, which can greatly simplify the pretreatment process. The assay can be extended to the in situ screening of other pollutant in food safety by changing the derivatization reagent.  相似文献

Monodisperse silica spheres that comprised a rhombic‐dodecahedral zeolitic imidazolate framework core–shell microsphere were applied in the sample pretreatment of navel orange. A rapid and efficient liquid chromatography with triple quadrupole tandem mass spectrometry method was established for simultaneously quantifying four plant growth regulators, 6‐benzylaminopurin, indole‐3‐acetic acid, indolepropionic acid, 3‐indolebutyric acid, in navel oranges. A satisfactory result was obtained, i.e., the peak area of the four plant growth regulators against concentration was linear with good correlation coefficients of 0.99987–0.99991. Under optimized conditions, the limits of detection were 3.0–59.4 μg/L for the four plant growth regulators. This method was applied to the simultaneous analysis of the four plant growth regulators in commercial samples, and all the detections were confirmed by acquiring transitions for each pesticide in the samples.  相似文献

A new multiresidue method has been developed and validated for the determination of more than 140 pesticide residues in cucumber and orange by gas chromatography coupled to triple quadrupole mass spectrometry (GC-QqQ-MS/MS) in a single run of 25.50 min. The triple quadrupole (QqQ) analyzer simultaneously operated in the selected reaction monitoring (SRM) and selected ion monitoring (SIM) modes, acquiring two or three transitions per compound. Samples were extracted by the application of a single-phase extraction of 10 g of sample with acetonitrile containing 1% of acetic acid, followed by a liquid-liquid partition formed by the addition of 4 g of MgSO(4) and 1 g of NaOAc. A dispersive solid-phase extraction (D-SPE) with primary secondary amine (PSA) was applied to clean up the extracts. A final concentration step was included in order to increase sensitivity in the instrumental analysis. The method was properly validated in each matrix in a wide dynamic range (10-400 microg kg(-1)): this work relies on a new quantification strategy by the use of two calibration curves to increase the dynamic range, which permitted reduction of sample dilutions and increase in sample throughput. Recovery was studied at three concentration levels (11.5, 50.0, and 150.0 microg kg(-1)), yielding values in the range 70-110% with precision values, expressed as relative standard deviation (RSD), lower than 20 and 25% for the intraday and interday precision, respectively. Limits of quantification (LOQs) were established at 10 microg kg(-1), the lowest maximum residue level (MRL) value set by the European Union in vegetables. A highly sensitive method was developed for simultaneously separating and identifying multiple compounds in radix curcumae. The determination of these compounds was achieved by combining supercritical fluid chromatography with drift tube ion mobility quadrupole time-of-flight MS. Related parameters were optimized: the RX-SIL column was used as the stationary phase, methanol was selected as the organic modifier, back pressure was 120 bar, back temperature was 60°C, the mobile phase flow rate was 1.75 mL/min, the makeup solvent was 0.2% formic acid/methanol with a flow rate of 0.7 mL/min. Under optimal conditions, multipolar compounds were separated. Furthermore, these compounds were identified by the values of collision sectional areas. The established method was verified by related parameters and exhibited good linearity, sensitivity, precision and accuracy. It could be extended to analyze other curcuminoids and sesquiterpenoids in natural products.  相似文献

We established a high-throughput and high-resolution analytical method based on supercritical fluid chromatography (SFC) coupled with mass spectrometry (MS) for the simultaneous profiling of diverse polar lipids in a mixture. Trimethylsilyl (TMS) derivatization was used for the analysis of ten polar lipids: phosphatidylglycerol (PG), phosphatidic acid (PA), phosphatidylinositol (PI), lysophosphatidylcholine (LPC), lysophosphatidylethanolamine (LPE), lysophosphatidylglycerol (LPG), lysophosphatidic acid (LPA), lysophosphatidylinositol (LPI), sphingomyeline (SM), and sphingosine-1-phosphate (S1P). Using the developed method, the peak tailings of PA, PI, LPA, LPI, and S1P improved, and the limit of detection of PG, PI, LPA, LPI, and S1P was enhanced by 12-, 40-, 510-, 39-, and 1490-fold, respectively. Next, in the analysis of sheep plasma, 20 minor species of PI, LPC, LPE, and SM, and 7 molecular species of LPA, LPI, and S1P were additionally analyzed. The relative ratio of the molecular species in each polar lipid was also found by quantification. Finally, the simultaneous and detail profiling of ten polar lipids was successfully performed by SFC/MS applying TMS derivatization. This developed method is particularly applicable to metabolomics, especially for targeting polar lipids.  相似文献

A method was developed for the determination of clenbuterol in meat using stable‐isotope‐dilution gas chromatography with mass spectrometry coupled with solid‐phase microextraction and on‐fiber derivatization. The samples were first homogenized with hydrochloric acid followed by protein deposition. After headspace solid‐phase microextraction and on‐fiber derivatization, the content of clenbuterol was measured with the aid of stable‐isotope dilution. The condition of solid‐phase microextraction was optimized by central composite design. The relative standard deviations, limit of detection, and recoveries for clenbuterol were 4.2–9.2%, 0.48 μg/kg, and 96–104%, respectively. The proposed method was satisfactory for analysis of real samples as compared with the Chinese standard method.  相似文献

Although several methods for the analysis of nitrogen compounds in diesel fuel have been described in the literature, the demand for rapid, sensitive, and robust analyses has increased in recent years. In this study, a comprehensive two‐dimensional gas chromatographic method was developed for the identification and quantification of nitrogen compounds in diesel fuel samples. The quantification was performed using the standard addition method and the analysis was conducted using comprehensive two‐dimensional gas chromatography coupled with fast quadrupole mass spectrometry. This study is the first to report quantification of nitrogen compounds in diesel fuel samples using the standard addition method without fractionation. This type of analysis was previously performed using many laborious separation steps, which can lead to errors and losses. The proposed method shows good linearity for target nitrogen compounds evaluated (m‐toluidine, 4‐ethylaniline, indole, 7‐methylindole, 7‐ethylindole, carbazole, isoquinoline, 4‐methylquinoline, benzo[h]quinolone, and acridine) over a range from 0.05 to 2.0 mg/L, and limits of detection and quantification of <0.06 and 0.16 mg/L, respectively, for all nitrogen compounds studied.  相似文献

Pesticides, widely applied in agriculture, can produce a variety of transformation products and their continuous use causes deleterious effects to ecosystem. Efficient and sensitive analytical techniques for enrichment and analysis of pesticides samples are highly required. Compared with other extraction methods, solid‐phase micro extraction is a solvent free, cost effective, robust, versatile, and high throughput sample preparation technique, especially for the analysis of pesticides from complicated matrices. Coupling of solid‐phase micro extraction with gas chromatography and mass spectrometry and liquid chromatography–mass spectrometry has been extensively applied in pesticide analysis. On the other hand, in recent years, combination of fast separation using solid‐phase micro extraction and rapid detection using ambient mass spectrometry is providing highly efficient pesticide screening. This article summarizes the applications of solid‐phase micro extraction coupled to mass spectrometry for pesticides analysis.  相似文献
