A solid‐phase extraction combined with a liquid chromatography‐tandem mass spectrometry analysis has been developed and validated for the simultaneous determination of 44 pharmaceuticals belonging to different therapeutic classes (i.e., antibiotics, anti‐inflammatories, cardiovascular agents, hormones, neuroleptics, and anxiolytics) in water samples. The sample preparation was optimized by studying target compounds retrieval after the following processes: i) water filtration, ii) solid phase extraction using Waters Oasis HLB cartridges at various pH, and iii) several evaporation techniques. The method was then validated by the analysis of spiked estuarine waters and wastewaters before and after treatment. Analytical performances were evaluated in terms of linearity, accuracy, precision, detection, and quantification limits. Recoveries of the pharmaceuticals were acceptable, instrumental detection limits varied between 0.001 and 25 pg injected and method quantification limits ranged from 0.01 to 30.3 ng/L. The precision of the method, calculated as relative standard deviation, ranged from 0.3 to 49.4%. This procedure has been successfully applied to the determination of the target analytes in estuarine waters and wastewaters. Eight of these 44 pharmaceuticals were detected in estuarine water, while 26 of them were detected in wastewater effluent. As expected, the highest values of occurrence and concentration were found in wastewater influent.  相似文献

We utilized ultra‐high performance liquid chromatography with tandem mass spectrometry and dispersive solid‐phase extraction to develop a new method for the detection of nine analytes (scopolamine, cephaeline, strychnine, hyoscyamine, brucine, hydrastine, ajmalicine, colchicine, and oleandrin) in herbal cosmetics. Acetonitrile/water and 2‐propylaminoethylamine were used to disperse and purify during the dispersive solid‐phase extraction step. The analytes were separated by a Waters UPLC HSS T3 column and detected through electrospray ionization source in the positive mode with multi‐reaction monitoring conditions. Under the optimal conditions, the calibration curves were linear in the range of 0.2–100.0 μg/L with the correlation coefficients higher than 0.995. The method limit of quantitation (S/N = 10) were 5.0 μg/kg for oleandrin and 1.0 μg/kg for the other eight alkaloids. The mean recoveries at three spiked concentration levels of 1.0–10.0 μg/kg were in the range of 86.9–116.5% with the intra‐day relative standard deviations (n  = 6) ranging from 2.4 to 8.8%, and inter‐day relative standard deviations ranging from 2.7 to 5.7%. This method is accurate, simple and rapid, and has been applied to the quality supervision of herbal cosmetics in Guangzhou.  相似文献

We have developed a method of liquid chromatography in tandem with mass spectrometry to monitor therapeutic levels of imatinib in plasma, a selective inhibitor of protein tyrosine kinase. After solid‐phase extraction of plasma samples, imatinib and its internal standard, imatinib‐D8, were eluted with Zorbax SB‐C18 at 60 °C, under isocratic conditions through a mobile phase consisting of 4 mm ammonium formate, pH: 3.2 (solution A) and acetonitrile solution B. The flow rate was 0.8 mL/min with 55% solution A + 45% solution B. Imatinib was detected and quantified by mass spectrometry with electrospray ionization operating in selected‐reaction monitoring mode. The calibration curve was linear in the range 10–5000 ng/mL, the lower limit of quantitation being 10 ng/mL. The method was validated according to the recommendations of the Food and Drug Administration, including tests of matrix effect (bias < 10%) and recovery efficiency (>80 and <120%). The method is precise (coefficient of variance intra‐day <2% and inter‐day <7%), accurate (95–108%), sensitive and specific. It is a simple method with very fast recording time (1.2 min) that is applicable to clinical practice. This will permit improvement of the pharmacological treatment of patients. A rapid confirmatory method has been developed and validated for the simultaneous identification, confirmation and quantitation of 11 nitroimidazoles in eggs by liquid chromatography tandem mass spectrometry (LC–MS/MS). The method is validated in accordance with Commission Decision 2002/657/EC and is capable of analysing metronidazole (MNZ), dimetridazole (DMZ), ronidazole (RNZ), ipronidazole (IPZ) and their hydroxy metabolites MNZ-OH, HMMNI (hydroxymethyl, methyl nitroimidazole), IPZ-OH. The method is also capable of analysing carnidazole (CRZ), ornidazole (ORZ), tinidazole (TNZ) and ternidazole (TRZ). MNZ, DMZ and RNZ have been assigned a recommended level (RL) of 3 μg kg−1 by the Community Reference Laboratory (CRL) in Berlin. The developed method described in this study is easily able to detect all the nitroimidazole compounds investigated at this level and below. Egg samples are extracted with acetonitrile, and NaCl is added to help remove matrix contaminants. The acetonitrile extract undergoes a liquid–liquid wash step with hexane; it is then evaporated and reconstituted in mobile phase. The reconstituted samples are analysed by liquid chromatography tandem mass spectrometry (LC–MS/MS). The decision limits (CCα) range from 0.33 to 1.26 μg kg−1 and the detection capabilities (CCβ), range from 0.56 to 2.15 μg kg−1. The results of the inter-assay study, which was performed by fortifying hen egg samples (n = 18) on three separate days, show the accuracy calculated for the various analytes to range between 87.2 and 106.2%. The precision of the method, expressed as %CV values for the inter-assay variation of each analyte at the three levels of fortification (3, 4.5 and 6.0 μg kg−1), ranged between 3.7 and 11.3%. A Day 4 analysis was carried out to examine species variances in eggs from different birds such as duck and quail and investigating differences in various battery and free range hen eggs.  相似文献

A simple and efficient multiresidue method using dispersive solid phase extraction and liquid chromatography coupled with tandem mass spectrometry was developed for the targeted analysis of indaziflam and its five metabolites (indaziflam‐diaminotriazine, indaziflam‐carboxylic acid, indaziflam‐triazine indanone, indaziflam‐hydroxyethyl, and indaziflam‐olefin) in pitaya samples (including roots, plants, flowers, peels, pulp, and whole fruit). The analytes were extracted with acetonitrile, and the extracts were purified using multiwalled carbon nanotubes. The method was validated using pitaya samples spiked at 0.5, 5, and 50 µg/kg, and the average recoveries varied from 61.1 to 103.7% with relative standard deviations lower than 12.7% (= 5). This method exhibited sufficient linearity within the concentration range of 0.1–100 µg/L. The limits of detection and quantification were in the ranges of 0.001–0.1 and 0.003–0.3 µg/kg, respectively. The method was successfully applied to analyze pitaya samples in Nanning, and no indaziflam or its metabolites were detected in the samples analyzed.  相似文献

A dispersive solid‐phase extraction coupled with ultra high performance liquid chromatography with tandem mass spectrometry method was developed and validated for the simultaneous determination of T‐2 toxin, penicillic acid, fumonisins B1, B2, and B3, aflatoxins B1, B2, G1, and G2, ochratoxin A, deoxynivalenol, 3‐acetyldeoxynivalenol, 15‐acetyldeoxynivalenol, and zearalenone in chestnut samples. The method was used to analyze 136 samples obtained from Shandong province in China. The mycotoxins were extracted using a dispersive solid‐phase extraction method and cleaned using an improved quick, easy, cheap, effective, rugged, and safe approach. The mycotoxins were then detected using a triple‐quadrupole mass spectrometer. The limits of detection and quantification ranged from 0.02 to 1 and 0.1 to 2 μg/kg, respectively. The recovery rates ranged from 74.2 to 109.5%, with relative standard deviations below 15%. A total of 71 samples were contaminated with seven mycotoxins at concentrations ranging from 1.2 to 105.5 μg/kg, with a number of samples exceeding the maximum limits set in the European regulations for mycotoxins in unprocessed chestnuts.  相似文献

A highly sensitive method was developed for the simultaneous separation and determination of organic and inorganic selenium species in rice by ion‐pairing reversed‐phase chromatography combined with inductively coupled plasma tandem mass spectrometry. To achieve a good separation of these species, a comparison between anion‐exchange chromatography and ion‐pairing reversed‐phase chromatography was performed. The results indicated that ion‐pairing reversed‐phase chromatography was more suitable due to better separation and higher sensitivity for all analytes. In this case, a StableBond C18 column proved to be more robust or to have a better resolution than other C18 columns, when 0.5 mM tetrabutylammonium hydroxide and 10 mM ammonium acetate at pH 5.5 were used as the mobile phase. Moreover, an excellent sensitivity was obtained in terms of interferences by means of tandem mass spectrometry in the hydrogen mode. The detection limits were 0.02–0.12 μg/L, and recoveries of five selenium species were 75–114%, with relative standard deviations ≤ 9.4%. This method was successfully applied to the analysis of rice samples. Compared with previous studies, the proposed method not only gave comparable results when used for measuring selenium‐enriched rice, but it can provide greater sensitivity for the detection of low concentrations of selenium species in rice.  相似文献

A novel in‐tube solid‐phase microextraction method based on a graphene oxide coated column was developed for the determination of triazines in waters. This column was prepared by the covalent modification of monolayer graphene oxide sheets onto the inner wall of a fused‐silica capillary. Scanning electron microscopy showed that the thickness of the graphene oxide coating was ~30 nm, with a porous, wrinkled membrane‐like structure. Its performance was evaluated through the extraction of triazines in water. Results showed that the coating was stable for at least 100 replicate extractions, and variety of multi‐columns was less than 10%. Flow rate, loading volume, pH, and ionic strength of samples played an important effect on the extraction. The high extraction efficiency was mainly attributed to π–π stacking and hydrogen bonding interactions. The in‐tube solid‐phase microextraction was used in the determination of triazines with liquid chromatography and tandem mass spectrometry, and the detection limits were 0.0005–0.005 μg/L for five triazine compounds. Further, the method was applied to the analysis of triazine herbicides in real samples including tap water, sea water, and river water, and the recoveries were 82.8–112.0, 85.4–110.5, and 81.6–105.9%, respectively, with RSDs of 2.7–7.1%.  相似文献

A highly sensitive method based on solid‐phase extraction and ultra high performance liquid chromatography with electrospray tandem mass spectrometry has been developed for simultaneous determination of five iodinated X‐ray contrast media in environmental water samples. Various solid‐phase extraction cartridges have been evaluated and a combination of LiChrolute EN and ENVI‐Carb solid phase extraction cartridges was selected for sample enrichment. The method was comprehensively validated on ground water, tap water, surface water, drinking water, and waste water by the conventional procedures: linearity, method detection limits, accuracy and precision, matrix effects. Good linearity (R2 > 0.999), low detection limits (0.4–8.1 ng/L), satisfactory recoveries (55.1–109.5%) and precision (0.8–10.0% for intra‐day precisions and 0.6–16.5% for inter‐day precisions) were obtained for all the target compounds. Iopamidol, iohexol, and diatrizoate in some matrices were affected by matrix effects, which were slightly eased by using the isotope‐labeled internal standard. The developed method was successfully applied for real samples collected in Shanghai, China, with detected concentrations up to 2200 ± 200 and 9000 ± 1000 ng/L for iohexol and iopamidol, respectively.  相似文献

In this study, a new method for the determination of 12 nitroimidazoles and their hydroxymetabolites (metronidazole, hydroxymetronidazole, dimetridazole, ronidazole, hydroxydimetridazole, ipronidazole, hydroxyipronidazole, carnidazole, ornidazole, secnidazole, ternidazole, tinidazole) in beeswax has been developed and validated. The optimized sample preparation procedure included melting and dilution of beeswax in a mixture of n‐hexane and isopropanol followed by extraction with 2% acetic acid. The extracts were purified on strong cation exchange based solid‐phase extraction cartridges and evaporated in a vacuum system with vortex motion. The separation and detection of the nitroimidazoles in the beeswax extracts were achieved within 12 min by liquid chromatography tandem mass spectrometry using a pentafluorophenyl analytical column and applying a gradient elution with acetonitrile and 0.01% acetic acid as mobile phases. The method performance characteristics were evaluated at three concentration levels (1, 2, and 5 μg/kg) and the method was found to be suitable for determination of all tested nitroimidazoles. The limits of detection and quantification were 0.2–0.5 and 0.5–1 μg/kg, respectively. The recoveries varied from 71.2 to 104.9% while the relative standard deviations were less than 13.8% under the intermediate precision conditions.  相似文献

A sensitive and robust multiresidue method for the simultaneous analysis of 114 pesticides in tobacco was developed based on solid‐phase extraction coupled with gas chromatography and tandem mass spectrometry. In this strategy, tobacco samples were extracted with acetonitrile and cleaned up with a multilayer solid‐phase extraction cartridge Cleanert TPT using acetonitrile/toluene (3:1) as the elution solvent. Two internal standards of different polarity were used to meet simultaneous pesticides quantification demands in the tobacco matrix. Satisfactory linearity in the range of 10–500 ng/mL was obtained for all 114 pesticides with linear regression coefficients higher than 0.994. The limit of detection and limit of quantification values were 0.02–5.27 and 0.06–17.6 ng/g, respectively. For most of the pesticides, acceptable recoveries in the range of 70–120% and repeatabilities (relative standard deviation) of <11% were achieved at spiking levels of 20, 100, and 400 ng/g. Compared with the reported multiresidue analytical method, the proposed method provided a cleaner test solution with smaller amounts of pigments, fatty acids as well as other undesirable interferences. The development and validation of the high sensitivity, high selectivity, easy automation, and high‐throughput analytical method meant that it could be successfully used for the determination of pesticides in tobacco samples.  相似文献

Cyadox is a novel antimicrobial growth‐promoter of the quinoxalines. For food safety and pharmacokinetic studies, a convenient, sensitive and reproducible LC‐ESI‐MS/MS method was developed for the simultaneous determination of cyadox and its major metabolites, quinoxaline‐2‐carboxylic acid, 1,4‐bisdesoxycyadox, cyadox‐1‐monoxide and cyadox‐4‐monoxide in chicken plasma. Plasma sample was subjected to a simple deproteinisation with acetonitrile. Analysis was performed on a C18 column by detection with mass spectrometry in multiple reaction monitoring mode. A gradient elution program with 0.2% formic acid, methanol and acetonitrile was performed at a flow rate of 0.2 mL/min. The decision limits (CCαs) of five analytes in plasma ranged from 1.0 to 4.0 μg/L, and the detection capabilities (CCβs) were <10 μg/L. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. The extraction recoveries of five analytes were between 87.4 and 93.9% in plasma at the spiked levels of 5 (10)–200 μg/L with the relative standard deviations <10% for each analyte. The developed method demonstrated a satisfactory applicability in real plasma samples.  相似文献

UV filters, contained in sunscreens and other cosmetic products, as well as in some plastics and industrial products, are nowadays considered contaminants of emerging concern because their widespread and increasing use has lead to their presence in the environment. Furthermore, some UV filters are suspected to have endocrine disruption activity. In the present work, we developed an analytical method based on liquid chromatography with tandem mass spectrometry for the determination of UV filters in tap and lake waters. Sixteen UV filters were extracted from water samples by solid‐phase extraction employing graphitized carbon black as adsorbent material. Handling 200 mL of water sample, satisfactory recoveries were obtained for almost all the analytes. The limits of detection and quantification of the method were comparable to those reported in other works, and ranged between 0.7–3.5 and 1.9–11.8 ng/L, respectively; however in our case the number of investigated compounds was larger. The major encountered problem in method development was to identify the background contamination sources and reduce their contribution. UV filters were not detected in tap water samples, whereas the analyses conducted on samples collected from three different lakes showed that the swimming areas are most subject to UV filter contamination.  相似文献

Summary Reversed-phase liquid chromatography with fluorescence detection (RPLC-FD) and with electrospray ionization (ESI) tandem mass spectrometric detection (RPLC-ESI-MS-MS), both coupled with automated solid-phase sample extraction, have been used for trace enrichment and analysis of octylphenol and nonylphenol (NP) in municipal wastewater. Quality data calculated for the methods were detection limits (LOD), quantitation limits, linearity, precision, and accuracy. For LC-ESI-MS-MS theLOD were below 0.08 μg L−1. Unequivocal detection of NP in wastewater samples was achieved by use of LC-ESI-MS and LC-ESI-MS-MS. In particular, application of the LC methods to wastewater samples revealed that the selectivity of LC-MS-MS was more than that of LC-FD for qualitative and quantitative analysis of complex matrices.  相似文献

A simple, reliable and sensitive liquid chromatography tandem mass spectrometry (LC‐MS/MS) protocol was developed and validated for quantification of bisoprolol in human plasma. The sample was pretreated with a simple procedure of protein precipitation and an isotope‐labeled d5‐bisoprolol was used as internal standard. The chromatographic separation was performed on a Capcell Pak C18 MG III column (100 mm × 2.0 mm, 5 µm). The protonated ion of the analyte was detected in positive ionization by multiple reaction monitoring mode. The mass transition pairs of m/z 326.3 → 116.3 and m/z 331.3 → 121.3 were used to detect bisoprolol and the internal standard, respectively. Linearity, accuracy, precision, recovery, matrix effect, dilution test and stability were evaluated during method validation over the range of 0.5–100 ng/mL. Acrylamide has been reported to be present in high-temperature processed foods and normal processed food intake could lead to significant acrylamide exposure. Acrylamide in vivo can be conjugated with glutathione in the presence of glutathione transferase. This conjugation product is further metabolized and excreted as N-acetyl-S-(propionamide)cysteine (NASPC) in the urine. NASPC could be considered a biomarker for acrylamide exposure. The objective of this study was to develop a highly specific, rapid and sensitive method to quantify urinary NASPC, serving as a biomarker for acrylamide exposure assessment. Isotope-labeled [13C3]NASPC was successfully synthesized and used as an internal standard. This urine mixture was directly analyzed using a newly developed liquid chromatographic/tandem mass spectrometric method coupled with an on-line clean-up system. The detection limit for this method was estimated as < 5 microg l(-1)(0.4 pmol) on-column. The method was applied to measure the urinary level of NASPC in 70 apparently health subjects. The results showed that the NASPC urinary level was highly associated with smoking. Smokers had a significantly higher urinary NASPC level (135 +/- 88 microg g(-1) creatinine) than non-smokers (76 +/- 30 microg g(-1) creatinine). A highly sensitive and selective LC/MS/MS isotope dilution method was successfully established. With an on-line clean-up system, this system is capable of routine high-throughput analysis and accurate quantitation of NASPC in urine. This could be a useful tool for health surveillance for acrylamide exposure in a population for future study.  相似文献

A method based on liquid chromatography with electrospray ionization high‐resolution mass spectrometry (Exactive Orbitrap) combined with solid‐phase extraction using a strong cationic exchange mixed‐mode sorbent has been developed for the determination of seven drugs of abuse, including two synthetic cathinones, as well as some of their metabolites in environmental water samples. The method provides low detection limits and a high confirmation power thanks to the diagnostic and two fragment ions monitored for each compound in high‐resolution mass spectrometry, providing six identification points for each analyte. The clean‐up step based on methanol in the extraction step adequately decreased the matrix effect, mainly for river and effluent water, and provided suitable process efficiency. Method detection and quantitation limits for environmental waters were at low nanogram per liter. The method was applied to analyze the samples of influent and effluent wastewater, as well as surface water. Codeine, methadone, and its metabolite were determined in all samples of wastewater and the metabolite of cocaine, benzoylecgonine, was found at the highest concentration.  相似文献

A dispersive liquid–liquid microextraction method for the simultaneous determination of 11 pharmaceuticals has been developed. The method is based on a microextraction procedure applied to wastewater samples from different regions of Hungary followed by high‐performance liquid chromatography with mass spectrometry. The effect of the nature of the extractant, dispersive solvent, different additives, and extraction time were examined on the extraction efficiently of the dispersive liquid–liquid microextraction method. Under optimal conditions, the linearity for determining the pharmaceuticals was in the range of 1–500 ng/mL, with the correlation coefficients ranging from 0.9922 to 0.9995. The limits of detection and limits of quantification were in the range of 0.31–6.65 and 0.93–22.18 ng/mL, respectively.  相似文献

An ultra high performance liquid chromatography with tandem mass spectrometry method was established for the rapid and simultaneous analysis of seven antiviral drugs, amantadine, rimantadine, memantine, moroxydine, imiquimod, oseltamivir, and acyclovir, in chicken liver, muscle, and egg. Homogenized samples were extracted with trichloroacetic acid and acetonitrile solutions and then purified by cation‐exchange solid‐phase extraction. The target drugs were analyzed by liquid chromatography with a UPLC BEH Amide column (2.1 mm × 100 mm, 1.7 μm) coupled with a tandem mass spectrometer operating in the positive multiple‐reaction mode. A perfectly linear relationship was obtained within the concentration ranges of 0.5–20 μg/L for acyclovir and 0.1–10 μg/L for the other six antiviral drugs. The average recoveries of the seven antiviral drugs using four addition levels in chicken liver, muscle, and eggs were 82.67–90.10, 82.30–92.27, and 81.98–93.77%, respectively, and the acceptable coefficients of variation were 5.18–9.88, 4.84–11.2, and 42.8–9.95%, respectively. The detection limits and detection capabilities of the analysis method for the seven antiviral drugs were in the ranges of 0.04–0.64 and 0.11–0.78 μg/kg, respectively. Additionally, an inter‐laboratory study among five laboratories further validated the method.  相似文献
