 提出了一种用高效液相色谱(HPLC)分离和用电感偶合等离子体质谱仪(ICP-MS)或火焰原子吸收光谱仪(FAAS)作元素专一检测器在线测定硒的化学形态的方法。在优化的HPLC条件下,用ESAⅢ阴离子色谱柱(250mm×4.6mm),以柠檬酸铵为流动相(5.5mmol/L,pH5.5,流速1.5mL/min),进样量100μL,分离和测定三甲基硒离子、硒代蛋氨酸、亚硒酸和硒酸盐只需8min。HPLC-FAAS在线分析4种硒化合物的检测限为p(Se)=1mg/L。  相似文献

建立了富硒黑木耳中硒代胱氨酸、硒代半胱氨酸、亚硒酸、硒蛋氨酸、硒酸5种硒形态的液相色谱-原子荧光光谱分析方法。通过链酶蛋白酶E酶解,结合超声提取后,选取Hamilton PRP-X100离子交换色谱柱(250 mm×4.1 mm,10μm),40 mmol/L的磷酸氢二铵为流动相,在16 min内,5种硒形态完全达到基线分离。5种硒形态在线性范围内相关系数R为0.9990~0.9999;加标回收率为76.1%~108%;检出限分别为硒代胱氨酸0.35μg/L、甲基-硒代半胱氨酸0.46μg/L、亚硒酸0.26μg/L、硒代蛋氨酸0.64μg/L、硒酸3.06μg/L;方法应用于富硒黑木耳中硒形态的分析,精密度高、重现性好、方法稳定、准确可靠,是测定富硒黑木耳中硒形态含量的有效方法。  相似文献

For selenium speciation analysis, the hyphenation of chromatographic separation with element-specific detection has proved a useful technique. A powerful separation system, which is capable of resolving several biologically and environmentally important selenium compounds in a single column, is greatly needed. However, that has been difficult to achieve. In this paper eight selenium compounds, namely, selenite [Se(IV)], selenate [Se(VI)], selenocystine (SeCys), selenourea (SeUr), selenomethionine (SeMet), selenoethionine (SeEt), selenocystamine (SeCM) and trimethylselenonium ion (TMSe+), were separated by using mixed ion-pair reagents containing 2.5 mM sodium 1-butanesulfonate and 8 mM tetramethylammonium hydroxide as a mobile phase. The separation of these anionic, cationic and neutral organic selenium compounds on a LiChrosorb RP18 reversed-phase column took only 18 min at a flow-rate of 1.0 ml/min with isocratic elution, and baseline separation among the six organic Se compounds was achieved. Inductively coupled plasma mass spectrometry (ICP-MS) was employed as element-specific detection. A comparison of ICP-MS signal intensity obtained with a Barbington-type nebulizer and with an ultrasonic nebulizer (USN) was made. Different signal enhancement factors were observed for the various selenium compounds when a USN was used. The speciation technique was successfully applied to the study on chemical forms of selenium in a selenium nutritional supplement. Selenomethionine was found to be the predominant constituent of selenium in the supplement.  相似文献

The coupling of a high-power nitrogen (N2) microwave-induced plasma (MIP) mass spectrometry--(MS) (1.3 kW) with high-performance liquid chromatography, connected with concentric nebulizer (CN), ultrasonic nebulizer (USN) and a hydride generation (HG) systems, for the optimization and determination of selenium compounds, has been carried out. The MIP-MS system fulfils the ideal requirement being an on-line real-time chromatographic detector for Se speciation analysis. Interchanging of MIP-MS system fabricated nebulizer (concentric) with an ultrasonic nebulizer increases about 3.4-12 (peak height) and 6.5-10 (peak area) times ion signals for the selenium compounds. The detection limits for selenate, selenite, trimethylselenonium ion (TmSe), selenomethionine (Semet) and selenoethionine (Seet) (in Milli-Q-water) obtained with the optimized HPLC-USN-N2MIP-MS system are 0.11, 0.14, 0.09, 0.14 and 0.10 microg L(-1), respectively, about 12-48 times lower than the HPLC-CN-MIP-MS and 1.5-4.4 (peak height) times lower compared to the HPLC-CN-inductively coupled plasma (ICP)-MS coupling. Considering peak area, the repeatability (R.S.D. for three successive analyses) and intermediate precision (R.S.D. for three successive analyses performed on three different days), achieved for five Se compounds are 0.8-5.6, and 1.1-5.9%, comparable with the HPLC-CN-ICP-MS, HPLC-HG-MIP-MS and HPLC-CN-MIP-MS systems. The combined HPLC-USN-N2MIP-MS has been adequately applied for the determination of Se compounds in certified National Institute for Environmental studies human urine CRM No. 18. The results reasonably agree with the HPLC-CN-ICP-MS values. This encouraging combination may be an alternative ion source of mass spectrometry for coming generation in regard to the selenium speciation analysis.  相似文献

Summary Separation of seven inorganic and organic selenium compounds, namely selenic acid [Se(VI)], selenous acid [Se(IV)], trimethylselenonium iodide (TMSe+), selenocystine (SeCys), selenomethionine (SeMet), selenoethionine (Seet), and selenocystamine (SeCM), has been performed on a LiChrosorb C 18 column by using mixed ion-pair reagents; 1-butanesulfonic acid and tetramethylammonium hydroxide. Flame atomic absorption spectrometry (FAAS) was used as an element-specific detector. The retention behaviors of selenium compounds in terms of several chromatographic parameters, such as pH of the mobile phase, the concentrations of ion-pair reagents, and the content of organic modifier (methanol) were investigated. It was found that the separation of both inorganic and organic selenium compounds can be achieved within 12 min with a mobile phase of 10 mM 1-butanesulfonic acid −4 mM tetramethylammonium hydroxide −4 mM malonic acid −0.05% methanol adjusted to pH 4.5 at a flow rate of 1.0 mL min−1. The results obtained in this study showed that the use of mixed ion-pair reagents is very useful to improve the separation of selenium compounds. The applicability of this technique for the speciation of selenium compounds in real samples was demonstrated by the determination of selenium compounds in a selenium nutritional supplement. The results were found to be in good agreement with those obtained by ion-exchange HPLC-ICP-MS.  相似文献

Different steps for the fluorimetric determination of Se in urine have been investigated. A HNO3---HClO4 (4:1) mixture is useful for urine digestion, and reduction of Se(VI) to Se(IV) is effectively carried out with HCl (6M). Selenium(VI) present after the digestion process constitutes 14.5–36.6% of total Se. An optimum pH of 1.80±0.05 and the addition of 1 ml of 2,3-diaminonaphthalene (DAN) (0.1%, w/v) are established in the formation of Se—DAN complex. Heating to 60°C, a time of incubation of 15 min is recommended to assure the complete formation of Se—DAN complex. A volume of 5 ml of cyclohexane and vigorous shaking for 45 sec is necessary for the extraction process. With this optimized method, the detection limit of selenium was 0.82 μg/l., within-day precision for a 50.0 μg/l. standard solution and urine (27.3 μg/l.) were 2.4 and 2.7% and between-day for the urine was 3.9% (33.9 μg/l.). Analytical recovery of 0.5 ml of Se standard (250 μg/l.) added to 1 ml of urine was 99.9±2.9% (95.8–104.4, n = 12). Normal levels of selenium excretion in urine obtained from healthy people were 27.9±8.7 μg/day (13.2–44.1), not observing significant differences (P < 0.05) between sexes.  相似文献

Increased sensitivity is obtained by introducing higher amounts of sample into the measurement system via high efficiency nebulizers (ultrasonic nebulizer, hydraulic-high pressure nebulizer according to Berndt and concentric high efficiency nebulizer according to Meinhard). Because this means also higher matrix effects a combination of ion chromatographic (IC-TMS) and thermal trace-matrix-separation by aerosol desolvation (T-TMS) is introduced. Isotope dilution (ID) proves to be the calibration most suitable to achieve the highest accuracy. First applications on the analysis of refractory metals (e.g. Ti, V, Nb, Ta) and non-metals (e.g. P, S, As, Se) showed recoveries of 60–105%, an imprecision of the recoveries of 2–50%, but an overall inaccuracy of only 0.1 to 4%.  相似文献

The behavior of selenium in thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS) was studied and the developed procedure was applied for selenium determination in biological materials after microwave-assisted sample digestion. A sample volume of 600 μL was introduced into the hot metallic Ni tube at a flow rate of 0.4 mL min−1 using water as carrier. The limit of detection obtained for Se was 8.7 μg L−1 (3sblank/slope, n = 10), which is 95-fold better than that typically obtained using FAAS. The applicability of the TS-FF-AAS procedure was evaluated for selenium determination in biological materials. Certified reference materials of pig kidney (BCR 186) and mussel (GBW 08571) were analyzed and a t-test had not shown any statistically significant difference at a 95% confidence level between determined and certified values for both materials. The procedure was successfully applied for determination of Se in pig kidney and shellfish. It was demonstrated that TS-FF-AAS improved the performance of FAAS (flame atomic absorption spectrometry) for determination of Se.  相似文献

Aqueous solutions and blood plasma spiked with selenite (Se(IV)), selenate (Se(VI)), selenomethionine (SeMet) or trimethylselenonium (TMSe) iodide were analyzed by Zeeman-corrected electrothermal atomic absorption spectrometry (ETAAS) using palladium as a chemical modifier, and by inductively coupled plasma mass spectrometry (ICP-MS). Using ETAAS, the sensitivities for Se(IV), SeMet and TMSe in aqueous solution were similar, whereas the sensitivity of Se(VI) was 63% of that value. In blood plasma, the ETAAS sensitivities of Se(IV) and Se(VI) were equal, whereas the sensitivities of SeMet and TMSe were 87 and 56%, respectively, of that value. In contrast, the ICP-MS sensitivities obtained for Se(VI), TMSe and SeMet were between 96 and 98% of that obtained for Se(IV) in aqueous solution and in blood plasma. It is concluded, that ICP-MS is superior to ETAAS as the problem of differences in sensitivity of the selenium species when using ETAAS are not prevalent when using the ICP-MS technique.  相似文献

An on-line FIA pretreatment with HBr/KBrO3, assisted by on-line focused microwave-induced digestion, has been coupled with hydride generation-atomic absorption spectrometry (HG-AAS) for final detection for total selenium determination. This total selenium determination is virtually independent of the different Se species investigated (selenite, selenate, selenomethionine, selenoethionine and selenocystine). Detection limits of 0.8 μg l−1 of Se can be achieved by AAS with precisions better than 5%. This continuous flow system for selenium determination allows a high sample throughput (about 30 samples h−1 can be analyzed) in which high automation can be achieved and constitutes a convenient real-time continuous detector for the different selenocompounds tested. Direct non-chromatographic speciation of inorganic selenium (selenite and selenate in their mixtures) is demonstrated by simple on-off operation of the focused microwaves connected in the flow system.

Validation of this simple on-line FIA system has been carried out by analyzing total Se recovered from spiked tap waters and by speciating mixtures of Se(IV) and Se(VI) spiked to the same samples. The fast conversion of Se compounds into volatile selenium could be considered as a sort of specific “general” detector for Se compounds which can be extremely useful for Se speciation by hybrid chromatographic techniques.  相似文献

Analytical methods for the speciation of nine selenium species (selenite, selenate, selenourea, trimethylselenonium ion, selenocystamine, selenocystine, selenocysteine, selenomethionine and selenoethionine) that are commonly encountered in biological and environmental samples were developed. Good separation was achieved by either a mixed ion-pair reversed phase chromatography (LiChrosorb RP 18, 2.5 mM 1-butanesulfonate-8 mM tetramethylammonium hydroxide-4 mM malonic acid-0.05% methanol, pH 4.5) or a conventional ion-pair reversed phase chromatography (Inertsil ODS, 10 mM tetraethylammonium hydroxide-4.5 mM malonic acid, pH 6.8) with on-line ICP-MS detection. Using a 20-μl sample loop, low detection limits around 1 ng ml−1 expressed as Se were achieved for the examined selenium species. The methods were used for the determination of selenoamino acids in a selenium nutritional supplement. The developed methods were found to be rather robust. No alteration of the separation was observed when the protease enzymatic extracts were analyzed without dilution. Both water extracts and enzymatic extracts were chromatographed first with the mixed ion-pair reversed phase chromatographic system, then the major chromatographic peaks were collected and analyzed by the second ion-pair reversed phase chromatographic system for a further verification of their identity. Selenomethionine was found to be the major selenium species in the supplement. A major unknown species, probably Se-adenosylhomocysteine, could be determined in the extracts. A biological reference material, Dolt-2, was also examined for the selenoamino acids. Selenocystine and selenomethionine could be detected in its enzymatic extract, suggesting that Dolt-2 may be used as a reference material for the identification of selenoamino acids in biological and environmental samples. As selenoethionine does not occur naturally in the investigated samples, it is added as an internal standard in this study.  相似文献

To avoid mass interferences on analyte ions caused by argon ions and argon molecular ions via reactions with collision gases, an rf hexapole filled with helium and hydrogen has been used in inductively coupled plasma mass spectrometry (ICP-MS), and its performance has been studied. Up to tenfold improvement in sensitivity was observed for heavy elements (m > 100 u), because of better ion transmission through the hexapole ion guide. A reduction of argon ions Ar+ and the molecular ions of argon ArX+ (X = O, Ar) by up to three orders of magnitude was achieved in a hexapole collision cell of an ICP-MS ("Platform ICP", Micromass, Manchester, UK) as a result of gas-phase reactions with hydrogen when the hexapole bias (HB) was set to 0 V; at an HB of 1.6 V argon, and argon-based ions of masses 40 u, 56 u, and 80 u, were reduced by approximately four, two, and five orders of magnitude, respectively. The signal-to-noise ratio 80Se/ 40Ar2+ was improved by more than five orders of magnitude under optimized experimental conditions. Dependence of mass discrimination on collision-cell properties was studied in the mass range 10 u (boron) to 238 u (uranium). Isotopic analysis of the elements affected by mass-spectrometric interference, Ca, Fe, and Se, was performed using a Meinhard nebulizer and an ultrasonic nebulizer (USN). The measured isotope ratios were comparable with tabulated values from IUPAC. Precision of 0.26%, 0.19%, and 0.12%, respectively, and accuracy of 0.13% 0.25%, and 0.92%, respectively, was achieved for isotope ratios 44Ca/ 40Ca and 56Fe/57Fe in 10 microg L(-1) solution nebulized by means of a USN and for 78Se/80Se in 100 microg L(-1) solution nebulized by means of a Meinhard nebulizer.  相似文献

An inductively coupled plasma mass spectrometer (ICP-MS) was used as an ion chromatographic (IC) detector for the speciation analysis of arsenic and selenium. The arsenic and selenium species studied included arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), selenite [Se(IV)] and selenate [Se(VI)]. Gradient elution using (NH4)2CO3 and methanol at pH 9 allowed the chromatographic separation of all species in less than 12 min. Effluents from the IC column were delivered to the nebulization system of ICP-DRC-MS for the determination of arsenic and selenium. The potentially interfering 38Ar40Ar+ and 40Ar40Ar+ at the selenium masses m/z 78 and 80 were reduced in intensity by approximately 3 orders of magnitude by using 0.6 mL min−1 CH4 as reactive cell gas in the DRC while an Rpq value of 0.3 was used. Meanwhile, arsenic was determined as the adduct ion 75As12CHH+ at m/z 89, which is more sensitive than 75As. The limits of detection for arsenic and selenium were in the range of 0.002–0.01 ng mL−1 and 0.01–0.02 ng mL−1, respectively, based on peak height. The relative standard deviation of the peak areas for five injections of 5 ng mL−1 As and Se mixture was in the range of 2–4%. The concentrations of arsenic and selenium species have been determined in urine samples collected locally. The major As and Se species in urines were AsB, DMA and probably selenosugar at concentration of 20–40, 15–19 and 17–31 ng mL−1, respectively. The recoveries were in the range of 94–105% for all the determinations. This method has also been applied to determine various arsenic compounds in two fish samples. In this study, a simple and rapid microwave-assisted extraction method was used for the extraction of arsenic compounds from fish. The arsenic species were quantitatively leached with an 80% v/v methanol solution in a focused microwave field during a period of 5 min.  相似文献

The accuracy of the determination of selenium in human body fluids by hydride-generation a.a.s. depends critically upon the sample decomposition-method used. Digestion with HNO3 alone gave low selenium recoveries, but with nitric, sulfuric and perchloric acids at a final temperature of 310°C gave results that agreed with those obtained by other techniques. The recovery of selenomethionine added to whole blood and of trimethylselenonium iodide added to urine was 97–104%. The average selenium values found for 6 healthy individuals were 88 μg l?1 in whole blood, 75 μg l?1 in blood plasma and 307 μg (kg Hb)?1 in erythrocytes. A detection limit of 5 μg l?1 Se in body fluids was found under routine conditions.  相似文献

A method for the determination of total selenium in serum samples by graphite furnace atomic absorption spectrometry was evaluated. The method involved direct introduction of 1:5 diluted serum samples (1% v/v NH4OH+0.05% w/v Triton X-100®) into transversely heated graphite tubes, and the use of 10 μg Pd+3 μg Mg(NO3)2 as chemical modifier. Optimization of the modifier mass and the atomization temperature was conducted by simultaneously varying such parameters and evaluating both the integrated absorbance and the peak height/peak area ratio. The latter allowed the selection of compromise conditions rendering good sensitivity and adequate analyte peak profiles. A characteristic mass of 49 pg and a detection limit (3s) of 6 μg 1−1 Se, corresponding to 30 μg l−1 Se in the serum sample, were obtained. The analyte addition technique was used for calibration. The accuracy was assessed by the determination of total selenium in Seronorm™ Trace Elements Serum Batch 116 (Nycomed Pharma AS). The method was applied for the determination of total selenium in ten serum samples taken from individuals with no known physical affection. The selenium concentration ranged between 79 and 147 μg l−1, with a mean value of 114±22 μg l−1.  相似文献

The optimization of the hyphenation between capillary zone electrophoresis (CZE) and inductively coupled plasma mass spectrometry (ICP-MS) was studied for the simultaneous determination of metalloid species in the environment. Arsenic (arsenite, arsenate, monomethylarsonic acid, dimethylarsinic acid), selenium (selenite, selenate, selenomethionine, selenocystine), antimony (antimonate) and tellurium (tellurite, tellurate) species were simultaneously separated using a 75-μm i.d. fused silica capillary using either a chromate or a phosphate electrolyte. Different nebulizers were tested for introduction in the detector. A V-groove nebulizer (the Babington) and two concentric micronebulizers (the MCN-100 and the MicroMist) were studied in order to improve resolution, sensitivity and reproducibility. The optimization of CE-ICP-MS interface operating parameters is discussed for each nebulizer–interface combination, and special attention is given to the position of the capillary inside the nebulizer. Different nebulizer gas and liquid sheath flow rates were studied in detail and they hardly affect electrophoretic resolution and peak width. The best analytical performance characteristics were obtained with the MicroMist nebulizer. Detection limits with this nebulizer were found to range between 6 and 58 μg l−1 depending on the species investigated using pressure injection and below 1 μg l−1 for most of the species with electromigrative injection. Analysis of soil extracts showed that it was possible to carry out this technique on real samples.  相似文献

A new on-line preconcentration flame atomic absorption spectrometry (FAAS) system for trace element determination was developed based on sorption of soluble metal complexes on the walls of a PTFE knotted reactor using flow injection techniques. The system was applied to the determination of cadmium in biological materials. Cadmium complexed with sodium diethyldithiocarbamate was sorbed on the inner walls of the reactor and eluted on-line by isobutyl methyl ketone. The retention efficiency was 81% at a sampling loading rate of 5.2 ml/min. The enhancement factor was 66 and the concentration efficiency was 61/min with a 50 sec preconcentration period, consuming 4.2 ml sample. A detection limit of 0.1 μg/l. Cd (3σ) was obtained with a sampling frequency of 55/hr. The precisions were 1.2% RSD for 20 μg/l. Cd (N = 11). Thiourea and ascorbic acid/phenanthroline were used to overcome interferences from copper and iron, respectively. The analytical results obtained for powdered rice and human hair standard reference materials were in good agreement with the certified values.  相似文献

An analytical method for the speciation of selenomethionine, selenocystine, selenite and selenate by high performance liquid chromatography (HPLC) with atomic spectrometric detection is presented. An organic polymeric strong anion exchange column was used as the stationary phase in combination with an aqueous solution of 6 mmol L–1 of salicylate ion at pH 8.5 as the mobile phase which allowed the isocratic separation of the four selenium analytes within 8 minutes. The separated selenium species were detected on-line by flame atomic absorption spectrometry (FAAS) or inductively coupled plasma mass spectrometry (ICP-MS). The signal-to-noise ratio of the FAAS detector was optimized using a hydrogen-argon entrained-air flame and a slotted-tube atom trap (STAT) in the flame. The limit of detection (3 σ) achieved by the HPLC-FAAS system was 1 mg L–1 of selenium (100 μL injections) for each of the four selenium species. More powerful selenium detection was achieved using an ELAN 5000 ICP-MS instrument. Selenium was measured at m/z = 82. The ICP-MS signal intensity was enhanced by a factor of 3–4 after addition of 3% methanol to the chromatographic mobile phase and by using an increased plasma power input of 1300 W. The limit of detection achieved under these conditions was 1 μg L–1 (100 μL injections). The HPLC-ICP-MS system was used for selenium speciation of selenite and selenate in aqueous solutions during a BCR certification exercise and for selenium speciation in the certified reference material, BCR No. 402 White Clover. Extraction experiments revealed that the selenium species in the biological material were extractable only in the presence of water in the extraction medium. The results indicated that selenate and a compound of unknown identity U were present in the plant sample.  相似文献

The stability of five selenium compounds, selenate, Se(VI), selenourea, SeUr, trimethylselenonium ion, TMSe(+), selenomethionine, SeMet, and selenoethionine, SeEt, at concentrations from 30-60 micro g L(-1) in a pooled human urine, stored in dark at -20 degrees C, 4 degrees C, or ambient temperature (ca. 25 degrees C), without addition of any stabilizing reagent was evaluated. The investigated Se species were determined independently by mixed ion-pair reversed-phase liquid chromatography with inductively coupled plasma mass spectrometric (ICP-MS) detection. The general trend is the lower the temperature used for storage, the higher the stability of Se species, when other conditions such as light, acidity, and container material are kept constant. On the basis of these results it is considered that the storage of urine samples at -20 degrees C for a short-term (within one month) is safe for Se speciation analysis. Long-term storage of urine samples for speciation analysis should, however, be undertaken with caution.  相似文献
