(1)H and (13)C NMR spectroscopic data for 5alpha-androstanes and halo-5alpha-androstanes with different substituents at positions C-3, C-9, C-11 and C-17 were examined and assigned by a combination of 1D and 2D NMR experiments. The substituent effects on the (13)C chemical shifts were compared with those of epi-androsterone, used as a reference compound. The coupling constants (n)J((19)F,(13)C) were measured for compounds 6, 8, 11 and 14.  相似文献

The 1H and 13C NMR data for 3-azabicyclo[3.3.1]nonanes having C-1 methylsuccinimidoanthranilate esters and C-6 methyl ethers were measured and assigned using 1D (DEPT) and 2D (COSY, HSQC, HMBC, NOESY) experiments. The total assignment of the (1)H- and (13)C-NMR spectra for TZT-1027 was carried out using various NMR methods (1D, 2D NMR). It was found that TZT-1027 exists in two different conformations resulting from the cis-trans isomerization of the amide bond at N-11 and C-12 in DMSO-d(6). The (1)H- and (13)C-NMR spectra of compound 1 and 2 comprised of the partial structure of TZT-1027 were also assigned to be TZT-1027. These assignments showed that compound 1 is in good agreement with TZT-1027 with regard to formation of the conformers.  相似文献

The microbial transformation of glycyrrhetinic acid(1) by Cunninghamella blakesleana CGMCC 3.970 led to the production of five new metabolites(2-6).The structures of the metabolites were determined by extensive spectroscopic(HR-ESIMS,1D and 2D NMR) data analyses.The involved reactions exhibited specific hydroxylations at C-24,C-7,and C-15,and oxidation at C-3.Moreover,compounds 2,5,and 6showed significant neural anti-inflammatory activity by inhibiting lipopolysaccharide-induced NO production in mouse microglia BV2 cells with IC_(50) values of 0.76,0.94,and 0.16μmol/L,respectively.  相似文献

The 1H and 13C NMR spectra of methyl-8-(2-furyl)-5-methyl-1,3-dioxo-3,3a,4,6,8,9,9a,9b-octahydrofuro[3,4-f]isoquinoline-7(1H)-carboxylate (1) and trimethyl 8-methyl-3-phenyl-3,4,4a,7-tetrahydro-2,5,6(1H)-isoquinolinetricarboxylate (2) at room temperature displayed doubling of the majority of signals, suggesting the presence of two rotational conformers (rotamers) in a ratio approximately 1:1.2 (in a mixture of CDCl3 and C6D6), approximately 1:1 (in CD2Cl2) and approximately 1:1.4 (in C6D6). On the basis of the temperature-dependent 1H NMR spectra of 1 and 2, the barrier to interconversion of the rotamers was calculated to be approximately 16 kcal mol(-1). The average chemical shifts and spin-spin coupling constants were analyzed for the resolution-enhanced 300 MHz 1H NMR spectrum of 1 at 333 K in CDCl3 solution. From analysis of the spin-spin coupling constants, it is proposed that the nitrogen-containing ring is in a chair conformation with C-2-H equatorial. Low- and room-temperature NOESY experiments in conjunction with theoretical ab initio calculations supported the hypothesis that the two rotamers interchange via rotation about the C-N bond. Spectral assignments of all proton and carbon resonances were made on the basis of one- and two-dimensional NMR experiments (DEPT, DQCOSY, NOESY, HETCOR and gHMBC).  相似文献

The (1)H and (13)C NMR spectra of 2,4,6,8-tetraaryl-3,7-diazabicyclo[3.3.1]nonan-9-ones (1-2), oximes (3-8) and O-benzyl oximes (9-12) were recorded. The chemical shifts were unambiguously assigned using 1D and 2D NMR spectral data. The results clearly indicate that the compounds exist in chair-boat conformation with equatorial and axial orientation of the aryl groups in the chair and boat forms, respectively. Since the molecules are flexible and dynamic in solution, the chair and boat forms are mutually interconvertible. In 3-12, because of the effect of oximation/oximination, all the protons in the heterobicyclic systems gave distinct signals except the benzylic protons of the boat form. In all synthesized compounds, the aryl group protons at C-6,8 are shielded by the aryl groups at C-2,4 and therefore appear in the lower frequency region than the aryl groups at C-2,4.  相似文献

Regioselective addition of allylthiol at the C-3 position adjacent to the nitrogen carrying the phenyl group of the 1,4-phenylenediamine moiety of compounds 1-4 was rigorously confirmed by 1D NOE difference in combination with gHMBC experiments. The structures of 1,4-phenylenediamines 1-4, allylsulfanyl-N1-alkyl-N4-phenyl-1,4-phenylenediamines 5-8 and cyclization products 9-14 were completely analyzed in both CDCl3 and DMSO-d6 solutions. The 1H and 13C NMR spectra of 10 and 11, which contain two chiral centers, exhibit duplication for several signals, indicating the existence of two diastereomeric forms. The full structures of 5 and 9 were unambiguously confirmed by x-ray crystallography. The 1H and 13C NMR spectra of all compounds were assigned using one- and two-dimensional NMR techniques (APT, DEPT, 1D NOE difference, COSY, NOESY, HETCOR, gHMQC and gHMBC).  相似文献

Seven new polyoxygenated steroids, hirsutosterols A-G (1-7), were isolated from the Formosan soft coral Cladiella hirsuta. Their structures were elucidated by spectroscopic methods, particularly in 1D and 2D NMR experiments. The absolute configurations of 1 and 5 were determined by Mosher's method. Sterols 4-6 possess hydroxy groups at C-9 and C-11 and might be oxidatively cleaved to the corresponding 9,11-secosterols. Hirsutosterol A (1) was found to exhibit a stronger cytotoxicity against a limited panel of cancer cell lines.  相似文献

Four novel proteasome inhibitors, TMC-95A-D (1-4) have been isolated from the fermentation broth of Apiospora montagnei Sacc. TC 1093, isolated from a soil sample. All of the molecular formulas of 1-4 were established as C(33)H(38)N(6)O(10) by high-resolution FAB-MS. Their planar structures were determined on the basis of extensive analyses of 1D and 2D NMR, and degradation studies. Compounds 1-4 have the same planar structures to each other, and are unique highly modified cyclic peptides containing L-tyrosine, L-aspargine, highly oxidized L-tryptophan, (Z)-1-propenylamine, and 3-methyl-2-oxopentanoic acid units. The absolute configuration at C-11 and C-36 of 1-4 was determined based on chiral TLC and HPLC analyses of their chemical degradation products. The ROESY analysis along with (1)H-(1)H coupling constants clarified the absolute stereochemistry at C-6, -7, -8, and -14 of the cyclic moieties. These studies revealed the relationships of 1-4 to be diastereomers at C-7 and C-36.  相似文献

The complete assignments of all the (1)H and (13)C NMR signals of three sesquiterpene lactones, 4-oxo-5(6),11-eudesmadiene-8,12-olide (1), 4-oxo-11-eudesmaene-8,12-olide (2) and (1(10)E)-5beta-Hydroxygermacra-1(10),4(15),11-trien-8, 12-olide (3), were carried out by various 2D NMR experiments. Compounds 1-3 were isolated from the roots of Inula helenium for the first time. Among them, 1 was identified as a new nor-sesquiterpene lactone, and 2 was isolated from a natural source for the first time. A comparative study of alkoxycarbonylation processes of both 19-nor-A-ring and A-ring stereoisomers of 1alpha,25-dihydroxyvitamin D3 analogues catalyzed by Candida antarctica lipase B (CAL-B) has been described. The presence of the methyl group in the A-ring at C-2, as in 3-6, has a determining role in the regioselectivity of the biocatalysis, mainly allowing the hydroxyl group at C-5 position to react. For the 19-nor-A-ring stereoisomers 7-10, which lack the C-2 methyl group, the configurations at C-3 and C-5 have a high influence in the selectivity exhibited by CAL-B. Thus, each couple of enantiomers showed opposing regioselectivities depending on the C-3 configuration. When C-3 possesses an (S)-configuration, enzymatic alkoxycarbonylations took place at the C-5-(R) or C-5-(S) hydroxyl groups. However, if the chiral centers at C-3 are (R), CAL-B alkoxycarbonylated the C-3-(R) hydroxyl group independently of the configuration at C-5. The corresponding carbonates are useful A-ring precursors of 1alpha,25-dihydroxyvitamin D3 analogues, selectively modified at the C-1 or C-3 positions. In addition, an improved synthesis of cis A-ring synthons 5 and 6 is described using a Mitsunobu methodology.  相似文献

In this investigation we report a complete assignment of (13)C, (1)H and (15)N solution and solid state chemical shifts of two bacterial photosynthetic pigments, bacteriochlorophyll (BChl) a and bacteriopheophytin (BPheo) a. Uniform stable-isotope labelling strategies were developed and applied to biosynthetic preparation of photosynthetic pigments, namely uniformly (13)C, (15)N labelled BChl a and BPheo a. Uniform stable-isotope labelling with (13)C, (15)N allowed performing the assignment of the (13)C, (15)N and (1)H resonances. The photosynthetic pigments were isolated from the biomass of photosynthetic bacteria Rhodopseudomonas palustris 17001 grown in uniformly (13)C (99%) and (15)N (98%) enriched medium. Both pigments were characterised by NMR in solution (acetone-d(6)) and by MAS NMR in solid state and their NMR resonances were recorded and assigned through standard liquid 2D (13)C-(13)C COSY, (1)H-(13)C HMQC, (1)H-(15)N HMBC and solid 2D (13)C-(13)C RFDR, (1)H-(13)C FSLG HETCOR and (1)H-(15)N HETCOR correlation techniques at 600 MHz and 750 MHz. The characterisation of pigments is of interest from biochemical to pharmaceutical industries, photosynthesis and food research.  相似文献

75%aqueous ethanol extract from the whole herbs of Selaginella doederleinii was isolated,and two new apigenin derivatives,doederflavones A(1) and B(2),together with ten known compounds(3-12) were characterized.Their structures were assigned by extensive spectroscopic methods including 1D/2D NMR and HR-ESIMS.Compounds 1-6 bear an aryl substituent at the C-8 or C-6 positions in ring A of apigenin skeleton.Compounds 1 and 2 were evaluated for their in vitro cytotoxicity against four human cancer cell lines A549,MCF-7,SMMC-7721,and LoVo,both of which exhibited significant cytotoxicity against A549 with IC_(50) values of 0.82 μmol/L and 1.32 μmol/L,respectively.  相似文献

(1)H and (13)C NMR spectra have been recorded for 3,5-bis[(E)-thienylmethylene]piperidin-4-one (1a), 3',3″-dimethyl-3,5-bis[(E)-thienylmethylene]piperidin-4-one (1b), 5',5″-dibromo-3,5-bis[(E)-thienylmethylene]piperidin-4-one (1c), their 1-methyl derivatives 2a-c and 3,5-bis[(E)-thienylmethylene]-2r,6c-diphenylpiperidin-4-one (3a). For selected compounds 2D spectra have been recorded. The spectral data are used to study the configuration and conformation of these molecules. The chemical shifts are discussed in light of steric, electronic and magnetic anisotropic effects. The magnetic anisotropic effects of thiophene ring and phenyl group are noteworthy. (1)H-(1)H COSY spectrum of 2b suggests that long-range (1)H-(1)H coupling, up to seven bonds, is possible in it. HMBC spectrum of 2b displays the magnetic nonequivalence of C-2 and C-6 and protons at these carbons.  相似文献

Two new C-3/C-3' biflavanones, chamaejasmenin D (1) and isochamaejasmenin B (2), were isolated from the roots of Stellera chamaejasme, together with five known biflavanones, chamaejasmenin A (3), chamaejasmenin B (4), neochamaejasmin A (5), sikokianin A (6), and chamaejasmenin C (7). Their structures were elucidated by spectroscopic methods, including 2D NMR techniques. Among these compounds, 1-3 demonstrated potent antimitotic and antifungal activity with minimum inhibitory concentration (MIC) values of 6.25, 6.25, and 3.12 microg/ml, respectively.  相似文献

The leaves of Morinda citrifolia, Linn. afforded a new benzofuran and a bis-nor-isoprenoid, blumenol C, hitherto unreported from this source. The structures of these have been elucidated as 5-benzofuran carboxylic acid-6-formyl methyl ester (1) and 4-(3'(R)-hydroxybutyl)-3,5,5, trimethyl-cyclohex-2-en-1-one (2) respectively through spectroscopic studies. The NMR data (including 1D, 2D techniques) and stereochemistry at C-3' of Compound 2 is also being reported for the first time.  相似文献

An analytical method for determination of the xanthate group distribution on viscoses based on liquid-state NMR spectroscopy was developed. Sample preparation involves stabilization of the xanthate group by allylation followed by derivatization of the remaining free hydroxyl groups at the glucose unit. The method was applied for studying (1) the γ-value (number of xanthate groups per 100 glucose units) of viscose, (2) the distribution of the xanthate groups on the anhydroglucose unit (AGU), and (3) changes of the xanthate group distribution during ripening. Results of the γ-value determination are well comparable with reference methods. Elucidation of the xanthate group distribution on the AGU gives the percentage at the C-6 position and a cumulative share of the positions C-2 and C-3. During ripening, xanthate groups at C-2 and C-3 degrade first, while xanthates at C-6 decompose at a slower rate.  相似文献

Experiments are described which prove the assignments of the α- and β-protons in the 1H NMR spectra of methyl phaeophorbide a ( 1 ) and methyl pyrophaeophorbide a ( 2 ). Because of the structural relationship between derivatives of the bacteriochlorophylls d ( 5 ) and 2 , dehydration of 5 results in a homologue mixture [2-(des-α-hydroxyethyl)-2-vinyl-bacteriomethyl phaeophorbide] d ( 6 ) of 2. Since the homologue substituents in 6 are located at C-4 and C-5 surrounding the β-H position, and since only one broad signal appears in its 1H NMR spectrum, this is assigned to the β-proton. This experiment proves that the sequence of increasing shielding is β, α and δ in 6 ; and, therefore, the same sequence applies to the 1H NMR spectra of 1 and 2 . This knowledge reveals that the product of electrochemical reduction of 1 in deuteromethanol is exclusively an α-chlorin-phlorin ( 8 ). In addition, the 1H NMR spectrum of the 2-vinyl derivatives of the bacteriomethyl phaeophorbides c ( 7 ) shows the same broad signal at lowest field as does that of 6 . The sequence of increasing shielding is therefore, β, α. The influence of the additional δ-methyl group in 7 on the ring current is explained.  相似文献
