建立电感耦合等离子体质谱法同时测定地质样品中镉、锗、钴含量的分析方法。样品用氢氟酸-硝酸混合溶液加热溶解,然后加热浓缩驱酸,再用硝酸溶液(1+1)复溶提取,在选定的仪器工作条件下进行测定。镉、锗、钴的含量分别在0.00~0.10,0.00~5.00,0.00~50.00 μg/g范围内与质谱峰强度呈良好的线性关系,相关系数均大于0.999,方法检出限分别为0.01,0.02,0.02 μg/g。用该方法对国家一级标准物质进行测定,测定值的相对误差均小于10%,测定结果的相对标准偏差为0.79%~8.45%(n=12)。该方法样品处理过程简便,检测效率高,适用于批量地质样品中镉、锗、钴的测定。 相似文献

Inductively coupled plasma mass spectrometry (ICPMS), particularly with sector field mass analyzers (SF-ICPMS), has emerged in the past several years as an excellent analytical technique for rapid, highly sensitive determination of transuranic elements (TRU) in environmental samples. SF-ICPMS has advantages of simplicity of sample preparation, high sample throughput, widespread availability in laboratories worldwide, and relatively straightforward operation when compared to other competing mass spectrometric techniques. Arguably, SF-ICPMS is the preferred technique for routine, high-throughput determination of 237Np and the Pu isotopes, excepting 238Pu, at fg-pg levels in environmental samples. Many research groups have now demonstrated the SF-ICPMS determination of 239 + 240Pu activities, 240Pu/239Pu and other Pu atom ratios in several different application areas. Many studies have examined the relative contribution of global fallout vs. local/regional Pu sources in the environment through measurement of 240Pu/239Pu and, in some cases, 241Pu/239Pu and 242Pu/239Pu. “Stratospheric fallout”, which was deposited from thermonuclear tests, conducted largely during the 1952–1964 time period, is characterized by a well-defined 240Pu/239Pu of ~ 0.18, while most other sources have different ratios. Examples of local/regional Pu sources are the Nevada Test Site, the Chernobyl plume, and accidents at Palomares, Spain and Thule, Greenland. The determination of Pu activities and atom ratios has stimulated much interest in the use of Pu as a marine tracer; several studies have shown that Pu is transported over long distances by ocean currents. 240Pu/239Pu ratios > 0.20 in sediments and seawater of the North Pacific are ascribed to ocean current transport of fallout from the Pacific Proving Ground. In nuclear forensics, much effort is focused on detection and fingerprinting of small amounts of TRU in environmental samples consisting of bulk material or individual isolated particles. Activity measurements of 239 + 240Pu, determined by SF-ICPMS, have the potential to supplement and/or replace 137Cs as a tracer of erosion, deposition, and sedimentation. Undoubtedly, the application of SF-ICPMS in TRU analysis will continue to expand, witness new developments, and generate interesting unforeseen applications in upcoming years. 相似文献

Chicken is the most consumed meat in North America. Concentrations of arsenic in chicken range from μg kg−1 to mg kg−1. However, little is known about the speciation of arsenic in chicken meat. The objective of this research was to develop a method enabling determination of arsenic species in chicken breast muscle. We report here enzyme-enhanced extraction of arsenic species from chicken meat, separation using anion exchange chromatography (HPLC), and simultaneous detection with both inductively coupled plasma mass spectrometry (ICPMS) and electrospray ionization tandem mass spectrometry (ESIMS). We compared the extraction of arsenic species using several proteolytic enzymes: bromelain, papain, pepsin, proteinase K, and trypsin. With the use of papain-assisted extraction, 10 arsenic species were extracted and detected, as compared to 8 detectable arsenic species in the water/methanol extract. The overall extraction efficiency was also improved using a combination of ultrasonication and papain digestion, as compared to the conventional water/methanol extraction. Detection limits were in the range of 1.0–1.8 μg arsenic per kg chicken breast meat (dry weight) for seven arsenic species: arsenobetaine (AsB), inorganic arsenite (AsIII), dimethylarsinic acid (DMA), monomethylarsonic acid (MMA), inorganic arsenate (AsV), 3-nitro-4-hydroxyphenylarsonic acid (Roxarsone), and N-acetyl-4-hydroxy-m-arsanilic acid (NAHAA). Analysis of breast meat samples from six chickens receiving feed containing Roxarsone showed the presence of (mean ± standard deviation μg kg−1) AsB (107 ± 4), AsIII (113 ± 7), AsV (7 ± 2), MMA (51 ± 5), DMA (64 ± 6), Roxarsone (18 ± 1), and four unidentified arsenic species (approximate concentration 1–10 μg kg−1). 相似文献

was used for the determinations. The analytical results of the reference materials agreed with the certified values. The precision
between sample replicates was better than 6% for all determinations. The method detection limit estimated from analyte addition

As a long-lived beta-emitting radioisotope of iodine, 129I is produced both naturally and as a result of human nuclear activities. At present time, the main part of 129I in the environment originates from the human nuclear activity, especially the releases from the spent nuclear fuel reprocessing plants, the 129I/127I ratios have being reached to values of 10−10 to 10−4 in the environment from 10−12 in the pre-nuclear era. In this article, we review the occurrence, sources, inventory, and concentration level of 129I in environment and the method for speciation analysis of 129I in the environment. Measurement techniques for the determination of 129I are presented and compared. An overview of applications of 129I speciation in various scientific disciplines such as radiation protection, waste depository, and environmental sciences is given. In addition, the bioavailability and radiation toxicity (dose to thyroid) of 129I are discussed. 相似文献

An analytical method for separation and determination of thallium species in water using high‐performance liquid chromatography with inductively coupled plasma mass spectrometry was developed. The composition and concentration of mobile phase, injection volume, and pH value were optimized respectively with an anion or cation exchange column. The results showed that Tl(I) and Tl(III) were effectively separated using anion exchange column Hamilton PRP‐X100, with the mobile phase consisting of 200 mmol/L ammonium acetate and 10 mmol/L diethylenetriaminepentaacetic acid (pH = 4.2). When using a Dionex cation exchange guard column, CS12A, 15 mmol/L HNO3, and 3 mmol/L diethylenetriaminepentaacetic acid as the mobile phase, Tl(I) and Tl(III) could be effectively separated. The detection limits of the methods were 3–6 and 9–12 ng/L, respectively. In a solution containing Fe ions and oxalic acid, a significant quantity of Tl(I) was oxidized. Fe ions and oxalic acid in the water samples did not interfere with high‐performance liquid chromatography‐inductively coupled plasma mass spectrometry measurement results. 相似文献

An analytical procedure for the reliable determination of Pd, Pt and Rh in plant samples by inductively coupled plasma-mass spectrometry (ICP-MS) was developed. An ultrasonic nebulizer (USN) was used for sample introduction to improve sensitivity. Under various synthetic plant sample matrix compositions, it was established experimentally that moderate amounts (0.2–2%) of dissolved solids decreased the analyte signals significantly. Internal standardisation with In (for Pd and Rh) and Ir (for Pt) proved to be essential for obtaining correct results. Five open digestion approaches, used for converting solid plant samples to aqueous solution, were also tested for the purpose, namely dry-ashing, dry-ashing followed by HF attack, wet digestion with H2O2–HNO3, wet digestion followed by HF attack and aqua regia digestion. Recovery tests in two spiked plant materials showed that only wet digestions must be used. With these ways, all PGEs could be reliably quantified by USN-ICP-MS without applying a separation or preconcentration step with a good precision (below 10% RSD). The aqua regia procedure was applied to the determination of PGEs in various plant matrices collected along a highway. Results showed that mosses were probably the best choice of samples to monitor the bioaccumulation of PGEs in time. 相似文献

A simple and convenient method for the digestion of animal tissues, lichens, and plants for 33 metals measured by sector field inductively coupled plasma mass spectrometry (SF-ICP-MS) was described. Microwave-assisted acid digestions were performed at atmospheric pressure by means of a multi-samples rotor designed for processing a large number of samples at once in screw-capped disposable polystyrene liners. The digested samples were filled up to final volume directly in the polystyrene liners ready for elemental quantification. Seven certified reference materials, namely BCR 184 (bovine muscle), BCR 186 (pig kidney), DORM-2 (dogfish muscle), BCR 422 (cod muscle), BCR 62 (olive leaves), BCR 100 (beech leaves), and BCR 482 (lichen) were analysed to verify the accuracy of the method. The linearity range, limit of quantification, precision, and recovery by addition of non-certified elements were also assessed. All elements, with the exception of Hg in BCR 184 and As in BCR 186, were above the quantification limit and blank concentrations, and good agreement existed between found and target values in bovine muscle, pig kidney, and cod muscle. Significant deviations were observed for Al, Co, Cr, Mn, and Ni in dogfish muscle and for Ca, Cr, Fe, and Hg in lichens and plants. The proposed digestion procedure offers a low contamination risk, simplicity, speed, low cost, and applicability in routine analysis, and the SF-ICP-MS method allowed metals from a fraction of ng?g?1 to hundreds of µg?g?1 to be quantified in one analytical run. 相似文献

Due to the lack of a stable technetium isotope, and the high mobility and long half-life, 99Tc is considered to be one of the most important radionuclides in safety assessment of environmental radioactivity as well as nuclear waste management. 99Tc is also an important tracer for oceanographic research due to the high technetium solubility in seawater as TcO4−. A number of analytical methods, using chemical separation combined with radiometric and mass spectrometric measurement techniques, have been developed over the past decades for determination of 99Tc in different environmental samples. This article summarizes and compares recently reported chemical separation procedures and measurement methods for determination of 99Tc. Due to the extremely low concentration of 99Tc in environmental samples, the sample preparation, pre-concentration, chemical separation and purification for removal of the interferences for detection of 99Tc are the most important issues governing the accurate determination of 99Tc. These aspects are discussed in detail in this article. Meanwhile, the different measurement techniques for 99Tc are also compared with respect to advantages and drawbacks. Novel automated analytical methods for rapid determination of 99Tc using solid extraction or ion exchange chromatography for separation of 99Tc, employing flow injection or sequential injection approaches are also discussed. 相似文献

A highly sensitive method was developed for the simultaneous separation and determination of organic and inorganic selenium species in rice by ion‐pairing reversed‐phase chromatography combined with inductively coupled plasma tandem mass spectrometry. To achieve a good separation of these species, a comparison between anion‐exchange chromatography and ion‐pairing reversed‐phase chromatography was performed. The results indicated that ion‐pairing reversed‐phase chromatography was more suitable due to better separation and higher sensitivity for all analytes. In this case, a StableBond C18 column proved to be more robust or to have a better resolution than other C18 columns, when 0.5 mM tetrabutylammonium hydroxide and 10 mM ammonium acetate at pH 5.5 were used as the mobile phase. Moreover, an excellent sensitivity was obtained in terms of interferences by means of tandem mass spectrometry in the hydrogen mode. The detection limits were 0.02–0.12 μg/L, and recoveries of five selenium species were 75–114%, with relative standard deviations ≤ 9.4%. This method was successfully applied to the analysis of rice samples. Compared with previous studies, the proposed method not only gave comparable results when used for measuring selenium‐enriched rice, but it can provide greater sensitivity for the detection of low concentrations of selenium species in rice. 相似文献

An inductively coupled plasma quadrupole mass spectrometer (ICP-QMS) equipped with a Dynamic Reaction Cell™ (DRC) was used to reduce the oxide interferences for the accurate determination of Ag and Cd in samples in the presence of excess Zr, Nb and Mo. The effect of the operating conditions of the DRC system was studied to obtain the best signal to noise ratios for and . The potentially interfering polyatomic ions determination were decreased in intensity to an insignificant level by using O2 as the reaction gas. Furthermore, a rejection parameter q (Rpq) of 0.6 was used to filter out unwanted precursors of interfering species from the ion beam to eliminate interferences created in the reaction cell. The detection limit was ca. 2 and 3 pg ml−1 for and , respectively, with the DRC ICP-MS method. The method was applied to the determination of Ag and Cd in NIST SRM 2709 San Joaquin soil reference material as well as NIST SRM 1568a rice flour reference material. Gas chromatography–mass spectrometry (GC-MS) method and a liquid chromatography–fluorescence (LC-FL) detection method using experimental design and optimisation approach were improved for the quantitative determination of nitrite and nitrate in biological, food and environmental samples. The obtained recoveries of nitrite and nitrate ions from samples based on both GC-MS and LC-FL results ranged from 98.5% to 98.9% for nitrite and 97.9% to 98.4% for nitrate. The precision of these methods, as indicated by the relative standard deviations (RSDs), was within the range from 2.4% to 3.6% for nitrite and 2.5% to 3.8% for nitrate, respectively. The limits of detection of nitrite and nitrate ions from samples based on GC-MS and LC-FL results ranged from 0.01 to 0.14 ng L?1 for nitrite and 0.02 to 0.71 ng L?1 for nitrate, respectively. The optimised isolation procedure by central composite design was successfully applied to real samples. The results revealed that the proposed procedure combined with GC-MS and LC-FL techniques is more sensitive, reliable and selective compared to the other methods available for the precise determination of trace levels of nitrite and nitrate in biological, food and environmental samples. 相似文献

A method based on the coupling microHPLC-microneb-ICPMS has been developed for Hg(II), MeHg+, EtHg+ and PhHg+ species. Gradient elution using methanol and l-cysteine at pH 3.0 allowed the chromatographic separation of all species in less than 13 min (total analysis time 15 min). The direct coupling of microLC to ICPMS through a Micromist nebulizer permits the analysis of environmental water without sample pretreatment and derivatization steps. Nebulizer type, organic modifier and column length were the main parameters tested. The methanol content and pH of the mobile phase greatly affected the retention time and sensitivity of the method. Key factors to obtain high signal to noise ratio, at concentrations below 1 μg L−1, were found to be the nebulization step and traces of Hg present in the complexing agent. A detailed optimization of carrier and make up gas flow rates have enabled the nebulization of the methanol gradient elution with good mass transport efficiency, low organic solvent loading into the plasma and excellent precision.The performance of the microHPLC-microneb-ICPMS method developed was evaluated on a surface water sample filtered (0.22 μm) and spiked with 0.5 μg L−1 (as Hg) of each species. Precision (R.S.D., n = 6) for all species of Hg varied from 0.5 to 2.1%. Detection limit, defined as three times the standard deviation (n = 6), ranged from 8 ng L−1 for EtHg+ to 32 ng L−1 for PhHg+ and was noticeably lower than those reported in previous LC-based methods. Accuracy was suitable with recoveries ranging from 85 to 100% when tested at two levels (0.5 and 10 μg L−1) in groundwater samples. Recovery was matrix affected when water samples of high salinity (depurated wastewater and seawater) were used. 相似文献

A simple, sensitive and rapid liquid chromatographic/electrospray ionization tandem mass spectrometric method was developed and validated for the quantification of lacidipine in human plasma using its structural analogue, amlodipine, as internal standard (IS). The method involves a simple single-step liquid-liquid extraction with tert-butyl methyl ether. The analyte was chromatographed on an Xterra MS C(18) reversed-phase chromatographic column by isocratic elution with 20 mM ammonium acetate buffer-acetonitrile (10:90, v/v; pH 6) and analyzed by mass spectrometry in the multiple reaction monitoring mode. The precursor to product ion transitions of m/z 456.4 --> 354.4 and m/z 409.3 --> 238.3 were used to measure the analyte and the I.S., respectively. The chromatographic run time was 1.5 min and the weighted (1/x(2)) calibration curves were linear over the range 0.1-25 ng ml(-1). Lacidipine was sensitive to temperature in addition to light. The method was validated in terms of accuracy, precision, absolute recovery, freeze-thaw stability, bench-top stability and re-injection reproducibility. The limit of detection and lower limit of quantification in human plasma were 50 and 100 pg ml(-1), respectively. The within- and between-batch accuracy and precision were found to be well within acceptable limits (<15%). The analyte was stable after three freeze-thaw cycles (deviation <15%). The average absolute recoveries of lacidipine and amlodipine (IS) from spiked plasma samples were 51.1 +/- 1.3 and 50.3 +/- 4.9%, respectively. The assay method described here could be applied to study the pharmacokinetics of lacidipine. 相似文献

24S-hydroxycholesterol (i.e., cerebrosterol, 24S-OH-Chol) is the main form of cholesterol elimination from the brain. Liquid chromatography–tandem mass spectrometry methods were developed for the quantification of the total and unesterified/unbound fractions of 24S-OH-Chol, its monosulfate, monoglucuronide, and diconjugate derivatives (24S-OH-Chol-3sulfate [3S], 24S-OH-Chol-24glucuronide [24G] and 24S-OH-Chol-3S, 24G, respectively) in human plasma. Linearity, precision, accuracy, and extraction recovery were validated within the typical physiological and pathological ranges of concentrations for each compound. The lower limit of quantifications was 2.00, 0.33, 0.26, and 0.74 ng/ml for 24S-OH-Chol, 24S-OH-Chol-24G, 24S-OH-Chol-3S, and 24-OH-Chol-3S, 24G, respectively. Extraction recovery values in total and unbound plasma fractions were also analyzed in murine and monkey plasma and varied from 73% in mouse to 113% in cynomolgus monkey. The methods could rapidly (less than 7 min) quantify individual compounds with high sensitivity, accuracy (bias ≤15%), and reproducibility (coefficient of variation [CV] ≤ 17%). Their clinical applications were validated by measuring levels of the 4 compounds in samples from 20 noncholestatic donors, 5 cholestatic patients suffering from primary biliary cirrhosis, and 10 patients suffering from biliary stenosis. Results highlight the abundance of 24S-OH-Chol in the total fraction and the abundance of 24S-OH-Chol-3S and 24G in the unbound ones. While the latter strongly accumulate in plasma fractions of cholestatic patients, levels of 24S-OH-Chol remained similar to those of healthy donors. Our results indicate that this approach is suitable for monitoring cerebrosterol and its conjugates in large-scale clinical studies. 相似文献

A new method was developed and fully validated for the quantitation of benazepril, benazeprilat and hydrochlorothiazide in human plasma. Sample pretreatment was achieved by solid-phase extraction (SPE) using Oasis HLB cartridges. The extracts were analysed by high-performance liquid chromatography (HPLC) coupled to a single-quadrupole mass spectrometer (MS) with an electrospray ionization interface. The MS system was operated in selected ion monitoring (SIM) modes. HPLC was performed isocratically on a reversed-phase porous graphitized carbon (PGC) analytical column (2.1 x 125.0 mm i.d., particle size 5 microm). The mobile phase consisted of 55% acetonitrile in water containing 0.3% v/v formic acid and pumped at a flow rate of 0.15 ml min(-1). Chlorthalidone was used as the internal standard (IS) for quantitation. The assay was linear over a concentration range of 5.0-500 ng ml(-1) for all the compounds analysed, with a limit of quantitation of 5 ng ml(-1) for all the compounds. Quality control (QC) samples (5, 10, 100 and 500 ng ml(-1)) in five replicates from three different runs of analyses demonstrated intra-assay precision (coefficient of variation (CV) < or =14.6%), inter-assay precision (CV < or = 5.6%) and overall accuracy (relative error less than -8.0%). The method can be used to quantify benazepril, benazeprilat and hydrochlorothiazide in human plasma, covering a variety of pharmacokinetic or bioequivalence studies. 相似文献

Besides liquid chromatographic (LC)/UV methods adapted to therapeutic drug monitoring, there is still a need for more powerful techniques that can be used for pharmacological research and clinical purposes. We developed an LC method coupled with tandem mass spectrometry (MS/MS) to separate, detect and quantify with high sensitivity the nucleoside analogues used in multitherapies (zidovudine, stavudine, zalcitabine, didanosine, lamivudine and abacavir) in plasma and in the intracellular medium. We worked on two essential issues: (i) the need to use two ionization modes in order to achieve the best sensitivity, which leads to the optimization of the chromatographic separation of drugs detected in the positive ionization mode and drugs detected in the negative ionization mode, and (ii) the need to optimize the extraction step in order to enhance sample recovery. The peripheral blood mononuclear cells were lysed in Tris buffer-MeOH. A clean-up procedure was performed by solid-phase extraction only for plasma samples. The LC separation was carried out on a Zorbax Stable Bond C(18) column followed by MS/MS analysis after electrospray ionization in either the negative or positive mode. The positive ionization mode was applied at the beginning of the run to detect zalcitabine and lamivudine, then the ionization mode was changed to negative for the detection of didanosine, stavudine, internal standard and zidovudine. The calibration range for all the analytes was 0.5-200 ng ml(-1). The recoveries were between 64 and 90%, with coefficients of variation (CVs) lower than 15%. The inaccuracy (bias) was +/-15% with CVs always lower than 12%. The analytes were stable at room temperature and in the extraction solvent for at least 24 h, after storage at -80 degrees C for 3 months, after three freeze-thaw cycles and in the injection solvent after 48 h at 4 degrees C. Together with the measurement of intracellular triphosphorylated metabolites thanks to the powerful plasma and intracellular assay method for intact drugs, it is possible to describe the behaviour of nucleoside analogues against HIV through plasma pharmacokinetics, cell membrane diffusion including drug transport involvement, and also the intracellular metabolism. 相似文献

In the present paper, a critical overview of the most commonly used techniques for the characterization and the determination of carbon nanotubes (CNTs) is given on the basis of 170 references (2000–2014). The analytical techniques used for CNT characterization (including microscopic and diffraction, spectroscopic, thermal and separation techniques) are classified, described, and illustrated with applied examples. Furthermore, the performance of sampling procedures as well as the available methods for the determination of CNTs in real biological and environmental samples are reviewed and discussed according to their analytical characteristics. A new high-performance liquid chromatographic/electrospray ionization tandem mass spectrometric (HPLC/ESI-MS/MS) method was developed for the simultaneous quantification of 5-fluorouracil (5FU), methotrexate (MTX) and cyclophosphamide (CP) in environmental samples. These compounds, commonly used in the treatment of cancer, are recognized as genotoxic. In order to estimate the occupational exposure of hospital personnel handling these drugs, wipe samples were taken from the working surfaces and directly analyzed (with trophosphamide as internal standard) using a reversed-phase capillary column and MS/MS detection. This is the first HPLC/MS/MS method for the simultaneous determination of 5FU, MTX and CP. The present method offers high sensitivity, with detection limits of 1.1 microg l(-1) for MTX and CP and 33.3 microg l(-1) for 5FU, avoiding any sample preconcentration procedure. Rapidity, specificity, high accuracy (mean values between 92.4 and 99.9%) and precision (mean RSD values between 3.4 and 12.1%) make the method suitable for the routine determination of these three antineoplastic drugs. 相似文献
