Imaging in Raman spectroscopy is a valuable tool for analytical chemistry. Although molecular characterization at micron level is achieved for many applications, it usually fails producing chemical images of micron size samples as expected in chemical, environmental and biological analysis. The aim of the work is to introduce the potential of super-resolution in vibrational spectroscopic imaging. This original chemometrics approach uses several low resolution images of the same sample in order to retrieve a higher resolution chemical image. It is thus possible to overcome in a certain way some physical and instrumentals limitations. To illustrate the methodology, sub-micronic details of a Si/Au sample are retrieved from low resolution images with different super-resolution algorithms. The better results are obtained with Iterative L2/Bilateral Total Variation regularization method. The use of a regularization procedure gives also better results since its first property is to preserve edges during the reconstruction of the super-resolved image. This concept of chemical image data processing should open new analytical opportunities.  相似文献

Being able to predict the final product yield at all stages in long-running, industrial, mammalian cell culture processes is vital for both operational efficiency, process consistency, and the implementation of quality by design (QbD) practices. Here we used Raman spectroscopy to monitor (in terms of glycoprotein yield prediction) a fed-batch fermentation from start to finish. Raman data were collected from 12 different time points in a Chinese hamster ovary (CHO) based manufacturing process and across 37 separate production runs. The samples comprised of clarified bioprocess broths extracted from the CHO cell based process with varying amounts of fresh and spent cell culture media. Competitive adaptive reweighted sampling (CoAdReS) and ant colony optimization (ACO) variable selection methods were used to enhance the predictive ability of the chemometric models by removing unnecessary spectral information. Using CoAdReS accurate prediction models (relative error of predictions between 2.1% and 3.3%) were built for the final glycoprotein yield at every stage of the bioprocess from small scale up to the final 5000 L bioreactor. This result reinforces our previous studies which indicate that media quality is one of the most significant factors determining the efficiency of industrial CHO-cell processes. This Raman based approach could thus be used to manage production in terms of selecting which small scale batches are progressed to large-scale manufacture, thus improving process efficiency significantly.  相似文献

Optical scattering by biological tissues largely deteriorates the efficiency of the Raman analysis of these tissues. To evaluate the effect of scattering on Raman depth profiles (RDPs), we developed a simple method using a thin-layered sample mimicking real tissues and a conventional Raman microscope. The sample comprised three layers: a silicon wafer, a thin aqueous film containing Intralipid particles as scatters, and a fused silica window; this design was used to mimic real skin tissues quantitatively. The multi-scattering effect, which deteriorates spatial resolution, was clearly observed as broadening of RDPs. Decrease in Raman intensity was also systematically examined as a function of both the concentration of the Intralipid particles and depth of the film, and evaluated using Lambert-Beer's law. The abovementioned observations can be quantitatively explained on the basis of the scattering cross-section and concentration of the Intralipid particles, indicating that the method is useful for the quantification of the deterioration of Raman measurements due to optical scattering.  相似文献

A rapid Raman spectroscopy protocol is reported to classify gasoline according to its distributor and to identify and quantify common adulterants. Gasoline from three distributors was collected from 19 stations in São Paulo, Brazil. Principal component analysis (PCA) showed specific clusters for each distributor, and partial least squares discriminant analysis (PLS-DA) correctly identified the origin of the samples. To evaluate the technique for the identification and quantification of the adulterants, authentic samples from each distributor were fortified at levels from 2.5 up to 25.0% (v/v) using ethanol, methanol, toluene, and turpentine to obtain 120 altered samples. PCA showed clear separation among the samples with the adulterants and PLS-DA precisely identified the adulterants (478 in 480 predictions by cross-validation), irrespective of the distributor and the concentration. One classification model was used to characterize all distributors. To quantify the adulterants, 36 multivariate calibration models were constructed using partial least squares (PLS), interval PLS, and PLS genetic algorithm for each distributor and for each adulterant. Cross-validation errors of less than 5.0% were obtained for all adulterants regardless of the distributor. Raman spectroscopy and multivariate analysis were shown to be powerful for rapid and inexpensive for the characterization of gasoline origin and the identification and quantification of common adulterants.  相似文献

Methods for chemical analysis at the nanometer scale are crucial for understanding and characterizing nanostructures of modern materials and biological systems. Tip‐enhanced Raman spectroscopy (TERS) combines the chemical information provided by Raman spectroscopy with the signal enhancement known from surface‐enhanced Raman scattering (SERS) and the high spatial resolution of atomic force microscopy (AFM) or scanning tunneling microscopy (STM). A metallic or metallized tip is illuminated by a focused laser beam and the resulting strongly enhanced electromagnetic field at the tip apex acts as a highly confined light source for Raman spectroscopic measurements. This Review focuses on the prerequisites for the efficient coupling of light to the tip as well as the shortcomings and pitfalls that have to be considered for TERS imaging, a fascinating but still challenging way to look at the nanoworld. Finally, examples from recent publications have been selected to demonstrate the potential of this technique for chemical imaging with a spatial resolution of approximately 10 nm and sensitivity down to the single‐molecule level for applications ranging from materials sciences to life sciences.  相似文献

Fast determination of milk fat content using Raman spectroscopy   总被引:1,自引:0,他引:1  
In our work, we have demonstrated the capability of VIS Raman spectroscopy in combination with partial least square regression (PLS) as a rapid technique for direct milk fat determination. Raman spectra of milk samples revealed contributions from proteins, but mainly from their fat content with different spectral characteristics. Three different methods of sample preparations were applied: (i) liquid milk contained in an open dish, (ii) dried milk droplets on glass plates covered with Al foil, and (iii) liquid milk contained in quartz cuvettes. Methods (i) and (ii) showed a good PLS model for milk fat prediction with low root mean square errors and high correlation coefficients. The main advantage of milk sample contained in the dish lies in its simplicity as well as the fact that the open container maximizes the signal of interest avoiding background contributions. Our results show that Raman spectroscopy is suited for in-line monitoring purposes.  相似文献

Ovarian cancer is the sixth most common cancer among women worldwide, and mortality rates from this cancer are higher than for other gynecological cancers. This is attributed to a lack of reliable screening methods and the inadequacy of treatment modalities for the advanced stages of the disease. FTIR and Raman spectroscopic studies of formalin-fixed normal, benign, and malignant ovarian tissues have been undertaken in order to investigate and attempt to understand the underlying biochemical changes associated with the disease, and to explore the feasibility of discriminating between these different tissue types. Raman spectra of normal tissues indicate the dominance of proteins and lower contents of DNA and lipids compared to malignant tissues. Among the pathological tissues studied, spectra from benign tissues seem to contain more proteins and less DNA and lipids compared to malignant tissue spectra. FTIR studies corroborate these findings. FTIR and Raman spectra of both normal and benign tissues showed more similarities than those of malignant tissues. Cluster analysis of first-derivative Raman spectra in the 700–1700 cm−1 range gave two clear groups, one corresponding to malignant and the other to normal+benign tissues. At a lower heterogeneity level, the normal+benign cluster gave three nonoverlapping subclusters, one corresponding to normal and two for benign tissues. Cluster analysis of second-derivative FTIR spectra in the combined spectral regions of 1540–1680 and 1720–1780 cm−1 resulted into two clear clusters corresponding to malignant and normal+benign tissues. The cluster corresponding to normal+benign tissues produced nonoverlapping subclusters for normal and benign tissues at a lower heterogeneity level. The findings of this study demonstrate the feasibility of Raman and FTIR microspectroscopic discrimination of formalin-fixed normal, benign, and malignant ovarian tissues.  相似文献

We demonstrated an on-line Raman measurement of polyethylene (PE) pellet density when it is flying in a sample line. While in flight, pellets are sparsely populated at spectral collection, a spectral collection strategy covering a large spatial volume (larger number of pellets simultaneously) is necessary to acquire reasonable Raman intensity. In addition, the Raman measurement must be less sensitive to pellet position, because position and distribution are uncontrollable in a flying condition. To fulfill these requirements, a wide area illumination (WAI) scheme capable of covering a large sample volume (illumination volume: 0.7 cm3) was used when the pellets were flying in a 2.5-cm-diameter sample line. In addition, a long focal length (250 mm) was used so that minor changes in pellet position would not significantly affect the resulting Raman spectral feature. Although Raman intensity substantially decreased due to the large void space among flying pellets, a correct spectral feature representing PE was successfully obtained without any significant spectral distortion. Using partial least squares (PLS) regression, the prediction error under flying conditions was 0.0009 g cm−3, which was comparable to that acquired when the pellets were packed (0.0008 g cm−3). When a conventional Raman scheme covering a smaller sample volume with a short focal length was used, the PE intensity decreased dramatically, and the resulting signal-to-noise ratio was not proper for quantitative analysis.  相似文献

Recently, Raman spectroscopy become a popular and potential analytical technique for the analysis of pharmaceuticals as a result of its advancement. The innovation of laser technology, Fourier Transform-Raman spectrometers with charge coupled device (CCD) detectors, ease of sample preparation and handling, mitigation of sub-sampling problems using different geometric laser irradiance patterns and invention of different optical components of Raman spectrometers are contributors of the advancement of Raman spectroscopy. Transmission Raman Spectroscopy is a useful tool in pharmaceutical analysis to address the problems related with sub-sampling in conventional Raman back scattering. More importantly, the development of surface-enhanced Raman scattering (SERS) has been a prominent advancement for Raman spectroscopy to be applied for pharmaceuticals analysis as it avoids the inherent insensitivity and fluorescence problems. As the active pharmaceutical ingredients (APIs) contain aromatic or conjugated domains with strong Raman scattering activity, Raman spectroscopy is an attractive alternative conventional analytical method for pharmaceuticals. Coupling of Raman spectroscopy with separation techniques is also another advancement applied to reduce or avoid possible spectral interferences. Therefore, in this review, transmission Raman spectroscopy, SERS, and SERS coupled with various separation techniques for pharmaceutical analysis are presented.  相似文献

Irregularly shaped and inorganic fullerene-like MoS2 compounds were characterized by Resonance Raman spectroscopy using an exciting line at 633 nm. It was shown that the relative intensity of the longitudinal acoustic mode at 226 cm−1 and its overtone strongly depends on the MoS2 crystallite size but not on the size of the particles made of agglomerated crystallites. This technique appeared as a promising probe to characterize in situ very small crystallites that are not observed by XRD.  相似文献

The conformations of dodecamethylcyclohexasilane Si6Me12 and undecamethylcyclohexasilane Si6Me11H have been investigated by ab initio calculations employing the B3LYP density functional with a 6-31+G(d) basis set. Local minima as well as transition structures were calculated with imposed symmetry constraints. For Si6Me12, three unique minima, which correspond to the chair, twist and boat conformations were located with relative zero-point-vibration-corrected energies of 0.0, 7.8 and 11.4 kJ mol(-1). A half-chair conformation with four coplanar silicon atoms connects the chair and twisted minima via an energy barrier of 16.0 and 8.2 kJ mol(-1), respectively. A second transition structure with a barrier of 3.9/0.3 kJ mol(-1) connects the twist with the boat structure. Solution Raman spectra of Si6(CH3)12 and Si6(CD3)12 fully corroborate these results. Below -40 degrees C, the symmetric SiSi ring breathing vibration is a single line, which develops a shoulder (originating from the twist conformer) at longer wavelengths whose intensity increases with increasing temperature. From a Van't Hoff plot, the chair/twist enthalpy difference is 6.6+/-1.5 kJ mol(-1) for Si6(CH3)12 and 6.0+/-1.5 kJ mol(-1) for Si6(CD3)12, which is in reasonable agreement with the ab initio results. Due to the low barrier, the boat conformation cannot be observed, because either the lowest torsional vibration level lies above it or a rapid interconversion between the twist and boat conformations occurs, resulting in averaged Raman spectra. For Si6Me11H, six local minima were located. The chair with the hydrogen atom in the axial position (axial chair) is the global minimum, followed by the equatorial chair (+1.9 kJ mol(-1)) and the three twist conformers (+5.3, +8.0 and +8.1 kJ mol(-1)). The highest local minimum (+11.9 kJ mol(-1)) is a C(s) symmetric boat with the hydrogen atom in the equatorial position. Two possible pathways for the chair-to-chair interconversion with barriers of 13.9 and 14.5 kJ mol(-1) have been investigated. The solution Raman spectra in the SiSi ring breathing region clearly show that below -50 degrees C only the axial and equatorial chairs are present, with an experimental deltaH-value of 0.46 kJ mol(-1). With increasing temperature a shoulder develops which is attributed to the combined twist conformers. The experimental deltaH-value is 6.9 kJ mol(-1), in good agreement with the ab initio results. Due to the low interconversion barriers, the various twist conformers cannot be detected separately.  相似文献

Accurate analysis of samples is very important for scientists working in many fields. XRF device is used very frequently especially in mine analysis. However, researchers are trying to reach accurate results with many different analysis methods. In addition to the known analysis methods, alternative research methods also guide the studies. In this study, two barite ore samples, collected from two regions of different nature (Denizli and Akda?madeni) by following specified sampling methods, were analyzed using Confocal Raman Spectroscopy (CRS) and Polarized Energy Dispersive X-Ray Fluorescence (PEDXRF) spectrometer. The first sample was from a metamorphic basement, and the second was from an alkali syenite rock unit. The main objective of this paper is to compare the optical characteristics of these two different barite samples collected from different regions under a polarized microscope, using CRS and PEDXRF. The results of polarized microscopy analysis showed that the barite taken from Denizli is associated with calcite. On the other hand, the barite taken from Akda?madeni is associated with galena, celestite, and quartz. Two different colors were observed in the barite samples. CRS and PEDXRF results showed that the barites collected from two regions differed in mineral association, chemical composition, and physical properties. The accuracy of the chemical analysis technique was ensured by following USGS standards, GBW 7109, and GBW-7309 Sediment. Barite ores were analyzed using HR-800 (HORIBA-Jobin Yvon) CRS and a polarized microscope (Leica DMLP). Thanks to this study, it has been shown that mineral analyzes can be performed with an accuracy close to XRF with Confocal Raman spectroscopy. Confocal Raman spectroscopy will also guide researchers for mineral analysis.  相似文献

The development of a classification system based on the Raman spectra of milk samples is proposed in present study. Such development could be useful for nutritionists in suggesting healthy food to infants for their proper growth. Previously, molecular structures in milk samples have been exploited by Raman spectroscopy. In the current study, Raman spectral data of milk samples of different species is utilized for multi-class classification using a dimensionality reduction technique in combination with random forest (RF) classifier. Quantitative and experimental analysis is based on locally collected milk samples of different species including cow, buffalo, goat and human. This classification is based on the variations (different concentrations of the components present in milk such as proteins, milk fats, lactose etc.) in the intensities of Raman peaks of milk samples. Principal component analysis (PCA) is used as a dimensionality reduction technique in combination with RF to highlight the variations which can differentiate the Raman spectra of milk samples from different species. The proposed technique has demonstrated sufficient potential to be used for differentiation between milk samples of different species as the average accuracy of about 93.7%, precision of about 94%, specificity of about 97% and sensitivity of about 93% has been achieved.  相似文献

In archaeometry, one of the main concerns is to extract information from an art object, without damaging it. Raman spectroscopy is being applied in this research field with recent developments in mobile instrumentation facilitating more routine analysis. This research paper evaluates the performances of five mobile Raman instruments (Renishaw RA100, Renishaw Portable Raman Analyser RX210, Ocean Optics RSL-1, Delta Nu Inspector Raman, Mobile Art Analyser - MArtA) in three different laboratories. A set of samples were collected, in order to obtain information on the spectral performances of the instruments including: spectral resolution, calibration, laser cut-off, the ability to record spectra of organic and inorganic pigments through varnish layers and on the possibilities to identify biomaterials. Spectra were recorded from predefined regions on a canvas painting to simulate the investigation of artworks and the capabilities to record spectra from hardly accessible areas was evaluated.  相似文献

Interactions of dimethoxycurcumin (1) a lipophilic bioactive curcumin derivative with dipalmitoyl phosphatidylcholine (DPPC) were investigated. The thermodynamic changes caused by (1) and its location into DPPC lipid bilayers were monitored by differential scanning calorimetry and Raman spectroscopy. The results reveal that (1) influences the thermotropic properties of DPPC lipid membrane causing abolition of the pretransition and broadening of the phase-transition profile and slightly decreases the Tm at increasing concentrations. The Raman height intensity ratios of the peaks I2935/2880, I2844/2880 and I1090/1130 are representative of the interaction of (1) with the alkyl chains and furnish information about the ratio between disorder and order that exists in the conformation of the alkyl chain. The intensity changes of the peak at 715 cm−1 indicates interaction between the choline head group and (1). The Raman spectroscopy results are in agreement with the thermal analysis results. Biologically active lipophilic molecules such as (1) should be studied in terms of their interaction with lipid bilayers prior to the development of advanced lipid carrier systems such as liposomes. The results of these studies provide information on the membrane integrity and physicochemical properties that are essential for the rational design lipidic drug delivery systems.  相似文献

Raman spectroscopic studies of four specimens from an important angel wall painting in need of conservation work in a medieval church have provided some information about the pigments and pigment compositions which will influence possible future preservation and restoration strategies. Excitation of the Raman spectra at 1,064 nm in macroscopic mode and at 785 nm in microscopic mode revealed that the white pigment on the angel's wings was a mixture of barytes with calcite and lead white in minor composition. Although the specimens provided were not directly associated with coloured regions of the painting, yellow and blue microcrystals were found and they were identified as chrome yellow and lazurite, respectively. Red and brown particles were identified as cinnabar/vermilion and haematite. Several green particles were also found but could not be identified. The green and blue crystals could be related to neighbouring coloured regions of the artwork and the yellow colour could be identified as a background to the angel figure. Particles of carbon were found to be dispersed throughout the specimens and can be ascribed to soot from candles, heating stoves or oil lamps providing lighting in the church. No evidence for biological deterioration was found from the spectra. The unusual pigment palette is strongly suggestive of a later date of painting than was originally believed but there is a possibility that an earlier rendition exists underneath. Following a review of the spectroscopic data, a more extensive sampling protocol is recommended, from which some stratigraphic evidence could identify the underlying plaster and possible artwork.  相似文献

Raman spectroscopy has been utilized to show the increase of single-walled carbon nanotubes (SWCNTs) content in commercial grade samples synthesized by the chemical vapour deposition (CVD) technique with a minimization of impurities using both hydrochloric acid treatment and surfactant purification. Surfactant purification methods proved to be the most effective, resulting in a three-fold increase in the percentage of SWCNTs present in the purified product as determined by Raman spectroscopy.  相似文献
