[structure: see text] Oligonucleotides with a novel, 2'-O-[2-[2-(N,N-dimethylamino)ethoxy]ethyl] (2'-O-DMAEOE) modification have been synthesized. This modification, a cationic analogue of the 2'-O-(2-methoxyethyl) (2'-O-MOE) modification, exhibits high binding affinity to target RNA (but not to DNA) and exceptional resistance to nuclease degradation. Analysis of the crystal structure of a self-complementary oligonucleotide containing a single 2'-O-DMAEOE modification explains the importance of charge factors and gauche effects on the observed antisense properties. 2'-O-DMAEOE modified oligonucleotides are ideal candidates for antisense drugs. 相似文献

[structure: see text] Oligonucleotides with a novel 2'-O-[2-(guanidinium)ethyl] (2'-O-GE) modification have been synthesized using a novel protecting group strategy for the guanidinium group. This modification enhances the binding affinity of oligonucleotides to RNA as well as duplex DNA (DeltaT(m) 3.2 degrees C per modification). The 2'-O-GE modified oligonucleotides exhibited exceptional resistance to nuclease degradation. The crystal structure of a palindromic duplex formed by a DNA oligonucleotide with a single 2'-O-GE modification was solved at 1.16 A resolution. 相似文献

Formation of 5-amino-3,4-di[(N,N-dimethylarnino)methylidenearnino]-2H-2-iminopyrroles 3 from the reaction of (Z)-N1-(2-amino-1,2-dicyanovinyl)-N2-substituted-formamidines 1 with dimethylformamide diethyl acetal has been shown to occur by initial formation of (Z)-N1-{l,2-dicyano-2-[N,N-dimethylamino)methylideneamino]vinyl}formamidines 8 (isolated), followed by base catalysed cyclisation and imi dazole ring opening by dimethyl amine. The kinetic product of the ring opening reaction is the 2,5-diimino2,5-dihydropyrrole derivatives 11, which have been isolated and characterized spectroscopically and by a single crystal X-ray analysis on the R = Ph derivative. In solution at room temperature the N-aryl derivatives undergo a rapid Dimroth rearrangement to give the thermodynamic ally more stable isomer 3 , but compound 11 (R = Me) is much more stable in solution. 相似文献

Synthesis of a series of 2′-O-[2-[(N,N-dialkylamino)oxy]ethyl]-modified 5-methyluridine nucleoside phosphoramidites and solid supports are described. Using these monomers, modified oligonucleotides containing phosphodiester linkages were synthesized in high yields. These modified oligonucleotides showed enhanced binding affinity to the complementary RNA (and not to DNA) and excellent nuclease stability with t1/2>24 h. The human serum albumin binding properties of modified oligonucleotides have been evaluated to assess their transport and toxicity properties. 相似文献

A synthesis is reported for N1-mono- and N1,N3-disubstituted uracil derivatives containing a terminal carbon-carbon double bond in the side-chain. Alkylation of vinyl 2-chloroethyl ether by uracil potassium salts leads to a mixture of 1-[2-(vinyloxy)ethyl] and 1,3-di[2-(vinyloxy)ethyl] derivatives while treatment of 2,4-bis(trimethylsilyloxy)pyrimidines by vinyl 2-chloroethyl ether leads exclusively to N1-monosubstituted products. Alkylation of cytosine by this chloroether gave 1-[2-(vinyloxy)ethyl]cytosine. Alkylation of 1-aryl-1H-1,2,4-triazol-3-ols with ethyl 2-bromopropionate under basic conditions resulted in the formation of 2-[(1-aryl-1H-1,2,4-triazol-3-yl)oxy]propionic acid, ethyl esters. No N-alkylated products were detected. Similar alkylation of 2-oxo-5-phenyl-1,3,4-thiazole and the corresponding 1,3,4-oxadiazole gave only N-alkylated derivatives. With 4-hydroxy-6-phenylpyrimidine and 2-oxo-4-phenylthiazole, both O- and N-alkylation occurred. Structure assignments were based on ir and 13C nmr spectral data. 相似文献

Regioselective synthesis of bis[(2-pyridylmethyl)oxy]tetrathiacalix[4]arenes was accomplished by a protection–deprotection method using benzyl groups as a protecting group. The conformational studies of distal-bis[(2-pyridylmethyl)oxy]thiacalix[4]arenes in solution and solid state are described. The two-phase solvent extraction data indicated that bis[(2-pyridylmethyl)oxy]tetrathiacalix[4]arenes show strong Ag+ (E%, 97%) affinity. In contrast, no significant E% is observed for K+. A good Job plots proves 1:1 coordination of 1,3-alternate
-3 with Ag+ cation. 1H-NMR Titration of 1,3-alternate-
3 with AgSO3CF3 also clearly demonstrates that a 1:1 complex is formed with retention of the original symmetry. The conformational changes of pyridine moiety from the original outward orientation of the ring nitrogen to the inside orientation toward the thiacalixarene cavity were observed in the process of Ag+ complexation. An attempted O-alkylation of the flexible macrocycle 1with 2-(chloromethyl)pyridine in the presence of Cs2CO3 under THF reflux afforded a mixture of twoconformers of tetra-O-alkylated product 4a in a ratio of 91:9 (cone-4a:1,2-alternate-4a)in 70% yield, while other possible isomers were not observed. In the case of Na2CO3, there was no reaction product,only the starting compound 1, whereasonly monoalkylated compound 3 was obtained when usingK2CO3 as the base. The distribution of cone conformer decreased in the case of O-alkylation of tetraol 1with 4-(chloromethyl)pyridine or benzyl bromide in the presence of Cs2CO3 in comparison with that ofO-alkylation with 2-(chloromethyl)pyridine, while the 1,2-alternate conformer increased in the same sequence. The larger Cs+might increase the contribution of the metal template effect, which can hold the 2-pyridylmethyl group(s) and theoxide group(s) on the same side of the tetrathiacalix[4]arene 1 through the cation-O- and -N-interaction in the caseof O-alkylation of tetraol 1 with 2-(chloromethyl)pyridine.Only when the template metal can hold the 2-pyridyl group(s) andthe oxide group(s) on the same side of the tetrathiacalix[4]arene is the conformation immobilized to thecone. The template effect of the cesium cation plays an important role in this alkylation reaction. The structuralcharacterization of these products is also discussed.The two-phase solvent extraction data indicated thattetrakis[(2-pyridylmethyl)oxy]thiacalix[4]arenes 4a show strong Ag+ affinity and a high Ag+ selectivity wasobserved for cone-4a. A good Job plot proves 1:1 coordination of cone-4a with Ag+ cation.1H-NMR titration of cone-4a with AgSO3CF3 also clearly demonstrates that a 1:1complex is formed with retention of the original symmetry. Reaction of 2-[(dimethylamino)methyl]aniline with butyllithium, followed by conversion with trimethylsilyl, triphenylsilyl, triphenylgermyl, trimethylstannyl, or tri-n-butylstannyl chloride, gives the corresponding substituted aniline. These compounds were further deprotonated by butyllithium and reacted with germanium, tin, and lead dichlorides, respectively, in both stoichiometric ratios 2:1 and 1:1, providing the target homo- ([2-(Me(2)NCH(2))C(6)H(4)(YR(3))N](2)M) and heteroleptic ([2-(Me(2)NCH(2))C(6)H(4)(YR(3))N]MCl) germylenes and stannylenes, where M = Ge, Sn, Y = Si, Ge, and R = Me, Ph. Unlike all of these cases, the heteroleptic plumbylene can only be obtained with this reaction when the amide is substituted by a trimethylsilyl moiety. Anilines substituted by trimethyltin or tri-n-butyltin moieties gave transmetalation products after the second deprotonation by butyllithium. The trimethyltin-substituted stannylenes could likewise not be obtained by hexamethyldisilazane elimination of (trimethylstannyl)-2-[(dimethylamino)methyl]aniline with 0.5 mol equiv of either bis[bis(trimethylsilyl)amido]tin or {bis[bis(trimethylsilyl)amido]tin chloride}. Products of these reactions are heterocubanes with compositions {[2-(Me(2)NCH(2))C(6)H(4)N]Sn}(4) and [2-(Me(2)NCH(2))C(6)H(4)N](2)(μ(2)-SnMe(2))(2), respectively, and Me(4)Sn or Me(3)SnCl. The structures of trimethylsilyl- and triphenylgermyl-substituted germylenes, stannylenes, and plumbylenes, as well as a number of their precursors, in the crystalline state, were investigated by X-ray diffraction and NMR spectroscopy in solution. To develop oligonucleotides containing new 2'-O-modified ribonucleosides as nucleic acid drugs, we synthesized three types of ribonucleoside derivatives modified at the 2'-hydroxyl group with 2-(methoxycarbonyl)ethyl (MOCE), 2-(N-methylcarbamoyl)ethyl (MCE), and 2-(N,N-dimethylcarbamoyl)ethyl (DMCE) groups, as key intermediates, via the oxa-Michael reaction of the appropriately protected ribonucleoside (U, C, A, and G) derivatives. Among them, the 2'-O-MCE ribonucleosides were found to be the most stable under basic conditions. To study the effects of the 2'-O-modification on the nuclease resistance of oligonucleotides incorporating the 2'-O-modified ribonucleosides and their hybridization affinities for the complementary RNA and DNA strands, 2'-O-MCE-ribonucleoside phosphoramidite derivatives were successfully synthesized and subjected to the synthesis of 2'-O-MCE-oligonucleotides and 2'-O-methyl-oligonucleotides incorporating 2'-O-MCE-ribonucleosides. The 2'-O-MCE-oligonucleotides and chimeric oligomers with 2'-O-MCE and 2'-O-methyl groups thus obtained demonstrated complementary RNA strands and much higher nuclease resistances than the corresponding 2'-O-methylated species. Finally, we incorporated the 2'-O-MCE-ribonucleosides into antisense 2'-O-methyl-oligoribonucleotides to examine their exon-skipping activities in splicing reactions related to pre-mRNA of mouse dystrophin. The exon-skipping assay of these 2'-O-methyl-oligonucleotide incorporating 2'-O-MCE-uridines showed better efficacies than the corresponding 2'-O-methylated oligoribonucleotide phosphorothioate derivatives. 相似文献
