The aim of this study was to explore the capability of spectroscopy in the visible (Vis) and short wavelength near-infrared (NIR) regions for the non-destructive measurement of wine composition in intact bottles. In this study we analysed a wide range of commercial wines obtained in Australia in different types of bottles (e.g. colours, diameters and heights), including different wine styles and varieties. Wine bottles were scanned in the Vis-NIR region (600–1,100 nm) in a monochromator instrument in transflectance mode. Principal component analysis (PCA) and partial least-squares (PLS) regression were used to interpret the spectra and develop calibrations for wine composition. Due to the relatively small number of samples available full cross-validation (leave-one-out) was used as validation. The coefficient of correlation in calibration and the standard error of cross-validation (SECV) were 0.67 (SECV: 0.48%), 0.83 (SECV: 4.01 mg L−1), 0.70 (SECV: 28.6 mg L−1) and 0.50 (SECV: 0.15) for alcohol content, total SO2, free SO2 and pH, respectively, in the set of wine samples analysed. These preliminary results showed that the assessment of wine composition by Vis and short wavelengths in the NIR is possible for either qualitative analysis (e.g. low-, medium- and high-quality grading), or for screening of composition during bottling and storage. Although low accuracy and precision were obtained for the chemical parameters routinely analysed in wine, calibration models for the chemical parameters were considered acceptable for screening purposes in terms of the standard errors obtained.  相似文献

Summary A new 80 μm Carboxen-polydimethylsiloxane (PDMS) fiber for solid phase microextraction (SPME) was tested for the enrichment of volatile organic compounds from water and air. Detection limits between 13 ng L−1 (CH2Cl2) and 0.1 ng L−1 (CHCl2Br and CHClBr2) for the combination: Carboxen-PDMS fiber and GC-ECD and between 35 ng L−1 and 45 ng L−1 (BTEX compounds) for the combination: Carboxen-PDMS and GC-FID using the headspace procedure were determined. Comparisons with the 100 μm PDMS fiber and further coatings show the advantages of the Carboxen-PDMS fiber with respect to extraction efficiency. Disadvantages of the new fiber compared with the 100 μm PDMS fiber are poorer repeatability and prolongation of equilibrium time. Distribution coefficients of the BTEX compounds between aqueous solution and SPME fiber coating were calculated and compared with the results of other researchers and with octanol-water partition coefficients.  相似文献

A rapid, accurate, and precise method is described for the determination of Pb in wine using continuous-flow hydride generation atomic fluorescence spectrometry (CF-HGAFS). Sample pretreatment consists of ten-fold dilution of wine followed by direct plumbane generation in the presence of 0.1 mol L−1 HCl and 1% m/v K3[Fe(CN)6] with 1% m/v NaBH4 as reducing agent. An aqueous standard calibration curve is recommended for Pb quantification in wine sample. The method provides a limit of detection and a limit of quantification of 0.3 μg L−1 and 1 μg L−1, respectively. The relative standard deviation varies between 2–6% (within-run) and 4–11% (between-run) at 3–30 μg L−1 Pb levels in wine. Good agreement has been demonstrated between results obtained by CF-HGAFS and direct electrothermal atomic absorption spectrometry in analyses of red and white wines within the concentration range of 9.2–25.8 μg L−1 Pb.  相似文献

A headspace solid-phase microextraction gas-chromatographic (HS-SPME-GC) procedure was used to determine the composition of the volatile fraction of white wine samples from several Spanish certified brands of origin (CBO). The compounds present were previously identified by gas chromatography−mass spectrometry (GC−MS) and quantitative determinations were carried out by GC-FID. Four CBO, Rueda, Ribeiro, Penedés, and Condado de Huelva, were studied. Rueda wines present the highest concentrations of ethyl acetate (55.86−125.27 μg mL−1), isoamyl acetate (0.91−6.72 μg mL−1), hexyl acetate (0.09−0.81 μg g mL−1), and 2-phenethyl acetate (0.14−0.66 μg mL−1). Compounds such as ethyl hexanoate (0.88−2.15 μg mL−1) and ethyl decanoate (0.29−0.96 μg mL−1) appeared in higher concentration in Ribeiro, Rueda, and Penedés samples. According to the results obtained and by applying pattern-recognition procedures differentiation of the considered CBO was attained. Principal-component analysis (PCA), linear discriminant analysis (LDA), and multilayer perceptrons neural networks (MLP-NN) were used as chemometric tools for pattern-recognition studies. Summary In this study a high-performance liquid chromatography method was developed for the determination of procyanidin dimers B1, B2, B3, B4 and procyanidin trimers C1 and T2 in white wine using fluorescence detection. This method allowed analysis of white wine with no prior treatment. Limits of quantification were form 12 μg.L−1 to 16 μg.L−1. Reproductibility data for replicate analysis was measured and the CV was less than 4.5%. This method was used for the determination of these compounds in six French white wines.  相似文献

A fed-batch culture system with constant feeding (glucose 80 g L−1, 0.25 ml min−1) was used to study the influence of glucose on cell dry weight and exopolysaccharides production from submerged Tremella fuciformis spores in a 5-L stirred-tank bioreactor. The results showed that high levels of cell mass (9.80 g L−1) and exopolysaccharides production (3.12 g L−1) in fed-batch fermentation were obtained after 1 h of feeding, where the specific growth rate (μ) and exopolysaccharides yield on substrate consumed (YP/S) were 0.267 d−1 and 0.14 g g−1. Unlike batch fermentation, maximal cell mass and exopolysaccharides production merely reached 7.11 and 2.08 g L−1; the specific growth rate (μ) and exopolysaccharides yield on substrate consumed (YP/S) were 0.194 d−1 and 0.093 g g−1, respectively. It is concluded that the synthesis of exopolysaccharides can be promoted effectively when feeding glucose at a late exponential phase.  相似文献

By alternate use of two different extraction solvents, n-hexadecane and benzyl alcohol, a headspace-solvent microextraction (HSME)–GC–FID/MS method has been established for characterization of the volatile components of an orange juice beverage. The method avoids two disadvantages of conventional HSME—difficulty identifying components obscured by the solvent peak and inefficient extraction of some of the compounds if one solvent only is used. The optimum conditions (droplet volume, equilibration temperature and time, extraction time, and ionic strength) were determined by the factor-rotation method. The volatile components of the beverage were mainly terpenes, terpenols, fatty acids, and alcohols, for example limonene (114.71 mg L−1), phellandrene (4.50 mg L−1), terpineol (p-menth-1-en-8-ol; 3.12 mg L−1), α-pinene (0.33 mg L−1), n-hexadecanoic acid (0.28 mg L−1), and terpinol (0.13 mg L−1).  相似文献

A novel method employing high-performance cation chromatography in combination with inductively coupled plasma dynamic reaction cell mass spectrometry (ICP–DRC–MS) for the simultaneous determination of the herbicide glyphosate (N-phosphonomethylglycine) and its main metabolite aminomethyl phosphonic acid (AMPA) is presented. P was measured as 31P16O+ using oxygen as reaction gas. For monitoring the stringent target value of 0.1 μg L−1 for glyphosate, applicable for drinking and surface water within the EU, a two-step enrichment procedure employing Chelex 100 and AG1-X8 resins was applied prior to HPIC–ICP–MS analysis. The presented approach was validated for surface water, revealing concentrations of 0.67 μg L−1 glyphosate and 2.8 μg L−1 AMPA in selected Austrian river water samples. Moreover, investigations at three waste water-treatment plants showed that elimination of the compounds at the present concentration levels was not straightforward. On the contrary, all investigated plant effluents showed significant amounts of both compounds. Concentration levels ranged from 0.5–2 μg L−1 and 4–14 μg L−1 for glyphosate and AMPA, respectively.  相似文献

The sensitivity and precision of headspace solid-phase micro extraction (HS-SPME) at an analyte solution temperature (T as) of +35 °C and a fiber temperature (T fiber) of +5 °C were compared with those for HS-SPME at T as and T fiber of −20 °C for analysis of the volatile organic compounds benzene, 1,1,1-trichloroethane, trichloroethylene, toluene, o-xylene, ethylbenzene, m/p-xylene, and tetrachloroethylene in water samples. The effect of simultaneous fiber cooling and analyte solution freezing during extraction was studied. The compounds are of different hydrophobicity, with octanol/water partition coefficients (Kow) ranging from 126 and 2511. During a first set of experiments the polydimethylsiloxane (PDMS) SPME fiber was cooled to +5 °C with simultaneous heating of the aqueous analyte solution to +35 °C. During a second set of experiments, both SPME fiber holder and samples were placed in a deep freezer maintained at −20 °C for a total extraction time of 30 min. After approximately 2 min the analyte solution in the vial began to freeze from the side inwards and from the bottom upwards. After approximately 30 min the solution was completely frozen. Analysis of VOC was performed by coupling HS-SPME to gas chromatography-mass spectrometry (GC-MS). In general, i.e. except for tetrachloroethylene, the sensitivity of HS-SPME increased with increasing compound hydrophobicity at both analyte solution and fiber temperatures. At T as of +35 °C and T fiber of +5 °C detection limits of HS-SPME were 0.5 μg L−1 for benzene, 1,1,1-trichloroethane, trichloroethylene, and tetrachloroethylene, 0.125 μg L−1 for toluene, and 0.025 μg L−1 for ethylbenzene, m/p-xylene, and o-xylene. In the experiments with T as and T fiber of −20 °C, detection limits were reduced for compounds of low hydrophobicity (Kow<501), for example benzene, toluene, 1,1,1-trichloroethane, and trichloroethylene. In the concentration range 0.5–62.5 μg L−1, the sensitivity of HS-SPME was enhanced by a factor of approximately two for all compounds by performing the extraction at −20 °C. A possible explanation is that freezing of the water sample results in higher concentration of the target compounds in the residual liquid phase and gas phase (freezing-out), combined with enhanced adsorption of the compounds by the cooled fiber. The precision of HS-SPME, expressed as the relative standard deviation and the linearity of the regression lines, is increased for more hydrophobic compounds (Kow>501) by simultaneous direct fiber cooling and freezing of analyte solution. Background contamination during analysis is reduced significantly by avoiding the use of organic solvents.  相似文献

The parameters of analytical procedures developed for direct ETAAS determination of Pb in wine are discussed. Atomic absorption spectrometers based on transversal and longitudinal Zeeman effect, wall and integrated platform atomization with two main approaches: (i) measurements in the presence of modifier and (ii) measurements without using any modifier are compared. The optimal temperature programs are defined according to the pre-treatment and atomization curves constructed in the presence of different types of wines. For all investigated instrumental systems, 1:1 dilution of wine sample with 0.2 mol L−1 HNO3 is recommended. Matrix interferences observed, call for standard addition calibration method for Pb quantification in wines. The detection limit (3σ) achieved for wine diluted in the ratio of 1:1 varied from 0.8 to 1.9 μg L−1 depending on the instrument used. The relative standard deviation for the concentration range of 10 to 80 μg L−1 Pb in wine is typically between 4–8%. The accuracy of the analytical procedures recommended was confirmed by comparing the results obtained with those found for wine samples previously digested with HNO3-H2O2 mixture, by added/found method and by parallel analysis using different instruments. A total of 66 wine samples from different regions of Macedonia were analyzed.   相似文献

A reversed-phase liquid chromatographic method, optimised for the separation of trans-, and cis-resveratrol, catechin, epicatechin, quercetin and rutin, is reported. Separation was achieved using a C18 column and a gradient elution with acetonitrile and 5% formic acid aqueous solution. The analyses required an equilibration period of 10 min and a run time of 25 min for completion. Identification was based on retention characteristics and by relative UV spectra, obtained by photodiode array detector and were compared with commercial standards. Analyses were performed without any sample pre-treatment. Detection was carried out by UV–Vis detector at three different wavelengths. The detection limit ranged from 0.16 μgm L−1 (cis-resveratrol) to 1.5 μgm L−1 (+)-catechin. Investigation was extended to quantitative determination of phenol compounds in Italian red wine and to investigate the stability of the six antioxidants.  相似文献

A rapid and inexpensive method for simultaneous quantification of terbumeton (TER), and its major potential metabolites (TED; terbumeton-desethyl, TOH; terbumeton-2-hydroxy and TID; terbumeton-deisopropyl) in soil bulk water (SBW) samples is proposed. The analytical method involves extraction–concentration from SBW samples using a graphitized carbon black (GCB) cartridge followed by their separation–detection by reversed-phase high-performance liquid chromatography analysis using a C18 column and a diode array detector. A mobile phase of acetonitrile−0.005 mol L−1 phosphate buffer (pH 7.0) (35:65, v/v) at a flow rate of 0.8 mL min−1 in isocratic elution mode has been used. After optimization of the extraction and separation conditions, this method can be used for the simultaneous determination of investigated compounds in the range of the international limits of 0.1 μg L−1. For TER the detection limit was 0.009 μg L−1 and it was 0.100, 0.550, and 0.480 μg L−1 for TED, TOH, and TID, respectively. The recoveries of TER, TED, TOH, and TID from SBW samples, measured at three levels of concentration range, were found to be between 48.0 and 102.0%. The intra-day precision measured by relative standard deviation (RSD) was always lower than 9.0%.  相似文献

A survey of contamination of surface and drinking waters around Lake Maggiore in Northern Italy with polar anthropogenic environmental pollutants has been conducted. The target analytes were polar herbicides, pharmaceuticals (including antibiotics), steroid estrogens, perfluorooctanesulfonate (PFOS), perfluoroalkyl carboxylates (including perfluorooctanoate PFOA), nonylphenol and its carboxylates and ethoxylates (NPEO surfactants), and triclosan, a bactericide used in personal-care products. Analysis of water samples was performed by solid-phase extraction (SPE) then liquid chromatography–triple-quadrupole (tandem) mass spectrometry (LC–MS–MS). By extraction of 1-L water samples and concentration of the extract to 100 μL, method detection limits (MDLs) as low as 0.05–0.1 ng L−1 were achieved for most compounds. Lake-water samples from seven different locations in the Southern part of Lake Maggiore and eleven samples from different tributary rivers and creeks were investigated. Rain water was also analyzed to investigate atmospheric input of the contaminants. Compounds regularly detected at very low concentrations in the lake water included: caffeine (max. concentration 124 ng L−1), the herbicides terbutylazine (7 ng L−1), atrazine (5 ng L−1), simazine (16 ng L−1), diuron (11 ng L−1), and atrazine-desethyl (11 ng L−1), the pharmaceuticals carbamazepine (9 ng L−1), sulfamethoxazole (10 ng L−1), gemfibrozil (1.7 ng L−1), and benzafibrate (1.2 ng L−1), the surfactant metabolite nonylphenol (15 ng L−1), its carboxylates (NPE1C 120 ng L−1, NPE2C 7 ng L−1, NPE3C 15 ng L−1) and ethoxylates (NPE n Os, n = 3-17; 300 ng L−1), perfluorinated surfactants (PFOS 9 ng L−1, PFOA 3 ng L−1), and estrone (0.4 ng L−1). Levels of these compounds in drinking water produced from Lake Maggiore were almost identical with those found in the lake itself, revealing the poor performance of sand filtration and chlorination applied by the local waterworks.  相似文献

Near infrared (NIR) spectroscopy was used to simultaneously predict the concentrations of malvidin-3-glucoside (M3G), pigmented polymers (PP) and tannins (T) in red wine. A total of 495 samples from 32 commercial scale red wine fermentations over two vintages using two grape varieties (Cabernet Sauvignon and Shiraz), and also including as additional variables two types of fermenters, two different yeasts, and three fermentation temperatures were used. Samples were scanned in transmission mode (400-2500 nm) using a monochromator instrument (NIRSystems6500). Calibration equations were developed from high performance liquid chromatography (HPLC) and NIR data using partial least squares (PLS) regression with internal cross validation. Using PLS regression, very good calibration statistics (Rcal2>0.80) were obtained for the prediction of M3G, PP and T with standard deviation (S.D.)/standard error in cross validation (SECV) ratio (residual predictive deviation, RPD)) ranging from 1.8 to 5.8. It was concluded that near infrared spectroscopy could be used as rapid alternative method for the prediction of the concentration of phenolic compounds in red wine fermentations.  相似文献

In this work, the hyphenation of the multisyringe flow injection analysis technique with a 100-cm-long pathlength liquid core waveguide has been accomplished. The Cl/Hg(SCN)2/Fe3+ reaction system for the spectrophotometric determination of chloride (Cl) in waters was used as chemical model. As a result, this classic analytical methodology has been improved, minimizing dramatically the consumption of reagents, in particular, that of the highly biotoxic chemical Hg(SCN)2. The proposed method features a linear dynamic range composed of two steps between (1) 0.2–2 and (2) 2–8 mg Cl L−1, thus extended applicability due to on-line sample dilution (up to 400 mg Cl L−1). It also presents improved limits of detection and quantification of 0.06 and 0.20 mg Cl L−1, respectively. The coefficient of variation and the injection throughput were 1.3% (n = 10, 2 mg Cl L−1) and 21 h−1. Furthermore, a very low consumption of reagents per Cl determination of 0.2 μg Hg(II) and 28 μg Fe3+ has been achieved. The method was successfully applied to the determination of Cl in different types of water samples. Finally, the proposed system is critically compared from a green analytical chemistry point of view against other flow systems for the same purpose.  相似文献

A simple micellar liquid chromatographic technique for deltamethrin determination was developed and validated. The method provided to be suitable for deltamethrin determination in pediculicide shampoo. Kromasil C18 column (150 mm×4.6 mm, 5 μm) and mobile phase −0.12 M sodium dodecyl sulfate with 9% (v/v) 1-butanol were used for deltamethrin separation. Detection wavelength was 265 nm. The retention time was about 15 min. Different validation parameters were evaluated. The specificity of the method was demonstrated. Linearity was established in the range 10–40 μg L−1. The limits of detection and quantitation were 1.06 and 3.22 μg mL−1, respectively. The method showed excellent accuracy (100.6%) and precision (repeatability) gave a relative standard deviation of less than 1%. The influence of the various method parameters (robustness study) was also studied.  相似文献

Contamination by Brettanomyces is a frequent problem in many wineries that has a dramatic effect on wine aroma and hence its quality. The yeast Brettanomyces/Dekkera is involved in the formation of three important volatile ethylphenols—4-ethylphenol, 4-ethylguaiacol and 4-ethylcatechol—that transmit an unpleasant aroma to wine that has often been described as ‘medicinal’, ‘stable’ or ‘leather’. This study proposes an in situ derivatisation and headspace solid-phase microextraction– gas chromatography coupled to mass spectrometry method to determine the three ethylphenols in red Brettanomyces-tainted wines. The most important variables involved in the derivatisation (acetic anhydride and base concentration) and the extraction (extraction temperature and salt addition) processes were optimised by experimental design. The optimal conditions using 4 mL of wine in 20-mL sealed vials were 35 μL of acetic anhydride per millilitre of wine, 1 mL of 5.5% potassium carbonate solution and 0.9 g of sodium chloride and the extraction was performed with a divinylbenzene–carboxen–poly(dimethylsiloxane) fibre at 70 °C for 70 min. Then, the performance characteristics were established using wine samples spiked with the ethylphenols. For all compounds, the detection limits were below the odour threshold reported in the literature and they were between 2 and 17 μg L−1 for 4-ethylguaiacol and 4-ethylphenol, respectively. Intermediate precision (as relative standard deviation) was acceptable, with values ranging from 0.3 to 12.1%. Finally, the method was applied in the analysis of aged Brettanomyces-tainted wines.  相似文献

A fully automated method using direct immersion solid-phase microextraction (DI-SPME) and headspace on-fiber silylation for simultaneous determinations of exogenous endocrine-disrupting chemicals (EDCs) and endogenous steroid hormones in environmental aqueous and biological samples by gas chromatography–mass spectrometry (GC-MS) was developed and compared to a previously reported manual method. Three EDCs and five endocrine steroid hormones were selected to evaluate this method. The extraction and derivatization time, ion strength, pH, incubation temperature, sample volume, and extraction solvent were optimized. Satisfactory results in pure water were obtained in terms of linearity of calibration curve (R 2=0.9932–1.0000), dynamic range (3 orders of magnitude), precision (4–9% RSD), as well as LOD (0.001–0.124 μg L−1) and LOQ (0.004–0.413 μg L−1), respectively. These results were similar to those obtained using a manual method, and moreover, the precision was improved. This new automated method has been applied to the determinations of target compounds in real samples used in our previous study on a manual SPME method. Exogenous octylphenol (OP), technical grade nonylphenol (t-NP), and diethylstilbestrol (DES) were at 0.13, 5.03, and 0.02 μg L−1 in river water and 3.76, 13.25, and 0.10 μg L−1 in fish serum, respectively. Natural steroid hormones estrone (E1), 17β-estradiol (E2), and testosterone (T) were at 0.19, 0.11, and 6.22 μg L−1 in river water; and in female fish serum E1, E2, and pregnenolone (PREG) were at 1.37, 1.95, and 6.25 μg L−1, respectively. These results were confirmed by the manual method. The developed fully automated SPME and on-fiber silylation procedures showed satisfactory applications in environmental analysis and the performances show improved precision and a reduced analysis time compared to the manual method.  相似文献

Multiple headspace solid-phase microextraction (MHS-SPME) with gas chromatography–mass spectrometry is proposed for quantification of nine volatile oak compounds in aged wines. These compounds are formed and extracted by wine when it is matured in oak barrels and are responsible for particular organoleptic properties and the high quality of these wines. Some important variables of the extraction process, for example volume of sample and extraction time, were studied. Extraction of 50 μL wine was performed with a divinylbenzene–Carboxen–polydimethylsiloxane fibre at 55 °C for 60 min. For calibration the same conditions were used, except that the wine was substituted by 50 μL of a standard solution in synthetic wine. The linearity, detection limits, and repeatability of the method were determined by use of standard solutions in synthetic wine. Detection limits were between 0.01 and 10 μg L−1 (for eugenol and furfural, respectively) and repeatability, expressed as relative standard deviation, was from 2 to 6%. The method was used to analyse six red wines and the concentrations obtained were statistically compared with those obtained by the standard addition method for the same wines.  相似文献
