One hundred samples of commercial bread purchased from January to October (2006) from retail bakeshops in five different cities (Rabat, Témara, Salé, Casablanca and Meknès) in Morocco were surveyed for the presence of ochratoxin A (OTA) using liquid chromatography coupled to fluorescence detection. The identification of OTA in positive bread samples was confirmed by methyl ester derivatization. Analytical results showed that forty eight (48%) samples were positive with OTA greater than the limit of quantification (LOQ = 0.051 ng/g). Levels of OTA in positive samples ranged between 0.14 and 149 ng/g. The average contamination of bread samples with OTA was 13 ± 1.5 ng/g. The highest frequency of positive samples (61.5%) and the most contaminated sample (149 ng/g) were found in bread commercialized in the Casablanca area. Twenty six of the positive samples exceeded the maximum level of 3 ng/g set by European regulations for OTA in cereals and derivatives. Based in the results presented in this study, the estimated daily intake of OTA from bread was 126 ng/kg bw/day. The present paper is the first ever drafted on the natural occurrence of OTA in bread consumed in Morocco. Data on the daily intake of OTA by the Moroccan population are also estimated for the first time.  相似文献

A total of 95 wine samples including 34 white, 10 rosé and 51 red wines originating from four different Turkish areas were analysed for ochratoxin A (OTA). An analytical method based on immunoaffinity column (IAC) for clean-up and high performance liquid chromatography with fluorescence detection (HPLC-FD) was used to determine OTA in wines. The limit of detection (LOD) was estimated as 0.006 ng ml− 1 for white wine and 0.010 ng ml− 1 for rosé and red wines. The limit of quantification (LOQ) was estimated as 0.020 ng ml− 1 in white wine and 0.030 ng ml− 1 in rosé and red wines. Recovery experiments were carried out with spiked samples in the range 0.1–1 ng ml− 1 of OTA. The average OTA recoveries from spiked white wine samples varied from 79.43% to 85.07%; while the mean recoveries for rosé and red wine samples were in the range of 77.48–83.96% and 76.61–83.55%, respectively. OTA was detected in 82 (86%) wine samples at levels of < 0.006–0.815 ng ml− 1, which were below the maximum allowable limit established by the European Community. The mean OTA concentration in red wines was slightly higher than in white and rosé wines. Furthermore, our data indicate that the geographic region of origin has strong influence on OTA level for white, rosé and red wines: wines originating from Thrace (n = 44, mean = 0.158 ng ml− 1) and Aegean (n = 28, mean = 0.060 ng ml− 1) regions of Turkey were more contaminated with OTA compared with wines originating from central (n = 15, mean = 0.027 ng ml−1) and east Anatolia (n = 8, mean = 0.027 ng ml− 1) areas. This study showed that the occurrence of OTA in Turkish wines is high, but at levels that probably leads to a non-significant human exposure to OTA by consumption of wines.  相似文献

Ochratoxin A (OTA) is a mycotoxin produced by fungi such as Penicillium verrucosum and Aspergillium spp. and has been found to have a variety of potentially deadly toxic effects. The favoured substrate for fungal growth and OTA production appears to be cereals and flour-based products, including bread. Due to the dietary relevance of bread for the Portuguese population, it is imperative that its OTA content remains well within safe quantities. As such, bread samples collected from commercial surfaces across the Algarve region and from the city of Bragança during the winter of 2007 were tested for OTA through extraction with immunoaffinity columns and quantification by liquid chromatography coupled with fluorescence detection. Although OTA content was found to be above the limit of quantification in approximately 60% and 50% of the analysed samples from Algarve and Bragança, respectively, all samples were found to be compliant with European Commission. OTA content reached maximums of 0.49 ng/g in Algarve and 0.43 ng/g in Bragança, and was thus below the maximum limit established by European legislation for bread of 3 ng/g. The results of the present study put the estimated daily intake of OTA from bread at approximately 0.26 ng/kg bw/day in Algarve and 0.38 ng/kg bw/day in Bragança, circa 1.5% and 2.0% of the TDI established by either the EFSA or the FAO/WHO, or over 4.5% and 6.5% if we consider the FAO/WHO advised bread consumption of 250 g/day. These results seem to suggest that, in these two Portuguese regions, OTA contamination is well under control and unlikely to represent a threat to consumer health.  相似文献

A total of 267 wine samples including 19 dessert, 186 red, 11 rosé and 51 white produced mostly in the years 1997–2002 in Italian and Hungarian regions were analyzed for ochratoxin A (OTA) using inmunoaffinity column (IAC) clean-up and HPLC with fluorimetric detection. None of Hungarian wine samples were contaminated with this mycotoxin. For Italian red wines, 84% of the samples were positive for OTA ranged from 0.01 to 4.00 ng/mL. Furthermore, OTA was detected in 63% of dessert, in 56% of rosé and in 19% of white wine samples ranged from 0.01 to 1.64, from 0.01 to 1.04 and from 0.01 to 0.21 ng/mL, respectively. A study of OTA daily exposure assessment in Italian wines was also carried out outlining a quite low contribution to the overall daily intake.  相似文献

Wine and grape juices were identified as one of the most important sources of ochratoxin A (OTA), a mycotoxin with diverse toxic effects that naturally appears in food and foodstuffs all over the world.The aim of this study was to assess the OTA levels in Portuguese wines through the application of a simple and accurate method based on liquid chromatography (LC) with direct injection, followed by fluorescence detection (FD).Randomly selected wine samples were used to evaluate the performance of direct injection as efficient, fast, inexpensive and safe sample preparation method. The proposed method was successfully validated. The limit of quantification (LOQ) was 1.0 μg/L and OTA recoveries from wine samples, spiked at the three fortification levels, were higher than 85.4%, with RSDs lower than 9.6% for both red and white wines. The presence of OTA was confirmed by methyl ester derivatization followed by LC analysis.Data on OTA levels were obtained for 60 Portuguese red and white wine samples. OTA was found in 12 samples, nine (26%) red wine samples and three (12%) white wine samples. Only one red wine sample and one white wine sample presented a contamination level above the LOQ, with 1.23 and 2.4 μg/L, respectively. It should be pointed out that this white wine sample exceeded the EC maximum permitted level of 2.0 μg/L. The safe dose established as 120 ng/kg body weight/week was not exceeded by the weekly intake estimated for the samples contaminated above the LOQ.  相似文献

Ochratoxin A (OTA) is a mycotoxin produced by fungi species belonging to the genera Aspergillus and Penicillium being isolated in alcoholic beverages. The aim of this work is developed and applied a procedure for the analysis of OTA in wines. An analytical method based on immunoaffinity column (IAC) for clean-up, liquid chromatography with fluorescence detection (LC-FD), and LC-FD after of OTA methylation was used to determine the occurrence of OTA in wines. Recoveries of this mycotoxin spiked to red wines at 0.5 ng/ml level were >90% with an average of relative standards deviations of 4%. Furthermore, 116 wine samples from designation of origin (DO) and three samples from food stores of Valencian Community (Spain) were examined for the occurrence of OTA being the levels of this mycotoxin ranged from <0.01 to 0.76 ng/ml. Finally, the estimated daily intake of OTA in this study was 0.15 ng/kg bw per day.  相似文献

A sensitive and accurate analytical method for the determination of ochratoxin A (OTA) in rice, based on extraction with phosphate-buffered saline/methanol, an immunoaffinity column (IAC) for clean-up, and high performance liquid chromatography with fluorescence detection (HPLC-FD), is described. The limit of quantification of the proposed method was 0.05 g kg–1. Recovery of OTA from rice samples spiked at 0.05 g kg–1 was 92%, with a within-day RSD of 5.4%. The proposed method was applied to 42 rice samples from Portugal and the presence of OTA was found in six samples at concentrations ranging from 0.09 to 3.52 g kg–1. The identification of OTA was confirmed by methyl ester derivatization and then HPLC analysis. The daily intake of OTA by the Portuguese population was also estimated.  相似文献

A fluorescence polarization (FP) immunoassay, based on a monoclonal antibody and an ochratoxin A (OTA)-fluorescein tracer, has been developed for rapid screening of OTA in red wine. Wine samples were diluted with methanol and passed through aminopropyl solid-phase extraction columns prior to the FP assay. Average recoveries from samples spiked with OTA at levels of 2.0 and 5.0 ng/mL were 79% with RDS of 11% (n = 6). The limit of detection of the FP immunoassay was 0.7 ng/mL OTA, and the whole analysis was performed in less than 10 min. The assay was tested on 154 red wine samples (naturally contaminated or spiked at level ranging from 0.1 to 5.0 ng/mL) and compared with an high-performance liquid chromatography/immunoaffinity column clean-up method, showing a good correlation (r = 0.9222). Their compliance with the European regulation (2.0 ng/mL OTA maximum permitted level) was correctly assessed for 70% of the analyzed samples of red wine, whereas confirmatory analyses were required for the remaining ones with OTA levels close to the regulatory limit. No false-negative or positive results were observed using the FP immunoassay. The proposed FP assay is a useful screening method for OTA in red wines, when high throughput is required, that could also be used for white and rosé wines, which are known to contain less interfering compounds such as polyphenols.  相似文献

The redox chemical behavior of americium and plutonium species in alkaline media has been studied, mainly two dis/reproportionation reactions: 2 Pu(VI) = Pu(V) + Pu(VII) and Am(III) + Am(V) = 2Am(IV). It was found that Am(VI) is readily reduced by the simple action of water, and that radiolysis has no effect. This is a marked departure from the behavior of Am in other media, where alpha particle self-irradiation generates redox active species that affect the Am oxidation state. Étude du comportement de Am et Pu en milieu alcalin. Le comportement redox de Am et Pu a été étudié en milieu alcalin, notamment les dismutations 2 Pu(VI) = Pu(V) + Pu(VII) et 2 Am(IV) = Am(III) + Am(V). Am(VI) est rapidement réduit par l’eau et la radiolyse n’a pas d’effet, contrairement à ce qui se produit dans d’autres milieux. Des indications ont été obtenues sur un composé de Am(VII). Ochratoxin A (OTA) is a secondary fungal metabolite produced by several moulds, mainly by Aspergillus ochraceus and by Penicillium verrucosum, that occurs in meat products. The aim of this work was to optimize an efficient extraction procedure for the determination of OTA in muscle tissue in order to assess its occurrence in meat samples. Three different apparatus, a Waring blender, a switching apparatus, and an ultrasonic processor, were evaluated to verify the efficiency of extraction. The analytical methods proposed involve the extraction with chloroform-orthophosphoric acid, cleanup through an immunoaffinity column, high-performance liquid chromatography/fluorescence detection for separation and identification of OTA, and confirmation with liquid chromatography/FD after methylation of OTA in muscle tissue. The limit of quantification of the proposed method was 0.04 μg kg−1. Recoveries of OTA, using switching apparatus, ranged from 90.3 to 103.2% for chicken muscle spiked at 2.4 and 0.48 μg kg−1, respectively, with a within-day relative standard deviation of 17 and 15.3%. The proposed method was applied to 38 chicken, swine, and turkey muscle samples and the presence of OTA was confirmed in five samples. Finally, the estimated daily intake of OTA in this study was between 23 pg kg−1 body weight per day for swine samples and 18 pg kg−1 body weight per day for turkey samples.  相似文献

Coffee is a beverage that people enjoy a lot in their daily lives and is an integral part of people's social life. In this study, the detection of ochratoxin A (OTA) in coffee was carried out by High-performance liquid chromatography with fluorescence (HPLC-FLD) detectors. Furthermore, the amount of ochratoxin A (OTA), Margin of exposure (MOEs), and Hazard quotient (HQ) in different types of coffees; instant, classic, and roasted coffee were calculated using Monte Carlo simulation (MCS) method. The average OTA concentration was in the rage of 3.6 to 26.6 µg/kg. The content of classic and instant coffee found to have.OTA, is below the maximum limit defined by the European Union legislation. The maximum limit for these two types of coffee is 10 µg/kg. The daily intakes of the OTA through classic and instant coffee were also found to be lower than the Tolerance daily intake proposed by Joint FAO/WHO Expert Committee on Food Additives (JECFA). MOEs (neoplastic effect) in adults was classic coffee (171026) > roasted coffee (15390) > instant coffee (8549) and also MOEs (non-neoplastic effects) in children was classic coffee (55790) > roasted coffee (5020) > instant coffee (2789). Consumers of instant coffee are at cancer risk based on neoplastic effects and also consumers of instant coffee and roasted coffee are at cancer risk based on non-neoplastic effects (MOEs lower than 10,000 value). HQ (nephrotoxic effect) in adults was instant coffee (0.132) > roasted coffee (0.097) > classic coffee (0.012). HQ due to consumption of coffee products was lower than 1, hence consumers are at safe non-cancer risk. Therefore, it is recommended reducing the concentration of mycotoxins in coffee products.  相似文献

The present work proposes a direct method based on slurry sampling for the determination of zinc and copper in human hair samples by multi-element sequential flame atomic absorption spectrometry. The slurries were prepared by cryogenic grinding and sonication of the samples. The optimization step was performed using univariate methodology and the factors studied were: nature and concentration of the acid solution, amount sample/slurry volume, sonication time, and particle size. The established experimental conditions are the use of a sample mass of 50 mg, 2 mol L− 1 nitric acid solution, sonication time of 20 min and slurry volume of 10 mL. Adopting the optimized conditions, this method allows the determination of zinc and copper with detection limits of 88.3 and 53.3 ng g− 1, respectively, and precision expressed as relative standard deviation (RSD) of 1.7% and 1.6% (both, n = 10) for contents of zinc and copper of 100.0 and 33.3 μg g− 1, respectively. The accuracy was checked and confirmed by analysis of two certified reference materials of human hair. The procedure was applied for the determination of zinc and copper in two human hair samples. The zinc and copper contents varied from 100.0 to 175.6 and from 3.2 to 32.8 μg g− 1, respectively. These samples were also analyzed after complete digestion in a closed system and determination by FAAS. The statistical comparison by t-test (95% confidence level) showed no significant difference between these results.  相似文献

Melatonin (N-acetyl-5-methoxytryptamine), the hormonal product of the pineal gland, has recently been reported as a multifaceted free radical scavenger and antioxidant. Melatonin production declines progressively with age. Therefore, supplemental administration or increased intake of melatonin from dietary sources may be beneficial in delaying age-related degenerative conditions. This study aimed to determine melatonin in commonly eaten bakery products based on wheat, rye or spelt flour formulations, which represented the most popular types of bread and roll in Europe. Melatonin was extracted from freeze-dried bread samples with methanol and its content was determined by micro-HPLC system coupled to triple quadrupole mass spectrometry (LC–MS/MS) of target analysis. Melatonin was identified in all types of bread and rolls, however, its content varied significantly, ranging from 0.26 ng/g to 3.21 ng/g of dry matter (DM). The best source of melatonin was mixed wheat-rye bread (average 3.02 ng/g DM) followed by rye bread (average 2.50 ng/g DM), whereas spelt bread, wheat and graham rolls were poor sources (below 0.54 ng/g DM). These findings may help consumers to select bread with high melatonin content. This study showed that micro-HPLC system coupled to triple quadrupole mass spectrometry can be successfully applied for the determination of melatonin in bakery products. Because of the relevance of melatonin as bioactive molecule with implications for food, nutritional sciences and human health, it was of interest to explore its levels in commonly daily taken bakery products.  相似文献

Ochratoxin A (OTA) is a secondary fungal metabolite produced by several moulds, mainly by Aspergillus ochraceus, A. carbonarius, A. niger and by Penicillium verrucosum. The present work shows the results of comparative studies using different procedures for the analysis of OTA in maize bread samples. The studied analytical methods involved extraction with different volumes of PBS/methanol, different extraction apparatus, and clean-up through immunoaffinity columns. The separation and identification were carried out by high-performance liquid chromatography with fluorescence detection. The optimized method for analysis of OTA in maize bread involved extraction with PBS:methanol (50:50), and clean-up with IAC column. The limit of quantification was 0.033 ng g−1. Recoveries ranged from 87% to 102% for fortifications at 2.000 and 0.500 ng g−1, respectively, within-day R.S.D. of 1.4% and 4.7%. The proposed method was applied to 15 samples and the presence of OTA was found in nine samples at concentrations ranging from nd to 2.650 ng g−1.  相似文献

The aim of this study was to estimate the contamination of grain coffee, roasted coffee, instant coffee, and cocoa purchased in local markets with ochratoxin A (OTA) and its isomerization product 2′R-ochratoxin A (2′R-OTA), and to assess risk of dietary exposure to the mycotoxins. OTA and 2′R-OTA content was determined using the HPLC chromatography with immunoaffinity columns dedicated to OTA. OTA levels found in all the tested samples were below the maximum limits specified in the European Commission Regulation EC 1881/2006. Average OTA concentrations calculated for positive samples of grain coffee/roasted coffee/instant coffee/cocoa were 0.94/0.79/3.00/0.95 µg/kg, with the concentration ranges: 0.57–1.97/0.44–2.29/0.40–5.15/0.48–1.97 µg/kg, respectively. Average 2′R-OTA concentrations calculated for positive samples of roasted coffee/instant coffee were 0.90/1.48 µg/kg, with concentration ranges: 0.40–1.26/1.00–2.12 µg/kg, respectively. In turn, diastereomer was not found in any of the tested cocoa samples. Daily intake of both mycotoxins with coffee/cocoa would be below the TDI value even if the consumed coffee/cocoa were contaminated with OTA/2′R-OTA at the highest levels found in this study. Up to now only a few papers on both OTA and 2′R-OTA in roasted food products are available in the literature, and this is the first study in Poland.  相似文献

The intake and tissue distribution of thorium (2 3 2Th) was studied in an urban (Bombay) population in India. From the analysis of 16 whole diet samples, the average daily intake through food was found to be 2 g (range 0.8–4.3 g·d–1). The estimated intake through drinking water and inhalation comes out to be 0.03 and 0.02 g per day. From the analysis of human autopsy tissue samples it is observed that the concentration ranges in lungs and bone are 1.5–16 g/kg and 0.2–9.0 g/kg fresh weight respectively. The average urinary concentration is 12 ng/1 (range 7–22 ng/l for 10 samples). Among the different body tissues, pulmonary lymph nodes were found to contain the highest concentration (geometric mean 53.4 g/kg, range 31.4–85.5 g/kg for 6 samples). Analysis of the samples was done by the neutron activation technique. 311.8 keV gamma photons of2 3 3Pa which is the activation product of2 3 2Th, were counted after chemical separation. A 54 cm3 intrinsic Ge detector coupled to 1024-channel analyser was used. Using the average lung content and the daily average intake values of thorium through inhalation, the clearance half-time from lung was estimated.  相似文献

Ultrasonic irradiation with the frequency of 20–22 kHz and absorbed acoustic power about 0.4 W ml–1 causes degradation of An(IV) tetrakis-β-diketonates, AnL4, where An(IV) is Th(IV), Np(IV), and Pu(IV), and HL is hexafluoroacetylacetone (HFAA) and dibenzoylmethane (HDBM), in hexadecane solutions in the presence of argon. The rate of the sonochemical process corresponds to a first-order kinetic law with respect to metal β-diketonate concentrations. The first-order rate constant of sonolysis increases with the increase in the volatility of the metal complexes. Solid sonication products consisted of a mixture of actinide carbides and partial degradation products, PDP, of initial metal β-diketonates. It is assumed that metal carbides are formed within the cavitating bubbles as a result of high-temperature process with participation of actinide(IV) β-diketonates and solvent vapours. L’irradiation ultrasonore avec une fréquence de 20–22 kHz et une intensité acoustique de 0,4 W ml–1 conduit à la décomposition de An(IV) tétrakis-β-dicétones, AnL4, où An(IV) est Th(IV), Np(IV) et Pu(IV), et HL est l’hexafluoroacétylacétone (HFAA) et le dibenzoylméthane (HDBM) en milieu hexadécane sous argon. La cinétique de décomposition sonochimique est d’ordre 1 par rapport à la concentration du complexe et la cinétique augmente avec la volatilité du complexe métallique. Les produits solides de décomposition sont un mélange de carbure d’actinides et de produits de décomposition partielle PDP. Il a été observé que les carbures d’actinide sont formés dans la bulle de cavitation compte tenu des fortes températures et de la présence simultanée du complexe métallique volatil et du solvant (hexadécane). In this work, headspace solid-phase micro-extraction (SPME) combined with gas chromatography-mass spectrometry (GC-MS) method for analysis of butyltin compounds in sediment samples was upgraded by the introduction of tandem mass spectrometry (MS/MS). Optimization and validation of this method based on an one step procedure, tetraethylborate in situ ethylation with simultaneous extraction by headspace SPME, combined with tandem mass spectrometry is described. A simple leaching/extraction step of mono-(M), di-(D) and tri-(T) butyltin (BT) compounds from the sediment is required as sample pre-treatment. The combination of the two techniques headspace SPME and MS/MS, led to very little matrix interference which permitted to attain limits of detection three or more orders of magnitude lower than those attained in previous methods: 0.3 pg g− 1 for MBT, 1 pg g− 1 for DBT and 0.4 pg g− 1 for TBT. Linear response range was from 0.02–1260 ng g− 1 for MBT, 0.07–1568 ng g− 1 for DBT and 0.04–2146 ng g− 1 for TBT and RSD < 15% was also obtained. The method was efficiently applied to a real sample sediment from Sado River estuary in Portugal, revealing the existence of BTs pollution, as the TBT level of 189 ± 15 ng g− 1 was much higher than the maximum established as provisional ecotoxicological assessment criteria.  相似文献

The development of actinide speciation methods in the environment is discussed. The results on speciation and migration of radionuclides in near-surface and deep conditions at the Krasnoyarsk mining and chemical plant and at the Production Association ‘Mayak’ (southern Ural) are reported. For surface conditions, the mobile plutonium fraction is mostly associated with inorganic complexes (carbonate) of the higher plutonium oxidation state, while less mobile fraction is associated with low-soluble Pu(IV) complexes with humic acids. The mobile fraction of americium consists of complexes with low molecular weight fulvic acids of non-specific nature with molecular weights lower than 10 kDa. Méthodes de spéciation des actinides en traces. Le développement de méthodes pour la spéciation des actinides dans l’environnement est discuté. Nous présentons des résultats concernant la spéciation et la migration des radionucléides dans des conditions de sub-surface et en milieux géologiques profonds à la mine de Krasnoyarsk et à l’usine chimique de Mayak (Oural du Sud). En conditions de sub-surface, la fraction mobile contient essentiellement des complexes inorganiques (carbonates) de Pu dans ses degrés d’oxydation élevés, tandis que la fraction moins mobile contient des complexes insolubles du Pu(IV), avec des acides humiques. La fraction mobile de Am comporte des complexes d’acides fulviques de nature non précisée, mais d’une masse molaire inférieure à 10 kDa. Contamination of food by mycotoxin occurs in minute/trace quantities. Nearly 92.5% of the cocoa samples present Ochratoxin A (OTA) levels at trace quantity. Hence, there is a necessity for a highly sensitive and selective device that can detect and quantify these organic toxins in various matrices such as cocoa beans. This work reports for the first time, a facile and label-free electrochemical impedimetric aptasensor for rapid detection and quantitation of OTA in cocoa beans. The developed aptasensor was constructed based on the diazonium-coupling reaction mechanism for the immobilization of anti-OTA-aptamer on screen printed carbon electrodes (SPCEs). The aptasensor exhibited a very good limit of detection (LOD) as low as 0.15 ng/mL, with added advantages of good selectivity and reproducibility. The increase in electron transfer resistance was linearly proportional to the OTA concentration in the range 0.15–2.5 ng/mL, with an acceptable recovery percentage (91–95%, RSD = 4.8%) obtained in cocoa samples. This work can facilitate a general model for the detection of OTA in cocoa beans based on the impedimetric aptasensor. The analysis can be performed onsite with pre-constructed and aptamer modified electrodes employing a portable EIS set up.  相似文献
