A cycloaddition strategy for the synthesis of highly enantioenriched 2,5-cis-disubstituted tetrahydrofurans has been developed. In the presence of catalytic Sn(OTf)2 or SnCl4, a range of aldehydes will undergo formal [3 + 2] cycloadditions with a scalemic donor-acceptor cyclopropane to form optically active heterocycles. Mechanistic studies support an unusual SN2 attack by the aldehyde on the activated cyclopropane. Through this mechanism, stereochemical information contained in the cyclopropane is effectively transferred to the tetrahydrofuran products.  相似文献

In this work, the one-step diastereoselective synthesis of cis-2,5-disubstituted tetrahydrofurans via Lewis acid catalyzed [3 + 2] cycloadditions of donor-acceptor (D-A) cyclopropanes and aldehydes is described. The scope and limitations with respect to both reaction partners are provided. A detailed examination of the mechanism has been performed, including stereochemical analysis and electronic profiling of both reactants. Experimental evidence supports an unusual stereospecific intimate ion pair mechanism wherein the aldehyde functions as a nucleophile and malonate acts as the nucleofuge. The reaction proceeds with inversion at the cyclopropane donor site and allows absolute stereochemical information to be transferred to the products with high fidelity. The mechanism facilitates the stereospecific synthesis of a range of optically active tetrahydrofuran derivatives from enantioenriched D-A cyclopropanes.  相似文献

The in situ reaction of protected dehydroamino acids with derivatives of vinyldiazomethane leads to good to excellent yields of vinyl cyclopropanes via 3 + 2 dipolar cycloaddition followed by N(2) extrusion. Chromatographic separation of the cyclopropane diastereomeric products, followed by characterisation by (1)H NMR and X-ray crystallography allowed the cis and trans diastereomers to be easily identified. Oxidative cleavage of the vinyl moiety then led directly to protected cyclopropane aspartic acid derivatives in three steps from commercially available materials. These compounds were converted to protected methylenephosphonate, difluoromethylenephosphonate and phosphoramidate analogues of [small beta]-aspartyl phosphate.  相似文献

New routes to transform donor-acceptor cyclopropanes in the presence of Lewis acids have been found. The reaction of dimethyl 2-phenylcyclopropan-1,1-dicarboxylate, a typical representative of this class of compounds, with an equimolar amount of anhydrous GaCl3 gives, depending on the reaction time, (2-phenylethylidene)malonate or styrylmalonate after hydrolysis, whereas in the presence of 15-20 mol % GaCl3 the starting cyclopropane undergoes dimerization to give the malonic derivative of 3,4-diphenylcyclopentane-1,1-dicarboxylate. In the presence of the GaCl3·THF complex, the same cyclopropane gives a substituted 4-phenyltetraline-2,2-dicarboxylate in high yield, whereas in the presence of SnCl4·THF it gives the dimer specified above along with products of chain oligomerization, the degree of which can be controlled by changing the concentration of the starting cyclopropane in solution.  相似文献

Work carried out in the past few years aimed at elucidating the mechanism of initiation of vinyl polymerization when a donor and an acceptor molecule, one or both of which may be vinyl monomers, is summarized. The emphasis of our investigation has been on polymerizable ether donors and strong electron acceptors which do not undergo polymerization, or the acceptor vinylidene cyanide. Alkyl vinyl ethers were polymerized in the presence of tetracyanoquinodimethane (TCNQ) and 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) in polar solvents. Observation of the ESR spectrum of the DDQ radical anion and the isolation of a 1:1 addition product of DDQ and alkyl vinyl ether when the two are mixed in a 1:1 ratio and quenched in alcohol support an initiation mechanism involving a coupling reaction of the donor monomer (radical cation) and the acceptor initiator (radical anion). The reaction of vinylidene cyanide (VC) with the vinyl ethers p-dioxene, dihydropyran, ethyl vinyl ether, isopropyl vinyl ether, and ketene diethylacetal in a variety of solvents at 25°C spontaneously afforded poly(vinylidene cyanide), the cycloaddition products 7,7-dicyano-2,5-dioxo-bicyclo[4.2.0] octane, 8,8-dicyano-2-oxo-bicyclo[4.2.0] octane, the 1,1-dicyano-2-alkoxycyclo-butanes, and 1,1-diethoxy-2,2,4,4-tetracyanohexane, respectively, and with the exception of p-dioxene, homopolymers of the vinyl ethers. In the presence of AIBN at 80°C, alternating copolymers were obtained in addition to the homopolymers and cycloaddition products, supporting the involvement of donor-acceptor complexes. The reaction of styrene with VC spontaneously formed an alternating copolymer in addition to the 1:2 head-to-head cycloaddition product, 1,1,3,3-tetracyano-4-phenylcyclohexane. Mixing VC with any one of the cyclic ethers tetrahydrofuran, oxetane, 2,2-dimethyloxirane, 2-chloromethyloxirane, and phenyloxirane resulted in the polymerization of both the VC and the cyclic ether to afford homopolymers of both. The cyclic ethers trioxane, 3,3-bis(chloromethyl)oxetane, and oxirane initiated the polymerization of VC, but did not undergo ring-opening polymerizations themselves. Other ethers such as 1,3-dioxolane, tetrahydropyran, and diethyl ether did not initiate the polymerization of VC. In these polymerizations, VC and the cyclic ethers polymerize via anionic and cationic propagation reactions, respectively.  相似文献

The reactions of diazo esters with 2-arylcyclopropane-1,1-dicarboxylates, the represen- tatives of donor-acceptor cyclopropanes (DACs), mediated by Sc(OTf)3, SnCl4, and GaCl3 proceeded with nitrogen elimination to give the C—C coupling products. No products of the formal [3+3] cycloaddition of diazo compounds to DACs were formed but the main reaction direction was addition of diazo ester to either 1,3- or 1,2-zwitterions generated upon Lewis acid-mediated cyclopropane ring opening giving rise to new 1,4- and 1,3-zwitterionic inter- mediates. The formed intermediates underwent further fragmentations and rearrangements to give substituted cyclopropanedi-, -tri-, and -tetracarboxylates. Mechanistic aspects of the observed reactions were discussed.  相似文献

A novel Lewis acid catalyzed [3+2] cyclodimerization of 2-arylcyclopropane-1,1-dicarboxylates is reported. It is the first example of a reaction wherein a donor-acceptor cyclopropane provides two carbons in a newly formed ring. The described cyclodimerization represents a general convenient approach to polyfunctionalized cyclopentanes.  相似文献

Catalyzed by Lewis acids, donor-acceptor cyclopropane 1,1-diesters reacted with enol silyl ethers to afford 1,6-dicarbonyl compounds in moderate to excellent yields. This supplied a mild carbon-carbon bond-forming method from the ring opening of cyclopropanes. A smooth tandem [3+2] cycloaddition/ring opening process has been clearly proved by an independent experiment.  相似文献

The reaction of (E)-1-(phenylseleno)-2-(trimethylsilyl)ethene (1) and vinyl ketones 2a-d in the presence of a chiral Lewis acid prepared from TiCl(4), Ti(O(i)Pr)(4), (R)- or (S)-1,1'-binaphthol (BINOL), and MS4A gave enantiomerically enriched cis cyclopropane products 3a-d. The enantiomeric excess and chemical yield varied depending on the ratio of TiCl(4) and Ti(O(i)Pr)(4) to 1. Reproducible results (43-47% ee/33-41% yields) for cis-1-acetyl-2-[(phenylseleno)(trimethylsilyl)methyl]cyclopropane (3a) were obtained using 1.1 equiv of TiCl(4), 0.54-0.65 equiv of Ti(O(i)Pr)(4), and 1.65 equiv of BINOL. The observed enantioselectivity was explained by consideration of the structure of the postulated intermediates, alkoxy titanium-carbonyl complexes, via ab initio MO calculations.  相似文献

Nileprost is a new prostacyclin analogue stabilized by introduction of the cyano group at its 5-position. The acidic and alkaline degradation and the structure determination of the degradation products were investigated. The degradation of nileprost is very slow in comparison with that of prostacyclin. Although prostacyclin is easily decomposed to 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) through the hydrolysis of a vinyl ether moiety, nileprost gives little of such hydrolysis product but many isomers and dehydrates. The structures of the products were determined by high-performance liquid chromatography (HPLC), negative ion fast atom bombardment mass spectra (N-FABMS) and nuclear magnetic resonance (NMR) studies. It was found that the vinyl ether moiety of nileprost is converted from 5Z form to 5E form in both media, and that the omega-side chain also undergoes transfer of hydroxyl group or dehydration in acid medium. These results indicate that introduction of the cyano group into the 5-position of prostacyclin is extremely effective for the stabilization of the vinyl ether moiety. Furthermore, on the basis of the structural elucidation, the reaction mechanism of nileprost in both media was clarified.  相似文献

Reactions of 2-alkoxypropenals with α-hydroxyamino oximes in neutral medium involve the aldehyde group of the former to afford both acyclic and cyclic azomethine oxides: N-(2-hydroxyiminoalkyl)-N-(2-alkoxy-2-propenylidene)amine oxides and 1-hydroxy-2,5-dihydroimidazole 3-oxides. The state of tautomeric equilibrium between the cyclic and acyclic products depends on the solvent nature and temperature. The reaction in acidic aqueous medium is accompanied by hydrolysis of the vinyl ether moiety in 2-alkoxy-propenals with formation of 2-oxopropionaldehyde which reacts with α-hydroxyamino oxime at the hydroxy-amino group to give substituted pyrazine 1,4-dioxides. The reaction of 2-alkoxypropenals with 1,2-bis-(hydroxyamino)cyclohexane leads to formation of 2-(1-alkoxyvinyl)-1,3-dihydroxyperhydrobenzimidazoles. The structure of the products was proved by IR, UV, and 1H and 13C NMR spectroscopy and X-ray analysis.  相似文献

Reductive cyclization of 4-hydroxymethyl-5,5-dimethyl(or pentamethylene)-2,5-dihydrofuran-2-ones by the action of sodium tetrahydridoborate gave bicyclic compounds in which the lactone ring is fused to a cyclopropane ring. Hydrolysis of the products with aqueous sodium hydroxide resulted in the formation of the corresponding disodium cyclopropane-1,1-dicarboxylates, which reacted with alkyl halides to produce the diesters. Acid hydrolysis of the fused systems was accompanied by opening of the cyclopropane ring with formation of 4-chloromethyl-5,5-dimethyl-2-oxotetrahydrofuran-3-carboxylic acid.  相似文献

High enantioselectivities (94-96% ee) were obtained for the inverse electron-demand 1,3-dipolar cycloadditions between cyclohexyl vinyl ether and 2-benzopyrylium-4-olate generated via Rh2(OAc)4-catalyzed decomposition of o-methoxycarbonyl-α-diazoacetophenone. The reactions were effectively catalyzed by Eu(OTf)3, Ho(OTf)3, or Gd(OTf)3 complexes (10 mol %) of chiral 2,6-bis[(4S,5S)-4,5-diphenyl-2-oxazolinyl]pyridine. The reactions with the other electron-rich dipolarophiles such as allyl alcohol, 2,3-dihydrofuran, and butyl-tert-butyldimethylsilylketene acetal showed moderate enanantioselectivities (60-73% ee). Good to high enantioselectivities (73-97% ee) were also obtained for the cycloadditions between 3-acyl-2-benzopyrylium-4-olates, generated from methyl 2-(2-diazo-1,3-dioxoalkyl)benzoates and butyl or cyclohexyl vinyl ethers, in the presence of binaphthyldiimine (BINIM)-Ni(II) complexes (10 mol %). Under similar conditions, the reaction between methyl 2-(2-diazo-1,3-dioxohexyl)benzoate and 2,3-dihydrofuran was highly endo-selective, and moderately enantioselective (70% ee). For the BINIM-Ni(II)-catalyzed reactions of cyclohexyl vinyl ether, the use of an epoxyindanone as the 3-acyl-2-benzopyrylium-4-olate precursor revealed that the chiral Lewis acid can function as a catalyst for asymmetric induction. The scope of the cyclic carbonyl ylides was extended to those generated from 1-diazo-2,5-pentanedione derivatives, which were reacted with butyl or TBS vinyl ether and catalyzed using the (4S,5S)-Pybox-4,5-Ph2-Lu(OTf)3 complex to give good levels of asymmetric inductions (75-84% ee).  相似文献

The ability of donor-acceptor cyclopropanes to (3 + 3)-cyclodimerize is disclosed. It has been found that Lewis acid-induced transformations of 2-(hetero)arylcyclopropane-1,1-dicarboxylates containing electron-abundant aromatic substituents led to the construction of six-membered cyclic systems. Depending on the substrate properties and the Lewis acid applied, three types of products can be obtained: (1) 1,4-diarylcyclohexanes, (2) 1-aryl-1,2,3,4-tetrahydronaphthalenes, and (3) 9,10-dihydroanthracenes.  相似文献

While the divinyl cyclopropane Cope rearrangement is well-known, and has been broadly applied in synthesis, examples of the aryl vinyl cyclopropane Cope rearrangement are less common and generally limited in scope or reaction yield. The aryl vinyl cyclopropane Cope rearrangement gives access to the benzocycloheptene scaffold, which is present in a variety of naturally occurring and medicinally relevant products. Herein we report a method to obtain either of two regioisomeric benzocycloheptene products via an aryl vinyl cyclopropane Cope rearrangement, featuring additive-controlled regioselectivity. Mechanistic studies indicate a dynamic equilibration of cyclopropane stereoisomers, followed by rearrangement of the cis diastereomer.  相似文献

Initiated by an organic molecule trifluoromethanesulfonimide (HNTf2) without any Lewis acid or Lewis base stabilizer, cationic polymerization of isobutyl vinyl ether (IBVE) takes place rapidly and the polymerization is proved to be in a controlled/living manner. The conversion of IBVE could easily achieve 99% in seconds. The product poly(isobutyl vinyl ether) is narrowly distributed and its molecular weight increases linearly with time and fits well with the corresponding theoretical value. This single‐molecular initiating system also works well in the living cationic polymerization of ethyl vinyl ether. HNTf2 is considered playing multiple roles which include initiator, activator, and stabilizer in the polymerization. Three isomeric nonconjugated dienes, o-, m- and p-(2-vinyloxyethoxy)styrenes, were selectively polymerized by anionic or radical initiators through the styryl double bond while leaving the vinyl ether moiety intact. The anionic-initiated polymeric products are of high molecular weight and narrow molecular weight distribution as characterized by membrane osmometry and gel-permeation chromatography, respectively. These polymers were subsequently crosslinked by cationic initiators via the vinyl ether moiety on the polymer side chains. Acid-catalyzed hydrolysis of the poly(2-vinyloxyethoxy)styrenes yielded their respective hydroxy-containing polymers, polyvinylphenoxyethanols. The latter were physically and spectroscopically identical to authentic samples prepared by radical polymerization of the corresponding vinylphenoxyethanols, which, in turn, were synthesized by hydrolysis of the (2-vinyloxyethoxy)styrenes. The polyvinylphenoxyethanols were shown to undergo many chemical transformations, such as esterification with 3,5-dinitrobenzoyl chloride, cyanoethylation, and urethane formation.  相似文献

以异戊二烯、甲基乙烯基酮为原料,氯化铝为催化剂,经Diels-Alder反应,合成得到1-甲基-4-乙酰基-1-环己烯,再用锍叶立德对羰基环氧化,专一地得到消旋体8,9-环氧烯,两步反应总得率为56%。从8,9-环氧烯出发,进一步合成得到萜类香料1,8-对二烯-10-醇和4-(4-甲基-3-环己烯-1-基)-4-戊烯醛。所有产物的结构经IR、NMR和MS证实。  相似文献

Three Lewis base variations of the synthetically useful aluminate [L x Li(TMP)((i)Bu)Al((i)Bu)2], where L is TMPH, Et3N or PhC(=O)N(i)Pr2, are reported, together with the reaction of the benzamide complex with 1,4-dioxane, which surprisingly leads to fragmentation of the cyclic ether and capture of its alkoxy vinyl ether residue within a novel dilithium dialuminium hexaalkyl aggregate.  相似文献

The bulky 2,5-dimethylthiophene (2,5-Me2T) reacts at 60 degrees C with TpMe2Ir(C2H4)2 to give a mixture of two TpMe2Ir(III) hydride products, 3 and 4, that contain in addition a thienyl (3) or a thienyl-derived ligand (4). For the generation of 3 only sp2 C-H activation is needed, but the formation of 4 requires also the activation of an sp3 C-H bond and the formation of a new C-C bond (between vinyl and thienyl fragments). In the presence of 2,5-Me2T, compound 4 reacts further to produce a complex thiophenic structure (5, characterized by X-ray methods) that derives formally from two molecules of 2,5-Me2T and a vinyl fragment. Compounds 3-5 can be readily protonated by [H(OEt2)2][BAr'4](Ar'= 3,5-C6H3(CF3)2), with initial generation of carbene ligands (in the case of 3 and 5) as a consequence of H+ attack at the beta-carbon of the Ir-thienyl unit. Free, substituted thiophenes, derived from the original 2,5-Me2T, may be isolated in this way.  相似文献
