Highly boron-doped diamond (BDD) electrode, have been examined for simultaneous detection of purine and pyrimidine bases in mild acidic media by using HPLC with amperometric detection. Cyclic voltammetry at as-deposited (AD) and anodically oxidized (AO) BDD were used to study the electrochemistry and to optimize the condition for HPLC applications. At AO BDD electrode, due to its higher overpotential of oxygen evolution reaction, well-defined anodic peaks were observed for the oxidation of purine and pyrimidine bases in acid medium, whereas at AD BDD the oxidation peak of thymine was overlapped with the anodic current of oxygen evolution. The chromatograms of adenine, guanine, cytosine, thymine and 5-methylcytosine mixture were well resolved by using a silica-based column and a solution of 5% acetonitrile in 100 mM ammonium acetate buffer (pH 4.25) as the mobile phase. The detection was carried out at AO BDD electrode at an applied potential of 1.6 V versus Ag/AgCl. Linear calibration curves were obtained within the concentration range from 0.1 to 10 μM with the limits of detection (S/N = 3) ranging from 26.3 to 162.1 nM, resulting in an order of magnitude higher sensitivities than those at conventional electrodes. HPLC analysis with diamond amperometric detector was successfully applied for determination of 5-methylcytosine in real DNA samples with high reproducibility. No deactivation of the electrode was found during cyclic voltammetric and HPLC measurements, indicating the high stability for analysis of biological samples.  相似文献

Monodispersed and highly stable gold nanoparticles with a diameter between 8 and 9 nm were synthesized in a weakly alkaline medium by chemical reduction of AuCl4 using 5-hydroxyl-1,4-naphthoquinone, and stabilized by the simultaneously formed poly(hydroxyl-1,4-naphthoquinone). The electrochemical properties of the resultant poly(hydroxyl-1,4-naphthoquinone) stabilized gold nanoparticles (AuNQ NPs) and its electrocatalytic activity for glucose oxidation in alkaline media were then investigated using a range of techniques, including dc cyclic, rotating disk electrode and Fourier transformed large amplitude ac voltammetry. The results demonstrate that these AuNQ NP modified electrodes exhibit excellent catalytic activity toward glucose oxidation in the potential region where the premonolayer oxidation process occurs. The overall catalytic glucose oxidation process was found to be mass transport controlled under the experimental conditions employed, allowing measurements to be conducted with a high reproducibility. The AuNQ NP modified electrodes showed a high sensitivity of 183 μA mM−1 cm−2 with a wide linear dynamic range of 0.5–50 mM and a detection limit of 61 μM. However, despite its excellent tolerance toward ascorbic acid, significant interference from uric acid was found with this AuNQ NP modified electrode.  相似文献

Nonenzymatic electrochemical determination of ethanol and glucose was respectively achieved using PdNi- and Pd-coated electrodes prepared by electrodeposition from the novel metal-free ionic liquid (IL); N-butyl-N-methylpyrrolidinium dicyanamide (BMP-DCA). BMP-DCA provided an excellent environment and wide cathodic limit for electrodeposition of metals and alloys because many metal chlorides could dissolve in this IL where the reduction potentials of Pd(II) and Ni(II) indeed overlapped, leading to the convenience of potentiostatic codeposition. In aqueous solutions, the reduction potentials of Pd(II) and Ni(II) are considerably separated. The bimetallic PdNi coatings with atomic ratios of ∼80/20 showed the highest current for ethanol oxidation reaction (EOR). Ethanol was detected by either cyclic voltammetry (CV) or hydrodynamic amperometry (HA). Using CV, the dependence of EOR peak current on concentration was linear from 4.92 to 962 μM with a detection limit of 2.26 μM (σ = 3), and a linearity was observed from 4.92 to 988 μM using HA (detection limit 0.83 μM (σ = 3)). The Pd-coated electrodes prepared by electrodeposition from BMP-DCA showed electrocatalytic activity to glucose oxidation and CV, HA, and square-wave voltammetry (SWV) were employed to determine glucose. SWV showed the best sensitivity and linearity was observed from 2.86 μM to 107 μM, and from 2.99 mM to 10.88 mM with detection limits of 0.78 μM and 25.9 μM (σ = 3), respectively. For glucose detection, the interference produced from ascorbic acid, uric acid, and acetaminophen was significantly suppressed, compared with a regular Pt disk electrode.  相似文献

Electrochemical glucose enzyme biosensors have been prepared on carbon film electrodes made from carbon film electrical resistors. Evaluation and characterisation of these electrodes in phosphate buffer saline solution has been carried out with and without pretreatment by cycling in perchloric acid or at fixed applied potential. Both pretreatments led to a reduction in the carbon surface oxidation peak and enabled better detection of hydrogen peroxide in the pH range of 5-7. Glucose oxidase enzyme was immobilised on the carbon surface by mixing with glutaraldehyde, bovine serum albumin and with and without Nafion. The performance of these two types of electrode was similar, that containing Nafion being more physically robust. Linear ranges were up to around 1.5 mM, with detection limits 60 μM, and pretreatment of the carbon film electrode at a fixed potential of +0.9 V versus SCE for 5 min was found to be the most beneficial. Michaelis-Menten constants between 5 mM and 10 mM were found under the different experimental conditions. Coating the immobilised enzyme layer with a thin layer of Nafion was found to give similar results in the determination of glucose to mixing it but with benefits against interferences for the analysis of complex matrices, such as wine. Potentialities, for a short-term-use or disposable sensors, are indicated.  相似文献

A novel, stable and sensitive non-enzymatic glucose sensor was developed by potentiostatically electrodepositing metallic Cu nanoparticles on graphene sheets. The electrochemical performance of the Cu-graphene sheets electrode for detection of glucose was investigated by cyclic voltammetry and chronamperometry. The Cu-graphene sheets electrode displayed a synergistic effect of copper nanoparticles and graphene sheets towards the oxidation of glucose in alkaline solution, showing higher oxidation current and negative shift in peak potential. At detection potential of 500 mV, the Cu-graphene electrode sensor presented a wide linear range up to 4.5 mM glucose with a detection limit of 0.5 μM (signal/noise = 3). In addition, the sensor responds very quickly (<2 s) with addition of glucose. Furthermore, the Cu-graphene sheets electrode exhibits high stability and selectivity to glucose, and the poisoning by chloride ion as well as interference from the oxidation of common interfering species (ascorbic, dopamine, uric acid and carbohydrate) are effectively avoided. The Cu-graphene sheets electrode allows highly selective and sensitive, stable and fast amperometric sensing of glucose, which is promising for the development of non-enzymatic glucose sensor.  相似文献

Boron-doped diamond (BDD) thin films with different crystal grain sizes were prepared by controlling the reacting gas pressure using hot filament chemical vapor deposition (HFCVD). The morphologies and structures of the prepared diamond thin films were characterized by scanning electron microscopy (SEM) and Raman spectroscopy. The electrochemical responses of K4Fe(CN)6 on different BDD electrodes were investigated. The results suggested that electron transfer was faster at the boron-doped nanocrystalline diamond (BDND) thin film electrodes in comparison with that at other BDD thin film electrodes. The prepared BDD thin film electrodes without any modification were used to directly detect glucose in the basic solution. The results showed that the as-prepared BDD thin film electrodes exhibited good selectivity for detecting glucose in the presence of ascorbic acid (AA) and uric acid (UA). The higher sensitivity was observed on the BDND thin film grown on the boron-doped microcrystalline diamond (BDMD) thin film surface, and the linear response range, sensitivity and the low detection limit were 0.25–10 mM, 189.1 μA mmo?1 cm?2 and 25 μM (S/N=3) for glucose in the presence of AA and UA, respectively.  相似文献

A novel copper incorporated self-assembled monolayers (SAMs) modified gold electrode (Cu/SAMs) for determination of glucose was developed by electrodepositing Cu particles on the SAMs of hexanethiol. The scanning electron microscopic (SEM) images showed that copper formed orbicular particles of nanosizes on the SAMs, which was much different from the fractal-like particles of copper formed at gold electrode. The Cu/SAMs film electrode exhibited high sensitivity to glucose oxidation and depressed responses towards some interferents of glucose in blood like uric acid and ascorbic acid. Under optimal working conditions, the oxidation current of glucose was proportional to the concentration of glucose in the range from 3.0 μM to 10 mM by amperometry with a low detection limit of 0.7 μM glucose (S/N = 3). This electrode was successfully applied to the determination of glucose in rat blood and the results were satisfactory.  相似文献

We have constructed a glucose biosensor by immobilizing glucose oxidase (GOD) on titanium-containing MCM-41 (Ti-MCM-41) modified screen-printed electrodes. The strategy of the sensing method is to monitor the extent of the decrease of the reduction current of O2 upon adding glucose at a selected potential. The detection can be done at the applied potential of −0.50 V and can efficiently exclude the interference from commonly coexisted substances. The constructed sensor has a high sensitivity to glucose (5.4 mAM−1 cm−2) and a linear response range of 0.10-10.0 mM. The detection limit is 0.04 mM at a signal-to-noise ratio of 3. The sensor also shows high stability and remains its catalytic activity up to 60 °C. The biocompatibility of Ti-MCM-41 means that this immobilization matrix not only can be used for immobilizing GOD but also can be extended to other enzymes and bioactive molecules, thus providing a promising platform for the development of biosensors.  相似文献

The development of all-solid-state potentiometric ion selective electrodes for monitoring of ascorbic acid, by using a screen-printed compatible solid contact is described. The applied methodology is based on the use of PVC membrane modified with some firstly-tested ionophores (triphenyltin(IV)chloride, triphenyltin(IV)hydroxide and palmitoyl-l-ascorbic acid) and a novel one synthesized in our laboratory (dibutyltin(IV) diascorbate). Synthesis protocol and some preliminary identification studies are given. A conductive graphite-based polymer thick film ink was used as an internal solid contact between the graphite electrode and the PVC membrane. The presence and the nature of the solid contact (plain or doped with lanthanum 2,6-dichlorophenolindophenol (DCPI)) seem to enhance the analytical performance of the electrodes in terms of sensitivity, dynamic range, and response time. The analytical performance of the constructed electrodes was evaluated with potentiometry, constant-current chronopotentiometry and electrochemical impedance spectroscopy (EIS). The interference effect of various compounds was also tested. The potential response of the optimized Ph3SnCl-based electrode was linear against ascorbic acid concentration range 0.005-5.0 mM. The applicability of the proposed sensors in real samples was also tested. The detection limit was 0.002 mM ascorbic acid (50 mM phosphate, pH 5 in 50 mM KCl). The slope of the electrodes was super-Nernstian and pH dependent, indicating a mechanism involving a combination of charge transfer and ion exchange processes. Fabrication of screen-printed ascorbate ISEs has also been demonstrated.  相似文献

A new methyl viologen-mediated amperometric enzyme electrode sensitive to glucose has been developed using carbon film electrode substrates. Carbon film electrodes from resistors fabricated by pyrolytic deposition of carbon were modified by immobilization of glucose oxidase through cross-linking with glutaraldehyde in the presence of bovine serum albumin. The mediator, methyl viologen, was directly immobilised with the enzyme together with Nafion cation-exchange polymer. The electrochemistry of the glucose oxidase/methyl viologen modified electrode was investigated by cyclic voltammetry and by electrochemical impedance spectroscopy. The biosensor response to glucose was evaluated amperometrically; the detection limit was 20 μM, the linear range extended to 1.2 mM and the reproducibility of around 3%. When stored in phosphate buffer at 4 °C and used every day, the sensor showed good stability over more several weeks.  相似文献

Optical biosensor arrays for rapidly determining the glucose concentrations in a large number of beverage and blood samples were developed by immobilizing glucose oxidase (GOD) on oxygen sensor layer. Glucose oxidase was first encapsulated in silica based gels through sol-gel approach and then immobilized on 96-well microarrays integrated with oxygen sensing film at the bottom. The oxygen sensing film was made of an organically modified silica film (ORMOSIL) doped with tris(4,7-diphenyl-1,10-phenanthroline) ruthenium dichloride (Ru(dpp)3Cl2). The oxidation reaction of glucose by glucose oxidase could be monitored through fluorescence intensity enhancement due to the oxygen consumption in the reaction. The luminescence changing rate evaluated by the dynamic transient method (DTM) was correlated with the glucose concentration with the wide linear range from 0.1 to 5.0 mM (Y = 13.28X − 0.128, R = 0.9968) and low detection limit (0.06 mM). The effects of pH and coexisting ions were systemically studied. The results showed that the optical biosensor arrays worked under a wide range of pH value, and normal interfering species such as Na+, K+, Cl, PO43−, and ascorbic acid did not cause apparent interference on the measurement. The activity of glucose oxidase was mostly retained even after 2-month storage, indicating their long-term stability.  相似文献

The gold nanostar@silica core–shell nanoparticles conjugated with glucose oxidase (GOx) enzyme molecules have been developed as the surface-enhanced Raman scattering (SERS) biosensor for label-free detection of glucose. The surface-immobilized GOx enzyme catalyzes the oxidation of glucose, producing hydrogen peroxide. Under laser excitation, the produced H2O2 molecules near the Au nanostar@silica nanoparticles generate a strong SERS signal, which is used to measure the glucose concentration. The SERS signal of nanostar@silica∼GOx nanoparticle-based sensing assay shows the dynamic response to the glucose concentration range from 25 μM to 25 mM in the aqueous solution with the limit of detection of 16 μM. The sensing assay does not show any interference when glucose co-exists with both ascorbic acid and uric acid. The sensor can be applied to a saliva sample.  相似文献

Herein, we demonstrated a facile strategy for preparing high-loading Au nanoparticles using thionine functionalized graphene oxide as the supporting material. Glassy carbon electrodes modified with the as-obtained nanocomposites showed remarkably electrocatalytic activity towards the oxidation of glucose, leading to an enzymeless glucose sensor with a wide linear range and a lower detection limit of 0.05 μmol/L.  相似文献

A microfluidic device was designed for amperometric determination of glucose by packing enzyme modified magnetic nanoparticles (MNPs) in its microchannel as an enzyme microreactor. Glucose oxidase was covalently attached to the surface of MNPs and localized in the microchannel by the help of an external magnetic field, leading to a tunable packing length. By changing the length of microreactor from 3 to 10 mm, the performance for glucose detection was optimized. The optimal linear range to glucose was from 25 μM to 15 mM with a detection limit of 11 μM at a length of 6 mm. The inter- and intra-day precisions for determination of 1.0 mM glucose were 0.8% and 1.7%, respectively, and the device-to-device reproducibility was 95.6%. The enzyme reactor remained its 81% activity after three-week storage. Due to the advantages of the device and fracture sampling technique, serum samples could be directly sampled through the fracture to achieve baseline separation from ascorbic acid, and proteins in the samples did not interfere with the detection. This work provided a promising way for pretreatment-free determination of glucose with low cost and excellent performance.  相似文献

An amperometric glucose biosensor was developed using an anionic clay matrix (layered double hydroxide (LDH), Ni/Al-NO3) for the immobilization of glucose oxidase (GOx). The biofilm was prepared by electrodeposition of the clay and GOx and subsequent cross-linking with glutaraldeyde. The Pt surface modified with the Ni/Al-NO3 shows a much reduced noise, giving rise to a better signal to noise ratio for the currents relative to H2O2 oxidation, and a linear range for H2O2 determination wider than the one observed for bare Pt electrodes. Under the optimised operative conditions, the performances of the biosensor have been evaluated by measuring the steady-state currents (at +0.45 V versus SCE) to increasing concentrations of glucose in “air saturated” 0.1 M phosphate buffer (pH 7.0). Both batch and flow injection modes were explored. The response to glucose was linear up to 8.0 and 12.0 mM, and the sensitivities were 7.7 ± 0.1 and 19.1 ± 0.2 mA M−1 cm−2, respectively. The current response of the biosensors does not significantly change for 15 consecutive days in batch and for 10 days in flow, at least, if stored at 4 °C in phosphate buffer, when not in use. The effects of interferants and applicability to fruit juices and soft drinks analysis of the biosensor were also investigated.  相似文献

The electrochemical oxidation of homocysteine was studied at as-deposited and anodized (oxidized) boron-doped diamond (BDD) thin film electrodes with cyclic voltammetry, flow injection analysis and high-pressure liquid chromatography with amperometric detection. At anodized boron-doped diamond electrodes, highly reproducible, well-defined cyclic voltammograms for homocysteine oxidation were obtained in acidic media, while as-deposited diamond did not provide a detectable signal. In alkaline media, however, the oxidation response was obtained both at as-deposited and anodized diamond electrodes. The potential sweep rate dependence of homocysteine oxidation (peak currents for 1 mM homocysteine linearly proportional to v(1/2), within the range of 0.01 to 0.3 V s(-1)) indicates that the oxidation involves a diffusing species, with negligible adsorption on the BDD surface at this concentration. In the flow system, BDD exhibited a highly reproducible amperometric response, with a peak variation less than 2%. An extremely low detection limit (1 nM) was obtained at 1.6 V vs. Ag/AgCl. In addition, the determination of homocysteine in a standard mixture with aminothiols and disulfide compounds by means of isocratic reverse-phase HPLC with amperometric detection at diamond electrodes has been investigated. The results showed excellent separation, with a detection limit of 1 pmol and a linear range of three orders of magnitude.  相似文献

We report a simple and rapid procedure that leads to incorporation of mediator and introduction of amine functionality onto the surface of screen-printed carbon electrodes (SPCE). The electrodes were doped with cobalt phthalocyanine (CoPc) by enhanced adsorption in a process that uses minimal amounts of this redox mediator as compared with CoPc loaded inks. The CoPc-doped SPCE showed a substantially increased sensitivity to hydrogen peroxide and thiocholine as compared to unmodified electrodes. This greatly facilitated their use as transducers for the construction of amperometric biosensors based on enzymes producing oxidizable products such as hydrogen peroxide or thiols. Immobilisation of enzymes including glucose oxidase, acetylcholinesterase and choline oxidase was achieved through their multi-contact electrostatic interaction with polyethyleneimine (PEI) which was electrodeposited on the surface of CoPc-doped electrodes in one step from ethanolic solution. The efficiency of enzyme immobilisation was shown to depend on the molecular weight of the PEI used, reaching a maximum for 25 kDa PEI. The biosensors shown sensitivity to glucose at 130 nA mM−1 (LOD 0.15 mM) and to acetylcholine at 70 nA mM−1 (LOD 0.10 mM) under +0.6 V. Detection of glucose has been demonstrated at +0.4 V with the sensitivity of 60 nA mM−1 and LOD of 0.33 mM. Possibility of the inhibition analysis of pesticides has been shown for acetylcholinesterase-based sensors.  相似文献

The electrochemical behaviors of formaldehyde (FA) at boron‐doped diamond (BDD) electrodes are investigated by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and linear scanning voltammetry (LSV) techniques. The CV results show that the oxidation reaction of FA is influenced by the hydroxyl concentration in the solution, and the peak current response with the FA concentration is linear at the range from 10 to 100 mM. The differential capacitance from EIS results indicate that the FA molecules adsorb at the BDD electrode surface at low potential (from 1.0 to 1.4 V). The kinetic studies have been examined with the various concentrations of FA, pH, and temperature. The activation energy of FA oxidation is also calculated. The results of kinetic study indicate that the adsorption of FA molecules at the BDD electrode is the rate‐determining step at low potential (from 1.0 to 1.40 V).  相似文献

A method using flow injection (FI) with amperometric detection at anodized boron-doped diamond (BDD) thin films has been developed and applied for the determination of tetracycline antibiotics (tetracycline, chlortetracycline, oxytetracycline and doxycycline). The electrochemical oxidation of the tetracycline antibiotics was studied at various carbon electrodes including glassy carbon (GC), as-deposited BDD and anodized BDD electrodes using cyclic voltammetry. The anodized BDD electrode exhibited well-defined irreversible cyclic voltammograms for the oxidation of tetracycline antibiotics with the highest current signals compared to the as-deposited BDD and glassy carbon electrodes. Low detection limit of 10 nM (signal-to-noise RATIO = 3) was achieved for each drug when using flow injection analysis with amperometric detection at anodized BDD electrodes. Linear calibrations were obtained from 0.1 to 50 mM for tetracycline and 0.5–50 mM for chlortetracycline, oxytetracycline and doxycycline. The proposed method has been successfully applied to determine the tetracycline antibiotics in some drug formulations. The results obtained in percent found (99.50–103.01%) were comparable to dose labeled.  相似文献

Electrodeposition of Pt-Pb nanoparticles (PtPbNPs) to multi-walled carbon nanotubes (MWCNTs) resulted in a stable PtPbNP/MWCNT nanocomposite with high electrocatalytic activity to glucose oxidation in either neutral or alkaline medium. More importantly, the nanocomposite electrode with a slight modification exhibited high sensitivity, high selectivity, and low detection limit in amperometric glucose sensing at physiological neutral pH (poised at a negative potential). At +0.30 V in neutral solution, the nanocomposite electrode exhibited linearity up to 11 mM of glucose with a sensitivity of 17.8 μA cm−2 mM−1 and a detection limit of 1.8 μM (S/N = 3). Electroactive ascorbic acid (0.1 mM), uric acid (0.1 mM) and fructose (0.3 mM) invoked only 23%, 14% and 9%, respectively, of the current response obtained for 3 mM glucose. At −0.15 V in neutral solution, the electrode responded linearly to glucose up to 5 mM with a detection limit of 0.16 mM (S/N = 3) and detection sensitivity of ∼18 μA cm−2 mM−1. At this negative potential, ascorbic acid, uric acid, and fructose were not electroactive, therefore, not interfering with glucose sensing. Modification of the nanocomposite electrode with Nafion coating followed by electrodeposition of a second layer of PtPbNPs on the Nafion coated PtPbNP/MWCNT nanocomposite produced a glucose sensor (poised at −0.15 V) with a lower detection limit (7.0 μM at S/N = 3) and comparable sensitivity, selectivity and linearity compared to the PtPbNP/MWCNT nanocomposite. The Nafion coating lowered the detection limit by reducing the background noise, while the second layer of PtPbNPs restored the sensitivity to the level before Nafion coating.  相似文献
