3‐Alkyl/aryl‐3‐ureido‐1H,3H‐quinoline‐2,4‐diones ( 2 ) and 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones ( 3 ) react in boiling concentrated HCl to give 5‐alkyl/aryl‐4‐(2‐aminophenyl)‐1,3‐dihydro‐2H‐imidazol‐2‐ones ( 6 ). The same compounds were prepared by the same procedure from 2‐alkyl/aryl‐3‐ureido‐1H‐indoles ( 4 ), which were obtained from the reaction of 3‐alkyl/aryl‐3‐aminoquinoline‐2,4(1H,3H)‐diones ( 1 ) with 1,3‐diphenylurea or by the transformation of 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones ( 3 ) and 5‐alkyl/aryl‐4‐(2‐aminophenyl)‐1,3‐dihydro‐2H‐imidazol‐2‐ones ( 6 ) in boiling AcOH. The latter were converted into 1,3‐bis[2‐(2‐oxo‐2,3‐dihydro‐1H‐imidazol‐4‐yl)phenyl]ureas ( 5 ) by treatment with triphosgene. All compounds were characterized by 1H‐ and 13C‐NMR and IR spectroscopy, as well as atmospheric pressure chemical‐ionisation mass spectra.  相似文献

The crystal structures of N‐aryl‐1,2,3,4‐tetrahydro‐3,3‐dimethyl‐2,4‐quinolinediones bearing methoxy‐ ( 1 ), methyl‐ ( 2 ), and chloro‐ ( 3 ) substituents in 2′‐position of the phenyl ring have been determined by X‐ray crystal structure analysis. The heterocyclic ring in 1–3 adopts an envelope conformation, with the smallest ring puckering in the ortho‐chloro derivative 3 . The N‐aryl ring is almost perpendicular with respect to the quinoline‐2,4‐dione ring. The corresponding dihedral angle values are 83.2(1)°, 80.0(9)°, and 83.4(2)° in 1, 2 and 3 , respectively. The hydrogen bond of C H⋅⋅⋅O type joins the molecules of the ortho‐methoxy derivative 1 into dimers. The supramolecular structure also contains two C H⋅⋅⋅π interactions that link the hydrogen‐bonded dimers into sheets. In ortho‐methyl derivative 2 , one C H⋅⋅⋅π interaction generates infinite chains, whereas two C H⋅⋅⋅O hydrogen bonds and three C H⋅⋅⋅π interactions in the ortho‐chloro derivative 3 form three‐dimensional framework. © 2008 Wiley Periodicals, Inc. 3‐Alkyl/aryl‐3‐amino‐1H,3H‐quinoline‐2,4‐diones react with alkyl/aryl isocyanates to give novel 3‐alkyl/aryl‐3‐ureido‐1H,3H‐quinoline‐2,4‐diones or 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones. In some cases, a mixture of both products was obtained and separated by fractional crystallization. All compounds were characterized by their 1H, 13C, ir and ms data and some of them also by 15N nmr data.  相似文献

Readily accessible acylamino(chloro)acetophenones, if treated with sodium rhodanide and α‐halogenocarbonyl compounds, provide 4‐acylamino‐5‐aryl‐2‐mercapto‐1,3‐oxazole derivatives which undergo recyclization on heating in polyphosphoric acid to give substituted 1,3‐thiazol‐2(3H)‐ones or 1,3‐thiazolidin‐2,4‐diones containing 2‐alkyl(aryl)‐5‐aryl‐1,3‐oxazol‐4‐yl residues at the N3 atom. © 2007 Wiley Periodicals, Inc. The treatment of α‐bromoalkyl aryl ketones and 2‐(propan‐2‐ylidene)hydrazine carbothioamide afforded 4‐aryl‐2‐(2‐(propan‐2‐ylidene)hydrazinyl)thiazoles via a Hantzsch‐thiazole synthesis, which reacted with 4‐aryl‐2,4‐diketoesters via a sequential Knorr‐pyrazole reaction to deliver a variety of aryl‐substituted ethyl 1‐(thiazol‐2‐yl)‐1H‐pyrazole‐3‐carboxylates in a one‐pot fashion with moderate to high yields. The key intermediates 4‐aryl‐2,4‐diketoesters, existing as its enolic lithium salt, were synthesized in situ by a high‐yield tert‐BuOLi‐mediated Claisen condensation of alkylphenones and diethyl oxalate. This class of elegant molecule comprises aryl groups on the two different heterocyclic cores, and the configurations of two representative molecules were determined by single crystal X‐ray crystallography.  相似文献

Syntheses of 2‐aryloxy/2‐chloro ethoxy‐2,3‐dihydro‐5‐benzoyl‐1H‐1,3,2‐benzodiaza‐phosphole 2‐oxides 3a–h were accomplished by reactions of equimolar quantities of 3,4‐diaminobenzophenone ( 1 ) with various aryl/chloroethoxy phosphorodichloridates 2a–g and 2h in the presence of triethylamine at 50–60°C. Compounds 3i–k were prepared by reacting 3,4‐diaminobenzophenone ( 1 ) with aryl thiophosphorodichloridates 2i–k under similar conditions. 3‐Alkyl/aryl‐3‐hydroxyquinoline‐2,4‐diones were reduced with NaBH4 to give cis‐3‐alkyl/aryl‐3,4‐dihydro‐3,4‐dihydroxyquinolin‐2(1H)‐ones. These compounds were subjected to pinacol rearrangement by treatment with concentrated H2SO4, resulting in 4‐alkyl/aryl‐3‐hydroxyquinolin‐2(1H)‐ones. When a benzyl (Bn) group was present in position 3 of the starting compound, its elimination occurred during the rearrangement, and the corresponding 3‐hydroxyquinolin‐2(1H)‐one was formed. The reaction mechanisms are discussed for all transformations. All compounds were characterized by IR, 1H‐ and 13C‐NMR spectroscopy, as well as mass spectrometry.  相似文献

Tuning of the physicochemical properties of the 1,3‐diphosphacyclobutane‐2,4‐diyl unit is attractive in view of materials applications. The use of arynes is shown to be effective for installing relatively electron rich aryl substituents into the open‐shell singlet P‐heterocyclic system. Treatment of the sterically encumbered 1,3‐diphosphacyclobuten‐4‐yl anion with ortho‐silylated aryl triflates in the presence of fluoride under appropriate conditions afforded the corresponding 1‐aryl 1,3‐diphosphacyclobutane‐2,4‐diyls. The air‐stable open‐shell singlet P‐heterocycles exhibit considerable electron‐donating character, and the aromatic substituent influences the open‐shell character, which is thought to be related to the property of p‐type semiconductivity. The P‐arylated 1,3‐diphosphacyclobutane‐2,4‐diyl systems can be further utilized as detectors of hydrogen fluoride (HF), which causes a remarkable change in their photoabsorption properties.  相似文献

Possible approaches to synthesis of 5‐methyl‐4‐oxo‐2‐(coumarin‐3‐yl)‐N‐aryl‐3,4‐dihydrothieno[2,3‐d]pyrimidine‐6‐carboxamides 4 have been discussed. It is shown that the preferable approach is cyclization of 2‐iminocoumarin‐3‐carboxamides 1 , utilizing 5‐amino‐3‐methyl‐N2‐arylthiophene‐2,4‐dicarboxamides 2 as binucleophilic reagents. The proposed procedure allowed us to easily obtain 4 in two stages, using common reagents. © 2007 Wiley Periodicals, Inc. 4‐Alkyl/aryl‐5‐nonanoyl/octadecanoyl‐2,4‐dihydro‐3H‐1,2,4‐triazoline‐3‐thiones were synthesized as potential antimicrobial agents. The course of synthesis included the reaction of nonanoyl/octadecanoyl hydrazines with selected alkyl/aryl isothiocyanates. The prepared thiosemicarbazides gave by cyclization the required 1,2,4‐triazoles. A number of synthesized compounds were subjected to in vitro testing against two gram‐positive, two gram‐negative bacteria and two fungi.  相似文献

Hypervalent‐iodine‐mediated oxidative coupling of the two aryl groups in either 2‐acylamino‐N‐phenyl‐benzamides or 2‐hydroxy‐N‐phenylbenzamides, with concomitant insertion of the ortho‐substituted N or O atom into the tether, has been described for the first time. This unusual metal‐free rearrangement reaction involves an oxidative C(sp2)? C(sp2) aryl–aryl bond formation, cleavage of a C(sp2)? C(O) bond, and a lactamization/lactonization. Furthermore, unsymmetrical diaryl compounds can be easily obtained by removing the tether within the cyclized product.  相似文献

3‐Hydroxyquinoline‐2,4‐diones 1 react with isocyanates to give novel 1,2,3,4‐tetrahydro‐2,4‐dioxoquinolin‐3‐yl (alkyl/aryl)carbamates 2 and/or 1,9b‐dihydro‐9b‐hydroxyoxazolo[5,4‐c]quinoline‐2,4(3aH,5H)‐diones 3 . Both of these compounds are converted, by boiling in cyclohexylbenzene solution in the presence of Ph3P or 4‐(dimethylamino)pyridine, to give 3‐(acyloxy)‐1,3‐dihydro‐2H‐indol‐2‐ones 8 . A series of salicylaldimine ligands were designed to promote palladium‐catalyzed Suzuki–Miyaura cross‐coupling reaction. After a screening process, a ligand with a bulky 2,4‐di‐tert‐butyl substituent on the salicyaldehyde backbone and cyclohexylamine moiety was found to serve as a good combination for this reaction in aqueous solutions of DMF. The protocol demonstrated a significant advance in the efficiency of the cross‐coupling of aryl bromides and aryl chlorides with arylboronic acids to produce the desired biaryl products. The Suzuki–Miyaura reaction of aryl bromides with benzeneboronic acid catalyzed by bis(chloro)(2‐pyridylquinoxaline)palladium(II) was investigated. The scope of the bis(chloro)(2‐pyridylquinoxaline)palladium(II) was determined in toluene at 80 °C using KOH as base. Using a 0.1% molar ratio of bis(chloro)(2‐pyridylquinoxaline)palladium(II) C1 as a catalyst, aryl bromides reacted with benzeneboronic acid to afford diaryl derivatives in excellent yield. This article reports a convenient and general method for the regioselective synthesis of a new series of 2‐alkyl(aryl)‐8‐methyl‐4‐trifluoromethyl‐7‐aminoquinolines in 86–93% yields, from cycloaromatization reactions of N‐(oxotrifluoroalkenyl)‐2,6‐diaminotoluenes in a strongly acidic medium polyphosphoric acid and absence of solvent. The enaminoketone intermediates were easily isolated from the reaction of 4‐alkoxy‐4‐alkyl(aryl)‐1,1,1‐trifluoroalk‐3‐en‐2‐ones [CF3C(O)CH═C(R)OR1, where R = H, Me, Ph, 4‐FPh, 4‐BrPh, 4‐MePh, and R1 = Me, Et] with 2,6‐diaminotoluene (2,6‐DAT) in methanol under mild conditions, in 46–70% yields. Another synthetic route also allowed the regioselective synthesis of 2‐aryl(heteroaryl)‐4‐methyl‐4‐trifluoromethyl‐7‐aminoquinolines from direct cyclocondensation reactions of 4‐alkoxy‐4‐aryl(heteroaryl)‐1,1,1‐trifluoroalk‐3‐en‐2‐ones with 2,6‐diaminotoluene in methanol under mild conditions, in 21–36% yields.  相似文献

5‐Substituted (amine, alkyl, aryl, heterocyclic) 4‐(1,3,4‐thiadiazol‐2‐yl)benzene‐1,3‐ diols were synthesized, and their antifungal properties were examined. The compounds were obtained by the one‐pot reaction of sulfinylbis((2,4‐dihydroxyphenyl)methanethione) with hydrazides or thiosemicarbazides. Their structures were identified from elemental, IR, 1H NMR, and MS spectra analyses. The activities of the derivatives against five phytopathogenic fungi in vitro were measured. The condensation of 5‐[4′‐(2″‐halo‐ethoxy)benzylidenyl]‐2,4‐thiazolidinediones 2 when separately carried with different 2‐amino thiazoles 3 and different sulphanilamides 4 in DMF, using potassium hydroxide in the presence of a catalytic amount of a phase transfer catalyst, gave new 5‐[4′‐(4″‐aryl‐thiazol‐2″‐yl‐aminoethoxy)‐3′/5′‐substituted benzylidenyl]‐2,4‐thiazolidinediones 5 and 5‐[4′‐(2″/4″‐sulphonamidophenyl aminoethoxy)‐3′/5′‐substituted benzylidenyl]‐2,4‐thiazolidinediones 6 derivatives in good yields, respectively. The structures of all new compounds were established from analytical and spectral data. All the reaction sequences were carried under microwave irradiation, as an efficient tool. © 2009 Wiley Periodicals, Inc.
