In the present study, we have made an effort to develop the novel synthetic antioxidants and antimicrobials with improved potency. The novel benzofuran‐gathered C‐2,4,6‐substituted pyrimidine derivatives 5a , 5b , 5c , 5d , 5e , 5f , 6a , 6b , 6c , 6d , 6e , 6f , 7a , 7b , 7c , 7d , 7e , 7f , 8a , 8b , 8c , 8d , 8e , 8f , 9a , 9b , 9c , 9d , 9e , 9f were synthesized by simple and efficient four‐step reaction pathway. Initially, o‐alkyl derivative of salicylaldehyde readily furnish corresponding 2‐acetyl benzofuran 2 in good yield, upon the treatment with potassium tertiary butoxide in the presence of molecular sieves. Further, Claisen–Schmidt condensation with aromatic aldehydes via treatment with thiourea followed by coupling reaction with different sulfonyl chlorides afforded target compounds. The structures of newly synthesized compounds were confirmed by IR, 1H NMR, 13C NMR, mass, and elemental analysis and further screened for their antioxidant and antimicrobial activities. The results showed that the synthesized compounds 8b , 8e , 9b , and 9e produced significant antioxidant activity with 50% inhibitory concentration higher than that of reference, whereas compounds 7d and 7c produced dominant antimicrobial activity at concentrations 1.0 and 0.5 mg/mL compared with standard Gentamicin and Nystatin, respectively.  相似文献

In this study, thiazole derivatives containing Schiff bases ( 7a , 7b , 7c , 7d , 7e , 7f , 8a , 8b , 8c , 8d , 8e , 8f , 9a , 9b , 9c , 9d , 9e , 9f ) were synthesized in moderate to high yields (49–94%) using the Hantzsch reaction with thiosemicarbazone derivatives ( 5a , 5b , 5c ) and 2‐bromo‐1‐phenylethanone derivatives ( 6a , 6b , 6c , 6d , 6e , 6f ). The structures of synthesized compounds were elucidated by IR, 1H NMR, 13C NMR, elemental analyses, mass spectroscopy and X‐ray diffraction analysis techniques. Moreover, the synthesized compounds were tested for their in vitro antifungal activity and most of them exhibited moderate to good activity against Fusariumoxysporumf.sp. lycopersici.   相似文献

A series of N‐alkylphthalazinone were synthesized by the reaction of phthalazin‐1(2H)‐one derivatives 1a , 1b , 1c with alkylating agents namely, propargyl, allyl bromide, epichlorohydrin, 1,3‐dichloro‐2‐propanol, 4‐bromobutylacetate, and 1‐(bromomethoxy)ethyl acetate to give the corresponding N‐alkylphthalazinone 2a , 2b , 2c , 3a , 3b , 3c , 5a , 5b , 5c , 6a , 6b , 6c , 7a , 7b , 7c , and 9a , 9b , 9c . Alkylation of phthalazin‐1(2H)‐thione to give a series from S‐alkylphthalazine 12 , 13 , 14 and thioglycosides 15 and 17 was performed. Deprotection of compounds 7a , 7b , 7c , 9a , 9b , 9c , 15 , and 17 resulted in the formation of the corresponding products 8a , 8b , 8c , 10a , 10b , 10c , 16 , and 18 . The structure of newly synthesized compounds was assigned by IR, 1H, 13C NMR, and elemental analysis. Some of these compounds were screened for antiviral and antimicrobial activity.  相似文献

The direct addition of Csp2–H bonds onto polar C=C, C=O, and C=N bonds is both synthetically and mechanistically important, because using aromatic C–H substrates in place of organometallic reagents provides a more direct and atom‐economical alternative to many important compounds without the pre‐generation of organometallic compounds from stoichiometric halides and the unavoidable generation of stoichiometric metal halide waste. In this account, we summarize our contributions to the transition‐metal‐catalyzed addition of aromatic C–H bonds to polar C=C, C=O, and C=N bonds via directing‐group‐assisted regiospecific reactions. These synthetic methods provide efficient access to benzylic alcohols, alkylbenzenes, 3‐substituted phthalides, N‐substituted phthalimides, N‐aryl benzamides, and indene derivatives from commercially available reagents. It is worth noting that valuable heterocycles such as 3‐substituted phthalides and N‐substituted phthalimides can be obtained in one step by this approach.

Four L ‐rhamnose–benzoxazinone compounds as epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors were designed and synthesized. All structures of the compounds were characterized by 1H‐NMR, 13C‐NMR, and high‐resolution mass spectrometry. The inhibition activities of the target compounds for the EGFR tyrosine kinase activity in vitro were determined. Compounds 6a , 6b , 6c , 6d displayed moderate activity in targeting EGFR.  相似文献

High‐spin FeII–alkyl complexes with bis(pyridylimino)isoindolato ligands were synthesized and their paramagnetic 1H and 13C NMR spectra were analyzed comprehensively. The experimental 13C—1H coupling values are temperature (T?1)‐ as well as magnetic‐field (B2)‐dependent and deviate considerably from typical scalar 1JCH couplings constants. This deviation is attributed to residual dipolar couplings (RDCs), which arise from partial alignment of the complexes in the presence of a strong magnetic field. The analysis of the experimental RDCs allows an unambiguous assignment of all 13C NMR resonances and, additionally, a structural refinement of the conformation of the complexes in solution. Moreover the RDCs can be used for the analysis of the alignment tensor and hence the tensor of the anisotropy of the magnetic susceptibility.  相似文献

Imidazo[4,5‐c ]pyrazole derivatives ( 3a–f , 4a–f , and 5a–f ) were efficiently synthesized by one‐pot three‐component reactions using CeO2–MgO as the catalyst. The synthesized compounds were characterized by IR, 1H NMR, 13C NMR, and mass spectroscopic analyses. The in vitro antimicrobial activity of the synthesized compounds against various bacterial and fungal strains was screened. Compound 3b was highly active [minimum inhibitory concentration (MIC): 0.5 μg/mL] against Gram‐positive Staphylococcus aureus , and compounds 3b , 3f , 4d , and 4e were highly active (MIC: 0.5, 2, 2, and 0.5 μg/mL, respectively) against Gram‐negative Pseudomonas aeruginosa and Klebsiella pneumoniae , relative to standard ciprofloxacin in the antibacterial activity screening. Compounds 3b and 4f were highly active (MIC: 4 and 0.5 μg/mL, respectively) against Aspergillus fumigatus and Microsporum audouinii in the antifungal activity screening compared with the clotrimazole standard.  相似文献

Novel functionalized bis‐thiazole derivatives ( 4a–d , 9a , b , 13a–e , and 16a–d ) were synthesized in good to excellent yields (70–90%) via the ring closure of benzo[d ]thiazol‐2‐amine and various α‐haloketones in the presence of carbon disulfide or aryl isothiocyanates as S‐nucleophiles. The reaction of 2‐heteroaryl‐substituted trimethinium salts (A, B, and C) with aromatic 1,2‐diamines ( a , b , c , d , and e ) in acetonitrile/acetic acid leads to 6,13‐disubstituted 1,4,8,11‐tetraaza[14]annulene derivatives ( 1 , 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , 10 , 11 , 12 , 13 , 14 ). The UV–vis spectral behavior of these compounds was examined in acetonitrile. Elemental analysis, IR, 1H‐NMR, 13C‐NMR, and mass spectra confirm the molecular structure of the newly synthesized compounds.  相似文献

We have synthesized eight possible diastereoisomers 3 a – h of the C79–C97 fragment of symbiodinolide ( 1 ) in a stereodivergent manner by utilizing a dithiane addition to the aldehyde as a key step. Comparison of the 13C NMR chemical shifts of the natural product 1 and the synthetic products 3 a – h indicated that the relative stereostructure of this fragment in symbiodinolide ( 1 ) is that represented in 3 a or f . We have stereodivergently synthesized eight possible diastereoisomers of the C94–C104 fragment 4 a – h , and we have compared their 13C NMR chemical shifts with those of the natural product, which established the relative stereochemistry of this fragment to be that described in diastereoisomers 4 a or e . By combining the stereostructural outcomes of the C79–C97 and C94–C104 fragments, we have proposed four candidate compounds of the C79–C104 fragment 2 a – d . We also synthesized diastereoisomers 2 a and b ( 2 a in the preceding article; Chem. Eur. J. 2015 , DOI: 10.1002/chem.201503880) by a Julia–Kocienski olefination and diastereoisomers 2 c and d by a Wittig reaction. By comparing the 13C NMR chemical shifts of natural symbiodinolide ( 1 ) with those of the synthetic products 2 a – d , we have reassigned the stereostructure of the C79–C104 fragment of natural product 1 to be that depicted in diastereoisomer 2 b .  相似文献

Long‐range 1H‐15N heteronuclear shift correlation methods at natural abundance to facilitate the elucidation of small molecule structures have assumed a role of growing importance over the past decade. Recently, there has also been a high level of interest in the exploration of indirect covariance NMR methods. From two coherence transfer experiments, A→B and A→C, it is possible to indirectly determine B?C. We have shown that unsymmetrical indirect covariance methods can be employed to indirectly determine several types of hyphenated 2D NMR data from higher sensitivity experiments. Examples include the calculation of hyphenated 2D NMR spectra such as 2D GHSQC‐COSY and GHSQC‐NOESY from the discrete component 2D NMR experiments. We now wish to report the further extension of unsymmetrical indirect covariance NMR methods for the combination of 1H‐13C GHSQC and 1H‐15N longrange (GHMBC, IMPEACH‐MBC, CIGAR‐HMBC, etc.) heteronuclear chemical shift correlation spectra to establish 13C‐15N correlation pathways.  相似文献

A series of novel [4‐(1,2,3‐thiadiazol‐4‐yl)phenoxy]methylene anchored 1,3,4‐triazoles ( 8a , 8b , 8c , 8d , 8e , 8f , 8g , 8h ) and 1,3,4‐thiadiazoles ( 9a , 9b , 9c , 9d , 9e , 9f , 9g , 9h , 9i ) were synthesized from thiosemicarbazide ( 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h , 7i , 7j ). The structures of these newly synthesized compounds were confirmed on the basis of IR, 1H‐NMR, mass spectral techniques, and elemental analysis. The in vitro antimicrobial screenings of the synthesized compounds were carried out against four bacterial pathogens, namely Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa and three fungal pathogens Candida albicans, Aspergillus niger and Aspergillus clavatus, using broth microdilution minimum inhibitory concentration method. The compounds 7d , 7j , 8a , 9a , 9b , and 9i exhibited promising antibacterial activity against the tested strains, whereas some compounds were found to be active against one of the tested bacterial strains.  相似文献

Molecular dynamics simulations of the structure H (sH) clathrate of tert‐butylmethylether show the prevalence of ether–water hydrogen bonding (see picture), absent in the neo‐hexane sH clathrate. This affects guest–cage dynamics and host–water dielectric relaxation dynamics. The 13C and 1H NMR relaxation times for both guests are measured, and the differences are explained in terms of guest–host interactions in the two clathrates.

Nine new 4‐substituted 2‐phenylimidazole derivatives have been synthesized by aldol condensation of 2‐phenylimidazole‐4‐carbaldehyde with various active methylene compounds. In case of non‐symmetric active methylene compound used, the stereospecific formation of only one cis‐trans isomer has been observed. The predominant formation of products with bulkier substituents standing opposite on double bond formed by aldol condensation has been proved. cis‐trans Isomerism of three unsymmetrically substituted products has been determined by 1H coupled 13C NMR experiments. 3‐[(2‐Phenylimidazol‐4‐yl)methylene]pentane‐2,4‐dione has been characterized by single crystal X‐ray structural analysis as well. Selected bond lengths and angles have proved the expected large mesomerical stabilisation in the molecule. The hydrogen bond in crystal phase has been observed.  相似文献

A water‐soluble sample (TM4b), extracted from sclerotia of Pleurotus tuberregium, was analyzed using elemental analysis, one‐ and two‐dimensional 1H and 13C NMR. The results indicated that TM4b was protein–polysaccharide complex, and the polysaccharide moiety was hyperbranched β‐D ‐glucan with residuals branched at C3, C2, C4, and C6 positions. A preparative size‐exclusion chromatography (SEC) column combined with nonsolvent addition method was used to fractionate TM4b, and nine fractions were obtained. Solution properties of TM4b in 0.15 M aqueous NaCl were studied using static laser light scattering and viscometry at 25 °C. The dependences of intrinsic viscosity ([η]) and radius of gyration (〈S2〉) on weight–average molecular weight (Mw) for TM4b in the Mw range from 1.89 × 104 to 2.58 × 106 were found to be [η] = 0.21M and 〈S2〉 = 3.63M. It indicated that TM4b existed as compact sphere conformation in the aqueous solution. Atomic force microscopy image further confirmed that the TM4b molecules exhibited globular shape in the solution. This work gave valuable information on fractionation and chain conformation characterization of the globular protein–polysaccharide complex. © 2007 Wiley Periodicals, Inc. A series of new isatin–mesalamine conjugates ( 9a – g ) were synthesized via conjugation of isatin ( 3a ) and its derivatives ( 3b – 3d , 4 , 5 , and 6 ) with mesalamine ( 7 ) by using chloroacetyl chloride as a bifunctional linker. Compounds 3a – 3d were prepared by employing Sandmeyer reaction. Compounds 4 , 5 , and 6 were obtained from isatin ( 3a ) via previously reported methods. The synthesized compounds were characterized by IR, mass, 1H NMR, and 13C NMR spectral techniques. Synthesized compounds ( 3a – d , 4 , 5 , 6 , and 9a – g ) were evaluated for in vitro antioxidant activity by DPPH assay method using ascorbic acid as standard. Hybrids 9b (IC50 = 368.6 ± 3.5 μM) and 9f (IC50 = 335.1 ± 2.9 μM) showed better antioxidant activity than its parent compounds such as 3a (IC50 = 556.8 ± 2.9 μM), 5 (IC50 = 511.9 ± 3.6 μM), and 7 (IC50 = 768.9 ± 2.7 μM). Acetic acid‐induced ulcerative colitis in rat model was chosen to examine the antioxidant potential of the synthesized hybrids ( 9b and 9f ) in the amelioration of ulcerative colitis. Colonic myeloperoxidase and malondialdehyde enzymes were used as biomarkers of anti‐ulcerative colitis activity. In the present study, hybrids 9b and 9f reduced the levels of colonic myeloperoxidase and malondialdehyde enzymes significantly (p < 0.05) when compared with control (colitic), at a dose (0.03 mM/12.5 mg/kg b.w. p.o.) (50%) less than that of its parent moieties mesalamine (0.16 mM/25 mg/kg) and isatin (0.16 mM/25 mg/kg). Thus, the molecular hybridization was proved to be significant in enhancing the activity of hybrids 9b and 9f by reducing the dose.  相似文献

Two novel compounds, 8–[2–(2–thienyl)vinyl]–10,10–dimethyl–10H–pyrido[1,2–a] indolium perchlorate ( 3a ) and 8–[2–(5–phenyl–2–thienyl)vinyl]–10,10–dimethyl–10H–pyrido[1,2–a]indolium perchlorate ( 3b ) were synthesized and characterized by IR, 1H–NMR, elemental analyses, and X–ray diffraction. Crystal structural analysis suggested that either 3a or 3b exhibited good coplanarity and rings and vinyl in the target molecule could make up a large conjugated system. Ultraviolet–visible absorption analysis indicated both 3a and 3b possessed large maximum absorptions, and 3b underwent a significant redshift (43.0 nm) in comparison with 3a .  相似文献

The syntheses of the zwitterionic spirocyclic λ5Si-silicates 6–9 are described. These chiral zwitterions contain a pentacoordinate (formally negatively charged) silicon atom and a tetracoordinate (formally positively charged) nitrogen atom. Compounds 6 · 1/2 HO(CH2)2OH, 7 , 8 · CH3CN, and 9 were studied by solution-state (1H, 13C, 29Si) and solid-state (29Si CP/MAS) NMR experiments. In addition, all compounds were structurally characterized by single-crystal X-ray diffraction. The dynamic behavior (Berry-type enantiomerization) of 7–9 in solution was studied by VT 1H NMR experiments. These experimental studies were completed by ab initio investigations of the related anionic model species 10–12 .  相似文献

High energy density materials with ethylene‐ and propylene bis(5‐nitroiminotetrazolate) as the anions are reported; all salts were fully characterized by IR, and 1H, 13C, and 15N NMR spectroscopy as well as elemental analyses. In addition, the heats of formation (ΔHf) and the detonation pressures (P) and velocities (D) were calculated.

A rapid access to 2,2′‐bithiazole‐based copolymers has been developed on the basis of the sequential palladium‐catalyzed C H/C X and C H/C H coupling reactions. To assemble a “copolymer” through homopolymerization, a type of symmetric A‐B‐A‐type building block is designed as the monomer and prepared via the regioselective C5 H arylation of thiazole. A PdCl2/CuCl‐cocatalyzed oxidative C H/C H homopolymerization has been established to afford the 2,2′‐bithiazole‐based copolymers with high Mn (up to 69400). The current protocol features atom‐ and step‐economy and exhibits a potential in the highly efficient construction of conjugated copolymers.
