Single lipid vesicles adsorbed on SiO(2) were manipulated using an atomic force microscope (AFM) operated in contact mode. For large force setpoints, single vesicles were either pushed sideways or ruptured by the tip, depending on the tip type (sharp or blunt) used, while for small force setpoints the vesicles were imaged by the tip. To extend the interpretation of and to guide the experiment, we have developed a generic model of the vesicle-tip-substrate system and performed Monte Carlo simulations, addressing the influence of force setpoint and tip speed and shape on the type of imaging or manipulation observed. Specifically, we have explored AFM-image height and width variations versus force setpoint, typical AFM images for small and large force setpoints, tip-induced vesicle strain versus force setpoint, typical vesicle shapes during pushing for different tip speeds, and the details of vesicle rupture induced by the tip.  相似文献

A tripod molecule with an azobenzene arm was designed as a single molecular tip for noncontact atomic force microscopy (NC-AFM). The azobenzene moiety showed photoisomerization that enabled measurements of the same position of the sample by different tip apexes with different interactions. Photoswitching behavior of the molecule synthesized and adsorbed on Au surfaces was examined and reversible switching between the trans- and cis forms was successfully confirmed by NC-AFM measurements.  相似文献

The cation complexation behavior of dibenzocrown ethers adsorbed on highly oriented pyrolytic graphite substrates was investigated by means of atomic force microscopy using probe tips modified chemically with ammonium ion by silane coupling. The specific adhesion force based on the intermolecular force between dibenzocrown ether and ammonium ion was observed via force curve measurements in ethanol at the interface between the substrate and tip. The observed specific force decreased in the presence of the alkali metal ion in solution, indicating that the cation in solution interferes with the complexation of the crown ethers adsorbed on the substrate with the ammonium ion immobilized on the tip. The blocking effect of metal ions in solution on the observed force depended on the sizes of both the blocking cation and crown ether ring, suggesting that the surface-adsorbed dibenzocrown ethers possess a selective cation-complexing ability similar to that in their bulk state and that the adhesion force measurements using cation-modified tips allow evaluation of the cation-complexing ability of crown ethers under cation-competitive conditions.  相似文献

We report the first observation of a phase transition in a nanoscale direct deposition process. This transition involves the melting of molecular ink layers in dip-pen nanolithography, and it is observed by measuring the temperature dependence of the growth rate of the deposited pattern. The results are interpreted using a diffusion equation approach in conjunction with a "double-molecular-layer" model of the adsorbed molecules on the atomic force microscope tip. The theory provides a qualitative explanation for the dependence of the pattern growth rate on solvent and adsorbed water as well as on temperature.  相似文献

Thin layers of cellulose I nanocrystals were spin-coated onto silicon wafers to give a flat model cellulose surface. A mild heat treatment was required to stabilize the cellulose layer. Interactions of this surface with polyelectrolyte layers and multilayers were probed by atomic force microscopy in water and dilute salt solutions. Deflection–distance curves for standard silicon nitride tips were measured for silicon, cellulose-coated silicon, and for polyelectrolytes adsorbed on the cellulose surface. Transfer of polymer to the tip was checked by running deflection–distance curves against clean silicon. Deflection–distance curves were relatively insensitive to adsorbed polyelectrolyte, but salt addition caused transfer of cationic polyelectrolyte to the tip, and swelling of the polyelectrolyte multilayers.  相似文献

An oxidized silicon tip and an oxidized silicon wafer were hydrophobized with phenyl groups, and the long-range attraction induced by hydrophobation is shown in force vs. distance curves. Friction force measurements performed by means of an atomic force microscope (AFM) under electrochemical conditions on a pure Au(111) electrode surface and one modified with a foreign metal are presented; after deposition of a (sub)monolayer copper on a Au(111) single crystal electrode a large increase of the friction force is observed compared to the pure Au(111) electrode surface; the extent of the increase not only depends on the copper coverage, but also on the normal load and may be explained by a higher energy dissipation due to motion of the sulfate anions adsorbed on the copper atoms induced by the AFM tip.  相似文献

The adsorption of mixtures of aqueous solutions of cationic hydroxyethylcellulose polymer JR400 and anionic surfactant, sodium dodecyl sulfate, using atomic force microscopy (AFM) has been studied. Samples with various compositions from different regions of the ternary phase diagram presented in our previous work were imaged by atomic force microscopy on freshly cleaved mica, and hydrophobically modified mica and silica in soft-contact mode. A series of "washing" (subsequent injection of compositions with gradually decreasing polymer/surfactant ratio) and "scratching" (mechanical agitation of the surface material with an AFM tip) experiments were performed. It was revealed that the morphology of the adsorbed layer altered in a manner following the changes in morphology in the bulk solution. These changes were evidenced in cluster formation in the layer. The results suggest that the influence of the surface was limited to the formation of the adsorbed layer where the local concentrations of polymer and surfactant were higher than those in the bulk. All further modifications were driven by changes in the mixture composition in bulk. Force measurements upon retraction reveal the formation of network structures within the surface aggregates that will greatly slow structural reequilibration.  相似文献

Using scanning probe microscopy techniques, at low temperatures and in ultrahigh vacuum, individual molecules adsorbed on surfaces can be probed with ultrahigh resolution to determine their structure and details of their conformation, configuration, charge states, aromaticity, and the contributions of resonance structures. Functionalizing the tip of an atomic force microscope with a CO molecule enabled atomic‐resolution imaging of single molecules, and measurement of their adsorption geometry and bond‐order relations. In addition, by using scanning tunneling microscopy and Kelvin probe force microscopy, the density of the molecular frontier orbitals and the electric charge distribution within molecules can be mapped. Combining these techniques yields a high‐resolution tool for the identification and characterization of individual molecules. The single‐molecule sensitivity and the possibility of atom manipulation to induce chemical reactions with the tip of the microscope open up unique applications in chemistry, and differentiate scanning probe microscopy from conventional methods for molecular structure elucidation. Besides being an aid for challenging cases in natural product identification, atomic force microscopy has been shown to be a powerful tool for the investigation of on‐surface reactions and the characterization of radicals and molecular mixtures. Herein we review the progress that high‐resolution scanning probe microscopy with functionalized tips has made for molecular structure identification and characterization, and discuss the challenges it will face in the years to come.  相似文献

Nanostructures of lysozyme molecules adsorbed to mica were generated by the tip of an atomic force microscope in contact, tapping, and force-distance mode in aqueous solution. In contact mode at high ionic strength and adjusted lysozyme concentration a monolayer of defined pattern and orientation could be formed by the scan process of the tip. A lysozyme monolayer with minimal pattern size of about 60 nm was achieved by line scan. At larger loading forces besides a monolayer also 3D-aggregates of lysozyme molecules could be generated. In force-distance mode the volume of 3D-aggregates grows with increasing generation time, lysozyme concentration in the bulk phase, loading force, and frequency of up- and down-movement of the substrate toward the fixed cantilever. In tapping mode 3D-aggregates could be generated as well. It is postulated that reduction of electrostatic interaction between the oppositely charged lysozyme molecules and mica surface by sufficient high ionic strength is essential for monolayer formation. It is discussed that for the underlying mechanism of monolayer generation in contact mode lysozyme molecules of the bulk phase adsorb to the tip, become pulled off and attach to the mica surface by the scan process of the tip.  相似文献

The effect of the aminoglycoside (streptomycin) incorporation on the nanomechanical properties of pure dipalmitoylphosphatidylcholine (DPPC) vesicles was studied using atomic force microscope (AFM) on mica surface. The vesicles were prepared by extrusion and adsorbed on the mica surface. The forces, measured between an AFM tip and the vesicle, presented that the breakthrough of the tip into the vesicles occurred two times. Each breakthrough represented each penetration of the tip into each bilayer. Force data prior to the first breakthrough were fitted well with the Hertzian model to estimate Young's modulus and bending modulus of the vesicles. It was found that the Young's modulus and bending modulus were not varied with the incorporation of AGs (streptomycins) up to the 1:1 AG/DPPC vesicle system. This result may suggest that the AGs do not lead to the disruption of DPPC packing.  相似文献

Glaser-like coupling of terminal alkynes by thermal activation is extensively used in on-surface chemistry. Here we demonstrate an intramolecular version of this reaction performed by atom manipulation. We used voltage pulses from the tip to trigger a Glaser-like coupling between terminal alkyne carbons within a custom-synthesized precursor molecule adsorbed on bilayer NaCl on Cu(111). Different conformations of the precursor molecule and the product were characterized by molecular structure elucidation with atomic force microscopy and orbital density mapping with scanning tunneling microscopy, accompanied by density functional theory calculations. We revealed partially dehydrogenated intermediates, providing insight into the reaction pathway.  相似文献

Physicochemical studies on the mechanisms of protein adsorption onto solid material surfaces have been extensively performed so far, mainly based on the analysis of factors such as the equilibrium adsorbed amount (adsorption isotherms), time-dependent change of adsorbed amount (adsorption kinetics), and conformational change of adsorbed protein. However, direct understanding of the strength of the molecular interaction between protein and the material surface has not been established yet. For this issue, the force measurement techniques of an atomic force microscope (AFM) using a protein-modified probe tip are recently becoming powerful tools to analyze the actual interaction forces between protein and material surfaces. In this mini review, we discuss the characteristics and interpretation of the AFM force-versus-distance curves (fd curves) obtained with the protein-modified probe tip, and the relationship between the forces measured from the fd curves and the driving forces in the natural process of protein adsorption. Relative degrees of each of the following contributions which determine the character of protein adsorption are clarified: (1) the intrinsic protein/material forces mediated by solvent, (2) the thermodynamic stability of protein/material adhesion interface, and (3) diffusion force of protein molecules. Within these driving forces, the latter two in particular are confirmed to play essential roles in determining the character of protein adsorption, based on the profiles of fd curves.  相似文献

ZnO film-based ultraviolet (UV) detector was fabricated by photoassisted peak force tunnel atomic force (PFTUNA) on fluorine tin oxide (FTO) substrate. The PFTUNA current in dark and in UV light was ~0.1 and 2.0 nA, respectively. The UV sensitivity (photocurrent/dark current) is more than 20. The response time and the recovery time are ~0.12 and 0.32 s, respectively. The UV sensing mechanism is that the holes will transport to the ZnO surface to capture the adsorbed oxygen ions to weaken the depletion layer under UV illumination. The PFTUNA current between the tip and the ZnO film is consistent with the Richardson–Schottky (RS) thermionic emission model.  相似文献

Liquid tapping atomic force microscopy was used to study the nonspecific adsorption of horse spleen ferritin at a bare gold surface at single molecule resolution. The majority of ferritin molecules adsorbed irreversible on gold surfaces in accordance with the random sequential adsorption (RSA) mechanism frequently used to describe irreversible adsorption processes. However, the time-resolved data also reveal events that go beyond the RSA model, i.e., lateral mobility and fragility of some molecules, resulting in desorption, chain formation, and subunit dissociation. Scanning effects of the AFM tip were observed, resulting in diminished protein coverage in the scanned area.  相似文献

We study the capillary force in atomic force microscopy by using Monte Carlo simulations. Adopting a lattice gas model for water, we simulated water menisci that form between a rough silicon-nitride tip and a mica surface. Unlike its macroscopic counterpart, the water meniscus at the nanoscale gives rise to a capillary force that responds sensitively to the tip roughness. With only a slight change in tip shape, the pull-off force significantly changes its qualitative variation with humidity.  相似文献

The micromechanical properties of pure and cholesterol modified egg yolk phosphatidylcholine (EggPC) vesicles prepared by sonication were studied by atomic force microscopy (AFM) on mica surface. The force curves between an AFM tip and an unruptured vesicle were obtained by contact mode. During approach, two repulsion regions with two breaks were observed. The slopes of the two repulsive force regimes for the pure EggPC vesicles are determined to be several times lower than that of EggPC/cholesterol vesicles. The elastic properties from force plot analysis based on the Hertzian model showed that Young's modulus (E) and the bending modulus (kc) of cholesterol-modified vesicles increased several-fold compared with pure EggPC vesicles. The significant difference is attributed to the enhanced rigidity of the EggPC vesicles as a result of the incorporation of cholesterol molecules. The behavior of cholesterol-modified vesicles upon adsorption is different from that in solution as revealed by mechanical properties. The results indicate that AFM can provide a direct method to measure the mechanical properties of adsorbed small liposomes and to detect the stability change of liposomes.  相似文献
